Indução de morte celular imunogênica por terapia fotodinâmica mediada por cloreto de alumínio-ftalocianina incorporado a nanoemulsão em células de câncer de mama murino

Tese (doutorado)—Universidade de Brasília, Instituto de Ciências Biológicas, Programa de Pós-Graduação em Biologia Animal, 2018. / A nanobiotecnologia, que aplica técnicas, princípios biológicos e nanotecnológicos para melhorar a compreensão e transformação dos biossistemas, é oferecida como proposta de inovação tecnológica para tratamento do câncer. Dentre os avanços da nanobiotecnologia nos diversos setores do campo da saúde, destacam-se aqueles aplicados à terapia fotodinâmica (TFD), modalidade terapêutica minimamente invasiva e cujo uso é clinicamente aprovado para o tratamento de diferentes tipos de doenças oncológicas e não oncológicas. A TFD é capaz não apenas de destruir o tumor devido à sua toxicidade direta às células cancerosas, mas também de causar infarto do tecido canceroso por destruir a microvasculatura tumoral, bem como por ativar o sistema imunitário contra antígenos tumorais. Diante disso, o objetivo principal deste estudo foi avaliar a indução de morte celular imunogênica (MCI) causada pela TFD mediada por nanoemulsão com cloreto de alumínio-ftalocianina (TFD-NE-AlFtCl) em células de câncer de mama murino in vitro e in vivo. Para tanto, foi realizada a produção de um novo lote da NE-AlFtCl, que foi caracterizado e teve a estabilidade para suas prorpiedades coloidais, fotofísicas e físico-química avaliada. Concomitantemente aos testes de estabilidade, a NE-AlFtCL foi utilizada para avaliação do perfil de internalização pelas células 4T1, bem como para a avaliação de qual via de endocitose estava sendo utilizada para internalização, além da investigação de sua colocalização subcelular. Após a avaliação dos perfis de internalização, da via endocítica e da colocalização, as células 4T1 foram submetidas a diferentes protocolos da TFD-NE-AlFtCl, variando os tempos e doses de energia (J/cm²). As células 4T1 foram avaliadas quanto à via de morte desencadeada pela exposição a TFD-NE-AlFtCl. Foi verificada também a emissão de padrões moleculares associados a dano (DAMP) —HSP70, HSP90, Calreticulina, HMGB1 e IL-1β—, fator-chave para a indução de morte celular imunogênica (MCI). Por fim, após a avaliação dos perfis de liberação desses DAMP, as células pré-tratadas com TFD-NE-AlFtCl foram utilizadas como vacina em camundongos da espécie Balb-C a fim de verificar a ativação de uma resposta antitumoral. Para tanto, após as vacinações, os animais foram submetidos ao enxerto de células 4T1 viáveis, e monitorados quanto ao surgimento de tumor, volume tumoral e sobrevida durante todo experimento. O tempo de 15 minutos foi suficiente para identificação do FS AlFtCl dentro das células 4T1 e essa internalização foi dependente de caveolinas. Os resultados obtidos quanto à internalização e à colocalização subcelular do FS AlFtCl nas organelas núcleo, lisossomos, mitocôndrias e no retículo endoplasmático (RE) demonstraram diferenças na retenção do FS AlFtCl. Sendo que, os tempos de 15, 30 e 45 minutos apresentaram uma maior quantidade de FS AlFtCl nas organelas supracitadas, quando comparados como os tempos de 60, 120 e 240 minutos. Os diferentes protocolos da TFD-NE-AlFtCl aplicados nas células foram eficazes quanto à diminuição da viabilidade das células 4T1. Vale ressaltar ainda que à medida em que o tempo e a dose de energia foram variados, houve variação do perfil de morte e da via de morte ocasionada (apoptose/necrose). Os tratamentos com TFD-NE-AlFtCl 9,01 nM 25 J/cm² apresentaram maior perfil de apoptose, o que refletiu numa maior emissão de DAMP: HSP70, HSP90, Calreticulina, HMGB1 e IL-1β, quando comparados com os demais protocolos. Esses resultados apresentados pelo tratamento com TFD-NE-AlFtCl 9,01 nM 25 J/cm² se refletiram também, na avaliação in vivo, sendo verificada uma maior resistência dos animais quanto ao surgimento do tumor, a apresentação de menores volumes tumorais e maior sobrevida. Contudo, o presente estudo conclui que os diferentes protocolos de TFD-NE-AlFtCl aplicados às células 4T1 foram capazes de fazer a emissão dos DAMP — sendo esse requisito chave para ação de uma ativação de MCI e uma ativação antitumoral — porém, essas emissões ocorreram de formas diferentes mediante as alterações de tempo de dose de energia. Conclui-se também que a aplicação da TFD-NE-AlFtCl 9,01 nM 25 J/cm² mais branda demonstrou melhores resultados quanto ao surgimento e desenvolvimento do tumor, além de apresentaremm uma maior taxa de sobrevida. / Nanobiotechnology, which applies techniques, biological principles and nanotechnologies that improve the understanding and transformation of biosystems, is offered as a proposal of technological innovation for the treatment of cancer. Among the advances in nanobiotechnology in the various sectors of the health field, the most important are those applied to photodynamic therapy (PDT), a minimally invasive therapeutic modality and whose use is clinically approved for the treatment of different types of oncological and non-oncological diseases. PDT is capable not only of destroying the tumor due to its direct toxicity to the cancer cells but also by causing infarction of the cancerous tissue by destroying the tumor microvasculature as well as by activating the immune system against tumor antigens. Therefore, the main objective of these studies was to evaluate the induction of immunogenic cell death (ICD) of nanoemulsion-mediated PDT with aluminum chloride-phthalocyanine (PDT-NE-AlFtCl) in murine breast cancer cells in vitro and in vivo. For this, the production of a new batch of NE-AlFtCl was carried out, which was characterized and had its stability for its colloidal, photophysical and physicochemical properties evaluated. In parallel to the stability tests, NE-AlFtCL was used to evaluate the internalization profile by 4T1 cells, as well as to evaluate which pathway of endocytosis was being used for internalization, and to evaluate its subcellular colocalization. After the evaluation of the internalization profiles, the endocytic pathway and colocalization, the 4T1 cells were submitted to different protocols of the PDT-NE-AlFtCl, varying the time and energy doses (J/cm²). 4T1 cells were evaluated for the path of death triggered by exposure to PDT-NE-AlFtCl. DAMPs—HSP70, HSP90, Calreticulin, HMGB1 and IL-1β—, a key factor for the induction of immunogenic cell death (ICD), were also verified. Finally, after evaluation of the release profiles of these DAMPs, cells that we pretreated with PDT-NE-AlFtCl were used as vaccine in mice of the Balb-C species in order to verify the activation of an antitumor response. To do this, after the vaccinations, the animals were submitted to the grafting of viable 4T1 cells and monitored for appearance of tumor, tumor volume and survival throughout the experiment. The time of 15 minutes was sufficient for identification of the FS AlFtCl within the 4T1 cells and this internalization was dependent on caveolinas. The results obtained on the internalization and the subcellular colocalization of FS AlFtCl in the nucleus, lysosomes, mitochondria and in the endoplasmic reticulum (ER) showed differences in the retention of FS AlFtCl. The times of 15, 30 and 45 minutes showed a higher amount of FS AlFtCl in the aforementioned organelles when compared with the times of 60, 120 and 240 minutes. The different protocols of PDT-NE-AlFtCl applied in cells were effective in decreasing the viability of the 4T1 cells. It is worth noting that as time and energy dose were varied, there was variation of the death profile and of the way of death (apoptosis / necrosis). Treatments with 9.01 nM PDT-25 J/cm² had a higher apoptosis profile, which reflected in a higher emission of DAMPs: HSP70, HSP90, Calreticulin, HMGB1 and IL-1β, when compared to the other protocols. These results presented by the treatment with PDT-NE-AlFtCl 9.01 nM 25 J /cm2 also reflected in the in vivo evaluation, which demonstrated a greater resistance of the animals regarding the appearance of the tumor, the presentation of smaller tumor volumes and greater survival rates. However, the present study concludes that the different PDT-NE-AlFtCl protocols applied to the 4T1 cells were able to make the DAMPs emission — this being a key requirement for the action of ICD activation and antitumor activation —; it is worth mentioning that these emissions occurred in different ways regarding modification in time and energy doses. It was also concluded that the application of the milder 9,01 nM 25 J / cm2 PDT-NE-AlFtCl demonstrated better results regarding the occurrence and development of the tumor, besides presenting better survival rates.
