This thesis deals with horse tourism and the current situation of horse tourism in Žďár nad Sázavou. The practical part is devoted to the research of identifying possibilities and equipment of the stables in the selected area and which activities in connection with horses are the most used and demanded ones by the public. The survey was conducted by using the questionnaires. The first questionnaire was created for stables in the selected area. The second questionnaire was dedicated to the public. The results are processed graphically and verbally. The aim of the thesis is to propose some possibilities of development for this type of tourism based on the results of the questionnaires. The proposals for development options of horse tourism in the selected area are described in the next part. In conclusion, it is a brief evaluation and commenting of the whole thesis.

The NAD+-linked malic enzyme from Ascaris suum has been studied with regard to its kinetic and catalytic properties. Possible relationships between these properties and the physiological functioning of the malic enzyme were examined.

The ADP-ribose elongation catalyzed by poly(ADP-ribose) polymerase (PARP) [EC 2.2.2.30] has been partially characterized utilizing mono (ADP-ribosyl)ated polyamines. Arginine methyl ester (AME)-(ADP-ribose) and agmatine (AGMT)-(ADP-ribose) were synthesized enzymatically with a eukarytic mono(ADP-ribosyl) transferase and cholera toxin, respectively.

Preeclampsia (PE), a hypertensive disease of pregnancy, occurring at or after gestational week 20. PE can have life threating consequences for both the mother and the baby. PE is a highly heterogenous disease which makes it challenging to identify any effective therapeutic interventions. We previously discovered three molecular subclasses of PE disease. One of these subclasses is characterized by heightened placental inflammation (inflammation-driven PE). Since it is a newly identified form of PE, we currently do not know about the molecular mechanisms driving this inflammation-driven form of PE. Interestingly, we have observed that placentas from this inflammatory PE subclass uniquely express higher levels of NAD+ consuming enzymes- PARPs - and thus exhibit a decrease in NAD+ content. NAD+ is a regulator of cellular energy metabolism and mitochondrial function. Several studies in the non-pregnant populations suggested that pro-inflammatory disease conditions can trigger hyperactivation of NAD+ consuming enzymes causing a depletion in total NAD+ content, leading to mitochondrial dysfunction and organ failure. Thus, we tested the hypothesis that NAD+ depletion causes placental mitochondrial dysfunction in the inflammatory subclass of PE and that boosting NAD+ could prevent development of this form of placental disease. We aimed to profile PARP activity, NAD+ availability, and mitochondrial health in human cases of all three PE subclasses. We examined the causal relationship between inflammation and dysregulated NAD+ signalling in both an in vitro human trophoblast culture model and in a rodent model of inflammation-driven PE. We also evaluated the therapeutic potential of NAD+ booster, nicotinamide riboside (NR) to improve placental health and function in the rodent model of inflammation-driven PE. Our results suggest that along with increased activity of PARP enzymes and decreased NAD+ levels, human inflammatory PE placentas also exhibit decreased levels of mitochondrial proteins and increased oxidative DNA damage. Using an in vitro human placental (HTR8 cell line) inflammation model we showed that increasing NAD+ under an inflammatory condition improved trophoblast mitochondrial and cellular function. Using an in vivo LPS induced rat model of inflammation-driven PE, we demonstrated that NAD+ boosting during pregnancy improved placental mitochondrial function, reduced inflammation and oxidative stress. This subsequently resulted in improved pregnancy outcomes demonstrated by reduced maternal blood pressure, increased placental/fetal weights and increased fetal survival in the LPS model. Overall, this study identifies targeting NAD+ signaling as a promising intervention for PE. NAD+ boosting through NR has been tested in non-pregnant human populations and found to be safe and effective in enhancing NAD+ levels. Thus, findings of this thesis lay the ground to test NAD+ boosting strategies in PE patients in near future.

Master thesis: Wedding in the town of region of Zdar nad Sazavou Author: Bc. Veronika Macková Supervisor: PhDr. Jaroslava Krupková, CSc. ABSTRACT This thesis is focused on the description of the current marriage in the district town of Zdar nad Sazavou. The thesis is a methodology of field research, where I describe the basic information on how to collect information in the field. This work incorporates two general chapters on marriage. One is about how it is legally defined marriage in the Czech Republic, and the second deals with the status of marriage in human life. An integral part of this thesis is also crucial appreciation of literature and sources and historical development and current form Zdarsky whole region. In this thesis the main chapters are solved on three: the bridal, wedding day and the next day after the wedding. Each chapter has several requirements under subchapters. Contemporary wedding in the district town Zdar nad Sazavou is the traditional model compares the captured in the 19th and 20th century. The main objective of this Diploma thesis covering the topic ``Late Romanic heritage sights situated in Posázaví/ in Týnec nad Sázavou and Poříčí nad Sázavou), was to sum up the literature touching those sights, to describe the historical buildings and to link them with the whole architectonic development in ancient Bohemia. In its first part, the Diploma thesis concludes the literature used for my descriptions, in the second part it describes selected buildings {--} chapel of St. Havel in Poříčí nad Sázavou, chapel of St. Petr in Poříčí nad Sázavou, rotunda of St.Václav in Týnec nad Sázavou and extinct Romanic church located in the same city. The thesis also touches building{\crq}s conditions, describes their reconstructions, inventory and ranks them chronologically into the context of ancient Romanic architecture in our country. Finally all my descriptions are also supported by the visual attachment, which is the last part of my work.

The site is located on the border of the old and new buildings – on one side, there is a Stalingrad housing estate from the 1950s built from brick houses in the socialist realism style. On the other side of the site are built new family houses, which are quickly expanding. The smaller part of the object is still used, the rest is abandoned. The house is surrounded by a large garden. The garden is defined by a fence and trees in a way, which creates a barrier, so the building does not communicate with surroundings in any way. The aim of the project is to create a centre of Stalingrad connected with surroundings and creating a meeting place for communities living in the area by using building interventions.

The subject of the diploma thesis is based on the specific intention of the town Žďár nad Sázavou for the reconstruction of the original kindergarten building at Okružní Street. The aim of the project is to create a multifunctional community center with operational and administrative facilities. The main starting point is the work with the existing building - reconstruction and extension of the original kindergarten building. Contrary to conventional new construction "on green meadows, this assignment emphasizes the use of existing values, thickening and strengthening of the existing city, as opposed to the contemporary colonization of the free landscape.
