Vitamin D Sub-set Analysis from the Flash Study

Vitamin D is important to the health of college students. The objective of our study was to measure sun exposure, skin pigmentation, vitamin D intake, and serum 25-hydroxyvitamin D (25[OH]D) in a subset of participants from The Following the Longitudinal Aspects of Student Health (FLASH) Study to determine the best predictors of 25(OH)D status. Participants were college-aged freshman who had their blood drawn in spring (Visit 1) and fall 2010 (Visit 2) at California Polytechnic State University (Cal Poly), San Luis Obispo, CA. (35.3°N). Vitamin D intake was measured using a 28-day food frequency recall questionnaire (specific to vitamin D foods and supplements) while questions specific to the frequency of milk and fish intake were accessed from the FLASH questionnaire. Sun exposure was measured using a 28-day recall questionnaire (time in sun and sun exposure index [SEI]) and questions (frequency of weekday/ weekend exposure) from the FLASH questionnaire. Skin pigmentation was measured using a reflectance spectrophotometer. Serum 25(OH)D was measured at a local pathology lab as measured by an IDS-iSYS. Means (SD) were as follows (n= 40): reflectance of the forehead was 61 (3.5) L* (Lightness) for Visit 1 and 61 (4.3) L* for Visit 2. Vitamin D intake was 308 (234) IU for Visit 1 and 316 (257) IU for Visit 2. Time outside was 81 (44) mins for Visit 1 and 76 (39) mins for Visit 2. Serum 25(OH)D was 85 (24) nmol/L for Visit 1 and 113 (28) nmol/L in Visit 2 which was significantly higher (p < 0.0001). The SEI was 53 (38) body surface area (BSA) exposed (m2) x mins for Visit 1 and 55 (34) m2 x mins Visit 2. Although 90% of participants in Visit 1 and 88% in Visit 2 were below the RDA guidelines for vitamin D intake (600 IU/day), 5% of participants in Visit 1 and none in Visit 2 had serum 25(OH)D serum levels < 50 nmol/L (the recommended level of sufficiency for bone health), demonstrating the importance of sun exposure to vitamin D status in these college students. To determine the strongest predictors of status we used regression analysis to predict serum 25(OH)D with skin reflectance, vitamin D intake, and sun exposure. We found that weekend sun exposure, fish intake, and forehead skin reflectance were the strongest predictors of serum 25(OH)D (R2= 0.50, p= 0.0010) demonstrating that simple questionnaires can help to predict serum 25(OH)D status.

Intuitive Eating, Attitudes to Food, and Body Size: A Comparison Between Nutrition Majors and Non-Majors

Dietitians are expected to deliver sound and scientifically objective advice to the general public, yet their personal beliefs and behaviors could influence delivery of nutrition care. Increased understanding of the personal attitudes and behaviors of dietitians concerning eating behavior and body image could help improve dietetic practice. Traditional nutrition education emphasizes cognitive eating, i.e., monitoring energy intake and comparing macronutrient intakes to the current acceptable ranges. Intuitive eating, however, promotes the release of cognitive eating in favor of greater attention to physiologic cues, or “body wisdom”. We hypothesized that nutrition students in a traditional curriculum would report eating less intuitively than non-nutrition majors. We surveyed 258 female undergraduate students (96 nutrition majors and 162 non-majors) at the University of Massachusetts Amherst. Using Mann-Whitney U tests, we assessed the differences between nutrition majors and non-majors in terms of: intuitive eating, as measured by the Intuitive Eating Scale-2 (IES-2); body mass index (BMI, kg/m2); magnitude of body dissatisfaction (actual weight – ideal weight); and dieting behavior. We also used non-parametric Spearman’s rho correlations and Chi-squared statistics to examine relationships between variables. A two-way between-groups analysis of variance was used to calculate statistical differences in intuitive eating scores between diet behavior and major. Contrary to our working hypothesis, we found that IES-2 scores were significantly higher in majors versus non-majors (p= 0.01) and significantly lower (pnormalor underweight BMI. These observations provide novel information indicating that nutrition undergraduate students, who have the intention of becoming registered dietitians, report that they eat more intuitively and have a lower degree of body dissatisfaction than do undergraduate students not majoring in nutrition. Additional research is needed to address issues related to body dissatisfaction and body weight.

An Assessment Evaluating the Possible Participation of a Virtual Nutrition Education Program for Older Adults

The importance of proper nutrition education among older adults is crucial in promoting overall physical health and management of chronic health conditions. However, access to such education has been limited, especially during the COVID-19 pandemic. With the current boom in social media popularity, as well as virtual communication portals such as Zoom, the question must be asked if nutrition education could be effectively implemented for the use of older adults through these electronic channels. The study design for this particular research was a cross sectional survey to inform development of an online nutrition education program for older adults in Northeast Tennessee. (Appendix B). The survey was distributed as a paper-based survey in seven senior centers and as an online Qualtrics survey distributed by all senior center directors in Northeast Tennessee. A total of 160 surveys were collected (61 paper-based, 99 online), with 150 surveys being complete. Frequencies showed that the majority of participants have internet access and use social media platforms, such as Facebook and YouTube, which was not affected by home location, such as rural versus urban. Most participants were interested in participating in a nutrition education program. The survey also revealed more information about seniors in Northeast Tennessee that will be helpful when developing this program.

RELATIONSHIPS BETWEEN BEHAVIORAL MEASURES AND PRODUCTIVITY IN FINISHING BEEF CATTLE

The relationship between measures of temperament, growth performance, and social hierarchy in finishing beef cattle were explored in two experiments. In experiment 1, high OCS (objective chute score) steers had periods of significantly higher ADG (P < 0.01), but OCS had no relationship with dominance ranking (P > 0.47). Conversely, slow exit velocity (EV) correlated with higher ranking (P ≤ 0.06), but EV had no relationship with performance (P > 0.37) in a competitive environment. Rank showed no relationship with performance (P > .58). In experiment 2, steers with fast EV had periods of decreased growth (P ≤ .06), intake (P ≤ .06), and gain:feed (G:F; P = 0.02). There were no interactions between EV, OCS, and monensin or between EV and monensin. Monensin and EV together, however, significantly impacted overall (days 0 – end) G:F (P = 0.02) and gain (P = 0.05). Overall, these studies further confirm the idea that EV affects performance as does OCS in concert with monensin. Moreover, it further confirms that different measures of temperament correlated to different aspects of performance and should not be lumped together under the general term “temperament” when describing its relationship with performance.

Efficacy of a Mobile Application for Improving Gait Performance in Community-Dwelling Older Adults

The United States is a rapidly aging nation. Older adults have higher rates of falls than any other age group. One in four older adults fall each year. Many of these falls are associated with sedentary lifestyles and decreased muscular strength effecting balance and gait performance. Physical activity (exercise) can reduce the risk of falls among older adults, yet adherence remains low. Exergames can increase adherence to interventions that promote health and physical activity. Social engagement can increase self-efficacy and motivation to exercise. By design, the Bingocize® health promotion mobile application (app) increases social engagement, while providing a multi-factorial fall prevention intervention. The purpose of this investigation was to evaluate the efficacy of the app to improve gait in community-dwelling older adults (N=38; mean age 72.42 years +12.58). Participants were clustered and randomly assigned to (a) experimental (n=20; using app with bingo game, health education and exercise) or (b) control (n=18; using app with bingo game, health education without exercise) condition. Each group completed a tenweek intervention that consisted of two- 45-60 minute sessions per week. Pre and post gait analysis, at self-selected (SS) and fast-walking speeds, measured using the GAITRite® Electronic Walkway (GWS). Gait analysis included parameters of velocity, cadence, step time, step length and width, and single and double support time. A mixedmodel ANOVA (p < .05) was used for statistical analysis. There were no main effects observed. Significant interactions (group x time) were observed at fast speed and SS speed compared to the control group. Significant interactions were observed at fast speed included velocity (λ = .886, F (1, 36) = 4.61, p = .039, 𝜼𝒑 𝟐 = .114); and step length (λ = .864, F (1, 36) = 5.64, p = .023, 𝜼𝒑 𝟐 = .136); and were observed at SS speed for single support time (λ = .887, F (1, 36) = 4.59, p = .039, 𝜼𝒑 𝟐 = .113). Post hoc analyses using paired and independent samples t-tests were conducted on gait variables with observed significant interactions. The independent samples t-test for Single Support Time (SS) post was significant (t (36) = 2.454, p = .019, two-tailed). None of the remaining post hoc analyses were significant. There was a meaningful detectable change (MDC) in mean velocity (>5 cm/s) over time, for both SS and fast walking speeds, within the experimental condition. MDC in gait speed ranges from 5 cm/s (small) to 10 cm/s (large). As for clinical significance, this should be considered a small, yet meaningful detectable change. It is the conclusion of the investigators, that the app, with the exercise intervention, can effectively produce a meaningful change in gait speed (5 cm/s), which has the potential for reducing the risk of falls in older adults. Identification of Non-Nuclear Receptors for 1,25-dihydroxyvitamin D<sub>3</sub> in Chick Kidney and Brain

1,25-dihydroxyvitamin D3 (1,25(OH)2D3) has been shown to mediate the rapid, non-nuclear stimulation of calcium and phosphate transport in chick intestine through binding to a receptor localized in the basal lateral membrane. By using an antibody to the N-terminus of the membrane receptor, studies were undertaken to determine whether a comparable protein exists in kidney and brain, and whether it is present in a particular subcellular fraction. The first step was to establish fractionation protocols to separate subcellular organelles as judged by marker enzyme analyses. Differential centrifugation and Percoll gradient fractions were prepared from chick kidney and brain whole homogenates by two methods (method 1 and method 2). Protein and marker enzymes were analyzed in each fraction to determine the distribution of organelles. By method 1, the organelles were not adequately separated. By method 2, chick kidney and brain were found to have the same order of organelle distribution: In the post-nuclear pellet (P2), fraction 1 was found to be enriched for the lysosomal marker acid phosphatase; fractions 2-5 were found to be enriched for the mitochondrial marker succinate dehydrogenase; fraction 8 was found to be enriched for the Golgi marker α-D-mannosidase; and fraction 9 was found to be enriched for the plasma membrane marker Na+,K+ ATPase. In Percoll gradients of microsomal membranes prepared from the 100,000xg pellet (P3), fractions 1-3 contained the endoplasmic reticulum marker enzyme activity glucose-6-phosphatase. Subsequently, sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDSPAGE) and Western blots were performed using the antibody to 1,25(OH)2D3 receptors in chick intestinal basal lateral membrane. The areas of the bands were scanned by computer, and analyzed quantitatively. After establishing a suitable protein concentration for Western analysis, differential centrifugation and Percoll gradient fractions were analyzed. Finally, the specific binding of [3H]1,25(OH)2D3 was determined in Percoll gradient fractions to assess whether the receptor is functional. Plasma membrane 1,25(OH)2D3 receptors were found in both chick kidney and brain cells. Golgi membranes also were found to have receptor activity, perhaps since this organelle packages proteins for delivery to other membranes. In kidney, fraction P27 demonstrated a very high receptor activity, and [3H]1,25(OH)2D3 specific binding assays showed these membrane receptors are functional. Although this fraction lacks traditional marker enzyme activity, it may contain endocytic vesicles. The physiological function and the mechanism of action of plasma membrane receptors in these two tissues remain to be determined.

Nutritional Ergogenic Aids: The Influences of Carbohydrate-Protein Supplementation During Endurance Exercise

Mixed results, in terms of performance benefits, have been found when comparing carbohydrate (CHO) and carbohydrate-protein (CHO-P) supplementation during endurance exercise. Thus this study assessed performance from three different supplements (CHO-P, CHO, double carbohydrate [CHO-CHO]) as compared to a placebo (PLA) during a time trial (TT) run. Twelve male recreational runners (age = 32.4 ± 9.5 yrs; body mass index [BMI] = 22.7 ± 1.5 kg/m2; VO2max = 59.68 ± 7.53 mL/kg body weight; 100% white) individually completed four, 12-mile TT runs, 7-10 days apart, at about 75% of the their race pace. Dietary and physical activity consistency within the 24-hour time period prior to each run was controlled via individualized diet and activity prescriptions based off of the diet consumed and the physical activity engaged in prior to TT 1. Throughout each TT run, participants consumed a 600 mL load of one of the four aforementioned supplements. Supplement order was counterbalanced with a latin-square design. Endurance performance was measured by time to complete the 12-mile run, and time to complete the last 1.2 miles of the run, where participants were instructed to run at maximal effort, 100% race pace. A main effect of time occurred during the TTs for perceived exertion (RPE) and heart rate (HR). RPE (Borge 10-point scale) significantly increased from the mid-point of the TT to completion of the run (4.7 ± 0.7, 9.7 ± 0.9, p < 0.001); HR significantly increased from the start of the run to the start of the maximal effort, and was significantly higher at completion of the effort (84.4 ± 14.5 bpm, 166.0 ± 8.3 bpm, 178.8 ± 7.4 bpm, p < 0.001). No significant difference was found in overall time to complete the 12-mile run or maximal effort between the supplements (PLA = 88.6 ± 11.6 min, CHO = 89.1 ± 11.3 min, CHO-P = 89.1 ± 11.8 min, CHO-CHO = 89.6 ± 11.9 min; PLA = 8.3 ± 1.2 min, CHO = 8.2 ± 1.2 min, CHO-P = 8.2 ± 1.2 min, CHO-CHO = 8.4 ± 1.5 min). These findings suggest that type of supplementation (CHO, CHO-CHO, CHO-P) consumed during an endurance exercise bout has no influence on enhancing endurance performance in male recreational runners during TT runs less than 100 minutes in length.

Home Food Availability, Dietary Intake and Child Weight Status

Background: Childhood obesity is a critical public health problem. There is a crucial need to identify environmental factors that either encourage or prevent obesogenic behaviors. The home food environment is one of the primary environments in which children are exposed to food. Therefore, it is crucial to study how the home food availability influences dietary intake and weight status. Objective: This study examines parental report of household food availability of fruits, vegetables, and milk, and its association with child weight status and child dietary intake of these foods. Methods: This is a cross-sectional study, based on surveys from 489 students in grades 3-12 their parents in 2009. Child participants were Destination ImagiNation® finalists. Child data collection included surveys based on the Youth Risk Surveillance Survey and anthropometric measurements of height and weight used to calculate child z-BMI. Parents self-reported their heights and weights and home food availability. Descriptive statistics were conducted and multiple linear regression was used to predict whether or not a relationship existed between home food availability, child weight status, and dietary intake of fruits, vegetables, and milk. Results: Overall, 12.5% of participants were overweight and 6.7% were obese. Approximately26% of participants consumed 5 or more servings of fruits and vegetables per day and 17% consumed 3 or more glasses of milk per day. However, a no significant relationship was found between home availability of fruits, vegetables, and milk and the reported intake of these foods. Although a significant association was also found between parent and child weight status, no significant association was found between home food availability and child weight status. Conclusions: Compared to national data, a larger proportion of this sample was classified as normal weight and consumed the recommended intake of fruits and vegetables and milk. Parents reported high availability of fruits, vegetables, and milk products. However, no significance was found between the reported availability of these foods and weight status or dietary intake of fruits, vegetables, or milk.

Changes to the Equine Hindgut Microflora in Response to Antibiotic Challenge

Antibiotics are important to equine medicine, but can cause detrimental side-effects including reduced feed intake, allergic reactions, and diarrhea. Antibiotic-associated diarrhea (AAD) is attributed to disruption of the hindgut microflora, permitting proliferation of pathogenic microbes. The objectives were to evaluate the effects of antibiotics on beneficial fecal bacteria, AAD-associated pathogens, microbial species richness and fermentation. Horses were assigned to treatment groups: control (no antibiotics, n=6), trimethoprim-sulfadiazine (oral, n=6), or sodium ceftiofur (IM, n=6). Fecal samples were taken during adaptation (3 wk), antibiotic challenge (1 wk), and withdrawal (1 wk). Fecal cellulolytics decreased by >99% during challenge and did not recover during withdrawal (P < 0.0001). Lactobacilli decreased by >60% during challenge (P = 0.0453). Salmonella spp. increased 94% with trimethoprim-sulfadiazine challenge (P = 0.0115). There was no detectable Clostridium difficile during adaptation or in any control horse. C. difficile increased (P < 0.0001) when horses were challenged, and remained elevated 7 d after withdrawal. There was no effect of challenge on in vitro digestibility or microbial species richness as evaluated by denaturing gradient gel electrophoresis (P > 0.05). These results indicate that antibiotics can disrupt the normal flora and allow proliferation of pathogens, even without affecting digestibility and causing AAD.

A Comparison of Job Responsibility and Activities between Registered Dietitians with a Bachelor's Degree and Those with a Master's Degree

Minimal educational requirements for Registered Dietitians (RDs) include a bachelor’s degree and practice program. Recently, a master’s degree was recommended. Studies have not established whether education affects employment. A secondary analysis of 2005 Dietetics Practice Audit data determined whether job responsibility, individuals supervised, and activities differed between 1,626 bachelor’s RDs (B-RDs) and 767 master’s (M-RDs) RDs, registered ≤5 years. Chi-square and ANOVA analyzed differences between B-RDs and M-RDs, at entry-level (0-3 years experience) and beyond-entry-level (3+-5 years experience). Beyond-entry-level B-RDs (31.8%) and entry-level M-RDs (31.9%) reported “supervisor/executive” responsibility more than entry-level B-RDs (26.5%; p=0.01). A higher percentage of M-RDs supervised (29.2%) than B-RDs (24.7%; p=0.02); however, B-RDs supervised more individuals (7.38 ± 4.89) than M-RDs (6.25 ± 4.87; t=2.32; p=0.021). A master’s degree has limited benefits; experience may affect responsibility, individuals supervised, and activities more than education.
