Promoter analysis of members of a plant defense-related LRR-RLK gene cluster in Arabidopsis thalianaMumm, Anina 15 July 2014 (has links)
M.Sc. (Biochemistry) / A 14-member, closely-spaced cluster of genes coding for leucine-rich repeat receptor-like kinases (LRR-RLKs) is located on chromosome 1 of Arabidopsis thaliana. Following on from previous microarray studies that found some of the members of this cluster to be upregulated in response to biotic stressors, including the bacterial elicitor flg22, the present study sought to confirm, using a luciferase-based protoplast assay, that flg22 does in fact induce the expression of the genes, and then to investigate the promoters of the genes. The promoters of At1g51790, At1g51850 and At1g51890 responded positively in this particular assay, and bioinformatic analyses determined that W-boxes are over-represented in the cloned regions. Mutational inactivation of individual W-boxes in the promoter of At1g51790 drastically reduced the flg22 response, except for the W-box closest to the start site, which seemed to increase both basal and flg22-inducible expression. In the promoter of At1g51850, mutational inactivation of either or both of its W-box dyads resulted in virtually no flg22 inducibility. The deletion of 6 W-boxes in the promoter of At1g51890, done via truncation, drastically reduced both its basal expression and its inducible response to flg22. These results provide evidence that W-box cis-elements are responsible for the upregulation of these LRR-RLKs in response to flg22. WRKYs -7, -11, -22,and -26 were found bioinformatically to have similar expression patterns to some of the genes in the cluster, and are thus good candidates to investigate as transcriptional regulators of the cluster in future studies.

Characterisation of AtPNP-A - a novel arabidopsis thaliana gene with role in water and salt homeostasisBastian, René January 2009 (has links)
Philosophiae Doctor - PhD / Plant natriuretic peptides (PNPs) are a novel class of extracellular, systemically mobile molecules that elicit a number of plant responses important in homeostasis and growth. Natriuretic peptides were first identified in vertebrates where they play a role in the regulation of salt and water balance. Subsequent experimental investigations have identified the presence of a natriuretic peptide hormone system in plants. While PNPs have been implicated in various physiological responses such as stomatal guard cell movements and regulation of net water uptake, its biological role has remained elusive. Here we have used co-expression and promoter content analysis tools to understand the biological role of the Arabidopsis thaliana PNP (AtPNP-A). The analysis of AtPNP-A and its co-expressed genes revealed that genes annotated as part of the systemic acquired resistance (SAR) pathway were over-represented, thus suggesting that AtPNP-A may function as a component of plant defense responses and specifically, SAR. The results further show that AtPNP-A shares many characteristics with pathogenesis related (PR) proteins in that its transcription is strongly induced in response to pathogen challenges, thus implying a newly described role for AtPNP-A in pathogen attack. Additional tissue expression analysis also indicated distinct localization of PNP activity in sepals and transcriptional meta-analysis showed that AtPNP-A may play a role in starch breakdown. Therefore, together with the finding that AtPNP-A plays a role in regulating phloem transport, we also hypothesize that AtPNP-A may play a role in phloem unloading in sepals to assist processes such as seed formation in plants. In plants, the second messenger, guanosine 3’,5’-cyclic monophosphate (cGMP) mediates a whole range of important processes including salinity tolerance, disease resistance, drought tolerance and responses to light. Since PNPs regulate water and salt homeostasis via a cGMP-dependent signaling pathways, it is thus important to analyse the transcriptome induced by the second messenger (cGMP) in Arabidopsis thaliana to give a better understanding of its mechanism of action. This study was also supplemented by the analysis of the gibberellic acid (GA) dependent transcriptome, since cGMP also plays a role its transcription pathway. This data analysis, together with promoter content investigation, revealed that genes upregulated after cGMP treatment and down-regulated in the GA insensitive mutant (ga1-3) were enriched with a GA response element (GARE), while no GARE enrichment were observed in genes up-regulated in the ga1-3 mutant. These findings suggest that GARE is indicative of GA-induced and cGMP-dependent transcriptional up-regulation. Gene ontology analysis confirmed previous reports that cGMP is involved in ion homeostasis and indicated that the transcriptional cGMP response is bi-polar in the sense that both genes up- and down-regulated in response to cGMP is involved in cation transport. Additionally, ab initio analysis of genes transcriptionally dependent on cGMP identified CHX8 as a hub gene and promoter content of CHX8 co-expressed genes show enrichment of the GARE motif. The fact that CHX8 has its highest expression levels during male gametogenesis and pollen tube growth, together with our findings, suggest that GA-induced and cGMP- dependent genes may play a key role in ion and water homeostasis in the male gametophyte. Finally, we propose that the type of analysis undertaken here can yield new insights into gene regulation networks and inform experimental strategies to unravel complex transcription regulatory systems under different developmental and stimulus specific conditions. / South Africa

A functional analysis of the response of the Southern Kalahari dune vegetation to land-use intensityGeldenhuys, Conrad 16 November 2011 (has links)
Natural and anthropogenic environmental variables pressures exert fitness pressures on our flora and vegetation and necessitate an understanding of the mechanisms of plant responses to those pressures. Overgrazing and desertification are examples of the stressors that affect plant community composition, on both large and small scales. In order to investigate the plant community response to these stressors it has been shown that it is more practical and informative to group plant species according to their functional characteristics into functional types than attempting to predict plant response solely based on previous range distribution data. The present study involved a survey of the dunefield vegetation of several privately owned farms as well as a communal farm in the southern Kalahari. The farms were subjectively chosen to represent a gradient from underutilized to overutilized. Plant community surveys were conducted on the farms and the dominant plant species were sampled and researched for selected functional traits to explore possible relationships between traits and grazing pressure. Ordination techniques were used to identify plant functional grazing response types. The identification of plant functional types through easy measurable traits will be a great aid in modelling and longterm environmental monitoring approaches. Of the three dunefield habitats, the dune street habitat was identified as the most vulnerable to overgrazing. A reduction in cumulative cover and an increase in bare surface cover indicated increased grazing pressure in the dune crest habitat. In the dune streets shrub cover and annual grass cover increased when veld was overgrazed. Functional analysis of 112 Kalahari plant species for a set of 47 plant traits showed that species clustering were strongly related to vegetative characteristics. The emerging clusters were strongly associated with major life forms, which comprised phanerophytes of different size classes, geophytes and herbaceous perennials, as well as herbaceous annuals. These clusters were identifiable even with the inclusion of reproductive traits. Selected vegetative traits are good indicators of plant response to land-use intensity since these traits lend competitive advantages to plant persistence under utilisation pressure. Plant height, spinescence, specific leaf area, leaf dry matter content, leaf sclerophylly and stem density were found to be examples of such traits. Phanerophyte and chamaephyte dominated functional groups were mostly associated with lower range condition and hemicryptophyte dominated groups with higher range condition, whereas herbaceous therophyte dominated groups did not show any specific association. / Dissertation (MSc)--University of Pretoria, 2011. / Plant Science / unrestricted

Allelopathic effects of bankrupt bush (Seriphium plumosum) on the germination ability of specific plant speciesMokou, Moketla Baltimore January 2016 (has links)

(Plant production)) -- University of Limpopo, 2016 / Seriphium plumosum is a declared indicator of bush encroachment, and poses a serious threat to the management of sustainable utilization in all grasslands. The successful invasiveness of S. plumosum is attributed to its competitive ability and high allelopathic potential. A trial was established at the University of Limpopo to investigate the interference between S. plumosum and four plant species, namely: Eragrostis curvula, E. tef, Panicum maximum and Lactuca sativa. Plant material of S. plumosum were collected and used to make infusions which were used on the receiver species.
The infusion inhibited the germination of all the receiver species, and it was highly significant (P ≤ 0.01), compared to control treatments where no inhibition occurred. All receiver species were sensitive to roots and shoots infusions, but the effect of shoots infusion differed significantly (P ≤ 0.01) from those of roots infusion. All receiver species were sensitive to both summer and winter collected materials, but plant material collected in winter had a bigger effect (P ≤ 0.01) than plant material from summer. All receiver species were sensitive to both fresh and stored plant material, but inhibition effects were not statistically significantly different (P ≥ 0.05).
All receiver species were sensitive to both fresh and stored soils collected in infested areas, but effects were not significantly different (P ≥ 0.05), while the effects of infested and un-infested soils differed significantly (P ≤ 0.01). All receiver species were sensitive to soils collected during summer and winter. Where infested soils were concerned, all receiver species were sensitive to infested soils, compared to control treatments where no effects occurred. Where stored infested soils were concerned, all receiver species were sensitive to both fresh and stored infested soils. It was concluded that both plant material of S. plumosum and soils from areas encroached by S. plumosum have a negative effect on seed germination of the four receiver species. Should a farmer control this species by means of cutting, it should be quickly removed to allow the gass to recover. The farmer must not expect quick recovery of grasses due to the presence of allelopathic substances in the soil. / Agri-seta

Celery mosaic 1s e virus disease of major economic importance in Utah. In yeare of severe epiphytotics. it has been a limiting factor in celery production. In 1947 losses due to this disease were estimated to exeed 8 per cent of the total crop (17). This estimate does not include losses resulting from reduction in grade or from plants discarded in the packing sheds. The celery acreage in Utah dropped from a high of 950 acres in 1946 to 750 acre. in 1947 and dropped again in 1948 to 420 acres. Leaders in the industry attributed this reduction in Acreage, in part at least, to losses incurred by the growers from celery mosaic.
