A cor vermelha brilhante característica do presunto Parma é resultante, principalmente, do pigmento Zinco-protoporfirina IX (ZnPP). A ZnPP é formada a partir da mioglobina por uma reação de transmetalação, catalisada pela enzima ferroquelatase (FECH), em que o íon de Fe(II) coordenado ao grupo heme é substituído pelo íon Zn(II). O presunto Parma apresenta uma maior estabilidade oxidativa em relação aos demais produtos cárneos curados além de não conter nitrito e nitrato, portanto, são considerados mais saudáveis. A utilização da FECH no processamento de carnes curadas pode permitir a produção de produtos cárneos curados mais saudáveis e em menor tempo. No presente trabalho a proteína ferroquelatase de Bacillus subtilis (BsFECH) foi expressa em células de E. coli BL21(DE3), purificada por cromatografia de afinidade ao níquel e exclusão por tamanho e caracterizada por dicroísmo circular, emissão de fluorescência do triptofano e cromatografia de exclusão por tamanho analítico. Em termos de estabilidade foi encontrado que altas concentrações de sal aumentam a estabilidade da proteína frente aos agentes denaturantes ureia e temperatura. A BsFECH produzida é capaz de ligar-se ao substrato modelo de porfirina (TPPS), conforme verificado por espectroscopia de UV-Vis, com uma Ka = 3,8x105 M-1 e é capaz de se associar à metamioglobina, conforme verificado por reação de cross-linking com dissuccinimidil suberato e avaliado por SDS-PAGE. A BsFECH aumenta significativamente a taxa de inserção de íons de zinco na TPPS e mostra uma cinética de saturação com uma constante de ligação aparente de Zn(II) ao complexo [BsFECH-TPPS] de 1,3x104 M e uma constante de primeira ordem de 6,6x10-1 h-1 para a dissociação do complexo ternário. A reação de troca ferro/zinco na mioglobina catalisada pela BsFECH é facilitada pela proteólise limitada da mioglobina com pepsina que abre um caminho para a reação de troca metálica com base na interação proteína-proteína entre o fragmento globina da mioglobina e a BsFECH. / The bright red color, characteristic of the Parma ham, results mainly of the pigment Zinc-Protoporphyrin IX (ZnPP). Functional design opportunities for water conservation through native landscape restoration in southwestern Idaho, U.S.A.

This creative project inspected the design of water conserving landscapes by restoring native vegetation. Water conserving design principles discovered in the background research were applied to the redesign of the Old Fort Boise Park in Parma, Idaho. Important issues that were explored included discussing the need for water conserving landscape designs that result in functional spaces that meet the needs of intended users. The main focus of water conservation was implemented by using native plants that typically require less water than non-native species. The project developed a park master plan that could demonstrate to Idaho residents that native plants, when carefully sited, result in lower maintenance, aesthetically pleasing visual qualities, environmentally sensitive performances and could be integrated into landscapes on private or public properties. Rather than viewing the Idaho native plants as unattractive and dull, this project explored the idea that under cultivation and as part of the site character a carefully designed water conserving naturalized landscape has a beauty unsurpassed by traditional landscapes. The park redesign also employed educational and interpretive systems wherein native plants were highlighted and identified. These educational and interpretive systems could assist park users, especially local residents, in familiarizing themselves with many of the beautiful native species that are common in the surrounding deserts, prairies, hills and mountains. The creative project examined one particular site, while it developed principles and concepts applicable to other sites throughout the Great Basin Region. / Department of Landscape Architecture

Engineering procesů veřejné správy - projekt PARMA / Process engineering of Public administration - the PARMA project

The Parma project deals with process (re)engineering of public administration. In the first phase the project focused on mapping the processes of a local authority. Then the key processes were set and the project moved to the reengineering phase. This thesis shows that the matter is much more complicated and goes beyond public administration as such. The process perspective allows to create many different processes coping with the same issue. The thesis, with the help of Prof. Boullier's work ("La Boussole écodémocrate"), offers a "philosophical" framework of public sector. This contribution is the foundation for future Parma project development. The thesis contains some propositions designed for the Parma project management improvement. It also presents the methodology used in the Parma project (MMABP -- Methodology for Modelling and Analysis of Business Processes), and gives some propositions for its improvement.

A huge difficulty in the forecast of flow is the seasonality presence, due to drought and flood periods in the year. Many scientists, with different methodologies, have been concerned with finding a best model, compared with the utilized by Brazil s system - Markovian Model. The Makovian Model, or selfregressive with order 1, is a Box & Jenkins methodology, and requires data handling to treat non-stationarity, or the use of regular models, requiring a hardly theoretical formulation for the statistical procedures. Therefore, the statistical models, autoregressive model with seasonality and Holt-Winters model, of treatment of temporal series are presented and, carried out the flow s analysis and forecast for three study groups, in two different (historical) horizons. The performance of the models was compared and the results showed that the proposed models presents better adjust than the model adopted by Brazilian system / No planejamento da operação energética, a análise e previsão de vazões são muito importantes. Uma grande dificuldade na previsão de vazões é a presença da sazonalidade, devido aos períodos de seca e cheia no ano. Muitos estudiosos, com metodologias diversas, têm se preocupado em encontrar um modelo de melhor ajuste, em comparação ao utilizado pelo sistema brasileiro, ou seja, o modelo auto-regressivo de ordem 1, que consiste numa metodologia de Box & Jenkins e exige manuseio nos dados para tratar a não-estacionariedade. O presente trabalho analisa e compara os modelo utilizados pelo sistema brasileiro (PAR), com modelo matemático que considera a sazonalidade dos dados (SAR) e o método de Holt-Winters e, modelos amplamente estudados como PARMA e ANFIS. O desempenho dos modelos foi comparado e os resultados mostraram que em muitos estudos os modelos PAR/PARMA e ANFIS apresentam melhor ajuste , no geral, em relação aos demais
