QEEG Correlates of Cognitive Deficits in Multiple Sclerosis During Targeted Cognitive Tasks

Multiple sclerosis (MS) is the most common neurological disorder of young adulthood and is often associated with cognitive impairment and emotional dysfunction. Due to the nature of the disease, the cognitive deficits in MS are often variable in their presentation, and consist of deficits in processing speed, attention, working memory, and executive functioning. The purpose of the present study was to explore common methods of documenting MS-related cognitive deficits, to elucidate the relationship between the cognitive deficits seen in MS and physiological markers of cognitive functioning (i.e., quantitative EEG), and to analyze the relationship between cognitive deficits and mood dysfunction in MS. There were 26 participants diagnosed with remitting-relapsing multiple sclerosis and 18 age, sex, and education matched controls. Results of cognitive testing indicated deficits in gross cognitive functioning, language, attention, processing speed, working memory, and executive functioning. A MANOVA encompassing group, task (PASAT and SPT) and load (light and heavy) showed significant group and load effects, but no main effect of task. The MS group performed worse than the controls and both groups performed better on the light load than the heavy load. Post hoc analysis indicated that performance on the PASAT 3 second trial was worse than on the PASAT 2 second trail compared to controls. Given that the PASAT 3 trial is theoretically easier than the PASAT 2 trial and that the PASAT 3 was administered first, the above results likely reflect learning effects. A Repeated Measures ANCOVA encompassing EEG and cognitive data (PASAT and SPT) indicated group-level differences on task performance, and suggested that at rest mean peak alpha frequency (PAF) is associated with performance on the PASAT, but not the SPT. EEG coherence during cognitive tasks was reduced between short-range connections in the theta, alpha, and beta frequency bins and enhanced in a limited number of long-range, anterior to posterior connections in the theta frequency bin in the MS group compared to controls. Finally, the MS participants had significantly more symptoms of depression and anxiety compared to normal controls. A hierarchical multiple regression analysis suggested that cognitive functioning is deleteriously affected by depression and anxiety. Overall, the results of this study substantiate the feasibility of utilizing QEEG as a physiological indicator of cognitive and cortical dysfunction in MS and show the importance of recognizing depression and anxiety and their contributions to cognitive deficits in individuals with MS.

Mate Choice in a Sexually Dimorphic Marine Bird, the Great Frigatebird (Fregata minor)

Darwin's theory of sexual selection explains the existence of sexual dimorphism, or within-species sex differences in shape, color, size, and behavior. In some cases, sexually dimorphic traits, especially extravagant male ornaments, seem maladaptive and thus in opposition to natural selection. The crux of Darwin's theory was that sexual selection arises from individual differences in reproductive success that result from competition for mates. In this dissertation, I investigated several aspects of sexual selection and the evolution of female mating preferences and male ornaments in the great frigatebird (Fregata minor). Frigatebirds as a group (family Fregatidae) are the most ornamented of any seabirds, and are among the most ornamented of any animal group. Their most prominent ornament is a gular (throat) pouch which becomes red in males during the breeding season, and which is inflated and displayed to females during courtship. Male courtship display also includes a warble vocalization and extension and trembling of the wings. I investigated the following issues concerning sexual selection and ornamentation in great frigatebirds: 1) the source of ornamental coloration in male great frigatebird gular pouches. I determined that this was a carotenoid-based color display; 2) the relationship of male mating success to gular pouch size and coloration. I determined that mating success was not related to the size or color of this ornament; 3) the relationship between male vocal display traits and female preferences. Again, I found no relationship between vocal display traits and female preferences, and finally, 4) the role of a major histocompatibility complex (MHC) locus in female mate choice. The MHC is a highly polymorphic multi-gene family associated with immune defense and has been proposed to play a role in mate choice. I found a significant disassortative mating pattern amongst mated pairs compared to random pairings based on MHC genotypes. In summary, I found no evidence for female mating preferences based on visual or auditory display traits associated with male ornamentation. However, I did find evidence for female mating preferences based on genetic dissimilarity at an MHC locus.

THE ACOUSTIC EMISSIONS PRODUCED BY ESCHERICHIA COLI DURING THE GROWTH CYCLE

The objective of this study was to determine if acoustic emissions (AE) generated by three strains of Escherichia Coli (5024-parent strain, 8279-mutant strain and 8279-random/unrelated strain) could be used to differentiate each strain during their growth cycle. An acoustic sensor with an operating range of 35 kHz-100 kHz was inserted into the growth vessel and attached to a selected channel to capture AE data. The growth vessel was loaded with 60 ml of tryptic soy broth (TSB) (0.25% fructose) media with alginate (1.1%) or without alginate and inoculated with 1% (108 CFU/ml) of an E. coli strain. The growth vessel was placed in a monitoring chamber and incubated at 32°C for 8-9 h. The AE’s generated by each strain were collected throughout the growth cycle. All strains grown in media with and without alginate generated AE’s within 5 min post inoculation. Strains grown in media without alginate generated stronger (P < 0.0001) absolute energy (ABSE) and higher peak frequencies (PFRQ’s), than in media with alginate. The AE’s generated by strains 5024 and 8237 were stronger and easily distinguished from those generated by strain 8279. Strain 8237 generated 12% stronger ABSE from the 3rd to 8th h and 51% stronger PFRQ intensities than strain 5024 during 0-8 h. However, strain 5024 generated 15% stronger ABSE and 31% higher PFRQ’s during the final hour of growth. Strain 5024 generated the highest PFRQ’s from 5-50 kHz, while strain 8237 generated higher frequencies from 100-500 kHz. Fourteen distinguishable differences (P< 0.05) in generated PFRQ’s, between strains 5024 and 8237, were also observed in every 5 kHz increments from 100-500 kHz. Of these differences, strain 8237 generated higher frequencies within eight of the kHz ranges, while strain 5024 generated higher frequencies within six other kHz ranges. These data suggests that all bacteria may generate different AE’s, thus producing a unique “fingerprint” of sound that will allow for its identification.
