Análise histológica e de microscopia eletrônica do desenvolvimento inicial de jaú (Zungaro jahu)

As coletas foram realizadas na Usina Hidrelétrica Engenheiro Souza Dias – Jupiá (CESP – Castilho,SP). As amostras foram coletadas em tempos pré-determinados da extrusão até à absorção total do vitelo, e destinadas ao processamento para análise em microscopia de luz, eletrônica de varredura e transmissão. O desenvolvimento embrionário do Z. jahu foi dividido em 7 estágios: zigoto, clivagem, mórula, blástula, gástrula, organogênese e eclosão. Durante o período larval observou-se o desenvolvimento dos principais órgãos, dentre eles: órgãos do sistema digestório, coração, arcos branquiais, sistema nervoso/sensorial, tireóide, pronefro e bexiga gasosa. Os ovos na extrusão apresentaram diâmetro médio de 1,38±0,06 mm, e as larvas eclodidas 3,79±0,11 mm. Os ovócitos e ovos, após a fertilização, apresentaram formato discóide, coloração amarela e uma única micrópila. Aos 10 min pós-fertilização (mpf) houve diferenciação dos pólos animal e vegetativo. A formação da célula-ovo ocorreu aos 15 mpf. Dos 20 aos 75 mpf ocorreram sucessivas clivagens até a fase de mórula observada entre 90 e 105 mpf. Em seguida identificaram-se as fases de blástula e gástrula, com 120 e 180 mpf, respectivamente. O final da fase de gástrula, 360 mpf, caracterizou-se pela formação do tampão vitelino. A partir disso, iniciou-se a diferenciação das regiões cefálica e caudal, o alongamento do embrião, culminando na eclosão de larvas transparentes com pontos de pigmentação após 780 mpf. A absorção total do vitelo foi observada com 60 horas pós-eclosão (hpe) a uma temperatura média de 28,75±0,59 °C.... / The aim of this study was to describe the early embrionary and larval development of Zungaro jahu, a Brazilian fish, commonly known as Jaú belonging to Siluriform Order and in danger of extinction. The collections were carried out at the Usina Hidrelétrica Engenheiro Souza Dias – Jupiá (CESP – Castilho/SP). The samples were collected in pre-established times at eggs extrusion until the yolk absorption and processed to analysis by light, scanning and transmission electron microscopy. Early embrionary development of Z. jahu was divided in seven stages: zygote, cleavage, morula, blastula, gastrula, organogenesis and hatching. During the larval period the main organs development was noted: digestive tract, heart, gills arches, nervous/sensorial systems, thyroid, kidney and swim bladder. The eggs dimensions in average diameters at extrusion were 1.38±0.06 mm and the larvae were 3.79±0.11 mm. The fertilized oocytes and eggs presented a discoid shape, yellowish colour and a single micropyle. At 10 minutes after fertilization (maf) there was the differentiation of animal and vegetative poles. The egg cell formation occurred at 15 maf. At 20 to 75 maf occurred successive cleavages until morula phase seen at 90 and 105 maf. Then blastula and the gastrula phases were identified at 120 and 180 maf, respectively. The end of gastrula phase at 360 maf was characterized by formation of the yolk plug. From this there was the differentiation of cephalic and caudal regions, embryo elongation, ending in the hatching of transparent larvae with pigmentation sites at 780 maf. The total yolk absorption was noted at 60 hours after hatching (hah) to an average temperature of 28.75±0.59 ºC.
