Influência clínica e tecidual do diazepam no periodonto de sustentação durante o movimento ortodôntico

Existem evidências de que o AMPc exerce importante papel como mediador intracelular do remodelamento ósseo induzido pela pressão mecânica. A amostra consistiu de 36 cobaias (Cavia porcellus), com 9 semanas de vida, pesando entre 400 a 450g e divididos em grupo controle, placebo e experimental. Foram injetados 2,5mg/Kg de diazepam pela via intraperitonial, a cada 12 horas, nos animais do grupo experimental. Os animais dos grupos placebo e experimental, submetidos à movimentação ortodôntica, foram sacrificados 3, 7 e 14 dias após a aplicação da força. O dispositivo ortodôntico foi confeccionado com fio da liga titânio molibdênio com 0. 016" de diâmetro, no qual foi incorporado mola helicoidal liberando 25g/f. Verificou-se que o diazepam afetou o remodelamento ósseo, aumentando em 70% a extensão do movimento dentário, porém com absorção radicular. Expressivas alterações teciduais caracterizadas pelo aumento no tamanho dos osteoclastos, infiltração de macrófagos, danos às demais células do periodonto e desorganização das fibras do ligamento também foram vistas no grupo experimental. / There were evaluated the effect of diazepam, a potent inhibitor of the cyclic AMP (cAMP) hydrolysis, on the tissue reactions related to orthodontic tooth movement. There is evidence suggesting that cAMP plays an important role as intracellular mediator of bone remodeling induced by mechanical stress. The sample consisted of thirty-six guinea pigs (Cavia porcellus), 9 weeks of age, ranging in weight from 400 to 450g, divided into control group, placebo group and experimental group. ln the concentration of 2,5mg/Kg, diazepam was injected intraperitoneally every 12 hours to the experimental group. The animals of the placebo and experimental groups, submitted to orthodontic movement, were sacrified at 3, 7 and 14 days after the application of orthodontic force. The appliance was made with a 0.016" titanium molybdenum alloy wire formed into helical springs delivering 25g/f. lt was verified that diazepam affected bone remodeling, increasing in 70% the extension of the dental movement, but with root resorption. Marked tissue alterations characterized by large osteoclasts, macrophagic infiltration, damage to the other periodontal cells and disorganization of the ligament fibers were also seen in the experimental group.
