Seed germination and early plant development is a crucial phase of plants' life. Multiple internal and external stimuli influence germination progress and have a serious impact on a plant's survival and vitality. Biotic and abiotic stimuli trigger a whole range of changes, both on molecular and developmental levels, but the complex molecular mechanisms regulating these responses are far from being resolved. This thesis reviews the seed germination process and outlines the role of external stimuli in its progress. The experimental part describes the development of a method for seed germination monitoring, provides new insight into the role of hydrogen peroxide in germination, and analyzes effects of cadmium ions, temperature, salt and drought on proteome and metabolome of germinating seeds of Hordeum vulgare. In total, 2000 proteins and 800 metabolites were identified. The analyses revealed over 95 putative abiotic stress markers, including 63 and 36 proteins and metabolites, respectively

Biosensoren werden oft für die Messung einzelner Substanzen in komplexen Medien verwendet, wie z.B. bei der Blutzuckerbestimmung. Sie bestehen aus einem physikochemischen Sensor, dem Transduktionselement, und einer darauf immobilisierten biologischen Komponente, dem Erkennungselement. In dieser Arbeit wurde als Transduktionselement eine Elektrode und als Biokomponente das Enzym „Meerrettich Peroxidase“ (engl. horseradish peroxidase, HRP) verwendet. Solche HRP-Elektroden werden für die Messung von Wasserstoffperoxid (H2O2) eingesetzt. H2O2 wird im Körper von weißen Blutkörperchen produziert, um Bakterien abzutöten, wird teilweise ausgeatmet und kann in kondensierter Atemluft nachgewiesen werden. Da viele weiße Blutkörperchen bei einer Chemotherapie abgetötet und dadurch die Patienten anfälliger für Infektionen werden, muss ihre Anzahl regelmäßig überwacht werden. Dazu wird zurzeit Blut abgenommen. Im ersten Teil dieser Arbeit wurde untersucht, ob eine Überwachung der Anzahl an weißen Blutkörperchen ohne Blutabnahme durch eine H2O2-Messung erfolgen kann. Ein direkter Zusammenhang zwischen der ausgeatmeten H2O2-Menge und der Zahl der weißen Blutkörperchen konnte dabei nicht festgestellt werden. Für empfindliche H2O2-Messungen mit einer HRP-Elektrode ist ein schneller Austausch von Elektronen zwischen der Elektrode und dem Enzym notwendig. Eine Vorraussetzung dafür ist eine kurze Distanz zwischen dem aktiven Zentrum des Enzyms und der Elektrodenoberfläche. Um einen kurzen Abstand zu erreichen wurden im zweiten Teil dieser Arbeit verschiedene poröse graphitähnliche Materialien aus pyrolysierten Kobalt-Porphyrinen für die Elektrodenherstellung verwendet. Dabei stellte sich heraus, dass eines der untersuchten Materialien, welches Poren von etwa der Größe eines Enzyms hat, Elektronen etwa 200mal schneller mit dem Enzym austauscht als festes Graphit. Die HRP selbst enthält in seinem aktiven Zentrum ein Eisen-Protoporphyrin, also ein aus vier Ringen bestehendes flaches Molekül mit einem Eisenatom im Zentrum. Reagiert die HRP mit H2O2, so entzieht es dem Peroxid zwei Elektronen. Eines dieser Elektronen wird am Eisen, das andere im Ringsystem zwischengespeichert, bevor sie an ein anderes Molekül oder an die Elektrode weitergegeben werden. Im letzten Teil dieser Arbeit wurde das Eisen durch Osmium ausgetauscht. Das so veränderte Enzym entzieht Peroxiden nur noch ein Elektron. Dadurch reagiert es zwar langsamer mit Wasserstoffperoxid, dafür aber schneller mit tert-Butylhydroperoxid, einem organischen Vertreter der Peroxid-Familie. / Biosensors are often used for the measurement of specific substances in complex media, e.g. glucose in blood. They consist of a physicochemical sensor, the transducer, onto which a biological component, the recognition element, is immobilised. In this work, an electrode was used as transducer and the enzyme “horseradish peroxidase” (HRP) as biological component. Such HRP electrodes are used for the measurement of hydrogen peroxide (H2O2). H2O2 is produced in the body by white blood cells to destroy bacteria, is partially exhaled and can be measured in breath condensate. Since a lot of white blood cells are destroyed during chemotherapy and patients get more prone to infections, their amount must be checked regularly. Currently blood samples are taken for this purpose. In the first part of this work it was investigated, if the amount of white blood cells can be checked without taking blood by measuring H2O2. A correlation between the amount of exhaled H2O2 and the number of white blood cells could not be found. For a sensitive H2O2 measurement with an HRP electrode a quick exchange of electrons between electrode and enzyme is needed. One condition for this is a short distance between the active centre of the enzyme and the electrode surface. In order to achieve a short distance, several porous graphite-like materials made of pyrolysed cobalt porphyrins where used in the second part of this work for the electrode production. It turned out that one of the tested materials, which had pores about the same size as the enzyme, did exchange electrons with the enzyme about 200 times faster than solid graphite. HRP itself contains an iron protoporphyrin, i.e. a planar molecule consisting of four rings with an iron atom in the middle, its active centre. When HRP reacts with H2O2, it takes two electrons from the peroxide. One of these electrons is stored at the iron, the other in the ring system, until they are passed on to another molecule or the electrode. In the last part of this work, the iron was exchanged with osmium. The modified enzyme takes only one electron from peroxides. Thus it reacts slower with hydrogen peroxide, but faster with tert-butylhydroperoxide, an organic member of the peroxide family.

Studium vlivu elektrolytů na stabilitu a efektivitu diafragmového výboje / Study of electrolyte influence on diaphragm discharge stability and efficiency

This thesis is focused on so-called diaphragm discharge, which is one kind of electric discharges in liquid, which belongs among so-called AOP´s techniques, still more used for water cleaning in the present. One of effectiveness and stability indicators of diaphragm discharge is generation of hydrogen peroxide. In theoretical part, detail principle description of electric discharge in liquid is situated. Further, properties of electrolyte are introduced and general spectrophotometric method of obtained sample determination is described. In experimental part, a full procedure of experiment is introduced. Next part containing results and discussions introduces particular results of individual measurements and their reasons. Final chapter is the end, which forms total summary and evaluation of all results. By the application of all chosen electrolytes in solution at diaphragm discharge formation of hydrogen peroxide has appeared. Inorganic and organic electrolytes were used. As inorganic electrolytes following salts were selected – solutions of halogenides, next sodium nitrate as a representative of nitrates, potassium dihydrogenphosphate as a representative of phosphates, etc. Representative of organic electrolytes was citric acid. The value of initial conductivity of electrolytes had the main influence on hydrogen peroxide formation. Electrolytes potassium dihydrogenphosphate and sodium sulphate the great influence on effectiveness and stability of the diaphragm discharge. Their rate constants reached maximum value by the application of solution with initial conductivity of approximately 400 mikrosiemens, particularly 0.0492 mmol/l.min and 0.048 mmol/l.min. On the contrary, low values of rate constant were achieved in electrolyte ammonium chloride at around the same initial conductivity – 0.0269 mmol/l.min. During experiments stainless steel and platinum electrodes were used. It was found that kind of electrode material hadn’t influence on generation of hydrogen peroxide. Hydrogen peroxide was formed only in the cathode space.

Main goal of thesis is demonstration and study of H2O2 photoproduction. Photosensitizer used in this study was 5,10,15,20-meso-tetrafenylporfyrin (TPP), studied were also its complexes with Cu2+ a Ni2+ . Properties of nanofibers were examined using UV-Vis molecular absorption spectroscopy, fluorescence spectroscopy and electron microscopy (SEM).

Abstract: The presented scientific work was conducted in two main directions. The first one is an investigation of the simple biomolecules (glycine and sarcosine) bonding to cerium oxide model films by surface science techniques: photoelectron and near-edge X-ray absorption spectroscopies. Adsorption chemistry and thermal stability of the molecules on the oxides were studied in relation to the oxidation state of ceria cations, film morphology, and molecular deposition method. The oxygen vacancies in the oxide were shown to affect the adsorption geometry of glycine and stimulate molecular decomposition. The polycrystalline oxide morphology provided stabilizing effect on the glycine adlayer. Sarcosine deposited in vacuum formed densely packed adlayer with the molecules directed outwards. Interestingly, the results revealed that molecular film deposited from the aqueous solution, in contrast to deposition in vacuum, induces continuous reduction of the cerium oxide during thermal annealing. Jihomoravský lignit jako zdroj uhlíkových nanočástic / South-Moravian lignite as a source of carbon nanoparticles

This Master’s thesis deals with isolating carbon nanoparticles from South Moravian lignite using "green" – environmentally friendly methods. This method consists of mechanical-chemical-thermal stressing of lignite using ultrasound, hydrogen peroxide and high temperatures. This work aims to develop recommendations on coal as a source of carbon nanoparticles and based on these recommendations, to design procedures for isolating nanoparticles from South Moravian lignite using environmentally friendly methods. The identified procedures were designed to meet carbon-based particles with an oxidized surface. From the results we can deduct that it is possible to use South Moravian lignite for the preparation of carbon nanoparticles. However, it will be necessary to modify the process further and also to optimize the purification of the obtained particles.

The aim of this master thesis is the preparation and characterization of plasma activated water (PAW) prepared by dielectric barrier discharge at atmospheric pressure in air for the future applications in agriculture and medicine. The theoretical part is focused on plasma activated water and its effects on seeds and plants. The work also deals with the preparation and application of PAW in the world. Various types of plasma discharge configurations suitable for its preparation are described here. In the experimental part of the master thesis, distilled and tap water were chosen for activation. The concentrations of active particles in PAW generated by DBD such as nitrites, nitrates and hydrogen peroxide were determined. The value of the concentration of active particles changes probably due to mutual interactions when nitrites are oxidized by hydrogen peroxide to nitrates. Physical properties of PAW such as conductivity and pH were also characterized. After the preparation of PAW, the conductivity increases due to the increase in the concentration of active particles formed by dissociation and ionization of water molecules by plasma. The particles presented in the air, which are dissociated and ionized by plasma, also diffuse into the liquid, and contribute to this increase. The decrease in pH also corresponds to an increase in the concentration of hydrogen ions and the formation of nitric acids. Another aspect of this work was to study plasma activated water during the first eight hours after its preparation and to determine its properties. We have found that PAW prepared from tap water retains its characteristic properties for at least 8 hours and distilled water for 6 hours. In the master thesis, the analysis of plasma discharge was performed using optical emission spectroscopy. OES measurements were performed in the wavelength range 300 to 800 nm. The second positive nitrogen system (N2 (C 3u) N2 (B 3g)) and the first positive nitrogen system (N2 (B 3g) N2 (A 3+u)) were identified in the overview spectrum. Molecular nitrogen ions, OH· radicals, atomic oxygen and atomic hydrogen can be observed in the spectrum, too. The dielectric barrier discharge was also captured using a high-speed camera, and the recording shows the occurrence of multiple discharges that are spatially as well as temporary unstable.

Aktivity hemocytů plovatkovitých plžů a jejich změny způsobené nákazou trichobilharziemi / Lymnaeid snails: hemocyte activities and their changes caused by Trichobilharzia infections

Molluscs as well as all other invertebrates rely on innate immune response only. Their internal defense system is capable of destroying most pathogens. However, there are some exceptions, e.g. some snails serve as intermediate hosts for some trematodes. Trematodes are able to develop inside these snails due to intervention in the snail internal defense system. The submitted thesis describes hemocyte activities of two lymnaeid snails, Lymnaea stagnalis a Radix lagotis, and the influence of Trichobilharzia regenti infection on R. lagotis hemocytes. Hemocytes of both species exposed to various chemicals produced different amounts of H2O2 and NO. The response varied between both lymnaeid species. The amount of circulating hemocytes was elevated in R. lagotis snails due to T. regenti infection. However, the infenction attenuated hemocyte activities monitored by us. Hemocyte basal NO production was decreased as well as phagocytosis of bacteria, cell adherence and pseudopodia formation. Toxicity of L. stagnalis plasma against T. regenti miracidia was also described. Mechanisms used by trematodes to interact with the snail internal defense system will help us to understand why one species is suitable for the develepment of the trematode whereas another closely related species kills it.
