Die Wasserlöslichkeit von Wirkstoffen ist von elementarer Bedeutung für deren pharmazeutische Verwendung. Im Rahmen dieser Arbeit wurde der Ansatz der thermodynamischen Betrachtung des Lösungsprozesses von einfachen Modellsubstanzen auf pharmazeutisch relevante Substanzen aus der Gruppe der Phenylethylamine übertragen und entsprechend erweitert. Der Lösungsprozess wurde auf Grundlage des Satzes von Hess in Ersatzschritte aufgeteilt, die dann mittels kalorimetrischer Bestimmung, quantenchemischen Rechnungen und MD-Simulation bestimmt wurden, um die limitierenden Faktoren der Löslichkeit zu beschreiben und so gezielte Ansätze für die Löslichkeitsverbesserung finden zu können. Der Lösungsprozess wurde zum einen durch die Schritte Sublimation und Solvatation und zum anderen durch Schmelzen und Mischen beschrieben. Für die einzelnen Schritte sind die thermodynamischen Größen Enthalpie, Entropie und Freie Enthalpie bestimmt worden. Dies war zum Teil experimentell mithilfe von kalorimetrischen Messungen möglich, andere Größen sind mit dem semiempirischen Solvatationsprogramm AMSOL oder mit COSMOtherm berechnet worden. Die so gewonnenen thermodynamischen Größen geben Aufschluss über das Lösungsverhalten der untersuchten Substanzen, die zum Teil trotz augenscheinlich hydrophilen Merkmalen schlecht löslich in Wasser sind. Zusätzlich wurden mit Hilfe von MD-Simulationen mit dem Programm AMBER die inter- und intramolekularen Wechselwirkungsmöglichkeiten untersucht. Die Ausbildung intermolekularer Wasserstoffbrückenbindungen mit Wasser wurde mit Radialen-Dichteverteilungs-Funktionen (RDF) analysiert. Die Möglichkeit der Ausbildung von intramolekularen Wasserstoffbrückenbindungen und deren Einfluss auf das Lösungsverhalten wurde über die Bindungsabstände und der Darstellung der polaren Oberflächen bestimmt. / The water solubility of drugs is of fundamental importance for their pharmaceutical application. Within the scope of this thesis the thermodynamic approach to describe the dissolution process was extended from simple model substances to drug like molecules. The chosen substances belong to the group of the phenylethylamines. The solution process was spitted into substitution processes in accordance to the Hess theorem. To describe the limiting factors and find a well-directed approach for solubility improvement these steps were analysed with calorimetric methods, quantum-chemical calculations and MD-simulations. The dissolution process itself was described by the steps sublimation and solvation as well as the steps melting and mixing. The thermodynamic characteristics enthalpy, entropy and free energy were determined for these steps. Some of these values were determined experimentally with calorimetric methods. Other values had to be calculated using the semiempirical solvation program AMSOL or with COSMOtherm. These thermodynamic characteristics describe the dissolution behaviour of the examined substances which are partly poor soluble in water despite their obviously hydrophilic properties. In addition inter- and intramolecular interactions were analysed by means of MD-simulations using the program AMBER. The formation of intermolecular hydrogen bonds with water molecules was determined with radial distribution functions (RDF). The possibility of formation of intramolecular hydrogen bonds and their influence on the dissolution process was examined by the determination of the binding distance and the visualisation of the polar surface.

No / 2-phenylethylamine is an endogenous constituent of the human brain and is implicated in cerebral transmission. This bioactive amine is also present in certain foodstuffs such as chocolate, cheese and wine and may cause undesirable side effects in susceptible individuals. Metabolism of 2-phenylethylamine to phenylacetaldehyde is catalysed by monoamine oxidase B but the oxidation to its acid is usually ascribed to aldehyde dehydrogenase and the contribution of aldehyde oxidase and xanthine oxidase, if any, is ignored. The objective of this study was to elucidate the role of the molybdenum hydroxylases, aldehyde oxidase and xanthine oxidase, in the metabolism of phenylacetaldehyde derived from its parent biogenic amine. Treatments of 2-phenylethylamine with monoamine oxidase were carried out for the production of phenylacetaldehyde, as well as treatments of synthetic or enzymatic-generated phenylacetaldehyde with aldehyde oxidase, xanthine oxidase and aldehyde dehydrogenase. The results indicated that phenylacetaldehyde is metabolised mainly to phenylacetic acid with lower concentrations of 2-phenylethanol by all three oxidising enzymes. Aldehyde dehydrogenase was the predominant enzyme involved in phenylacetaldehyde oxidation and thus it has a major role in 2-phenylethylamine metabolism with aldehyde oxidase playing a less prominent role. Xanthine oxidase does not contribute to the oxidation of phenylacetaldehyde due to low amounts being present in guinea pig. Thus aldehyde dehydrogenase is not the only enzyme oxidising xenobiotic and endobiotic aldehydes and the role of aldehyde oxidase in such reactions should not be ignored.
