Caracteriza??o gen?tica e evolu??o dos v?rus dengue no Estado do Rio Grande do Norte

The etiologic agent is Dengue Virus (DENV), it is a single positive-stranded RNA virus of the family Flavivirus, genus Flaviviridae. Four serotypes are known, DENV-1, DENV-2, DENV-3 and DENV-4. One of the most important characteristic of these viruses is the genetic variability, which demands phylogenetic and evolutionary studies to understand key aspects like: epidemiology, virulence, migration patterns and antigenic characteristics. The objective of this study is the genetic characterization of dengue viruses circulating in the state of Rio Grande does Norte from January 2010 to December 2012. The complete E gene (1485 pb) of DENV1, 2 e 4 from Brazilian (Rio Grande do Norte) patients was sequenced. Phylogenetic analysis was performed using MEGA 5.2 software, Tamura-Nei model and Neighbor-Joining trees were inferred for the datasets. In Brazil, there is just one DENV-1 genotype (genotype V), one DENV-2 genotype (Asian/American) and two DENV-4 genotypes (genotypes I and II). Brazilian strains of DENV-1 are subdivided in two different lineages (BR-I and BR-II), the Brazilian strains of DENV-2 are subdivided in four lineages (BRI-IV) and genotype II of DENV-4 is subdivided in three Brazilian lineages (BRI-III). The viruses isolated in RN belong to lineage BR-II (DENV-1), BR-IV (DENV-2) and BR-III (DENV-4).The Caribbean and near Latin American countries are the main source of these viruses to Brazil. Amino acids substitutions were detected in three domains of E protein, this makes clear the necessity of studies that associate epidemiological and molecular data to better understand the effects of these mutations. This is the first study about genetic characterization and evolution of Dengue viruses in Rio Grande do Norte, Brazil / A dengue ? considerada a doen?a transmitida por artr?podes mais importante do mundo devido ao elevado n?mero de pessoas que correm o risco de contra?-la, principalmente em regi?es tropicais e sub-tropicais do planeta. O agente etiol?gico da doen?a ? o v?rus dengue (DENV), um v?rus de RNA fita simples e polaridade positiva, pertencente ? fam?lia Flavivridae e ao g?nero Flavivirus. S?o conhecidos quatro sorotipos distintos: DENV-1, DENV-2, DENV-3 E DENV-4. Uma das suas principais caracter?sticas ? a elevada variabilidade gen?tica que torna clara a necessidade da realiza??o de estudos filogen?ticos e evolutivos com o objetivo de compreender aspectos essenciais como: epidemiologia, virul?ncia, padr?es de migra??o e caracter?sticas antig?nicas. O objetivo deste estudo foi realizar a caracteriza??o gen?tica dos v?rus dengue circulantes no estado do Rio Grande do Norte durante o per?odo de Janeiro de 2010 a Dezembro de 2012. Realizou-se o sequenciamento do gene do envelope (1485pb) dos sorotipos DENV-1 (N=6), DENV-2 (N=6) e DENV-4 (N=4) isolados de casos cl?nicos de diferentes munic?pios do estado do Rio Grande do Norte. A an?lise filogen?tica foi realizada utilizando o programa Mega v5.2, m?todo de Neighbor-Joining e modelo Tamura-Nei. No Brasil, foi constada a circula??o de apenas um gen?tipo de DENV-1 (gen?tipo V), um gen?tipo DENV-2 (asi?tico/americano) e dois gen?tipos de DENV-4 (gen?tipos I e II) no Brasil. As cepas brasileiras de DENV-1 est?o dividas em duas linhagens temporalmente distintas (BR-I e BR-II), as cepas brasileiras de DENV-2 est?o subdividas em quatro linhagens (BRI-IV) e o gen?tipo II de DENV-4 apresentou tr?s linhagens de cepas brasileiras (LI-III). Os v?rus isolados no Rio Grande do Norte pertencem a linhagem BR-II (DENV-1), BR-IV (DENV-2) e BR-III (DENV-4). A fonte de importa??o destes v?rus para o Brasil ? principalmente o Caribe e pa?ses pr?ximos da Am?rica Latina. Foram detectadas substitui??es de amino?cidos ao longo dos tr?s dom?nios da prote?na E, tornando clara a necessidade da realiza??o de estudos que associem dados epidemiol?gicos e moleculares para melhor compreens?o dos efeitos dessas muta??es.
