The effects of the synthetic strigolactone GR24 on Arabidopsis thaliana callus culture

Thesis (MSc)--Stellenbosch University, 2012. / ENGLISH ABSTRACT: Plant growth promoting substances (PGPS) are emerging as useful tools in the investigation of important plant growth traits. Two PGPS, smoke-water derived from burning plant material and a synthetic strigolactone analogue, GR24, have been reported to regulate a wide variety of developmental and growth processes in plants. These PGPS are beginning to receive considerable attention in the area of improving plant biomass yield and production. Variation in growth between plants is a major impediment towards the complete understanding of the intrinsic processes that control biomass production. Callus cultures of the model plant Arabidopsis thaliana could overcome some of these hindrances. However, the suitability of these callus cultures as a model system for plant biomass production must be established first. This study aimed at using A. thaliana callus cultures as a platform to study the plant growth promoting activities of smoke-water and GR24. The first part of this study was conducted to develop an optimal protocol for inducing A. thaliana callus formation. Wild-type A. thaliana Col-O, as well as strigolactone deficient and insensitive mutants (max1-1, max2-1, max2-2, max3-9 and max4-1) were cultured for callus induction. Hypocotyl and leaf explants were cultured onto MS media supplemented with different hormone concentrations of 2,4-D and kinetin (2:2 mg/L 2,4-D:kinetin and 0.5:0.05 mg/L 2,4-D:kinetin). Both media proved suitable for callus induction of all genotypes, with max1-1 showing the highest efficiency (83.33% and 92.22%) of callus induction. Calli were then used as a platform for future investigations into the effects of smoke-water and GR24. Secondly, this study examined the effects of smoke-water and GR24 on wild-type A. thaliana Col-O callus. Basic physiological studies were conducted to determine if these two compounds would positively affect callus growth, as was shown in previous studies using whole plants. Calli cultivated on MS media containing the two different hormone concentrations were transferred onto the same fresh MS medium, supplemented with either smoke-water or GR24. Growth promotion by smoke-water and GR24 in calli was characterized by a significantly increased mass (biomass). Calli were additionally transferred onto MS medium containing either auxin only or kinetin only and supplemented with GR24 or smoke-water. In the auxin only system, increased mass was recorded for both GR24 and smoke-water treatments, while these two compounds seemed to reduce growth in the kinetin only system. The positive growth stimulatory effect observed for the auxin only system could be attributed to the synergistic relationship between auxin and strigolactones, whilst the reduced mass in the latter system could be due to the antagonistic interaction between strigolactones and cytokinins. Finally, this study has discovered a dual role of strigolactones in biomass accumulation and adventitious root formation for Arabidopsis thaliana callus. On an auxin- and cytokinin-free MS medium supplemented with GR24, calli of Arabidopsis thaliana strigolactone deficient mutants (max1-1 and max4-1) and the wild-type Col- O, but not the strigolactone response mutant (max2-2), showed enhanced biomass accumulation. In addition to this, the max4-1 mutant and wild-type Col-O demonstrated enhanced adventitious rooting, which was not apparent in max2-2. Together these data suggested that the biomass accumulation and the adventitious rooting activities of GR24 in Arabidopsis thaliana calli are controlled in a MAX2- dependent manner. The interaction between strigolactone, auxin and cytokinin signalling pathways in regulating these responses appears to be complex. Gene expression profiling showed regulation of stress-related genes such as B-box transcription factors, CALCINEURIN B-LIKE and RAP4.2 Genes encoding hormones associated with stress (ABA, ethylene) and defence mechanisms (JA) were upregulated. Expression of stress related genes indicated clues on some kind of stress mediation that might be involved during the regulation of the rhizogenic response. Conversely, smoke-water treatment could not enhance the biomass of the calli and nor could it induce adventitious rooting in the absence of auxin and cytokinin. This observation strongly emphasized the distinct roles of these two compounds, as well as the importance of the interaction and ratio of auxin and cytokinin in callus growth. This study has demonstrated a novel role of strigolactones in plant growth and development, i.e. enhancement of biomass production in callus cultures. Secondly the enhanced adventitious rooting ability is in agreement with recently published literature on the role of strigolactones in regulating root architecture. In vitro callus production is advantageous to plant sciences. It creates an opportunity for increasing plant material for cultivation and offers the use of cell cultures that accurately mimic specific growth responses. It could greatly contribute to the study of intricate regulatory and signalling pathways responsible for growth and development in plants. Because the regulation of plant biomass production is very complex and the molecular mechanisms underlying the process remain elusive, it is of paramount importance that further work be done in order to gain more in-depth insights and understanding of this aspect and subsequently improve efficiency and returns when applying biotechnology tools on commercially important crop plants. / AFRIKAANSE OPSOMMING: Verbindings wat plantgroei bevorder (PGBV) het as nuttige alternatief ontstaan om plant groei te ondersoek. Rook-water, afkomstig van verbrande plant material, en ‘n sintetiese strigolaktoon analoog, GR24, wat ‘n α, β-onversadigde furanoon funksionele groep in gemeen het, is vir die regulering van ‘n wye verskeidenheid ontwikkelings- en groei prosesse in plante verantwoordelik. Tans ontvang hierdie PGBVs aansienlik aandag in die area van die verbetering van plant biomassa opbrengs en -produksie. Die variasie in groei tussen plante is ‘n groot hindernis om die intrinsieke prosesse wat biomass produksie beheer, volledige te verstaan. Deur gebruik te maak van kallus kulture van die model plant Arabidopsis thaliana kan van hierdie hindernisse oorkom word. Tog moet die geskiktheid van kallus kulture as ‘n model sisteem vir plant groei biomass produksie eers gevestig word. Die doel van hierdie studie was om A. thaliana kallus kulture as ‘n platform vir die studie van die plantgroei bevorderingsaktiwiteite van rook-water en GR24 te gebruik. Laastens het hierdie studie het ‘n gelyktydige rol van strigolaktone vir biomassa akkumulasie en bywortelvorming in Arabidopsis thaliana kallus ontdek. Kallus van A. thaliana strigolaktoon afwesige mutante (max1-1 en max4-1) en die wilde tipe Col-0 (maar nie die strigolaktoon reagerende mutant (max2-2) het op ‘n ouksien en sitokinien vrye MS medium wat GR24 bevat ‘n verhoogde biomassa akkumulasie getoon. Hierdie studie toon op ‘n nuwe rol van strigolaktoon in plant groei en ontwikkeling, d.w.s die verhoogde biomassa produksie in kallus kulture. Tweedens, die verhoogde bywortelvormingsvermoë is in ooreenstemming met literatuur wat onlangs gepubliseer is i.v.m die rol van strigolaktone in die regulering van wortel argitektuur. Die in vitro produksie van kallus is voordelig in plant wetenskappe. Dit skep ‘n geleentheid vir die vermeerdering van plant materiaal vir kultivering en bied die gebruik van selkulture wat spesifieke groei reaksies op ‘n merkwaardige wyse akkuraat namaak. Dit kan grootliks bydra tot die studie van die delikate regulatoriese en sein transduksie paaie wat vir groei en ontwikkeling van plante verantwoordelik is. Aangesien die regulering van plant biomassa produksie baie kompleks is en die molekulêre meganismes vir die proses onbekend bly is dit van grootskaalse belang dat meer werk gedoen word om ‘n meer in diepte insig en kennis van die aspekte en gevolglike verbetering van effektiwiteit en wins te kry deur die toepassing van biotegnologiese metodes op die gewas plante wat van kommersiêle belang is.

Distribuição e caracterização química da fertilidade dos solos do estado do Amazonas.

The floodplains (locally designated as Várzea ), includes areas with approximately 60,000 km2 with different fertility gradients, due to the deposition of sediments of varied mineralogical origin, organic composition and particles sizes, derived from the Andes Mountains and falling of rivers banks. The soils of Amazon are characterized by small availability of nutrients. The present study was carried out to provide more substantial data about soil fertility of Amazonas State, Brazil. Results from 3340 soil samples from all 62 counties of the State and that were collected during the past 30 years were used. In the absence of geographical coordinates, it was assumed that sampling points were uniformly scattered over each county. Chemical attributes (P, K, Ca, Mg, Al, H+Al and base saturation) showed high variability, exception made to pH and CEC. The soils of region were mainly characterized by high acidity (pH), high level of exchangeable Al and H+Al, intermediate content of organic matter and low contents of P, K, Ca and Mg. Overall, base saturation was lower than 20%, value considered low for most crops. Studies, in genetic improvement native plants with higher potential of production and soil fertility are necessary, for sustainable agriculture in Amazonas State, Brazil. / Os solos da Amazônia tropical úmida, onde se localiza o Estado do Amazonas, são constituídos por cerca de 75% Latossolos (Oxisols) e Argissolos (Ultisols) e formado basicamente por duas unidades geomorfolócias Terra Firme denominação regional, com relevo variando de ondulado a levemente ondulado, localizados em áreas que não sofrem influência da enchentes dos rios com predominância de solos ácidos com altos teores de alumínio tóxico e ocupam 80% da região. As várzeas com aproximadamente 60.000 km2 com diferentes gradientes de fertilidade, devido a deposição de sedimentos de variadas origens mineralógicas, composição orgânica e tamanho de partículas variados derivados das Cordilheiras dos Andes e dos barrancos dos rios (fenômeno conhecido como terras caídas). Os solos são caracterizados pela baixa disponibilidade de nutrientes. O presente trabalho teve por objetivo fornecer dados mais substanciais sobre a fertilidade do solo do Estado do Amazonas. Foram usados os resultados das análises de 3340 amostras de terra, abrangendo os 62 municípios do Estado, as quais haviam sido coletadas durante 30 anos. Na ausência de coordenadas geográficas, foi assumido que as amostragens foram realizadas aleatoriamente no município. Exceto o pH e a CTC, os atributos químicos dos solos (P, K, Ca, Mg, Al, H+Al e saturação por bases) apresentaram grande variação na distribuição. Os solos, na sua maioria, foram caracterizados pela elevada acidez, teores intermediários de MOS, baixos de P, K Ca e Mg e altos de H e H+Al trocável. Em geral, a saturação por bases foi menor que 20%, valor considerado baixo para a maioria das culturas. São necessários estudos, com ênfase ao melhoramento genético de plantas nativas com grande potencial de produção e à fertilização para a utilização sustentável dos solos do Amazonas para fins agrícolas.

Mathematical Models of <i>Zea mays</i>: Grain Yield and Aboveground Biomass Applied to Ear Flex and within Row Spacing Variability

Field studies were conducted during the summers of 2007 and 2008 at the Agricultural Research and Education Complex, Western Kentucky University, Warren County, KY and commercial production fields in Caldwell County, KY, Warrick County IN, and Vanderburgh County, IN. The goals of these studies were to further validate the Duncan grain yield model, the Russell aboveground biomass model, and to study the effect of inconsistent spacing within rows on Zea mays L. yield. Plant spacing other than uniform decreases grain yield and profitability. The population experiments conducted at the Warren County location were a randomized complete block design with three planting densities, three varieties (c.v. DeKalb DKC6547, DeKalb DKC6346, DeKalb DKC6478) in 2007 and (DeKalb DKC6478, DeKalb DKC6342, and DeKalb DKC6544) in 2008, and three replications. Seeds were planted in rows 76 cm apart and 9.1 m long with four rows per plot in a no-till system on a Crider Silt Loam with pH of 6.8 and 1.5% organic matter. The effect of variable within row spacing was evaluated in commercial production fields by randomly selecting five adjacent rows of 5.3 meters in length at each location. Grain yield for each row was then curve fitted both linearly and exponentially. Minimizing interspecies competition was essential to evaluating the effects of competition within Zea mays L. A burn-down application of 2,4-D and glyphosate was used prior to planting. The most common weeds in the plots were Sorghum halepense L. (johnsongrass), Trifolium repens L. (white clover), and Taraxacum officinale L. (common dandelion) . Glyphosate was reapplied throughout the growing season due to reemergence of S. halepense and Ipomoea hederacea Jacq. (ivyleaf morningglory). The weight of each ear was recorded and one row from each plot was randomly selected to shell. The moisture content was measured from a subsample twice each row using an electrical conductivity moisture meter. The mean of the two moisture readings was used as the moisture content from the plot. Cob weights from shelled ears were recorded to determine the grain/cob mass ratio. This ratio was used to project the grain weight for the remaining harvested rows. Duncan’s grain yield model and Russell’s biomass model were curve fitted to the data for areas of 0.00040 hectares at the p < 0.05 significance level or greater in all population density plots. Individual plant grain masses were curve fitted to Duncan’s model with p < 0.05 significance in 3 out of 15 plots. Grain mass was negatively correlated (R < 0) with standard deviation of within row spacing in 14 of 15 plots. A linear fit to this trend was significant in only 2 of 15 plots. The Duncan yield curve and the Russell aboveground biomass model fit all 6 genotype by environment interactions for 2007 and 2008 to the α = 0.05 level of confidence when evaluated over a 5.3 meter length on 76.2 cm wide rows. Individual plants fit linearly at α = 0.05 in 9 out of 15 plots. Individual plants fit the Duncan yield curve at α = 0.05 in 4 out of 15 plots. Standard deviation of within row spacing fit grain yield loss significantly at &#; = 0.05 in two of 15 plots. The individual plant spacing and local population density collectively fit nine plots significantly at α = 0.05 or better.

Considering this, two experiments were organized in a greenhouse, located in the Universidade Federal Rural do Semiárido UFERSA, located in the town of Mossoró, RN, from January 2009 until June 2010, with the objective of determining the best grafting method for the production of seedlings of tamarind and pitombeira. We studied the types of grafting: grafting in top cleft, cleft grafting in the top simple English way , grafting in the top complicated English way , grafting budding side and plate cleft. The design used in the experiments was a randomized complete block with five treatments (grafting types). The experiment with tamarind had seven repetitions; each parcel consisted of 14 seedlings, a total of 98 per treatment, adding 490 in the experiment. Variables were evaluated: percentage of grafting success, shoot length, root length, branch length, number of leaves, number of branches, stem diameter, shoot dry matter, root dry matter, Total matter dry. The data were subjected to analysis of variance and compared through the Tukey test at 5% significance using the software SISVAR. The rates of graft takes were submitted to angular transformation of arcsen &#8730; x/100, and data on the number of branches in &#8730; x + 0.5. In order to promote the statistic, treatment of pimple on board was disregarded, considering that this treatment has zero index of plant take. The grafting methods in top cleft, cleft grafting in the English top grafting complicated and simple English way in the top, are respectively the best methods of grafting for the tamarind. The test was conducted with pitombeira with four replications and each plot was composed of twenty seedlings in a total of 400 seedlings on the experiment, in which was observed zero rate of grafting take / Devido às amplas perspectivas de mercado para as culturas do tamarindeiro e pitombeira, torna-se indispensável a avaliação de métodos que permitam a propagação em escala comercial. Diante disso, foram desenvolvidos dois experimentos em casa de vegetação, localizada no Campus da Universidade Federal Rural do Semiárido UFERSA, situada na cidade de Mossoró-RN, no período de janeiro de 2009 a junho de 2010, com o objetivo de determinar o melhor método de enxertia para a produção de mudas de tamarindeiro e de pitombeira. Foram estudados os tipos de enxertia: garfagem no topo em fenda cheia, garfagem no topo à inglesa simples, garfagem no topo à inglesa complicada, garfagem em fenda lateral e borbulhia em placa. O delineamento utilizado nos experimentos foi o de blocos casualizados, com cinco tratamentos (tipos de enxertia). O experimento com tamarindeiro foi constituído por sete repetições, sendo cada parcela composta por 14 mudas, num total de 98 por tratamento, perfazendo 490 no experimento. Foram avaliadas as variáveis: percentagem de pegamento dos enxertos, comprimento da parte aérea, comprimento do sistema radicular, comprimento de ramos, número de folhas, número de ramos, diâmetro do colo, matéria seca da parte aérea, matéria seca do sistema radicular, matéria seca total. Os dados obtidos foram submetidos à analise de variância e comparados através do teste de Tukey ao nível de 5% de significância utilizando o software SISVAR. Os índices de pegamento dos enxertos foram submetidos à transformação angular do arcsen &#8730;x/100, e os dados de número de ramos em &#8730;x + 0,5. Para fins da realização da estatística, o tratamento de borbulha em placa foi desconsiderado, tendo em vista que este tratamento apresentou índice zero de pegamento. Os métodos garfagem no topo em fenda cheia, garfagem no topo à inglesa complicada e garfagem no topo à inglesa simples são, respectivamente, os melhores métodos de enxertia para o tamarindeiro. O ensaio com pitombeira foi realizado com quatro repetições sendo cada parcela composta por vinte mudas, num total de 400 mudas no experimento, no qual foi verificado índice zero de pegamento dos enxertos

Variables were evaluated: percentage of grafting success, shoot length, root length, branch length, number of leaves, number of branches, stem diameter, shoot dry matter, root dry matter, Total matter dry. The data were subjected to analysis of variance and compared through the Tukey test at 5% significance using the software SISVAR. The rates of graft takes were submitted to angular transformation of arcsen &#8730; x/100, and data on the number of branches in &#8730; x + 0.5. In order to promote the statistic, treatment of pimple on board was disregarded, considering that this treatment has zero index of plant take. The grafting methods in top cleft, cleft grafting in the English top grafting complicated and simple English way in the top, are respectively the best methods of grafting for the tamarind. The test was conducted with pitombeira with four replications and each plot was composed of twenty seedlings in a total of 400 seedlings on the experiment, in which was observed zero rate of grafting take / Devido às amplas perspectivas de mercado para as culturas do tamarindeiro e pitombeira, torna-se indispensável a avaliação de métodos que permitam a propagação em escala comercial. Diante disso, foram desenvolvidos dois experimentos em casa de vegetação, localizada no Campus da Universidade Federal Rural do Semiárido UFERSA, situada na cidade de Mossoró-RN, no período de janeiro de 2009 a junho de 2010, com o objetivo de determinar o melhor método de enxertia para a produção de mudas de tamarindeiro e de pitombeira. Foram estudados os tipos de enxertia: garfagem no topo em fenda cheia, garfagem no topo à inglesa simples, garfagem no topo à inglesa complicada, garfagem em fenda lateral e borbulhia em placa. O delineamento utilizado nos experimentos foi o de blocos casualizados, com cinco tratamentos (tipos de enxertia). O experimento com tamarindeiro foi constituído por sete repetições, sendo cada parcela composta por 14 mudas, num total de 98 por tratamento, perfazendo 490 no experimento. Foram avaliadas as variáveis: percentagem de pegamento dos enxertos, comprimento da parte aérea, comprimento do sistema radicular, comprimento de ramos, número de folhas, número de ramos, diâmetro do colo, matéria seca da parte aérea, matéria seca do sistema radicular, matéria seca total. Os dados obtidos foram submetidos à analise de variância e comparados através do teste de Tukey ao nível de 5% de significância utilizando o software SISVAR. Os índices de pegamento dos enxertos foram submetidos à transformação angular do arcsen &#8730;x/100, e os dados de número de ramos em &#8730;x + 0,5. Para fins da realização da estatística, o tratamento de borbulha em placa foi desconsiderado, tendo em vista que este tratamento apresentou índice zero de pegamento. Os métodos garfagem no topo em fenda cheia, garfagem no topo à inglesa complicada e garfagem no topo à inglesa simples são, respectivamente, os melhores métodos de enxertia para o tamarindeiro. O ensaio com pitombeira foi realizado com quatro repetições sendo cada parcela composta por vinte mudas, num total de 400 mudas no experimento, no qual foi verificado índice zero de pegamento dos enxertos

The diploma thesis deals with the possibilities of sewerage and wastewater disposal in the recreational centre near Vřesovice. The purpose of the diploma thesis is the elaboration of variant solutions of sewerage, including the proposal of possibilities of wastewater treatment, economic evaluation of variants, their subsequent comparison and recommendation of the most suitable variant. The diploma thesis deals with the design of variants of wastewater treatment with subsequent discharge into surface waters. Finally, the advantages and disadvantages of the individual variants of sewerage are summarized with a comparison of costs and subsequent recommendation of the variant.

The diploma thesis is focused on proposal of workplace modification according to the lean manufacturing methodology (DMAIC). The target is to minimize the wasting and to increase the production efficiency based on LEAN tools implementation. The theoretical part of the thesis defines the basic terms, such as primarily lean production, DMAIC, wasting. In the practical part there is at problem analysis using the DMAIC method, then the proposal for improvement is solved, that contains reduction of wasting, process of the project, timetable, costs and implantation of the solution, finally there is evaluated the benefit.
