An investigation into the rapidly induced chemical responses of Myrica gale to insect herbivory

Carlton, Robert R. January 1990 (has links)
The effect of natural herbivory on the secondary metabolism of Myrica gale was investigated. In a field experiment herbivory was found to elicit changes in both the leaf phenolics and the density of volatile oil glands. Lygocoris spinolai, a capsid bug, accounted for most of the observed herbivory. One set of plants (the controls) were kept free of insect herbivores while a second set sustained capsid bug herbivory. The terpenoid and phenolic profile of leaves from each set was obtained on a weekly basis using GLC and IIPLC respectively. GC-MS was used to identify constituents of the volatile oil. UV, ¹H and ¹³C NMR, El-MS and FAB-tlS were used to identify the phenolics. Capsid bug damage induced a cpialitative change in the phenolic content of the leaves. This induction revealed a new compound, kaempferol-3-(2,3-diacetoxy-4-p-coumaroyl)rhamnoside, which was isolated and identified. The leaves also proved to contain other flavonoids, the concentrations of which were not affected by herbivory. Artificial damage was observed to elicit a quantitative change in leaf phenolics, thus having a different effect than herbivore damage. Herbivore damage also induced quantitative changes in the volatile oil by eliciting gland production whilst having no effect on the composition of the oil. The induced flavonoid and the volatile oil were tested for fungal growth inhibitory properties. Both were found to have marked antifungal activity at low concentrations when tested against fungal species isolated from the leaves of M. gale in the field. The observed phenomena are discussed in terms of defenses against herbivores and pathogens. The relationship between the nitrogen economy of M. gale and leaf chemistry is also discussed.

Identificação e caracterização de genes induzidos por Diatraea saccharalis (Lepidoptera: Crambidae) em cana-de-açúcar / Identification and characterization of genes induced by Diatraea saccharalis (Lepidoptera: Crambidae) in sugarcane

Medeiros, Ane Hackbart de 04 September 2008 (has links)
Plants respond to insect damage by induction and accumulation of a large set of defense proteins. An investigation was undertaken to study the sugarcane transcriptional changes following Diatraea saccharalis damage. The first approach was a detailed study about the induction of two isoforms of a sugarcane homologue of a barley wound inducible gene, barwin, named sugarwin (sugarcane wound-inducible). Induction of sugarwin transcripts occurs in response to mechanical wounding, D. saccharalis feeding and methyl jasmonate treatment. Their expression is restricted to the site of damage. Sugarwins are members of the late wound-inducible genes. The subcellular localization of the signal peptide fused to the gfp (green fluorescent protein) shows that these proteins are secreted. Although the exact function of the barwin domain has not been completely elucidated, antipathogenic activities has been described for a number of homologues. Multiple sequence alignment of barwin domain-containing sugarcane proteins and of mono and dicotiledoneous proteins reveals high similarity, suggesting that their function is conserved among species. This is the first report of a barwin-like protein induced by herbivory. The activity of this type of proteins against insects has never been studied. Based on the results presented here, it can be concluded that SUGARWINS are part of the sugarcane defense response strategy. The second approach to study the sugarcane response to D. saccharalis damage was the large-scale analysis, using DNA macroarrays, of serine proteases and serine protease inhibitors differently expressed in response to herbivory. While the protease inhibitors function in defense is well-established, the involvement of proteases in defense has been recently proposed. The transcript monitoring of sugarcane serine proteases in response to herbivory revealed several candidate genes for further functional studies. One of the greatest applications of these results is the identification of genes for use in biotechnological strategies to improve sugarcane insect resistance.

Soybean, [Glycine max (L.) Merrill], is one of the most important crops for Brazil, with high productivity and strong participation in the foreign market. Soybeans represent important nutritional source for humans and their derivatives are used in animal nutrition or even renewable energy production, as biofuel. However, production of this crop may be affected negatively by the attack of insect pests, such as the whitefly, Bemisia tabaci (Gennadius) biotype B (Hemiptera: Aleyrodidae). This insect is currently indicated as an important pest for the soybean crop, due to the increasing infestations and difficulty in control. Its management is mostly done with successive spraying of synthetic insecticides that promote rapid selection of resistant individuals. Thus, it is important to evaluate less aggressive alternatives, aligned with the integrated pest management (IPM), with emphasis on the use of resistant cultivars and resistance inducers, as silicon. Monitoring sucking insects feeding using the EPG technique (Electrical Penetration Graph) has been helping in the characterization of categories and resistance factors of plants against insect attacks. In this study, the feeding behavior of B. tabaci biotype B was assessed in resistant and susceptible genotypes of soybean in the presence and absence of silicon using EPG. In the first stage, four genotypes of soybean bearers of different categories of resistance against the whitefly were assessed (KS-4202, tolerance; IAC-19, antixenosis; UX-2569-159, antibiosis e PI-227687, susceptible). At the same time, the types of trichomes present on the abaxial surface of the leaflets were characterized, in order to establish possible correlations with the feeding behavior of the whitefly. The results confirm the presence of antixenotic factors in the genotype IAC-19 (low duration of feeding in the phloem phase). The high density of trichomes in the genotype PI-227687 was revealed as an antixenotic factor (delays the beginning of probing). KS-4202 was suitable for whitefly feeding, expected behavior in tolerant genotypes. In UX-2569-159 (antibiosis previously reported) resistance is likely more related to expression of antixenosis (high period of phloem feeding). In the second stage, the feeding behavior of B. tabaci biotype B was evaluated in two soybean genotypes with different levels of susceptibility to the insect (IAC-19, bearer of antixenosis and Conquista, susceptible) in the presence and absence of silicon. The results confirmed the presence of antixenotic factors in the genotype IAC-19 and suggest that the use of this mineral, through foliar application, accentuated the existing antixenosis factors in this genotype (increased time until probing and reduction in time of phloem feeding). Silicon interfered on the time until probing in Conquista, suggesting increment of physical barriers on the epidermis. / CNPq: 130280/2015-1

<b>Developing Sustainable Carrot Weevil Management Informed by Genetic and Behavioral Studies of Populations in the Great Lakes Region of the United States</b>

Emily J Justus (17583957) 09 December 2023 (has links)
<p dir="ltr">Specialty crop growers in the Great Lakes region of the United States and Canada have been struggling for decades to manage the carrot weevil (<i>Listronotus oregonensis</i>). In carrot, parsley, and celery production, growers may experience 40-100% yield loss, despite the intensive use of insecticides (Boivin, 1999; Jasinski, 2008; Telfer et al., 2019). To improve sustainable management of this native pest in commercial vegetable production, we must address existing knowledge gaps in carrot weevil mate-finding behavior, population genetic diversity and gene flow, and grower adoption and efficacy of integrated pest management (IPM) strategies, like cultivar selection, to combat this pest. To date, these factors have not been evaluated in the carrot weevil system, yet together they have significant implications for developing novel monitoring tools, grower decision making, and the potential evolution of insecticide resistance in this damaging pest.</p><p dir="ltr">To address knowledge gaps in grower adoption and the efficacy of IPM strategies, such as cultivar selection, I investigated the influence of carrot cultivar on the oviposition preference of carrot weevil in the laboratory and quantified the diversity of insect assemblages on carrots in Ohio and Indiana. In tandem, I delivered an online survey to specialty crop growers in Indiana and Ohio to measure their knowledge and use of insect-resistant cultivars as a part of an IPM strategy. I found that carrot cultivar did not impact carrot weevil oviposition preference, but rather the number of petiole trichomes was positively correlated with the cumulative number of egg scars found on a given carrot cultivar. Similarly, cultivar did not influence the diversity of insect assemblages associated with carrot foliage, but hemipterans were the dominant group found across cultivars. Moreover, insect diversity across all carrot cultivars was high, with Shannon-Weiner diversity indices ranging from 2.14 to 2.45.</p><p dir="ltr">I analyzed survey responses from vegetable growers regarding their knowledge, current use, and future use of insect-resistant cultivars and found no significant differences between conventional or organic growers, in either Indiana or Ohio, or across experience levels regarding their knowledge and current or future use of insect-resistant cultivars. Although a greater number of organic growers were aware of insect-resistant cultivars, they were not incorporated into their IPM plans. In contrast, half of responding conventional growers indicated implementing insect-resistant cultivars in current IPM plans. Growers indicated that more information about the efficacy, costs, and benefits of insect-resistant vegetable cultivars would be most helpful in informing their decision to incorporate this IPM tactic on their farms.</p><p dir="ltr">To address knowledge gaps regarding carrot weevil mate-finding behavior, I investigated behavioral response to conspecific frass using Y-tube bioassays and the volatiles of frass samples were analyzed using gas-chromatography mass-spectrometry (GC-MS) as starting points to assess the presence of volatile chemical cues that might function as sex or aggregation pheromones. I found that male and female weevils oriented strongly towards volatiles from male frass. However, conspecifics were not attracted to mixed or female frass, suggesting male frass contains behaviorally relevant chemical cues that are unique or present in greater abundance than in female frass. When I analyzed the chemical abundance and composition of the male frass volatiles, I found 16 common compounds across all samples and no compounds were unique to the male volatile blend. Specifically, (1S)-6,6-dimethyl-2-methylene-Bicyclo[3.1.1]heptane and (1R)-2,6,6-Trimethylbicyclo[3.1.1]hept-2-ene were present in significantly higher abundance in extracts of males frass compared to those of female frass.</p><p dir="ltr">Finally, to address knowledge gaps in the genetic diversity and gene flow of carrot weevil populations in the Great Lakes Region of the U. S., I used single nucleotide polymorphisms (SNPs) to compare genetic diversity and structure of agricultural populations of carrot weevil in Michigan and Ohio. I found limited genetic differentiation between three carrot weevil populations collected from celery and parsley fields located in Michigan and Ohio. I also found high inbreeding coefficients and heterozygote deficiency in all populations, suggesting that all carrot weevil populations are locally confined. These results suggest significant gene flow between populations, which has implications for management, specifically the mitigation and management of insecticide resistance.</p><p dir="ltr">Together, the results from my dissertation can be applied to the improvement of carrot weevil IPM across the Great Lakes region of the United States and Canada. Further investigations into the impact of physical characteristics, specifically petiole trichomes, on carrot weevil oviposition behavior can inform the breeding and development of resistant carrot cultivars. Results from my behavioral bioassay studies represent a critical first step towards the identification of an aggregation or sex pheromone, which would greatly improve detection of resident carrot weevil populations when used in conjunction with current monitoring traps. My results also suggest that monitoring and mitigation of insecticide resistance in carrot weevil populations should be considered at the regional scale, at minimum, with significant gene flow between carrot weevil populations within the Great Lakes Region.</p>

Qualidade de sementes e caracterização de genótipos de soja visando à resistência ao complexo de percevejos / Seed quality and characterization of soybean genotypes aiming resistance to stink bugs complex

Silva, Cibele Aparecida Teixeira da 18 February 2016 (has links)
A group of stink bugs composed by Euschistus heros (E.), Piezodorus guildinii (West.), and Nezara viridula (L.) causes several physiological disturbs in soybean during the field production, such as: delayed physiological maturity, leaf retention, yield loss, and decreased seed quality and germination potential. The reaction of soybean genotypes to stink bugs complex is an important approach for the crop breeding, seed production, and development of new resistant lines. This work aimed to compare the production and seed quality parameters, besides the plant defense under conditions of stink bugs attack. Assays were carried out at the Experimental Station of Anhumas (Department of Genetics, USP, ESALQ) with the soybean plants submitted to natural infestation by stink bugs, at 2012/13 and 2013/14 crop seasons, with absence and presence of chemical control of the insects, in a randomized block design with five replicates. The data were submitted to both ANOVA and MANOVA analysis. At the first season, 24 genotypes were evaluated as for the agronomic and plant resistance traits: seed formation period, plant life cycle, plant height, lodging index, agronomic value, number of pods per plant, mass of 1000 seeds, yield, foliar retention, and mass of healthy seeds. At the second season, eight genotypes were evaluated by the same agronomic and plant resistance traits of the first season, except the number of pods per plant. The pods damage index and seed physiological quality (germination, accelerated aging, emergence, electrical conductivity, and tetrazolium test) traits were evaluated only in the second season. The pods and seeds of LQ1050 and CD215 genotypes were collected between the R5 and R6 stages and had the isoflavones levels evaluated in conditions of stress by stink bugs attack. Through the monitoring of stink bugs population, it was possible to notice the increase of the insects in the seed formation period, with higher population density in the 2012/13 season. In the first season, the minimum and maximum masses of healthy seeds were taken as the main criterion to select susceptible and resistant genotypes, respectively, to the stink bugs attack, for the seed quality evaluation in the second season 2013/14. High yield genotypes do not ensure resistance to the stink bugs complex and, therefore, seeds with high physiological quality. We have noticed the genetic variability among the genotypes as for the physiological quality evaluated through the germination and vigor tests, at the second season. Traits used for the physiological quality can be correlated with the plant resistance to the stink bugs complex. Coumestrol and glyceollin, phytoalexin compounds, were identified in pods of the contrasting genotypes, but not in the seeds.

Qualidade de sementes e caracterização de genótipos de soja visando à resistência ao complexo de percevejos / Seed quality and characterization of soybean genotypes aiming resistance to stink bugs complex

Cibele Aparecida Teixeira da Silva 18 February 2016 (has links)
