Nutrition manipulation during development and its impact of metabolic homeostasis in the adult offspring

Latest epidemiological data suggests 1.5 billion adults worldwide are either overweight or obese. With increasing weight and obesity, adipocytes increase in size. The enlargement of adipocytes has been associated with low grade chronic inflammation via elevated adipokine secretion. Previous epidemiological studies in humans and experimental studies in animals have shown that during different periods of pregnancy (gestation) the offspring that are born to maternal nutritional manipulation are more susceptible to developing metabolic diseases in later adult life. Therefore, the aim of this thesis was to investigate the role of maternal nutritional manipulation on adipose tissue depots and in particular the consequences the effect on markers of adipokine secretion. Studies were conducted on both large and small animals (i.e. sheep and rats). Sheep studies focused on mid to late and late gestation periods of maternal nutritional restriction. Rat studies concentrated on long term fructose feeding during pregnancy and its effect on both the mother and offspring. Gene expression analysis identified an up-regulation in inflammatory related genes in pericardial and subcutaneous adipose tissue in the sheep studies. This was also seen in the rat studies with protein and gene expression displaying an up-regulation of inflammatory and metabolic related genes and proteins. The main conclusion of my thesis is that after following maternal nutrient restriction, females appear to be much more sensitive to inflammatory and metabolic adaptations compared to males, possibly due to sex hormones playing a role. Whilst fructose feeding during pregnancy concluded the possibility of homeorhesis playing a protective role against potentially detrimental inflammatory pathways being activated in the mothers, the offspring however displayed signs of low level chronic inflammation in the retroperitoneal depot from early infancy to later adult life.

The role of social capital in influencing the response capacity of farmers to bovine tuberculosis

Bovine tuberculosis (bTB) is one of the principal concerns currently facing the livestock industry in England. The disease has spread dramatically in recent years and is costing the country millions of pounds each year. Tens of thousands of cattle are being slaughtered annually; a huge financial and emotional burden to affected farmers. While various measures to control the disease have been taken, none have been successful in bringing it under control. Instead bTB continues to spread unabated. The essence of the bTB problem is that it necessitates industry buy-in in order to implement disease control measures. It is therefore not simply an issue of regulation. Current government bTB control policy emphasises communication and cooperative working across the government and the farming industry, coupled with cost and responsibility sharing. However, previous studies have shown that relationships between farmers and the government are already strained, engendered by a sense of distrust and a lack of confidence. Although some social science work has been conducted within the field of disease control and particularly bTB, the majority focuses on farmers‘ attitudes towards government policy and disease control. However, in order to implement successful disease control measures it is necessary to explore the ways in which farmers currently respond to bTB, and how their responses may be recognised by, and incorporated into, successful policy. While previous research has identified the important role of the wider social context in influencing farmers‘ attitudes and behaviour, no studies have yet provided an in-depth analysis of farmers‘ social networks in relation to bTB. In response, this study uses the lens of social capital to explore the various social ties which enhance or constrain farmers‘ capacity to respond to bTB. An iterative, mixed methods approach is adopted across two phases of research. The first incorporates twenty in-depth qualitative farmer interviews, exploring various aspects of bTB risk and response strategies as well as the core features of social capital. This informs a second, quantitative phase, in which data are iii gathered through a self-completion postal survey of 374 farmers in the South West of England. A farmer segmentation model is developed using factor and cluster analysis and two farmer groups are identified. The first group represents vulnerable farmers who are concerned about the negative impacts of bTB, and who are internally focused with respect to their networks. Characteristically, they exhibit strong relationships with others from within the farming community. In comparison, the second group are more resilient and less concerned about the impacts of bTB on their farm business. These farmers are externally focused, mainly seeking information from the government, the National Farmers‘ Union and their vet. The role of various forms of social capital is explored and an important distinction between the two farmer groups is found. Vulnerable farmers tend to be members of close networks of other farmers (bonding social capital), while resilient farmers are more likely to enjoy positive relationships with those from outside the farming community including vets (bridging social capital) and the government (linking social capital). However, while the research findings suggest that bridging and linking social capital can positively influence farmers‘ attitudes towards bTB, they do not necessarily lead to positive disease control behaviour. Statistical analysis of the data reveals no significant differences between the farmer groups in terms of their uptake of biosecurity measures, which represents an important disease avoidance strategy. A disjuncture between farmers‘ attitudes and their behaviour is therefore identified. The research concludes that investment in social capital between the government and farmers should form a core area of policy through providing opportunities for consistent and regular contact, allowing for the development of trusting and productive relationships. The current situation, characterised by low levels of trust and limited uptake of recommended disease control measures by farmers, indicates incoherence with contemporary policy discourses. A better understanding of the role of social capital in influencing farmer attitudes and behaviour will enable policy makers to increase the ability of farmers to respond to bTB risk, either through disease avoidance or through more effective management and coping mechanisms.

The olfactory basis for attraction of the bollworm Helicoverpa armigera (Hubner) (Lepidoptera: Noctuidae) to host-plant flowers

The objective of this work was to investigate whether or not olfactory clues play a role in host plant location by the polyphagous moth, Helicoverpa armigera. Volatiles collected from flowers of African marigold, Tagetes erecta, and sweet pea, Lathyrus odouratus, were found to elicit electroantennographic (EAG) responses from the antennae of female H. armigera. Compounds active in GC-EAG analyses of T. erecta floral headspace samples, identified by GC-MS and comparison of retention times on polar and non-polar GC columns with authentic standards, were (E)-myroxide, benzaldehyde, (f)-linalool, phenylacetaldehyde and (-)- piperitone. EAG-active compounds in L. odouratus floral headspace samples were identified as diacetone, (-)-linalool, phenylacetaldehdyde and benzyl alcohol. Increases in upwind flight to air entrained extracts of floral odours indicated that these cues caused attraction when presented to female H. armigera. A synthetic T. erecta blend comprising benzaldehyde, (f)- linalool, phenylacetaldehyde and (+)-limonene gave significant increases in upwind flight approaches. Limonene (either (+)- or (-)-) was found to be important for the behavioural response despite having low EAG-activity. There was no significant difference in upwind flight response to odours from the live flower and the synthetic floral blend. Significant increases in upwind flight were also obtained when insects were presented with a synthetic L. odouratus blend which contained the four EAG-active compounds identified from GCEAG studies. In field trapping experiments in Israel there was a significant difference in H. armigera catches in traps with a standard 4-component T. erecta lure compared with unbaited traps over the whole season. Mean H. armigera catch per trap per night (both sexes) over the whole season in unbaited traps, floral odour traps, pheromone traps and light traps were 0.004,0.11,8.8 and 1.35 respectively. The floral baited traps were non-selective catching large numbers of Ilymenoptera and Diptera as well as other moth species. Field trapping experiments in Pakistan indicated that the floral lure was significantly attractive to Earias spp. and other Lepidoptera although very few H. armigera were caught due to low population density. Olfactory cues are discussed in relation to host-plant finding behaviour of H. armigera. They are involved in early stages of host seeking behaviour prior to alighting on the plant and stimulate searching behaviour.

Non-invasive monitoring of environmental Mycobacterium bovis shedding in wild European badger (Meles meles) populations

The herd-level incidence of Mycobacterium bovis has been increasing in the United Kingdom (UK) and Republic or Ireland (RoI) for the past thirty years, resulting in substantial economic and animal welfare issues. Failure to control this pathogen in cattle is in part due to European badgers (Meles meles), a wildlife reservoir that are responsible for a proportion of transmission of M. bovis to cattle. Monitoring infection in badger populations is currently limited due to the need to trap badgers, which requires highly trained field staff and is expensive. In addition, although contact with infected badger faeces is a potential transmission route to cattle, very little is known about the extent and variability of the environmental pool of M. bovis shed by badgers. In this project we evaluated the suitability of using environmental badger faeces and a quantitative PCR (qPCR) assay to diagnose and monitor M. bovis in badger populations and described the extent of this environmental pool of potential infection. The first study identified that intensive environmental faecal sampling and analysis with qPCR is at least, if not more, sensitive at diagnosing M. bovis in badger populations than the currently used immunoassays. This study also identified that even within a high prevalence population, the levels of shedding of M. bovis in faeces are highly variable between groups and between seasons, suggesting that there may be heterogeneity in transmission risk throughout the year. Using this non-invasive qPCR method to monitor the first field trial of oral BCG vaccination identified a trend of decreasing levels of M. bovis in faeces with increasing vaccination levels however, these results failed to reach statistical significance, highlighting the importance of adequate sample sizes when implementing this method. Finally, characterisation of the gut and faecal microbiota from animals shedding M. bovis in faeces confirmed that the source of faecal M. bovis is most likely sputum that has been expelled from the lungs, and not from colonisation of the gut. The work presented here suggests that this non-invasive monitoring method can be applied to examine the variable pool of M. bovis over periods of time and large areas, providing an epidemiological tool which has the potential to be implemented to monitor infection in badger populations and disease intervention strategies.

Pigeon geographies : aesthetics, organisation, and athleticism in British pigeon fancying, c.1850-1939

This thesis provides new ways of thinking about human-bird encounters under domestication, providing the first substantive geographical study of ‘pigeon geographies’. It explores the spaces, practices, and human-pigeon relationships involved in pigeon showing and long-distance pigeon racing in Britain, from the mid-nineteenth century up until World War Two. The growth of fancy pigeon exhibitions was part of a wider Victorian passion for domesticating animals, at a time when human bodies were also subject to increasing aesthetic and moral scrutiny. Long-distance pigeon racing emerged at the end of this period, organised competitive sport more generally seen as an important means of moral improvement and identity expression. Like many other competitive pastimes in the second half of the nineteenth century, then, institutional bodies were formed to manage the expansion of showing and long-distance racing. The Pigeon Club and the Marking Conference were formed in 1885 to oversee British pigeon exhibitions, whilst the National Homing Union, formed in 1896, governed British long-distance pigeon racing. Both pastimes facilitated the formation of social worlds around varieties of domestic pigeon (Columba livia) and their respective practices. Whilst these pastimes historically had strong concentrations of male working-class followers – particularly in the north-west and north-east – they were both widespread throughout Britain and spanned all socio-economic classes, although accounts of female fanciers were rare. Through the exhibition of pigeons, fanciers debated and defined aesthetics, formulating breeding standards for each fancy breed, and questioning the ways in which pigeons were manipulated – sometimes contentiously – to produce the ‘ideal’. Long-distance pigeon racers, on the other hand, sought to understand and hone their birds’ athletic abilities, becoming entangled in scientific debate about homing, as well as geographical questions about the conduct and regulation of their sport. Racers were also drawn into aesthetic debates, exhibiting their racing birds during the off-season, the show pen becoming a fascinating frontier between showing and racing. Through the organisation of the spaces and practices that made up the fabric of these pastimes, pigeon showing and long-distance racing reconfigured both humans and their birds, the two becoming closely intertwined through collaborative encounters.
