Surface plasmon enhanced effects in photonic biosensors. / CUHK electronic theses & dissertations collectionJanuary 2008 (has links)
Detection of oligonucleotide target has been performed with a sandwich assay scheme. We compare the detection performance of strategies using probe oligonucleotide with or without gold nanoparticles (Au-NPs, 20nm) capped on 3'. Our experimental results reveal that while the DNA detection implemented with NIS can provide high sensitivity, both dynamic range and detection limit can be amplified with the aid of Au-NPs on 3' of the probes. The current detection limits of NIS with and without Au-NPs are 0.4 femtomole and 1 nanomole respectively. (Abstract shortened by UMI.) / Finally, this work presents a systematic study of the surface-enhanced Raman-scattering (SERS) properties of nanoparticle island substrates (NIS) and their application for oligonucleotide target detection. To effectively implement SERS on NIS and identify an optimal condition for DNA detection, the relationship between extinction maximum (lambdamax) and SERS enhancement factor (EF) will be explored in detail. This work demonstrates high S/N ratio SERS spectra can be achieved with NIS that has lambdamax located within a spectral window (∼60nm) defined by the excitation wavelength (514nm) and the scattered Raman wavelength. The highest EF measured is about 4x10 8 with a thickness of Ag being 50 A. / In addition, a surface plasmon enhanced ellipsometry (SPEE) biosensor scheme based on the use of a photoelastic modulator (PEM) has been explored. We showed that the polarization parameters of a laser beam, tan psi, cos Delta and ellipse orientation angle &phis;, can be directly measured by detecting the modulation signals at the 1st and 2nd harmonics of the modulation frequency under a certain birefringence geometry. This leads to an accurate measurement of refractive index variations within the evanescent field region close to the gold sensor surface, thereby enabling biosensing applications. Our experimental results confirm that the new scheme offers a decent detection limit of 2x10-7 refractive index unit (RIU) or 5ng/ml of biomolecule solute concentration without any compromise in dynamic range. / We have demonstrated that the sensitivity limit of intensity-based SPR biosensors can be enhanced when we combine the contributions from phase with that of amplitude instead of just detecting the amplitude or phase variation only. Experimental results indicate that an enhancement factor of as much as 20 times is achievable, yet with no compromise in measurement dynamic range. While existing SPR biosensor systems are predominantly based on the angular scheme, which relies on detecting intensity variations associated with amplitude changes only, the proposed scheme may serve as a direct system upgrade approach for these systems. / We have developed a novel design of multi-pass surface plasmon resonance (SPR) biosensor with differential phase interrogation based on multi-pass interferometry. This new configuration provides an intrinsic phase amplification effect of over two-fold by placing the SPR sensor head in a signal arm of the interferometer so that the interrogating optical beam will traverse the sensor surface infinite number of times. Experimental interferometers based on the Michelson and Fabry-Perot configurations have been employed to experimentally verify this amplification effect through the comparison with the Mach-Zehnder configuration. Results obtained from the salt-water mixtures, antibody-antigen, and protein-DNA binding reaction have confirmed the expected phase measurement enhancement. / Yuan, Wu. / Adviser: H. P. Due to their ability to route and manipulate photon at the nanoscale, SPs have been widely used in nano- and bio-photonics including biosensor, surface enhanced Raman scattering, optical tweezers, light emitting diodes, solar cell, and metamaterials, etc. However, although it is known that system geometry, material, wavelength, excitation condition are important in governing the behaviors of SPs as well as matter/SPPs interactions, how they work together in producing the final outcome is largely unknown. More importantly, there seems to be a "missing link" between these extrinsic parameters and the properties of SPs such that, for example, it is still not quite understood the exact roles of geometry play in controlling the SPs and their related properties. / In this thesis, we aim at bridging the link between extrinsic parameters and SPs properties based on temporal coupled-mode theory (CMT). CMT provides the knowledge of intrinsic parameters such as SPs radiative and absorption rates and their relationships with the phase, amplitude, and directionality of SPs. Because decay rates are strong function of system parameters, excitation condition, etc, connection between the extrinsic parameters and the SPs properties can now be established. With the decay rates and other accessories in hand, we study the fundamental of SPs from another perspective. For example, we examine how two degenerate SPs modes couple to yield a pair of dark and bright modes and find not only the radiation damping of SPs but also the absorption are modified. In addition, we study the decay length and the group velocity of SPs in real- and momentum- space to elucidate the interplay between the SPs losses and the wavevector-resolved projected density-of-optical-states (PDOS). We also explore the rate matching (i.e. critical coupling) for optimizing the phase-jump in SPs resonance sensing and achieve high figure of merit in sensing performance. In the last decade, SPs effects gained widespread attention for their potential application in photonic devices, sensing, surface-enhanced fluorescence, especially Surface-Enhanced Raman Scattering (SERS). Many published results have confirmed the expected strengths of SERS, hence making it possible for SERS to become a next generation ultra-sensitive biosensing platform, which may take the form of various nano-structures in order to achieve optimized hot spots. While the wavelength of SPs is closely related to material dielectric properties and has limited scope for further reduction, it is of critical importance to ensure that SPs are being generated with the highest intensity before any further application advancement is possible. Meanwhile, plasmonics has aroused longstanding interests among researchers to realize nanophotonic devices. For example, ordered arrays of closely spaced metallic nanoparticles (MNP) have been employed to transport optical signals via near-field coupling below the diffraction limit. However, radiation and absorption losses in these waveguides can be serious. New concepts for novel plasmonic devices are essential. / In light of these issues, this thesis focuses on tailoring SPs from the viewpoints of structural and material properties with the following objectives: / (1) To develop a new class of simple plasmonic devices based on tailoring of propagating surface plasmons (PSPs) or cooperation between PSPs and localized plasmon resonance (LPRs) to offer significant field focusing and intensity enhancement. It can serve a wide range of applications, including high field related biomolecular sensing and detection as well as non-linear optical effects. / (2) To design low loss nanophotonic wave guides based on gain medium, which may offer real opportunity for practical nanophotonic devices. To obtain a theoretical interpretation of relationship between surface plasmon resonance and host environment where the plasmonic structure embedded. This study should provide further insight towards sensing and device design. / We have achieved the following results in this project: / (1) A novel SERS excitation source based on focusing of surface plasmons around the center hole of a metal disk for cascaded enhancement is put forward and studied theoretically. The device offers intense SERS excitation with quasi-uniformity and horizontal polarization over a comparatively large hole. As revealed by fmite-difference time-domain (FDTD) method, the intensity spectra and the characteristics of the near field for the wavelength range of 650-1 000 nm exhibit a number of enhancement modes. Electric field intensity of the optimal mode enhances the SERS signal inside the hole by over four orders. An analytical model was also developed to gain precise interpretation on FDTD results. Our model also reveals the possibility of achieving eight orders of enhancement by optimizing the scale of the disk. Our results indicate that much higher electric field enhancement in hollow metal disks (HMDs) can only be possible when we have a hole at the centre and the direction of the focusing field is parallel to the surface of the plasmonic device. This is because of the presence of an insulating gap at the center, that higher level of electric field can exist as electrons are not allowed to flow pass the gap. On the other hand, in the case of a solid metal disk, the flow of mobile electron will tend to dampen the amplitude of the arriving SPs. In addition to generation of highly optimized hot spots for SERS, the large active hole also offers potential applications in fluorescence enhancement and nonlinear spectroscopy. / In addition to HMDs, we also develop a kind of highly optimized hot spots based on diffraction coupling between LPRs via gain-assisted PSPs. Thus derived device was theoretically analyzed. The process of diffraction coupling is achieved via localization of light by LPRs, LPRs-PSPs interplay and PSPs transfer. Our study shows that by incorporating optical gain to PSPs, a very strong boost of the electromagnetic enhancement of LPRs can be expected from a lasing process. We find that with a practical gain level, the enhancement factor of local electric field intensity can be larger than 10⁷. Hence, we offer an ideal configuration to realize high-field dependent single molecule SERS and also a newly applied physical scheme for nano-Iaser. / (2) We propose a low-loss nanoscale wave guide based on gain-assisted plasmonic resonance MNP chain. We demonstrate that by employing a gain material or even an appropriate dielectric for the host environment, waveguide loss can be reduced dramatically. A highly efficient pseudo-orthonormal basis expansion (POBE) method for obtaining the complex dielectric spectra of the low-loss transmission has been developed. Eigenmode analysis revealed the physical origin of those low-loss wave guiding modes, which opens the possibility to achieve waveguiding other than using conventional dipolar resonances of individual particles. A scheme based on electron beam lithography and chemically synthesized nanoparticles has been proposed to fabricate the device. Such plasmonic waveguides may serve as building blocks for making nanoscale optical devices especially for integrated nanophotonic circuits. Meanwhile, the originally developed POBE method, which reveals the general physical mechanism of SPs, can be used to further explore optimized gain-assisted plasmonic structures to design favorable nanophotonic devices. / This thesis begins with an old problem: light focusing and guiding in macroscopic scale with traditional dielectric, and sum up finally with SPs focusing and guiding in nanoscale with gain material and plasmonic material. Localized surface plasmon resonances of gold nanocrystals: refractive index sensitivity, plasmon coupling and photothermal conversion. / CUHK electronic theses & dissertations collectionJanuary 2010 (has links)
Assembly of noble metal nanocrystals gives rise to extraordinary plasmonic properties that are distinct from those of isolated ones. We have prepared clusters that are composed of two-dimensionally-ordered gold nanocubes on flat substrates and investigated their plasmonic properties. It is found that the plasmon resonances of the nanocube clusters are highly dependent on both the number and ordering of the nanocubes in the clusters. FDTD calculations reveal that the rich plasmon modes in the clusters originate from the interparticle couplings in the cluster and the couplings between the entire clusters and the substrate. / I believe that my research work has provided an in-depth fundamental understanding of the localized surface plasmon resonances of gold nanocrystals and will have a number of implications for the applications of metallic nanostructures in optics, optoelectronics, and biotechnology. / Noble metal nanocrystals have attracted much interest due to their rich optical properties, which arise from the localized surface plasmon resonances, the collective oscillations of free electrons confined on the nanoscale. Under resonant excitation by light, noble metal nanocrystals exhibit extremely large light scattering and absorption, as well as large near-field enhancements. These fascinating properties bring about a variety of applications, including plasmonic sensing, plasmonic waveguiding, surface-enhanced Raman scattering, plasmon-enhanced fluorescence, photothermal cancer therapy, and plasmonic-enhanced energy harvesting. Among various noble metal nanocrystals, gold nanocrystals exhibit high chemical stability and large biological compatibility. Moreover, their plasmon resonance wavelengths can be synthetically tuned from the visible to near infrared spectral regions. In this thesis, a systematic study on the localized surface plasmon resonances of gold nanocrystals is presented, both experimentally and theoretically. / Photothermal conversion of gold nanocrystals can be applied in the areas of photothermal polymerization, photothermal imaging, drug release from capsules, and photothermal therapy. We have investigated the photothermal conversion properties of different gold nanocrystals and their composites. The studies show that the plasmon wavelength, particle volume, shell coating, and assembly of gold nanocrystals all play important roles in their photothermal conversion efficiency. / The refractive index sensitivity of gold nanocrystals is a key factor in their practical sensing applications. I will first introduce the systematic studies on the dependence of the index sensitivity on the shapes and sizes of gold nanocrystals that have varying plasmon resonance wavelengths. The index sensitivity has been found to generally increase as the plasmon resonance wavelength for a fixed nanocrystal shape becomes longer and as the curvature of the nanocrystals gets larger. I have further studied the dependence of the index sensitivity on the different shapes of gold nanocrystals that have the same longitudinal plasmon resonance wavelength. The refractive index sensitivities have been found to vary with the nanocrystal shape. Finite-difference time-domain (FDTD) calculations have been performed on these nanocrystals to reveal the origin of this dependence. A linear relationship is found between the index sensitivity and the product of the electric polarizability with the curvature. On the basis of these studies, a novel plasmonic optical fiber device has further been fabricated to detect small changes in the local dielectric environment. / When fabricating plasmonic devices, such as waveguides, optical switches, plasmonic sensors, and plasmon-enhanced solar cells, one needs to attach metal nanocrystals onto different substrates. The interactions between gold nanocrystals and the substrates can strongly modify the plasmonic responses of the nanocrystals and therefore need to be taken into account when designing of various plasmonic devices. We have further investigated the coupling between gold nanocrystals and substrates with different dielectric properties, including insulating, semiconducting, and metallic ones. It is found that the substrates play an important role in both the scattering patterns and scattering spectra of the supported gold nanocrystals. Kinetics of insulin - insulin receptor interaction using a surface plasmon resonance (SPR)Subramanian, Kannan January 2014 (has links)
Type 2 diabetes or adult onset diabetes, has been a global epidemic for the past two decades, and the number of new cases accelerates every year. Insulin resistance is one of the major factors behind this, wherein the insulin receptor, which signals to regulate glucose levels, based on the hormone insulin, loses its sensitivity. Obesity is one other major concern which is caused due to the improper balance between the caloric intake and the energy utilized. Gastric bypass surgeries (GBP) are performed to avert obesity. However, a beneficial side-effect is that the state of insulin resistance is reset to near baseline levels within a few days after the procedure. The reason behind this remains unexplained, with possible humoral effects, hypothesized to occur after the bariatric procedure.
In this work, high-five insect cell line was utilized to recombinantly produce full length insulin receptors (IR). However commercially sourced IR ectodomains (eIR – soluble version of the full length IR with the completely extracellular α subunits along with extracellular and transmembrane regions of the β subunit), were used to study the interaction. Measuring the kinetics of IR-insulin interactions is critical to improving our understanding of this disease. In this study, a multiplex surface plasmon resonance (SPR) assay was developed for studying the interaction between insulin and the eIR. A scaffold approach was used in which anti-insulin receptor monoclonal antibody 83–7 (Abcam, Cambridge, UK) was first immobilized on the SPR sensorchip by amine coupling, followed by eIR capture. The multiplex SPR system (ProteOn XPR36TM, Bio-Rad Laboratories, Hercules, CA) enabled measurement of replicate interactions with a single, parallel set of analyte injections, whereas repeated regeneration of the scaffold between measurements caused variable loss of antibody activity. The main approach was to replicate the physiological IR-insulin interaction using this assay. It was also observed that insulin at higher concentrations tend to form dimers and hexamers in solution.
This was tested using size exclusion chromatography analysis and proved to be true. Therefore an alternative analyte with the similar binding properties and affinity of insulin and at the same time with reduced self- association characteristics was explored. Lispro, the analogue of insulin with reduced self-association properties (generated by swapping of residue 28 and 29 with Lys and Pro respectively) was finally used to study the interaction with eIR.
Interactions between recombinant human insulin with eIR-A (A isoform of the insulin receptor ectodomain) followed a two-site binding pattern (consistent with the literature), with a high-affinity site (dissociation constant KD1 = 38.1 ± 0.9 nM) and a low-affinity site (KD2 = 166.3 ± 7.3 nM). The predominantly monomeric insulin analogue Lispro had corresponding dissociation constants KD1 =73.2 ± 1.8 nM and KD2 =148.9 ± 6.1 nM, but the fit to kinetic data was improved when conformational change factor was included in which the high-affinity site was converted to the low-affinity site. Kinetics of interaction of insulin with eIR-A and eIR-B isoforms were then compared. eIR-A bound insulin with apparently higher affinity (with both the binding sites) when compared with eIR-B. This was again consistent with literature that IR-A had two-fold higher affinity for binding insulin than IR-B.
The assay was further extended to study the effect of external factors such as glucose, visfatin on this interaction. Glucose (the main biomolecule which is regulated by the IR-insulin interaction) was tested, if it had any direct effect on the interaction. It was observed that glucose did not have any effect on eIR-insulin interactions. Visfatin, an adipocytokine which has been highly debated in literature for its insulin mimetic effects and IR binding properties, was then tested. The standard assay did not provide much insights as the reference channel immobilized with 83-7 monoclonal antibody to the receptor had much affinity for visfatin, leading to non-specific binding and negative responses.
Therefore, in an alternative methodology was used - visfatin, Lispro and insulin were immobilized on separate channels along with bovine serum albumin immobilized on reference channel and eIR isoforms used as analyte to study the effect of visfatin on IR. This study showed that visfatin, a higher molecular weight protein compared to insulin, bound both the eIR isoforms. This is consistent with literature that visfatin binds IR at a site distinct from insulin, but the assay described here could not confirm the fact that it mimicked the signalling carried out by IR-insulin binding. Further studies are required to interpret the kinetics of visfatin-eIR interaction.
To my knowledge, this is the first SPR assay developed to study eIR-insulin interactions in real-time. This could potentially be extended to study the interaction of insulin with full length insulin receptors and the effect of humoral and other external factors on the interaction, without the need for insulin labelling.

Surface plasmon resonance (SPR) biosensors have attracted great attention in scientific research in the past three decades. Extensive studies on the immobilisation of biorecognition elements have been conducted in pursuit of higher sensitivity, but trialled formats have focussed on a thin layer modification next to the plasmon film, which usually requires in situ derivatization. This thesis investigates an 'off-chip' immobilisation strategy for SPR biosensing using silica particles and considers the implications of a particle-modified evanescent field on the signal amplitude and kinetics, for an exemplar affinity binding between immobilised IgG and its anti-IgG complement. Submicron silica particles were synthesized as carriers for the bio-recognition elements. They were then immobilised to form a sub-monolayer on the gold film of an SPR biosensor using two methods: thiolsilane coupling and physical adsorption aided by mechanical pressure. The bio-sensitivity towards an antigen/antibody interaction was lower than an SPR biosensor with an alkanethiolate SAM due to the difference in ligand capacity and position in the evanescent field. The binding kinetics of antigen/antibody pair was found to follow the Langmuir model closely in a continuous flow configuration but was heavily limited by the mass transport from the bulk to the sensor surface in a stop-flow configuration. A packed channel configuration was designed with larger gel particles as ligand carriers, packed on top of a gold film to create a column-modified SPR biosensor. This sensor has comparable bio-sensitivity to the previous sub-monolayer particle-modified systems, but the binding and dissociation of the analyte was heavily dependent on mass transport and binding equilibria across the column. A bi-directional diffusion mechanism was proposed based on a two-compartment mass transport model and the expanded model fitted well with the experimental data. The column-modified sensor was also studied by SPR imaging and analyte band formation was observed and analysed. Using the lateral resolution, a multiplexing particle column configuration was explored, and its potential in distinguishing a multicomponent analyte.

In this thesis study, the surface/interface plasmon excitations in different Si nanostructures were revealed through the EELS study in TEM/STEM. In the case of the planar boundary such as the wedge-like specimen, the spatially resolved EELS results disclose the dependence of the intensity and the position of the interface plasmon peak on the sample thickness. In the case of the Si-core/ SiO2-shell nanoparticles, we found that the SP/IP peak will firstly red-shifts with the increase of the SiO2 shell thickness and eventually levels off . As the aspect ratio of the Si nanoparticles increases, (from spherical particle to nanorod and nanowire), the SP/IP will split into two branches: transverse and longitudinal modes. We also found the intensity ratio of the transverse/longitudinal mode excitations depends on the diameter of the Si core size in the nanostructures. In the one-dimensional interacting Si nanoparticle chains, the Si nanoparticles were embedded in the SiO 2 shell, the splitting of the SP excitation into transverse and longitudinal modes was also observed. As the inter-particle distance reduces to several nanometers, the coupling of the IP excitation between the adjacent particles becomes significant, and results in the local field enhancement in-between the two particles. This is directly visualized using EFTEM imaging in TEM/STEM. / Surface/interface plasmons (SP/IP) are the plasmons confined at specific boundaries, describing the surface/interface charge density oscillation. They are generated when the scattered electromagnetic wave with its scattering vector component parallel to the boundary propagates along the surface/interface. Study of surface plasmon resonance in noble metals such as gold and silver nanoparticles have started decades ago, and recent interests are focused on the plasmonic properties of individual nanoparticles, as enabled by the size/shape control in the nanostructure growth and advances made in the characterization methodologies. Besides the noble metals, semiconductor such as silicon also attracts much attention for its plasmonic behavior. The surface/interface plasmon resonance frequency of Si-based nanostructures occurs at relatively higher energies (compared to Au and Ag), making it a perfect system to be studied using electron energy loss spectroscopy (EELS) based techniques. When performed in a scanning transmission electron microscope (STEM), such a technique enjoys excellent spatial resolution, and can map the local plasmonic properties of individual nanostructures. / The plasmon excitation depends sensitively on not only the material dielectric properties but also the geometrical configurations of the material. In the present thesis work, silicon-based nanostructures with planar, spherical, and cylindrical boundaries were investigated using both experimental and theoretical approaches, with focus on the plasmon oscillation originating from the Si/SiO 2 interface.
