Simplified modeling of shear tab connections in progressive collapse analysis of steel structures

Recent tragedies involving the collapse of several large and prominent buildings have brought international attention to the subject of progressive collapse, and the field of structural engineering is actively investigating ways to better protect structures from such catastrophic failures. One focus of these investigations is the behavior and performance of shear tab connections in steel structures during progressive collapse events. The shear tab, a simple connection, is typically modeled as a perfect pin in standard design, but in progressive collapse analysis, a much more accurate model of its true behavior and limits is required. This report documents the development of a simple yet accurate shear tab model and its use in understanding the behavior and limits of shear tab connections in column removal scenarios. Particular attention is paid to the connections’ axial force limit state, an aspect of behavior that is typically unimportant in standard design. / text

Influence des procédés industriels sur l'allergénicité des aliments / Effect of industrial process on food allergy

La prévalence de l’allergie alimentaire est en constante augmentation. Parallèlement les consommations alimentaires et les aliments eux même évoluent. L’aliment, de plus en plus soumis aux traitements industriels, peut présenter une allergénicité modifiée. L’objet de cette thèse sera d’évaluer l’influence des différents procédés industriels sur l’immunoréactivité des protéines alimentaires modifiées. Les différentes études présentées dans l’ouvrage aborderont l’effet de la texturation sur les protéines de soja ou de l’extrusion sur les protéines de lin (traitement thermique), l’effet de l’incorporation de nouveaux ingrédients dans l’alimentation moderne (inuline, lin). L’allergénicité des protéines recombinantes (process biotechnologique) sera abordée au travers de deux études : l’une proposera les protéines recombinantes de l’arachide comme outil diagnostique, en particulier pour le test d’activation des basophiles ; L’autre cherchera à démontrer l’innocuité allergénique d’un lysozymes recombinant humain, qui sera utilisé en alimentation humaine. L’étude de ces propriétés allergéniques sera réalisée par les tests in vitro (prick tests et test de provocation orale) et par les tests in vitro (IgEs). Des études par immuno-empreinte (immunoblot et dot blot) et par spectrométrie infrarouge permettront d’identifier les protéines modifiées. Enfin des tests cellulaires (test d’activation des basophiles) seront également réalisés. / The prevalence of the food allergy is in constant increase. At the same time food consumptions and food they even evolve. The food, more and more subjected to the industrial treatments, can present a modified allergenicity. The aim of this thesis will be to estimate the influence of the various industrial processes on the immunoreactivity of modified food proteins. The various studies presented in the work will approach the effect of the texturisation on proteins of soy or the extrusion on flaxseed proteins (heat treatment), the effect of the incorporation of new ingredients in the modern food (inuline, flaxseed). The allergenicity of recombinant proteins (biotechnological process) will be approached through two studies: the one will propose peanut recombinant proteins as a diagnostic tool, in particular for the basophil activation test (BAT); other one will try to demonstrate the allergenic harmlessness of a recombinant human lysozyme, which will be used in human food. The study of these allergenic properties will be realized by the in vivo tests (prick tests) and by the in vitro tests (IgEs). Studies by immunoblot and by infrared spectrometry will allow identifying modified proteins. Finally cellular tests (BAT) will be also performed.

HistoryLane : Web Browser History Visualization Method

With the evolution of Internet, users exploit it increasingly intensively for achieving their goals: sending emails, playing games, watching videos, reading news. They do it through Web browsers, that may vary in exact implementation, but have similar core functionalities . One of these core functionalities is the access to a browsing history. However, as browsing patterns are getting more complex, the traditional history tools become insufficient. Visualization of browsing history might be helpful in that case. In this study we propose a novel approach for browsing history visualization, named HistoryLane, which ts the parallel browsing paradigm, common for modern browsers. The main goal of HistoryLane is enabling the user to gain insight into his own or into other users' parallel browsing patterns over time. Principles of HistoryLane visualization approach are formulated based on recommendations, found during structured literature review. These principles constitute the base for a prototype, which was implemented as a Fire- fox extension. To evaluate the e ffectiveness of HistoryLane we conducted a survey and a quantitative experiment. The results of the evaluation show that HistoryLane is perceived by users as effective and intuitive method for browsing history visualization.

A comparison of double clip angle shear connections to shear tab connections in industrial applications

Kramer / In structural steel connection design, simple shear connections are one of the most common connection types utilized. The industry, especially from the side of the engineer, tends to lean toward using Double Clip Angle Connections as the default standard for simple shear connections. A double clip angle connection is a connection consisting of two angles transferring the shear forces from one member to the next either through bolts or welds. The design of Double Clip Angle Connections is efficient and the connections themselves are easy to fabricate. However, benefits to utilizing other types of shear connections exist. Many of these benefits are seen in the fabrication shop or during erection and construction. This is especially true of single shear plate or shear tab connections when applied to open structure design. Shear tab connections consist of a single plate that transfers the shear forces from one member to the next with bolts or with welds. The design of shear tab connections can be a more involved process than the design of double clip angles. Sometimes the shear plate or shear tab has to be longer than is typical. This is called an extended shear plate connection. These extended shear plates can bring other variables into the design that typically don’t occur with Double Clip Angle Connections such as bending of the plate or the need for multiple bolt columns. However, with proper planning and detailing, the benefits and savings experienced in the fabrication or construction phase may outweigh what can be seen as a more laborious design task. The purpose of this report is to identify the possible benefits achieved in using each of these connections, highlight the differences in the design approach for each, and use a study model to compare the outcome of using one connection over another in the design of a typical open structure. Double clip angles are typically the most efficient approach when speed of design and simplicity of fabrication are the desired outcomes. However, shear plate or shear tab connections have the potential to provide safer erection alternatives and materials savings if used in appropriate ways and with the right applications.

První část DP se věnuje mapování teoretické části, jejíž střípky jsou rozesety v mnoha cizojazyčných zdrojích. Je zde provedena podrobná analýza nehmotného majetku a jeho oceňování pro potřebu alokace kupní ceny. Druhá část je věnována praktické aplikaci první části na příkladech, nikoli nepodobných praxi.

Development and comparison of three immunoassay formats to screen for total anti-adeno-associated virus serotype 2 antibodies in human serum using the Gyrolab immunoassay platform

Recombinant adeno-associated virus vectors are one of the most promising gene delivery tools for applications within gene- and cell therapy. The high level of wild-type adeno-associated virus infections in humans is a limitation due to the pre-existing immunity against the vector or its transgene product. An important tool to develop effective and safe therapies is the ability to measure the pre-existing immune responses against the virus capsids in humans. This master thesis at Gyros Protein Technologies aimed to investigate if the Gyrolab immunoassay system can be used to screen for pre-existing anti-capsid immunity in human sera by optimizing and evaluate three different assay formats: an indirect assay, a generic anti-AAV adsorption assay and a bridging assay. The evaluation focused on immunity against adeno-associated virus serotype 2. All immunoassay formats performed well and depending on application, the different formats offers different advantages. The generic anti-AAV adsorption assay offers the ability to easily screen for several viral serotypes without having to label the capsid, and the bridging assay provides high sensitivity. When screening 31 individual human sera, 58% were positive using the indirect assay and the generic anti-AAV adsorption assay and 65% using the bridging assay format. Provided, is automated and high throughout immunoassays where 16 individuals can be screened in one-two hours. It is shown that all three immunoassay formats can be used to screen for anti-adeno-associated virus antibodies, even though further optimization, cut off development and a larger data set is needed to obtain a fully sophisticated screening tool.

'Tab' figurines and social identity at La Blanca

This thesis examines a special group of Middle Preclassic (900-600 BC) figurines excavated at La Blanca, an early Mesoamerican site on the Pacific Coast of Guatemala. Figurines at La Blanca are ubiquitous and derive from both elite and non-elite household contexts. Because of their widespread distribution, archaeologists associate figurines with daily practice and household ritual in ancient Mesoamerica. They represent a rare opportunity to examine materializations of the human body across social strata, and because their depositional contexts do not seem to suggest ritual care, their context of use remains enigmatic. With the dawn of the Middle Preclassic period, the community at La Blanca was at the center of a dramatic transition: in addition to the reconfiguring of political, social, and economic structures, the nature of personhood was profoundly transformed during this period. I argue that figurines were actively involved in the ongoing negotiation of social identity and personhood at La Blanca during this important transitional period. I specifically discuss a group of figurines from La Blanca called 'tab' figurines, which are remarkable for their exaggerated sexual characteristics and distinct approach to depicting the human form. I examine the 'tab' figurine assemblage in depth and examine how aspects of their context, form, and function helped their makers negotiate social identity at La Blanca. / text
