Biotic Factors and Temperature Tolerances via Critical Thermal Methodology in Goldfish

CTMinimum and CTMaximum were measured in 620 goldfish to determine if biotic factors, in particular starvation, condition factor and size, affect temperature tolerance. Twenty-eight days of starvation adversely affected both upper and lower temperature tolerance. Condition factor was related to upper but not lower temperature tolerance.

Screening ornamental pepper (Capsicum annuum L.) cultivars for temperature tolerance using pollen, physiological and seed germination parameters

Temperature affects reproductive potential, aesthetic and commercial value of ornamental peppers. In experiment one, temperature effects were assessed in 12 ornamental cultivars using in vitro pollen germination and tube length, and the physiological parameters, cell membrane thermostability, chlorophyll stability index and canopy temperature depression. In experiment two, seed germination rate and maximum seed germination response to temperature were assessed. Cumulative temperature response indices (CTRI) for pollen, seed, and physiological parameters were derived and used to classify cultivars for temperature tolerance. CTRI based on pollen parameters showed significant, but poor correlation with physiological parameters. CTRI based on seed parameters showed significant correlation with CTRI-physiological parameters. It is concluded that screening using pollen parameters will be ideal for reproductive temperature tolerance while seed and physiological parameters will be suitable for screening vegetative temperature tolerance. Identified tolerant cultivars are potential candidates for breeding programs to develop heat and cold tolerant ornamental pepper genotypes.

Midseason cold tolerance screening for the NSW rice improvement program

The current rice varieties grown by Australian farmers are susceptible to low temperature events particularly during the reproductive stage of plant development. The best management practices of sowing within the recommended time period and maintaining deep water (20–25 cm) through the microspore development stage only offer limited protection. There is a need to develop more cold tolerant varieties and to do so requires the development of low-temperature screening capacity for the NSW rice breeding program. This study looked at the requirements of adapting a controlled-temperature glasshouse facility to enable screening for tolerance to low temperatures during the reproductive stage of rice development. The investigations were grouped into two areas; 1) the physical aspects of the low temperature facility including the location of plants within the facility and within the tubs used to grow the plants and whether these can influence the reliability of the screening and 2) the biological effects of nitrogen (N) concentration in the plant at panicle initiation (PI) and plant susceptibility to low temperatures, and whether growth stage of the plant relative to PI at the start of low temperature treatment influenced floret sterility. A series of nine experiments were conducted at the Deniliquin Agricultural Research and Advisory Station glasshouse facility using up to five rice varieties selected for their divergence in low-temperature tolerance. One other experiment was conducted in a different facility. The modified glasshouse facility in Deniliquin was effective in providing the targeted screening environment of 27°C day and 13°C night temperature regime. There was however a smaller than expected effect of the low temperature exposure in some of the experiments with sterility following low temperature ranging from 9.9% to 27.7%. There was also a higher than expected level of sterility in the controls (i.e. not exposed to low temperature) with sterility levels up to 58% recorded in one experiment. The causes of these overall effects are not known. Notwithstanding these overall effects there were a number of findings that are important for developing a reliable screening facility. The spatial arrangement of the plants within the low temperature facility effected the level of sterility highlighting the need for experimental design to consider spatial variation. The existence of edge effects was identified within the tubs used to maintain permanent water on the potted plants whereby the outer plants in the tubs were less damaged by the low-temperature treatment. The overall N level in the leaf tissue was low even at the equivalent rate of 250 kg N ha-1 and there was only a very modest and inconsistent response in N concentration at PI to N application rates ranging from 0 to 250 kg ha-1. However, the method of growing the plants in pots and of nitrogen fertiliser application did not alter the N concentration. The concentration was the same when N was added either, on the same day as permanent water application, or three days prior to permanent water application. Also restricting the direction of water movement through the pots and therefore the soil within the pots did not alter the amount of N in the plants at PI. The low plant N concentrations were consistent across two glasshouses in which soil from the same source was used suggesting a soil limitation. A soil test identified that the soil phosphorus (P) was at a level at which plants have responded to P application under field conditions, and the loamy texture of the soil had an associated low cation exchange capacity in comparison to medium to heavy clay soil types commonly associated with rice growing. These factors may have reduced the N retention and uptake and, in part, explain the low injury from the low temperature exposures. In the variety Millin, there was no significant effect of timing of the exposure on sterility until it commenced 12 to 15 days after PI. This is a significant finding for a breeding program that must expose lines of unknown phenological development. It means that even though there are small differences in the rates of development, there is no large effect of this on sterility. However, this response was not seen in the other varieties tested and thus requires further validation. It was difficult to induce repeatable levels of floret sterility in this series of experiments most likely due to the low N concentrations in part due to the properties of the soil used to grow the plants. This highlights the importance of standardising all cultural aspects in order to provide uniform and repeatable screening information. The spatial effects highlight importance of experimental design for effective exposure to low temperature treatments, incorporation of the capacity for spatial analysis in the statistical design, the use of standard variety checks for floret sterility after low temperature treatment, and the determination of N concentration in plant tissue prior to exposure.

Escaping the Arrhenius Tyranny: Metabolic Compensation during exposure to high temperature in Daphnia

Poikilothermic organisms experience trade-offs associated with life at different temperatures caused by incompatible physiological and biochemical demands caused by temperature extremes. As the result many such organisms exhibit acclamatory effects, adjusting their metabolism and physiology to recently experiences temperatures. One such acclamatory effect is the metabolic compensation that allows an organism to withstand increases in temperature by decelerating biological rates below Arrhenius expectations, presumably reducing energetic demand and reducing stress. Daphnia magna is resilient across a wide temperature range, and if acclimated to mildly stressful temperatures first, exhibits longer survival in lethal temperatures. Certain genotypes of Daphnia also exhibit higher acute thermal tolerance than others, indicating the presence of genetic variation and local adaptation in heat tolerance. This study examined the effect of ambient temperature (5°C-37°C) and acclimation history (2 generations at 10°C or 25°C) on the oxygen consumption rates of 8 genotypes of Daphnia (4 with high acute temperature tolerance, and 4 low). There are nonlinear decelerations of Daphnia respiratory rates across a temperature gradient when acclimated to 25°C or following short 8-hour acclimation to measurement temperatures. Furthermore, Daphnia exposed to a near-lethal temperature (35°C) with a subsequent 24-hour recovery period at their native 25°C-acclimation temperature shows no indication of respiratory damage. Genotype showed no difference in metabolic compensation, indicating the process is genetically constrained. Regulation of mitochondrial and membrane function are promising areas to further characterize the mechanism of metabolic compensation found in this study.

Temperature Tolerance of Freshwater Fish Exposed to Water-Borne Cadmium

Lethal toxicity values (96 h LC50; mg Cd/L) for the test species were similar: Lepomis cyanellus, 11.52; Notropis lutrensis, 6.62; Pimephales promelus, 3.58. However the effects of cadmium concentration and exposure time on temperature tolerance varied between species. Neither cadmium concentration nor exposure time had a significant effect on the CTM of green sunfish. Both cadmium concentration and exposure time had a significant effect on the CTMs of red shiners and fathead minnows. By day 10 mean CTMs were 2.3 t- 4.5 C (red shiners) and 4.2 to 5.7 C (fathead minnows) lower than control CTM. These results suggest a potential problem in cadmium contaminated systems for high environmental temperatures to stress or kill fish.

Acidophilic bacteria are able to tolerate acidic environment and also contribute to the lowering of environmental pH value, implying potential applications in metal-leaching technology extracting metals from tailings and electronic wastes. In this study, we conducted a sampling campaign in abandoned Tin mine in Ha Thuong, Thai Nguyen province, to isolate acidophilic bacteria and to study physiological characteristics of the isolated bacteria. As a result, two acidophilic bacteria were successfully isolated and identification by 16S rDNA gene sequences showed that the two bacteria are similar to Acidithiobacillus ferrooxidans and Acidithiobacillus thiooxidans (98% and 94 % of similarity, respectively). Both strains are tolerant of pH in the range of 3 and have the ability to grow optimally at temperatures of 30°C. / Vi khuẩn ưa axit có ý nghĩa ứng dụng trong công nghệ tách rút kim loại từ quặng đuôi và ngay cả từ rác thải linh kiện điện tử. Trong nghiên cứu này, chúng tôi đã tiến hành thu mẫu ở mỏ thiếc bỏ hoang ở Hà Thượng, Thái Nguyên nhằm phân lập được nhóm vi khuẩn ưa axit và qua đó nghiên cứu một số đặc điểm sinh lý của vi khuẩn này. Kết quả chúng tôi đã phân lập được hai chủng vi khuẩn ưa axit. Định dạng bằng nhận diện trình tự gen 16S rADN cho thấy hai vi khuẩn này có độ tương đồng là 98% với vi khuẩn Acidithiobacillus ferrooxidans và 94% Acidithiobacillus thiooxidans. Temperature and Polyunsaturated Fatty Acid’s Effect on Daphnia magna Reproduction

Organisms adapt to their environments by adjusting their biochemistry and physiology; such adaptation is limited by resource availability and physiological constraints. The freshwater crustacean Daphnia magna inhabits a wide range of environments and must survive and reproduce within a range of temperatures. One limit to low-temperature adaptation is thought to be the availability of unsaturated fatty acids necessary to maintain proper fluidity of cellular membranes. D. magna maintained at 10 ºC on a diet poor in unsaturated fatty acids have been observed to produce clutches that fail to develop. However, this has not been observed on a diet rich in unsaturated fatty acids or at a higher temperature regardless of diet. Clonal variation is commonly seen in D. magna life history traits, including heat tolerance, and was also investigated. D. magna were kept at two temperatures and fed two algal diets that differ in unsaturated fatty acid content. To investigate the role of fatty acid composition on the reproductive success of D. magna, fatty acids were extracted from adults and eggs. Of the twenty-one clones studied, no clonal variation was seen in the ability to produce successful clutches at 10 °C on a diet poor in unsaturated fatty acids. Gas chromatography revealed significant differences in 20-carbon fatty acids and suggest a parent-offspring conflict over a limited resource.

FR‐H3 : a new QTL to assist in the development of fall-sown barley with superior low temperature tolerance

Fall-sown barley will be increasingly important in the era of climate change due to higher yield potential and efficient use of water resources. Resistance/tolerance to biotic and abiotic stresses will be critical. Low temperature is an abiotic stress of great importance. Resistance to barley stripe rust (incited by Puccinia striifomis f. sp. hordei) and scald (incited by Rhynchosporium secalis) will be important in higher rainfall areas. Simultaneous gene discovery and breeding will accelerate the development of agronomically relevant germplasm. The role of FR-H1 and FR-H2 in low temperature tolerance (LTT) has been well documented. However the question still remains: is LTT due only to FR-H1 and FR-H2 or are there other, undiscovered, determinants of this critical trait? We developed two doubled haploid mapping populations using two lines from the University of Nebraska (NE) with superior cold tolerance and one line from Oregon State University (OR) with good malting quality and disease resistance: NB3437f/OR71 (facultative x facultative) and NB713/OR71 (winter x facultative). Both were genotyped with a custom 384 oligonucleotide pool assay (OPA). QTL analyses were performed for LTT, vernalization sensitivity (VS), and resistance to barley stripe rust and scald. Disease resistance QTL were identified with favorable alleles from both NE and OR germplasm. The role of VRN-H2 in VS was confirmed and a novel alternative winter allele at VRN-H3 was discovered in the Nebraska germplasm. FR-H2 was identified as a determinant of LTT and a new QTL, FR-H3, was discovered on chromosome 1H that accounted for up to 48% of the phenotypic variation in field survival at St. Paul, Minnesota, USA. The discovery of FR-H3 is a significant advancement in barley LTT genetics and will assist in developing the next generation of fall-sown varieties. / Graduation date: 2012
