Foram coletadas sessenta e nove amostras. Usando técnicas citogenética tradicionais e protocolos de hibridização in situ (FISH). Foram realizados inferência bayesiana e analise de máxima parcimônia usando genes mitocôndrias ATP synthase 6 (ATPase-6), cytochrome c oxidase subunit 1 e o gene nuclear recombination activating gene 2 (RAG2). As populações de Hoplias malabaricus do Alto e Médio Magdalena mostram o cariomorfo 2n=42A, e as populações do Baixo Magdalena apresenta o cariomorfo 2n=40C. Os dados moleculares mostram dois haplogrupos: O Haplogrupo I está composto pelo Alto, Médio Magdalena + Dagua (Drenagem do Pacífico) e o Haplogrupo II formado pelo baixo Magdalena + Bacias do Caribe (San Jorge, Ranchería, Atrato e Sinú) + Orinoco. A biogeografia histórica de H. malabaricus na Colômbia mostra grandes diferencias ecológicas entre o Alto Magdalena+Medio Magdalena + Dagua y Atrato + Baixo Magdalena + Orinoco + Sinú + Ranchería. / The aim of this study was to reconstruct the historical biogeography of Hoplias malabaricus populations in the river basins of Colombia and Northern South America using cytogenetic and molecular data, to provide an assessment of those populations in the Neotropical Region. Sixty-nine samples were collected. Cytogenetic techniques included standard and fluorescent in situ hybridization (FISH) protocols. Bayesian inference and maximum parsimony analyses were performed using mitochondrial genes ATP synthase 6 (ATPase-6), cytochrome c oxidase subunit 1 and the recombination activating gene 2 (RAG2) nuclear gene sequences. Hoplias malabaricus populations from the Upper and Middle Magdalena River showed the 2n=42A karyotype, whereas the population from the Lower Magdalena River had the 2n=40C karyotype. Molecular data showed two haplogroups: Haplogroup I composed of Upper, Middle Magdalena + Dagua (Pacific Coast drainage) and Haplogroup II formed by Lower Magdalena + Caribbean river basins (San Jorge, Ranchería, Atrato, and Sinú) + Orinoco. The biogeographical history of H. malabaricus in Colombia reflects deep ecological differences between the Upper +Middle Magdalena + Dagua River in the Pacific, and the Atrato River + Lower Magdalena + Orinoco + Sinú + Ranchería.
