Cada período experimental teve duração de 21 dias, sendo sete dias para adaptação e 14 dias para coletas de amostras. As rações experimentais, isoprotéicas, foram formuladas para conter, na base da matéria seca, 60% de silagem de milho, 40% de concentrado e 0; 0,7; 1,4; e 2,1% de uréia, correspondentes aos teores de proteína bruta na forma de compostos nitrogenados não-protéicos (NNP) de 2,22; 4,18; 5,96; e 8,09%. Foi determinado o tempo necessário para a adaptação e medição do consumo e da produção de leite e avaliados a produção e a composição do leite, os consumos e as digestibilidades aparentes de matéria seca (MS), matéria orgânica (MO), fibra em detergente neuto (FDN), extrato etéreo (EE), carboidratos totais (CHOT) e proteína, o consumo de nutrientes digestíveis totais (NDT) e a produção de proteína microbiana, utilizando-se a excreção total de derivados de purina (DP) e as excreções de DP e de uréia obtidas a partir de coletas de urina de 24 horas e spot. As amostras de silagem de milho, concentrado e sobras foram coletadas diariamente e as de fezes, no 15 o e 20 o dia de cada período experimental. As o amostras spot de urina foram obtidas no 15 dia e as amostras de urina de o 24 horas, no 20 dia de cada período experimental. Amostras de sangue foram coletadas no 15 o dia de cada período experimental para avaliar as o concentrações de creatinina e uréia. As amostras de leite, obtidas no 15 e o 20 dia de cada período experimental, foram analisadas para gordura, proteína, alantoína e uréia. Para todos os tratamentos, sete dias foram suficientes para adaptação. Ao se elevarem os níveis de NNP na ração, os consumos de MS, MO, PB, CHOT e NDT diminuíram. Entretanto, as digestibilidades aparentes totais de MS, MO, FDN, EE, PB e CHOT não foram afetadas. A produção de leite corrigida ou não para 3,5% de gordura, as quantidades de gordura e proteína e os teores de proteína no leite tiveram comportamento linear negativo com o aumento dos teores de NNP nas rações; os teores de gordura e a eficiência alimentar não sofreram alterações. As estimativas do volume urinário e das excreções de uréia, alantoína, ácido úrico, purinas totais, purinas absorvidas e N-microbiano não diferiram daquelas obtidas pela coleta de 24 horas. / The experiment was realized in Cataguases, MG, using 16 Holstein cows allotted to four balanced 4 × 4 Latin square design, in accordance with the milking period. Each experimental period have a duration time of 21 days, with seven days of adaptation and 14 days of collection time. The four isoprotein experimental diet were formulated to contain in the dry matter base 60% of corn silage, 40% of concentrate and 0.0; 0.7; 1.4 and 2.1% of urea, corresponding to 2.22, 4.18, 5.96, and 8.09% of crude protein in the non protein nitrogen (NPN) based compounds. The milk composition and production, intake and apparent digestibilities of dry matter (DM), organic matter (OM), neutral detergent fiber (NDF), ether extract (EE), total carbohydrate (CHOT) and protein. The total digestible nutrient (TDN) intake were evaluated. The microbial protein production estimate using the total purine derivatives excretion (PD) and the comparison between the PD excretion and urea obtained from 24h and spot urine collection were also evaluated. Samples of corn silage concentrate and orts were daily collected and the feces samples collection at 15 th and 20 th day xiof the experimental period. The urine spot samples were obtained at 15 th day and the 24 hours urine samples at 20 th day of the experimental period. The th blood samples were collected at 15 of each experimental period to evaluated th the creatinine and urea concentration. The milk samples obtained at 15 and th 20 day of the experimental period were analyzed for fat, protein, allantoin and urea. Seven days of adaptation were reasonable for all treatments. The intake of DM, OM, CP, CHOT and TDN decreased as the NPN increased in the diets. However, the total apparent digestibilities of DM, OM, NDF, EE, CP and CHOT were not influenced by the NPN level in the diets. The 3,5% corrected fat milk production or not, the fat and protein yield and the milk protein content showed a negative linear behavior as the NPN increase in the diets, and the fat content and feed efficiency did not alter. The estimation of urinary volume, excretion of urea, allantoin, uric acid, total and absorved purines and microbial N did not differ from that obtained by the 24 hours urine collection. The plasma and milk urea nitrogen concentration linearly increase as the NPN levels increase in the diets.
