The aim of this thesis was to contribute to the standardization and validation of expiratory flow parameters independent of NO testing, particularly alveolar concentration (CANO) and bronchial NO flux (JawNO) in children and adolescents with allergic respiratory diseases. The thesis was created in the time of increased interest of medicine professionals in non-invasive examination methods and targeted therapeutic influence of inflammation in small airways in asthmatics. After the standardization and validation of clinical benefits, the measurement of concentrations of nitric oxide (FENO) in the air exhaled through the mouth with the speed of 50 ml/s was validated for the evaluation of eosinophilic airway inflammation in patients with asthma and started to be widely used in clinical practice. Currently, there is an ongoing research focused on a new method for FENO measurement at several flow rates, which uses physiological models of NO transfer in exhaled air, allowing to quantify the amount of NO in an indirect way and to monitor the intensity of the inflammatory processes in distal and proximal airways non- invasively. The main goal in reversing the allergy epidemic is the development of effective prophylactic strategies. Early life events, such as exposures to microbes, have a major influence on the development of balanced immune responses. Due to their ability to interact with host immune system and to modulate host immune responses probiotics, mainly bifidobacteria and lactobacilli have been used with some success in prevention of allergic disease. In order to be referred to as probiotic, bacterial strain has to undergo rigorous testing. We have selected three new Lactobacillus (L.) strains out of 24 human isolates according to their antagonistic activity against pathogenic bacteria, resistance to low pH and milieu of bile salts. Safety of these strains was proven upon intragastric administration to mice; moreover, we have shown their ability to shift cytokine Th1 - Th2 balance towards non-allergic Th1 response in isolated splenic cells. Allergen specific prophylaxis using probiotics as vehicles for mucosal delivery of recombinant allergen is an attractive concept for development of well-tolerated and effective allergy vaccines. 7 Abstract The thesis is focused on the pollen of selected allergenic plants. Thesis is structured into two parts., pollination and fertilization , development of the pollen grain , its structure and chemical composition. The end of the theoretical part, dedicated to allergies, is divided into subsections dealing with First, teoretical part describes the flower and its morphology, discusses the pollen grain, stamen anatomical structure immunity and allergies, naming and registration of allergens and cross- pollen allergy . The practical part contains an analysis of curriculum for students of secondary schools. An integral part of the work is a survey of teachers in secondary schools, in which method of questioning was used. The aim of this questioning was to determine the presence of solved issue in terms of representation of pollen and pollen allergens in textbooks for secondary schools , as well as the inclusion of this issue in the official curriculum, under the National Programme for the Development of Education in the Czech Republic. Based on the results of a questionnaire survey of secondary school teachers, teaching materials have been developed in the form of two worksheets for teaching botany and guidance on laboratory exercises in plant biology.

The work is divided into a theoretical and practical part. The theoretical part of the work is focused on explaining the basic concepts in allergology, types of allergies are presented, their symptoms and treatment. Furthermore, the work describes the types of prevention and the task of a nursing in allergology. In the practical part of the work three goals were set up. The first goal was to determine the role of a nurse in secondary prevention in patients / clients with an allergic disease. The second objective of the research was to determine whether differences exist by nurses in the provision of secondary prevention in patients / clients with an allergic disease at selected sites. The third objective was to determine how nurses prepare patients / clients for the adherence to a treatment regimen associated with their allergic disease. To achieve these objectives a qualitative research method of semi-structured interviews has been selected. The research group consisted of ten nurses, two nurses working in the allergology ambulance at the hospital and four nurses from private ambulances in České Budějovice, two nurses from private ambulances and one from the allergology ambulance at the hospital in Písek, and one nurse from the private allergology ambulance in Strakonice.

The aim of my thesis is the introduction of the systemic mastocytosis and discussion about results of allergen immunotherapy in patients with systemic mastocytosis and its preventive effects against recurrent anaphylactic reactions. Patients with systemic mastocytosis are more prone to severe systemic anaphylactic reactions after Hymenoptera stings than in the general insect venom allergic population patients without elevated basal tryptase. This severe reaction can result in the death of the patient, it is important to prevent it prophylactically. The medication of choice in insect venom allergic patients is hyposensibilization therapy - allergen immunotherapy, which uses venom allergens of causal Hymenoptera (honey bee, yellow jacket). The thesis aims to summarize the results obtained so far about the appropriateness of this treatment in patients with systemic mastocytosis, side-effects during VIT, optimal dosing schedule and duration of treatment in these patients.

The task of this thesis is to elaborate study of insect bite hypersensitivity incidence by the Old Kladrubian white horses. At the beginning of the thesis, problems of congenital disorders in general are summarized, such as origin of the disorders and the forming factors. Further, the insect bite hypersensitivity in horses as an allergic disorder with genetic predisposition is described. Original research concerns with analysis of all actual data of insect bite hypersensitivity incidence in population of Old Kladrubian horse and with analysis of selected horses´ progeny. Incidence of the disorder depends significantly on father of the foal and year of birth. No difference in morbidity between genders of foals was found. At the end the practical breeder´s advices are summarized.

We observe significant increase of herbal remedies used in paediatric patients in last decades. The goal of this work was set to examine available information about herbal safety. The list of herbs that are dangerous or inappropriate to use in children age was made. The herbs were grouped into simple classification system of herbal safety. In addition to chapters describing herbal safety the work contains knowledge about their general use in paediatrics phytotherapy, the pros and cons of their use. To gather the data of web research and scientific literature were employed. The analysed data were published between 1968-2017 and were collected using PubMed, ScienceDirect and Google Scholar. In my thesis I deal with the pollen of allergenic plants, the origin and proces of allergy to plant pollen and the inclusion of these topics in the teaching of plant biology in high schools. In the theoretical part I focus on topic of pollen allergy. First, I generally describe allergy and its origin, causes of allergic diseases, environmental factors and the development of allergic disease. I also briefly address the naming and registration of allergens and I also mention the so-called cross-allergy. Great emphasis is placed on pollen allergy, which is devoted to a larger unit, divided into individual subchapters dealing with the physiological function of the nose, followed by an allergic reaction of type I, an allergic reaction that occurs on the nasal mucosa. Furthermore, subchapters mentioning the classification of allergic rhinitis, diagnosis and pharmacotherapy, including additions to the drug groups used in the treatment of pollen allergy. Another larger unit is devoted to the structure of the plant's blossom as a source of allergen, which includes the anatomy of the blossom parts and a description of the pollen grain.
