The thesis examines the nursing care of children diagnosed with bronchial asthma. The thesis has three objectives. Firstly, it aims to identify differences in the care of asthmatic children in hospitals and outside hospital-type medical facilities; it also seeks to point out the most common problems of asthmatic children in each age category. Lastly, it examines the awareness of asthma amongst children and parents. Both quantitative and qualitative research was carried out to achieve the above objectives, using questionnaire and semi-structured interview techniques, respectively. The research part of the thesis was divided into two parts (two research samples). The first research sample consisted of children diagnosed with asthma and their parents, the other experimental group included nurses from selected hospitals and children's sanatoria/spas.

Bread has been one of the basic foodstuffs for more than 30,000 years. It´s prepared mainly by baking, where gluten plays an important role. Gluten is important for holding the gas in the dough to achieve the desired structure, texture and volume. At a time when agriculture began to develop, a disease of gluten began to emerge. One of the best known is the intolerance of gluten, the so-called celiac disease, which affect sapproximately 1 % of the world's population. The cause is gluten-rich proteins (prolamins and glutelins), found in wheat, rye, barley and oats. The second known diseaseis gluten allergy, which is often confused with celiac disease. The difference consist in a different autoimmune reaction to the presence of glucose, when the immunization on glucose produces IgE immunoglobulin and does not damage the small intestine mucosa The only possible treatment for patiens with a dinase is a strict gluten-free diet. Gluten-free breads and pastries are one of the main foods for celiac and allergy sufferers. Preparing a dough without gluten is difficult. Proper selection of raw materials plays an important role in meeting sensory properties. The aim of the diploma thesis was to prepare suitable recipe senriched with selected sources of antioxidants. In total, six different recipes were prepared, both from cereal gluten-free flour and legumes flour. Curcuma and red onion skin were selected sources of antioxidants. In the practical part a sensory evaluation of all samples of bread is performed. There was a positive evaluation of the questionnaire itself. When to Cheb the suitability of raw materials and procedures for dough preparation and baking. Based on the DPPH and FRAP tests, antioxidant aktivity was evaluated for all gluten-free bread. Bread senriched with red onion skin have the highest antioxidant activity in both basic bread and basic bread with legume flour. Thus, it can be said that onion skin effectively increase antioxidant activity. In the final part, a financial evaluation of all six types of gluten-free bread was made on the basis of raw materials used as compared to those already purchased in the shop. Based on this finding, it can be said that home preparation is a cheaper option.

The theme of the diploma thesis is allergens in the teaching of natural science, biology and chemistry. The subject and its aim is to find out the presence of allergens in selected classes of natural sciences (natural sciences, biology, chemistry) as well as the level of awareness of problems of allergies and allergens from teachers and pupils of elementary schools and grammar schools and to compare whether teachers of selected subjects take into account Creating climate climate for allergic pupils. The thesis consists of theoretical and practical part. The first part summarizes all theoretical knowledge, including the theme of allergies and allergens. It deals with the concept of allergy from the historical point of view, it characterizes the general importance of allergies and its connection with the immune system, it presents individual risk factors influencing the origin and development of allergic disease. A large part of the theoretical part describes allergic diseases including their manifestations, diagnosis and follow-up treatment, and also the individual types of allergens that are classified according to their effects on the organism. Due to high incidence, medical and socioeconomic burden and impact on individual quality of life and productivity, allergic disorders are a crucial issue for 21st century immunology. Much still remains to be elucidated, particularly regarding the very early processes in allergy development. In order to introduce timely, effective preventive measures, novel, more reliable predictive factors of allergy risk also need to be established. Dysregulation of proper balance between the branches of immune response, particularly unwarranted dominance of Th2, is the underlying cause of allergy. After birth, new immune balance needs to be established to prepare the neonate for adequate reactivity towards newly encountered environmental stimuli. Regulatory T cells (Treg) play a central role in finely setting this balance and inducing tolerance towards harmless environmental antigens, including allergens. Interactions with external factors, most importantly microbiota, modulate this process during the early postnatal "window of opportunity." Analysis of cord blood Treg of children of allergic mothers uncovered decreased presence of function-associated surface markers and lower production of IL-10. Reces on chronic disorders that occur at elementary schools. It mainly talks about formulation, structure, procedure, analysis of effects, and preventiv of the chronic disorders. Detailed focus is on identifying school children suffering from some chronic diseases such as asthma, diabetes, eczema, or flu. Reports and cooperation on an "Asthma, eczema, diabetes, flu" project were used to detect the suffering children. Both, healthy and sick children cooperate to achieve set goals within the project. Lastly, the matter of this thesis also includes prevention - recommendation to parents, whose children suffer from the diseases mentioned above. Theoretical observations having reference to chronic diseases, structure of reports, methods of evaluating the effects, preparation of the project and recommendation to parents whose children suffer from chronic disease are applied to completed report examination, project implementation, and discussion with parents.

The increasing incidence of children suffering from allergic diseases could be caused by sensitization of immature immune system during the intrauterine development. Several important scientific papers have demonstrated the ability of cord blood cells to respond by elevated proliferation activity after stimulation by common allergens. Following these findings, present study follows the production of cytokines which play a role in the pro- and anti-allergenic tuning of the immune system. Umbilical cord blood cells were stimulated with polyclonal activators (phytohaemagglutinin) and common allergens (ovalbumin, timothy grass, birch, mite). Subsequently, cytokine production was monitored using selected methods that reflect different stages of cell activation - at the level of mRNA by quantitative real time PCR (qRT-PCR), by flow cytometry detection of the presence of intracellular cytokines in different cell subpopulations and by ELISA measurement of cytokines in CBMC culture supernatants. The results obtained point to a very weak ability of these common allergens (timothy grass, birch, mite, ovalbumin) to stimulate CBMC to produce cytokines observed by all of these methodological procedures. This diploma thesis was focused on the study of lysozyme and lysozyme-like proteins, either of similar function (antibacterial) or molecular weight (14 - 17 kDa), of synanthropic acaroid mites. In general, animals utilize lysozymes for defensive (antimicrobial) or digestive purposes but also as a digestive enzyme. Some chitinases or other enzymes that act similarly to lysozyme can be utilized for similar purposes. Chitinases belong to house dust mite allergens. One of major mite are historically named lysozyme-like proteins which name relates to their size similar to lysozyme. Bacteriolytic activity has also 14.5 kDa (UniprotKB Q8MWR6) protein. The species selected for the study were domestic mites Dermatophagoides farinae, D. pteronyssinus and Lepidoglyphus destructor. Presence of lysozyme was detected by direct detection with polyclonal antibody using immunohistochemistry and dot blots. Immunohistochemistry proved presence of lysozyme epitopes in the feces of D. farinae, D pteronyssinus a L. destructor. Dot blot analysis demonstrated the presence of imunoreactivity of antibody in spent growth medium extracts (SGME) of all three species. This implies that lysozyme is synthesized in the midgut. Title: Occurrence and Prevention of Asthma in Youth Division Cross-country Skiers Objectives: The aim of this thesis is to determine the incidence of asthma in youth division cross-country skiers and its prevention. Methods: This thesis will comprise the method of survey and content analysis of documents. Hypotheses: No.1 Assume that asthma will have lower prevalence in youth division cross- country skiers than in adult cross-country skiers. No.2 Assume that the majority of the surveyed cross-country skiing coaches met with asthmatic symptoms in their athletes. Results: We found that 12% of youth division athletes suffer from asthma and 24% from allergies. Allergy is one of the most common diseases. Identification of early prognostic markers pointing to an increased risk of allergy development is therefore of increasing importance. Cord blood represents an easily attainable clinical material for searching for prognostic markers signalizing future allergy development. Proportions of Th1 cytokines, Th2 cytokines and regulatory cytokines were tested in cord blood of children of allergic mothers (children in relatively high risk of allergy development) in comparison with cord blood of children of healthy mothers (low risk children). Also the activities of lymphocytes, dendritic cells (DC) and regulatory cells (Tregs) were compared in children of healthy and allergic mothers. The generally increased activity of both in vitro stimulated and non-stimulated mononuclear cord blood leukocytes was proved in children of allergic mothers in comparison with low risk children. The increased activity of DC of high risk children was detectable only after polyclonal stimulation. Significantly less pronounced functional properties of cord blood Tregs were found in children of allergic mothers when compared with children of healthy mothers. This diploma thesis is a loose continuation of the bachelor thesis titled Impact of the Advertising Campaign on the Sales of Products of Pharmaceutical Companies and its Impact on the Consumer. The theoretical part was appropriately expanded to include new findings mentioned in modern literature in order to identify all protective factors that may affect the use or purchase of nasal hygiene products. Attention is paid to the associated processes such as marketing, marketing communications and consumer behavior of the customer. In connection with the subsequent analysis health issues are also mentioned. In the practical part, the success rate of individual advertising campaigns in the years 2007 - 2010 on Nasivin and Sterimar in connection with sales that occur subsequent to influenza, non- influenza diseases and allergic diseases morbidity in the population of the Czech Republic is evaluated. Buying habits of a customer ? a possible Nasivin consumer, what medication he/she uses to treat rhinitis, and what brands of nasal decongestants he/she knows are analyzed. In connection with the product Nasivin and Sterimar, the relationship of a customer and a pharmacist, with regard to the product brand and practices in recommendations, is analyzed. The data were obtained from several sources. The company Merck Serono provided me with information on advertising campaigns and sales results in 2007 - 2010. The market research agency Median provided the data for the target customer analysis. The third data source was the agency STEM / MARK, from which I received background data on consumers' buying habits and practices of pharmacists when recommending products. The fourth source of data was the State Institute of Public Health with information relating to respiratory and allergic diseases. By the carried out analysis a close correlation between the sale of products and the occurrence of respiratory diseases, but not allergic diseases, was proved. The company knows well how to target customers and their needs and it respects the products seasonality. It should focus primarily on acquiring new customers and stop the existing customers´ move to competition. In the discussion and the conclusion the issues of health, the Czech health care system and the prevention of civilization-related diseases are mentioned.
