Annonaceae family, has been a promising source of aporphine alkaloids and derivatives. The
antiprotozoal activity of this class of substances, mainly anti-leishmania activity, has aroused
the interest of several research groups around the world. In the present work were carried out
the analysis from alkaloidal fractions of the leaves, twigs and barks of the trunk from
Unonopsis duckei and Unonopsis guatterioides by mass spectrometry, doing an investigation
in the alkaloid profile in order to obtain promising structures with activity against Leishmania.
The analysis of MSn and LC/APCI/MS of the alkaloidal fractions of U. guatterioides
confirmed the presence of some alkaloids previously reported in the literature such as
anonaine, asimilobine, liriodenine and lysicamine. It was possible identify the nornuciferine
alkaloid which was characterized for the first time in the Unonopsis genus, and suggest the
presence of azafluorenone type alkaloid. The spectrometric investigation of U. duckei
revealed the presence of aporphine and oxoaporphine alkaloids which were already reported
in this genus, as well as guided the isolation of the proaporphine alkaloid glaziovine and the
mixture of glaucine and norglaucine alkaloids. These compounds are inedit in the Unonopsis
genus. The triterpene polycarpol was isolated from both species, observed U. duckei like the
best yield. In the phytochemical investigation of the apolar extracts of U. guatterioides was
possible to identify a mixture of sitosterol/stigmasterol steroids. The biological analyses of U.
duckei and U. guatterioides alkaloidal fractions showed that four of them were considered
highly active, one of them was moderatly active and only one was not showed active against
Leishmania amazonensis. Among the substances evaluated, the alkaloid glaziovine showed
the most effective activity against Leishmania amazonensis. / O gênero neotropical Unonopsis, um dos 130 gêneros pertencentes à família
Annonaceae, tem se mostrado uma fonte promissora de alcalóides aporfínicos e derivados. A
atividade antiprotozoário desta classe de substâncias, principalmente a atividade
antileishmania, tem despertado o interesse de diversos grupos de pesquisa ao redor do mundo.
No presente trabalho foram realizadas as análises das frações alcaloídicas das folhas, galhos e
cascas do tronco de Unonopsis duckei e Unonopsis guatterioides por meio de espectrometria
de massas, investigando seu perfil alcaloídico com vistas à obtenção de estruturas promissoras
em atividade antileishmania. Através das análises de MSn e LC/APCI/MS das frações
alcaloídicas de U. guatterioides foi possível confirmar a presença dos alcalóides anonaina,
asimilobina, liriodenina e lisicamina, já reportados na literatura da espécie, identificar pela
primeira vez no gênero o alcalóide nornuciferina, além de sugerir a presença de um alcalóide
do tipo azafluorenona, previamente reportado na espécie U. lindimanii. A investigação
espectrométrica de U. duckei revelou a existência de alcalóides aporfínicos e oxoaporfínicos
já reportados no gênero, bem como conduziu ao isolamento do alcalóide aporfínico glaziovina
e de uma mistura de glaucina e norglaucina, sendo todos esses alcalóides inéditos no gênero.
Foi realizado o isolamento do triterpeno policarpol de ambas as espécies, sendo o melhor
rendimento observado em U. duckei. A investigação dos constituintes menos polares de U.
guatterioides levou ao isolamento de misturas de sitosterol/estigmasterol. Das frações
alcaloídicas de U. duckei e U. guatterioides analisadas, quatro foram consideradas altamente
ativas, uma ativa e apenas uma não se apresentou ativa frente a Leishmania amazonensis.
Entre as substâncias avaliadas, o alcalóide glaziovina apresentou alta atividade frente a
Leishmania amazonensis.

Bocageopsis pleiosperma Maas, a specie with restricted distribution to the
Amazon Region. The plant material (leaves, twigs and bark) was collected in the
Adolpho Ducke Forest Reserve and submitted to hydrodistillation to obtain the
essential oils and acid-base treatment to getting the alkaloidal fractions. In the
study of the essential oils from leaves, twigs and bark it was identified 51
compounds, being β-bisabolene observed as major compound. The analysis by
n
ESI-IT-MS of alkaloidal fractions in analytical scale showed several ions of m/z
pair. These ions had their fragmentation patterns studied. Also through analysis
n
by ESI-IT-MS it was possible to identified the alkaloidal fraction of the leaves as
a promising source for the isolation of new compounds in the genus Bocageopsis.
From the fractionation alkaloidal fraction of the leaves of B. pleiosperma obtained
in preparative scale, it was isolated and characterized by nuclear magnetic
resonance and unidimensional methods (NMR 1D/2D) and mass spectrometry
(MS), alkaloids of aporphine type, proaporphine, oxoaporphine,
benzyltetrahydroisoquinoline and β-carboline. Among these were isolated
nornuciferine alkaloids, isoboldine, asimilobine and tetrahydroharmine and
obtained as a mixture of two and three compounds, the reticulin and laurotetanine
alkaloids; anonaine and liriodenine; estefarine, isoboldine and N-
methyllaurotetanine. All alkaloids are being described for the first time in the
Bocageopsis genus, being tetrahydroharmine alkaloid described for the first time
in Magnoliales order. The essential oils showed low antimicrobial activity against
C. albicans and P. aeruginosa. / O presente trabalho descreve o estudo fitoquímico e biológico de
Bocageopsis pleiosperma Maas, uma espécie com distribuição restrita à Região
Amazônica. O material botânico (folhas, galhos e cascas) foi coletado na
Reserva Florestal Adolpho Ducke e submetido à hidrodestilação para obtenção
dos óleos essenciais e tratamento ácido-base para obtenção das frações
alcaloídicas. Através do estudo dos óleos essenciais das folhas, galhos e cascas
foi possível identificar 51 compostos, sendo observado β-bisaboleno como
n
composto majoritário. A análise por ESI-IT-MS das frações alcaloídicas em
escala analítica evidenciou diversos íons de m/z par. Estes íons tiveram seus
n
padrões de fragmentação estudados. Através da análise por ESI-IT-MS foi
possível identificar a fração alcaloídica das folhas como promissora para o
isolamento de substâncias inéditas no gênero Bocageopsis. A partir do
fracionamento da fração alcaloídica das folhas de B. pleiosperma, obtida em
escala preparativa, foi possível isolar e caracterizar por métodos de ressonância
magnética nuclear uni e bidimensionais (RMN 1D/2D) e espectrometria de
massas (EM), alcalóides do tipo aporfínico, proaporfínico, oxoaporfínico,
benziltetraisoquinolínico e β-carbolínico. Dentre esses foram isolados os
alcaloides nornuciferina, isoboldina, asimilobina e tetrahidroharmano e obtidos
em mistura de dois e três compostos, os alcaloides reticulina e laurotetanina;
anonaína e liriodenina; estefarina, isoboldina e N-metillaurotetanina. Todos os
alcaloides estão sendo descritos pela primeira vez no gênero Bocageopsis,
sendo o alcaloide tetrahidroharmano descrito pela primeira vez na ordem
Magnoliales. Os óleos essenciais apresentaram baixa atividade antimicrobiana
contra C. albicans and P. aeruginosa.

Considerando o grave quadro das doenças negligenciadas, no Brasil e no mundo, e as limitações do tratamento empregado, na atualidade, torna-se urgente a pesquisa de novos fármacos, que sejam mais ativos e seguros. Para tanto, a busca de moléculas-protótipo, a partir de espécies vegetais, tem sido importante estratégia. Neste contexto, foi realizado o estudo de Annona crassiflora Mart. (Annonaceae), cujos alcalóides totais (AT) demonstraram promissora atividade antiprotozoária in vitro, em estudo anterior. Em paralelo, outras espécies de Annona mostraram atividade antimicobacteriana in vitro, igualmente, tendo motivado o presente estudo. Cinco das vinte frações alcaloídicas obtidas, por cromatografia em coluna, a partir dos AT das folhas, apresentaram atividade anti-Leishmania in vitro, tendo causado 100% de morte das formas promastigotas de Leishmania (L.) infantum chagasi. Além disto, três delas foram ativas frente ao Mycobacterium tuberculosis. O isolamento de dois alcalóides noraporfínicos foi realizado, por fracionamento biomonitorado em coluna cromatográfica, seguido de cromatografia líquida de alta eficiência (CLAE) semipreparativa. As estruturas dos compostos isolados foram elucidadas empregando-se as análises espectroscópicas de ressonância magnética nuclear, mono e bidimensionais, e por CLAE acoplada à espectrometria de massas (CLAEIES-EM2). Um dos alcalóides foi identificado pela primeira vez, nesta espécie, sendo que o outro apresentou estrutura inédita. Ambos demonstraram significativa atividade anti-Leishmania in vitro (CE50≤ 10 µ g/ mL) frente às formas promastigotas de L. (L.) infantum chagasi [MHOM/BR/1972/LD]. O primeiro teve maior índice de seletividade (IS: 7,4), em relação à citotoxicidade em células do tecido conjuntivo NCTC Clone 929 de camundongos. Frente ao Mycobacterium tuberculosis (ATCC 27294) e ao M. smegmatis (ATCC 35798), os alcalóides isolados foram inativos (CIM ≥ 128 µg/ mL). O óleo volátil das folhas foi analisado por cromatografia gasosa acoplada à espectroscopia de massas (CG-EM), tendo sido identificados 41 constituintes, prevalecendo os sesquiterpenos (81,7%) em relação aos monoterpenos (0,8%). Entre os compostos majoritários encontrados no óleo, citam-se os sesquiterpenos α-amorfeno (43,6%), E-cariofileno (17,7%) e o germacreno (5,3%). Nos testes de atividade anti-Leishmania in vitro frente às formas promastigotas de quatro espécies do parasita, o óleo foi mais ativo em L. (L.) infantum chagasi (CE50: 25,97 µg/ mL). Nas formas tripomastigotas do Trypanosoma cruzi mostrou atividade 8,5 vezes superior àquela do fármaco-padrão benznidazol (CE50: 5,31 µg/ mL). Os resultados obtidos ratificaram a importância da prospecção da flora, em particular de A. crassiflora, como fonte potencial de compostos bioativos, que venham a constituir novos fármacos, como alternativa à restrita terapêutica existente para o tratamento das doenças negligenciadas. / Neglected diseases are a serious health problem in Brazil and worldwide. The available drugs are limited in effectiveness with a high toxicity. There is an urgent need of more safe and bioactive compounds. The search of new molecules from plant species is a well known and important strategy to achieve this goal. In a previous work, Annona crassiflora Mart. (Annonaceae) showed a promising antiprotozoal activity. Beside this, other Annona species presented an interesting antimicobacterial action. In this bio-guided study, after the column fractionation of the leaves total alkaloids, in vitro tests were performed and five from twenty fractions were highly active (100% deaths) against promastigotes of Leishmania (L.) infantum chagasi, and only three were active against Mycobacterium tuberculosis. After purification of the bioactive fractions, two noraporphine alkaloids were isolated by HPLC and identified by the usual mono and bidimensional spectroscopic techniques. One of them was isolated from the first time from this species. The other one is a novel chemical entity. Both compounds presented anti- Leishmania activity (CE50 ≤ 10 µg/ mL) against L. (L.) infantum chagasi [MHOM/BR/1972/LD]. The first one showed a higher selectivity index (SI: 7.4) considering its mice connective tissue cells toxicity [NCTC Clone 929]. However, both were inactive against Mycobacterium tuberculosis (ATCC 27294) and M. smegmatis (ATCC 35798) (CIM ≥ 128 µg/ mL). In the leaves volatile oil 41 compounds were identified. The sesquiterpenes were in majority (81.7%), followed by monoterpenes (0.8%). The sesquiterpenes α-amorphene (43.6%), E-caryophyllene (17.7%) and germacrene (5.3%) were the main constituents. The oil was little effective against the four tested Leishmania species and slightly more active against L. (L.) infantum chagasi (CE50: 25.97 µg/ mL). However, it was highly active against the trypomastigotes of Trypanosoma cruzi (CE50: 5.31 µg/ mL) showing to be 8.5 times more active than benznidazol. These results stimulate a deeper investigation of those alkaloids as antiprotozoal agents, confirming the importance of the plant species metabolites as a source of new bioactive molecules and their potential as future drugs.
