Coffea arabica e Coffea canephora são responsáveis por 70% e 30% da produção comercial, respectivamente. Análise citogenética indicou que C. arabica é uma planta alotetraploide autógama formada por uma hibridação (1 milhão de anos atrás) entre os diplóides C. canephora e Coffea eugenioides. C. eugenioides é uma espécie silvestre que cresce em maiores altitudes próximo das bordas de florestas e produz poucas e pequenas sementes com baixo teor de cafeína. Por outro lado, C. canephora é alógama e cresce melhor em terras baixas, é também caracterizada por maior produtividade, maior tolerância a pragas e maior teor de cafeína, mas tem uma bebida considerada de qualidade inferior em comparação com a produzida por C. arabica. Durante a última década, algumas iniciativas de pesquisa têm sido lançadas para produzir dados genômicos e transcritômicos de algumas espécies de café. Esta coleção de ESTs representa uma boa visão do transcriptoma de C. arabica e C. canephora, sendo um importante recurso para análise molecular dessas espécies. Este trabalho teve como objetivo obter mais informações sobre algumas espécies do gênero Coffea, incluindo a estrutura dos genes, análise de expressão e identificação de genes e famílias gênicas que são específicos ou expandidos em café. Além disso, também foi proposto estudar a regulação da expressão gênica nos genes homeólogos da alotetraploide C. arabica. A fim de investigar estes conjuntos de dados de EST foram realizadas duas montagens: (i) a primeira montagem com cada espécie individualmente, com o objetivo de fazer uma análise comparativa entre as C. arabica, C. canephora e outras culturas, e (ii) com as duas espécies de café juntas, permitindo a identificação de SNPs entre C. arabica e C. canephora, e avaliar questões evolutivas em C. arabica. A identificação dos transcritos diferencialmente expressos e novas famílias gênicas foram utilizados como ponto de partida para a correlação de características de desenvolvimento e de perfis de expressão gênica em Coffea sp.. Domínios de proteínas e análises de Gene Ontology sugerem diferenças significativas entre os dados das espécies de café analisadas, principalmente em relação a síntese de açúcares, ligação de proteínas a nucleotídeos, retrotransposons e proteínas de resposta a estresse. A ferramenta OrthoMCL identificou as famílias de proteínas específicas ou predominante de café quando comparado com outras cinco espécies de plantas. Usando as discrepâncias de alta qualidade encontradas em ESTs sobrepostos de C. arabica e C. canephora, os perfis de diversidade de seqüência foram avaliados em ambas as espécies e utilizados para deduzir a contribuição de C. canephora e C. eugenioides na transcrição de C. arabica. A identificação de genes homeologous de C. arabica aos genomas ancestrais permitiu analisar as contribuições de expressão gênica de cada subgenoma. Nós sugerimos que este fenômeno tem uma questão importante na expressão dos genes e fisiologia de Coffea. / Abstract: Coffee is one of the most important crops in the world, being worldwide consumed and having significant participation in under development economies. Coffea arabica and Coffea canephora are responsible for 70% and 30% of commercial production, respectively. Cytogenetic analysis established that C. arabica is an autogamous alotetraploid formed by a recent (1 mya) hybridization between the diploids C. canephora and Coffea eugenioides. C. eugenioides is a wild species which grows in higher altitudes near forest edges, and produces few berries with small beans of low caffeine content. On the other hand, C. canephora is alogamous and grows better in lowlands. It is also characterized by higher productivity, more tolerance to pests, and higher caffeine content, but it has an inferior beverage compared with C. arabica. During the last decade, research initiatives have been launched to produce genomic and transcriptomic data about Coffea spp. This EST collection represents a good overview of C. arabica and C. canephora transcriptome, being appropriate as a resource for Coffea molecular analysis. This work aimed to obtain further information about Coffea spp. gene structure and expression and to identify genes that are specific or expanded in coffee plants. Moreover, it also intended to study the homeologous gene expression regulation in the alotetraploid C. arabica. In order to investigate these data two different EST assemblies were performed: (i) with each species individually, aiming the comparative analysis between the C. arabica, C. canephora and other crops; and (ii) with both coffee species together, allowing the identification of SNPs between C. arabica and one of its direct ancestors C. canephora and the examination of evolutive issues in C. arabica. The identification of differentially expressed transcrip ts and new gene families offered a starting point for the correlation of gene expression profil es and Coffea sp. development traits. Protein domain and Gene Ontology analyzes suggested significant differences between the data of coffee species analyzed, mainly in relation to complex sugar synthases, nucleotide binding proteins, retrotransposons and stress response. OrthoMCL tool identified specific or prevalent coffee protein families when compared with other five plant species. Using the high quality discrepancies, found in overlapped ESTs from C. arabica and C. canephora, sequence diversity profiles were evaluated within both species and used to deduce the transcript contribution of the C. canephora and C. eugenioides ancestors in the C. arabica. The assignment of the C. arabica homeologous genes to the ancestral genomes allowed us to analyze gene expression contributions of each subgenome. As metodologias utilizadas possibilitaram a obtenção do cariograma das três espécies com cromossomos definidos e resolução adequada para caracterização de cada par de homólogos, aparentemente semelhantes quando obtidos por meio de técnicas convencionais. A análise citogenética revelou que C. canephora possui um par de cromossomos metacêntricos (m) e 10 pares submetacêntricos (sm). Foi observada a presença de satélites no braço curto do cromossomo 6, e o comprimento absoluto de seus cromossomos variou de 1,58 a 3,30 μm. A citometria de fluxo foi usada para estimar a quantidade de DNA (valor 2C), na fase G0/G1, em Coffea. Foram analisadas amostras das espécies de C. canephora (2x), C. eugenioides (2x), C. racemosa (2x) e C. arabica (4x); do Híbrido de Timor, híbrido natural triplóide entre C. arabica e C. canephora (4x); dos supostos híbridos interespecíficos naturais entre C. racemosa e C. arabica, denominados UFV 557; e de plantas resultantes do retrocruzamento entre C. arabica e UFV 557. A diferença na quantidade do conteúdo de DNA entre espécies diplóides foi de até 0,50 pg = 45% (Coffea racemosa 1,11 pg; Coffea eugenioides 1,40 pg e Coffea canephora cv. ‘Robusta’ 1,61 pg). Considerando-se os valores intraespecíficos (Coffea canephora 1,43 a 1,61 pg e Coffea arabica 2,40 a 3,04 pg), estas diferenças foram de 0,18 pg (12,59%) e 0,64 pg (26,67%), respectivamente. A citometria de fluxo também revelou-se uma ferramenta importante no monitoramento e identificação de híbridos interespecíficos de Coffea. A citométria de imagem foi empregada para determinação do índice de densidade óptica de preparações de pontas de raízes esmagadas e coradas com reação de Feulgen. O método prófase/telófase (mensuramento de 10 núcleos em prófase e 10 em telófase por lâmina) foi empregado nesta pesquisa. Assim, valores 4C e 2C, referentes ao núcleos em prófase e telófase, foram estimados, e as razões entre os genomas de Coffea arabica cv. Catuaí Vermelho (2n=44) e Coffea canephora cv. Conillon (2n=22) e entre Coffea arabica cv. Mundo Novo e cv. Catuaí Vermelho puderam ser estabelecidas. As estimativas do índice entre as cultivares obtidas pelo método de citometria de imagem foram comparadas com valores obtidos previamente usando a citometria de fluxo e apresentaram diferenças inferiores a 2,27%. / New cytogenetic techniques together with methods of computer image analyses were utilized to set up the karyogram of C. canephora (2n=2x=22), C. arabica (2n=4x=44), and C. eugenioides (2n=2x=22). Thereby, morphologically appropriate chromosomes were obtained for cytogenetic characterization and identification. These methodologies brought forth a karyogram of the three species with defined chromosomes and a suitable resolution for the characterization of each homologous pair, which, when obtained by conventional techniques, are apparently similar. The cytogenetic analysis indicated that C. canephora owns one metacentric (m) and 10 submetacentric chromosome pairs (sm). Satellites were localized on the short arm of chromosome 6, while the absolute chromosomes length varied between 1,58 and 3,30 μm. Flux cytometry was used to estimate the quantity of the DNA (value 2C) in the G0/G1 phase, for Coffea. Samples of the following species were analyzed: C. canephora (2x), C. eugenioides (2x), C. racemosa (2x), and C. arabica (4x); the Timor Hybrid, natural triploid hybrid of C. arabica and C. canephora (4x); the suspected interspecific natural hybrids of C. racemosa and C. arabica, designated UFV 557; and the backcrosses of C. arabica and UFV 557. The difference in the content quantity of the DNA among the diploid species reached 0,50 pg = 45% (Coffea racemosa 1,11 pg, Coffea eugenioides 1,40 pg, and Coffea canephora cv. ‘Robusta’ 1,61 pg). These differences were 0,18 pg (12,59%) and 0,64 pg (26,67%), respectively, in the intra-species values (Coffea canephora 1,43 to 1,61pg and Coffea arabica 2,40 to 3,04 pg). Flux cytometry proved to be of great usefulness to monitor and identify the interspecific hybrids of Coffea. Picture cytometry was employed in the determination of the optical density index in slide preparations of root tips pressed and dyed by the Feulgen reaction. The prophase/telophase method (measurement of 10 nuclei in prophase and 10 in telophase per slide) was used in this study. The values 4C and 2C of the nuclei in prophase and telophase were estimated, and the genome ratios of Coffea arabica cv. Catuaí Vermelho (2n=44) and Coffea canephora cv. Conillon (2n=22), as well as of Coffea arabica cv. Mundo Novo and cv. Catuaí Vermelho could be established. In a comparison between the estimate indices of the cultivars obtained by the picture cytometry method and the values established previously by flux cytometry, the differences lay below 2,27%. / Tese importada do Alexandria

A técnica RAPD mostrou-se adequada para a estimação de distâncias genéticas (complemento de Jaccard) entre acessos de Coffea, obtendo-se nível de reprodutibilidade de 76,88%. O número de locos não influenciou a formação dos grupos principais, mas influenciou a ordenação dos acessos dentro dos subgrupos. Para a caracterização molecular de 18 clones diferenciadores de raças de Hemileia vastatrix foram utilizados 35 primers da Operon Technologies, Inc. Os 35 primers identificaram 158 locos polimórficos. O agrupamento, utilizando o método UPGMA, foi realizado com base na matriz de valores de dissimilaridades (complemento de Jaccard), e os grupos formados foram compatíveis com as informações disponíveis na literatura sobre a origem genealógica. A técnica RAPD foi eficaz na caracterização dos clones diferenciadores, produzindo marcas específicas para cada clone. O mapa parcial de ligação gênica para Coffea arabica L. foi construído a partir de uma população segregante RC 1 obtida do cruzamento entre Mundo Novo (IAC 464-18) e Híbrido de Timor (CIFC 2570), sendo este último utilizado como genitor recorrente. Foram obtidos 93 marcadores RAPD, dos quais 87 (93,5%) apresentaram segregação 1:1 (P>0,01), quatro (4,4%) segregação 2:1 (P>0,05) e dois (2,2%) segregação 5:1 (P>0,05). Para construção do mapa, foram utilizados os 87 marcadores que segregaram 1:1, sendo que cinco não mostraram -se ligados aos grupos formados. Oitenta e dois marcadores RAPD resultaram em oito grupos de ligação cobrindo 540,6 cM. O s grupos obtidos tiveram uma boa densidade de marcadores, exceto dois grupos. O maior intervalo entre dois marcadores foi 36.4 cM, e 94,5% dos intervalos não excederam a 20 cM. O tamanho dos grupos de ligação apresentou alta correlação com o número de marcadores (r=0,887), indicando distribuição aleatória dos marcadores nos grupos. O número de grupos de ligação é inferior ao correspondente número haplóide de cromossomos (22) e, portanto, o genoma da espécie foi parcialmente explorado e muitas regiões ainda não foram identificadas. / Fifty two arbitrary primers were used to evaluate the reproductibility and the influence of the marker numbers RAPD (Random Amplified Polymorphic DNA) in estimating the genetic distances among 40 accesses of the Coffea genus from the germplasm bank of UFV/EPAMIG (including 4 different species). The RAPD technique was shown to be appropriate for estimating the genetic distances (Jaccard complement) among Coffea accesses, as a reproductibility level of 76.88% is obtained. The loci numbers did not influence the formation of the main groups, however it influenced the ordering of the accesses within subgroups. For molecular characterization of 18 coffee differentials to identify Hemileia vastatrix races , 35 primers from Operon Technologies, Inc were used. The 35 primers identified 158 polymorphic loci. The grouping by the UPGMA method was accomplished based on dissimilarity values matrix (Jaccard complement), and the formed groups were compatible to the information available in literature on genealogical origin. The RAPD technique was effective in characterization of the coffee differentials, so producing specific marks for each clone. The partial genetic linkage map for Coffea arabica L. was built from a segregative RC 1 population obtained from crossbreeding between Mundo Novo (IAC 464-18) and Timor Hybrid (CIFC 2570), with the latter used as recurrent genitor. The following were obtained: 93 RAPD markers, from which 87 (93.5%) showed segregation 1:1 (P>0.01), four (4.4%) segregation 2:1 (P>0.05), and two (2.2%) segregation 5:1 (P>0.05). For map construction those 87 markers segregating 1:1 were used, and five showed no linkage to the formed groups. Eighty two RAPD markers resulted into eight linkage groups covering 540.6 cM. The obtained groups showed to have a satisfactory marker density, exception for two groups. The wider range between two markers was 36.4 cM, and 94.5% of the ranges did not exceed to 20 cM. The size of the linkage groups highy correlated to the number of markers (r=0.887), indicating random distribution of the markers within groups. The number of the linkage groups is lower to the corresponding haploid number of chromosomes (22), therefore the species genoma was partially explored, and many areas were not still identified. / Tese importada do Alexandria

Nesse sentido, a água residuária oriunda do processamento dos frutos do café (ARC) pode ser utilizada na agricultura como fertilizante natural, respeitando a concentração dos nutrientes nela contidos. Este trabalho teve como objetivo utilizar dois solos como meio de tratamento da ARC, possibilitando a remoção de material orgânico e inorgânico presentes nesses efluentes. Para isso, fez-se ensaio de colunas de solos, sendo aplicados a eles a ARC. A ARC foi coletada no período de julho e agosto e as amostras de solos foram retiradas em profundidade de 0 a 40 cm, em dois solos classificados como Latossolo Vermelho Amarelo, apresentando texturas diferentes, sendo o solo um de textura média e o solo dois de textura arenosa. Foram coletadas amostras do lixiviado da ARC e foram feitas determinações de pH, condutividade elétrica (CE), demanda química de oxigênio (DQO), P, Ca, Mg e K. Após a percolação da ARC em colunas de solos, as mesmas foram encaminhadas ao laboratório a fim de proceder as determinações de P, K, Ca, Mg, pH, matéria orgânica do solo (MOS), soma de bases, capacidade de troca de cátions (CTC) em pH 7, CTC efetiva e índice de saturação em bases. Foi possível concluir que a ARC contribuiu para a alteração das concentrações de pH e Mg, aumento da CTC em pH 7 e da concentração de K, diminuição do índice de saturação em bases e da concentração de Ca, sendo que as concentrações de P, MOS, CTC efetiva e soma de bases não foram significativamente alteradas pela adição da ARC. Para o solo de textura arenosa, a ARC contribuiu para a alteração do pH e da concentração de K, diminuição da concentração de Ca, MOS, soma de bases e índice de saturação em bases, aumento da CTC em pH 7, sendo que as concentrações de P, Mg e CTC efetiva não foram alteradas significativamente pela adição da ARC. Com relação às águas de drenagem decorrentes da aplicação da ARC em colunas de solo de textura média, pode-se concluir que houve aumento do pH, CE e da concentração de Ca, diminuição da DQO, da concentração de Mg e P, sendo que a concentração de K não foi significativamente alterada. Quanto às águas do lixiviado da ARC nas colunas de solo de textura arenosa, pode-se inferir que houve aumento do pH, da concentração de Ca e Mg, diminuição da CE, DQO e em alguns momentos da concentração de P, sendo que a concentração de K não foi significativamente alterada / Reuse of crop residues in agriculture is a promising alternative, which allows sustain agroenvironment providing improvements to the soil-plant system. This sense, the wastewater coming from the processing of coffee fruits (ARC) can be used as natural fertilizer in agriculture, respecting the concentration of nutrients it contains. This study had as objective use two soils as a means of treatment of ARC, allowing the removal of organic and inorganic material present in these effluents. For this, made to test soil columns, being applied to them the ARC. The ARC was collected during july and august and soil samples were removed in depth from 0 to 40 cm in two soils classified as Yellow Red Latosol, with different textures, being one soil of medium texture and two soil of arenaceous texture. Samples were collected from the leachate of the ARC and were made the measurements of pH, electrical conductivity (EC), chemical oxygen demand (COD), P, Ca, Mg and K. After percolation of the ARC in soil columns, they were directed to the laboratory to proceed the determinations of P, K, Ca, Mg, pH, soil organic matter (SOM), total bases, cation exchange capacity (CEC) at pH 7, effective CEC and base saturation index. It was concluded that the ARC contributed to the change in the levels of pH and Mg, CEC at pH 7 and concentration of K, reduced the base saturation index and Ca concentration, and the concentration of P, SOM, effective CEC and total bases was not significantly altered by the addition of the ARC. For arenaceous soil, the ARC has contributed to the change of pH and concentration of K, reduction in the Ca concentration, SOM, total of bases and base saturation index, increased CEC at pH 7, and the concentrations P, Mg and effective CEC were not significantly altered by the addition of the ARC. With respect the drainage water from the application of ARC in columns of the medium textured soil can conclude that increasing pH, EC and Ca concentration, reduced the COD, the concentration of Mg and P, and the concentration of K was not significantly changed. As the waters of the leachate from the ARC in the columns of arenaceous soil can bein ferred that there was an increase of pH, Ca and Mg concentration, reduced of the EC, COD and at times the P concentration, and the concentration of K was not significantly altered

Influenciando assim a produ??o de metabolitos secund?rios, inclusive os relacionados a forma??o das estruturas anat?micas, metabolitos de defesa e fotoss?ntese. Objetivou-se no presente trabalho avaliar os efeitos da deriva simulada de glyphosate sobre a anatomia e fisiologia de cultivares de caf? arabica. Para isso, fez-se tr?s avalia??es diferenciadas visando avaliar modifica??es primeiramente morfoanat?micas, depois concentra??es de fen?is totais, flavon?ides, cafe?na e por ultimo, floresc?ncia, concentra??es de clorofilas e quantifica??es de est?matos. O experimento foi instalado em esquema fatorial (3 x 5), com tr?s cultivares de caf? MGS Travessia, Oeiras MG 6851 e Catua? IAC 144, e cinco doses de glyphosate (0,0; 57,6; 115,2; 230,4 e 460,8 g ha-1), em delineamento de blocos casualizados, com quatro repeti??es. Trinta dias ap?s a aplica??o, foram coletadas doze folhas rec?m-expandidas do ?ltimo ramo plagiotr?pico do cafeeiro, quatro folhas para as analises anat?micas, quatro para as analises de concentra??es de fen?is, flavonoides, cafe?na e as quatro ultimas para a quantifica??o estom?tica. Nas analises morfoanat?micas observou-se os seguintes resultados: ocorreram sintomas de intoxica??o como estreitamento foliar e clorose nas folhas mais jovens das plantas. Com o aumento da dose de glyphosate, a cultivar Catua? sofreu redu??o na espessura foliar total, enquanto, que para as demais ocorreu incremento nesta caracter?stica. Para as vari?veis epiderme adaxial, epiderme abaxial e par?nquima lacunoso, ocorreu decr?scimo de espessura, contudo ocorreu aumento da espessura do par?nquima lacunoso com aumento das doses de glyphosate. Pode-se concluir que as tr?s cultivares quando submetidas ? deriva de glyphosate sofrem modifica??es morfoanat?micas. Com a redu??o do par?nquima pali??dico ocorre redu??o da taxa fotossint?tica e consequente redu??o do crescimento e produ??o do caf?. Para as analises de concentra??es de fen?is, flavon?ides e cafe?na, observou-se os seguintes resultados: com aumento das doses de glyphosate houve aumento na concentra??o de fen?is totais foliares at? a dose de 115,2 g ha-1 de glyphosate, para as tr?s cultivares, acima dessa dose as concentra??es de fen?is foram reduzidas. No entanto, com aumento das doses de glyphosate a concentra??o de flavon?ides diminuiu at? a dose de 115,2 g ha-1 de glyphosate, para as cultivares Travessia e Oeiras, acima dessa dose a concentra??o de flavon?ides para essas duas cultivares aumentaram. Ocorreu aumento da concentra??o de flavon?ides para a cultivar Catua? quando aplicado a dose de 115,2 g ha-1 de glyphosate, para doses mais altas ocorreu diminui??o da concentra??o de flavon?ides totais. Com aumento das doses de glyphosate, as concentra??es de cafe?na apresentaram comportamento similar entre as cultivares Oeiras e Catua?, ocorrendo diminui??o da concentra??o de cafe?na at? a dose de 115,2 g ha-1 de glyphosate e aumento a concentra??o de cafe?na para doses superiores. Observou-se que houve aumento na concentra??o de cafe?na para cultivar Travessia a partir de 115,2 g ha-1 de glyphosate. De acordo com o teste histoqu?mico todas as tr?s cultivares estudadas apresentaram maiores concentra??o de fen?is no par?nquima pali??dico ap?s a aplica??o da deriva de glyphosate. Conclui-se que, quando submetidas a subdoses de glyphosate, as cultivares analisadas apresentaram modifica??es bioqu?micas para as concentra??es de fen?is totais, flavon?ides totais e cafe?na. Com baixas doses 57,6 e 115,2 g/ ha?? de glyphosate, ocorreu aumento da concentra??o de fen?is totais aumentando assim a capacidade de defesa das cultivares, entretanto com o aumento das doses de glyphosate ocorreu redu??o da concentra??o de fen?is totais. A produ??o de flavon?ides e cafe?na possuem efeitos variados para as doses de glyphosate podendo aumentar e diminuir a concentra??o de forma independente, pois os metabolitos s?o supridos por outras rotas. Nas analises fisiol?gicas, observou-se os seguintes resultados: os valores da vari?vel fluoresc?ncia m?xima (Fm) decresceram de acordo com o aumento das doses de glyphosate, a cultivar Travessia apresentou maior decr?scimo que as demais cultivares. Para efici?ncia fotoqu?mica m?xima (Fv/Fm) ocorreu com o aumento do estresse de acordoo com o aumento das doses aplicadas do glyphosate. As concentra??es de clorofila a, b e total sofreram decr?scimo de acordo com o aumento das doses aplicadas de glyphosate. Para as vari?veis, ?ndice estom?tico e densidade estom?tica ocorreram um decr?scimo de valores de acordo com o aumento da dose aplicada de glyphosate. Glyphosate is the most used herbicide in the world for being of high efficiency and cheaper. Its action is on the enzyme 5enolpyruvylshikimate that is responsible for producing tyrosin, phenylalanine and tryptophan. It inhibits their production, consequently influencing the production of secondary metabolites, mainly the ones related to anatomical structures, metabolites of defense and photosynthesis. This study was carried out in order to evaluate effects of drift simulation of glyphosate on anatomy and physiology of Arabica coffee cultivars. There were three different stages of evaluations: first morphoanatomical modifications, then concentrations of total phenol, flavonoids and cafeine, and finally fluorescence, chlorophyll concentrations and stomata quantification. The experiment was arranged in a factorial scheme (3 x 5), with three coffee cultivars (MGS Travessia, Oeiras MG 6851 and Catua? IAC 144), at five glyphosate rates (0.0; 57.6; 115.2; 230.4 and 460.8 g ha-1), in a randomized block design, with four replicates. Thirty days after application, twelve recently expanded leaves from the last plagiotropic branches were collected: four for anatomical analysis, four for analysis of concentrations of total phenol, flavonoids and cafeine and four to quantify stomata. Morphoanatomical analysis showed intoxication symptoms such as foliar narrowing and chlorosis on the younger leaves were observed. Increasing glyphosate rate, the cultivar Catua? reduced its total foliar tickness, while to the others it was increased. Adaxial and abaxial epiderms and spongy parenchyma had their thickness decreased, however spongy parenchyma increased at higher rates of glyphosate. It could be concluded that the three cultivars under glyphosate drift shows morphoanatomical modifications. When palisade parenchyma is reduced, consequently photosynthetic rate, growth and coffee production reduce. Analyzing concentrations of total phenol, flavonoids and caffeine, it was observed that phenol concentration increased when glyphosate rates were increased up to 115.2 g ha-1 in all three cultivars, showing reduction at higher rates. For Travessia and Oeiras cultivars, the flavonoid concentration decreased at higher glyphosate rates, starting its increase at rates higher than 115.2 g ha-1. For Catua?, flavonoid concentration increased at 115.2 g ha-1 being reduced with higher rates. Regarding caffeine concentration, it was similar to Oeiras and Catua?. This concentration decreased at 115.2 g ha-1 and got higher at superior rates. The cultivar Travessia showed higher caffeine concentration at rates higher than 115.2 g ha-1. According to the histochemical tests, all the three cultivars present higher phenol concentration on paladise parenchyma after application of glyphosate drift. It can be conclude that, when submitted to subrates of glyphosate, cultivars shows biochemical modifications to all concentrations evaluated. At 57.6 and 115.2 g ha-1 total phenol increased providing a better capacity of defense to cultivars, however, when increasing these rates, total phenol concentration decreased. Flavonoids and caffeine production have varied effects on glyphosate rates. They can increase or decrease independently because metabolites are fulfilled by other ways. Through physiological analysis, it could be observed that values of maximum fluorescence decreased with higher glyphosate rates, and Travessia cultivar presented highest decrease. Chlorophyll concentrations a, b and total decreased according to rates increase, being harder to chlorophyll a. Values of stomatal index and stomatal density decreased when increasing glyphosate rate, being harder to cultivar Travessia. It can be concluded that subrates of glyphosate cause damage on photosynthesis because of the reduction on chlorophyll concentration and fluorescence intensity. Reduction of stomata number is also one of the features that can explain the influence promoted by glyphosate on photosynthesis.

For this, coffee plants were grown in hydroponic solution for 50 days after application (DAA) of the herbicide. The study was conducted in a randomized block design (RBD) with a 2x4 factorial design with the first factor related to the route of foliar and root absorption and the second to Assessmentmethodology effect of glyphosate doses used: 0, 115.2, 230.4; and 460.8 g ha-1 corresponding to 0, 8, 16 and 32% of the dose of 1440 g ha-1 isopropylamine salt formulation with 7 replicates. Symptoms of poisoning were characterized by chlorosis and narrowing of the leaf blade 50 and reached 70% for foliar and root uptake, respectively. With increasing rates of the herbicide to 460.8 g ha-1 occurred reductions in stem length, leaf number, leaf area, the leaf mass ratio, the root shoot ratio, the length of roots and masses dried leaves, stem, roots and all. The number of leaves, leaf area, dry weight of root, stem, leaf, every and leaf weight ratio were lower when the herbicide was absorbed by the leaves. Absorption pathways and reduced rates of glyphosate did not alter the behavior of K in the plant, but impaired performance of N, P, Ca and Mg, with all the effects more pronounced when the herbicide was absorbed by the air in relation to root uptake. Conclude that the process of absorption and reduced rates of glyphosate not interfere with the performance of the K young coffee in hydroponic conditions, but compromise the behavior of N, P, Ca, Mg, reducing the growth of these plants. The uptake or translocation of glyphosate and is most effective when absorbed through the leaves.

No entanto, em muitos locais, o café é cultivado a pleno sol, com produções satisfatórias. O objetivo deste trabalho foi, pois, identificar potenciais mecanismos de fotoproteção, bem como avaliar como eles se ajustam diurna e sazonalmente. As plantas foram cultivadas no campo, sob dois níveis de irradiância (50 e 100% da luz natural incidente), analisando-se folhas das faces do renque completamente expostas à luz, em três épocas contrastantes: agosto e dezembro de 2003 e outubro de 2004. A fotossíntese líquida e a condutância estomática apresentaram valores baixos e similares nas plantas de ambos regimes de luz em todas as épocas, especialmente à tarde. Tendência semelhante foi observada para a atividade da carboxilase/oxigenase da ribulose-1,5-bisfosfato, amostrada ao meio-dia. Observou-se fotoinibição crônica em agosto e uma discreta fotoinibição dinâmica em dezembro e em outubro nas plantas de ambos tratamentos. As concentrações de clorofilas e carotenóides totais foram menores em agosto nas plantas de ambos regimes de luz. O ângulo foliar foi sempre maior nas plantas a pleno sol em relação às sob sombra. Na maioria das amostragens, a taxa de transporte de elétrons foi semelhante em plantas de ambos tratamentos. Em laboratório, observou-se, na medida em que incrementava a irradiância, redução na fração de energia utilizada na fase fotoquímica da fotossíntese e aumentos concomitantes e consistentes na fração dissipada na forma de calor e na fração da energia não utilizada na fase fotoquímica e tampouco dissipada como calor. Isto foi observado até a irradiância actínica de 1500 μmol(fótons) m - 2 -1 s sem, contudo, verificar-se um padrão claro entre plantas a pleno sol e sob sombra. Não se observaram diferenças entre essas plantas acerca das atividades das enzimas antioxidantes: dismutase do superóxido (SOD), catalase (CAT), peroxidase do ascorbato (APX) e redutase da glutationa (GR). Comparativamente, maior atividade da SOD e da APX ocorreram em outubro, da CAT, em outubro/dezembro, enquanto a da GR foi similar entre as épocas avaliadas. Verificou-se tendência de maiores níveis de ascorbato nas plantas a pleno sol que naquelas sob sombra, mas apenas em agosto tais diferenças foram significativas. De modo geral, danos celulares de pequena magnitude, caracterizados por aumentos discretos no extravasamento de eletrólitos e acúmulo de aldeído malônico, foram mais evidentes em agosto e em outubro em relação a dezembro, embora similar entre plantas de ambos tratamentos. Em suma, nas condições deste experimento, o cafeeiro apresentou alta plasticidade fisiológica de seu aparelho fotossintético às variações da irradiância. / Coffee is native to shady environments, a fact that has been associated with a general belief that it is a shade species. However, in many places, coffee grows well without shade and even out-yields shaded coffee. The aim of this work was to identify potential mechanisms of photoprotection as well as to evaluate how they are able to adjust diurnally and seasonally. The plants were grown in the field under two irradiance levels (50 and 100% of incident natural light). Sampling and measurements were made using outer leaves from the sun-faced sides of the coffee hedgerow in three contrasting times: August and December 2003 and October 2004. Regardless of the treatments, net photosynthesis and stomatal conductance were very low, particularly in the afternoon. A similar trend was observed for the activity of ribulose-1,5-bisphosphate carboxylase/oxygenase, as analyzed at midday. Chronic photoinhibition was found in August, whereas a slight, dynamic photoinhibition was observed in December and October, irrespective of light treatments. Concentrations of chlorophylls and total carotenoids were smaller in August than in December or October, regardless of light conditions. Leaf angle was always steeper in plants under full sunlight in relation to those under shade. On most occasions, electron transport rate was similar in plants under both irradiance treatments. Under laboratory conditions, there was a reduction in the fraction of absorbed light used in photochemistry which was accompanied by increases in the fraction of absorbed light dissipated as heat as well as in that fraction neither used in photochemistry nor dissipated thermally. This was observed until applying an actinic irradiance of 1500 μmol (photons) m -2 s -1 , but no clear difference between sun leaves and shaded ones was noted, however. Activities of key antioxidant enzymes, superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), and glutathione reductase (GR), were similar between plants from both light treatments. Comparatively, larger activity of SOD and APX occurred in October, that of CAT in October/December, while GR activity was similar among the evaluated periods. A trend for higher reduced ascorbate pool was observed in plants grown in the open than in those grown under shade, but only in August such differences reach statistical significance. In a general way, discrete cellular damages, characterized by slight increases both in electrolyte leakage and malondialdehyde accumulation, were more evident in August and in October as compared to December, but such damages were similar between plants from both irradiance treatments. Summing up, the photosynthetic apparatus of the coffee tree, under the present experimental conditions, showed a high physiological plasticity to varying irradiance. / Dissertação importada do Alexandria

Isolamentos de Colletotrichum sp. foram feitos a partir de material coletado em quatro estados: Minas Gerais, São Paulo, Espírito Santo e Paraná. A partir dos isolamentos foram obtidas 180 culturas monospóricas, mantidas em sílica-gel, visando à preservação. Entre as 180 culturas inoculadas, em ramos com três rosetas de frutos verdes, houve diferença na incidência observada, tanto nos ramos com ferimento como naqueles sem ferimento, dez dias após a inoculação. A incidência máxima obtida foi de 69,5%, entretanto a testemunha, atomizada com água estéril, também manifestou sintomas e abundante esporulação de Colletotrichum gloeosporioides, resultado novamente observado quando da repetição do experimento. Isso gerou dúvidas sobre a eficiência da sanitização dos ramos e frutos verdes coletados no campo. As amostras passaram por um processo de esterilização superficial, utilizado nas etapas para isolamento de organismos endofíticos. Pôde-se observar que, mesmo superficialmente estéreis, os tecidos apresentavam abundante esporulação de C. gloeosporioides após seis dias de incubação em condições assépticas. Testes de patogenicidade foram então realizados, com e sem ferimento, utilizando-se três isolados, os quais foram inoculados em plantas oriundas de cultura de tecido, com três meses de idade, não sendo observados sintomas por mais de 20 dias após a inoculação. Porém, conseguiu-se recuperar o fungo das plantas inoculadas, mesmo depois de lavadas em água corrente, álcool 70% e cloro ativo 2%. Isso mostra que o organismo penetrou nos tecidos, entretanto sem manifestar sintomas. A partir desses resultados, concluiu-se que testes de patogenicidade de C. gloeosporioides devem ser conduzidos em plantas de cultura de tecido, a fim de se assegurar a sanidade do material a ser inoculado, visto que, mesmo após esterilização superficial do tecido, o organismo pode permanecer latente. Neste trabalho, obtiveram- se indícios de que C. gloeosporioides apresenta relação endofítica nos tecidos de cafeeiro (Coffea arabica). / This work had the objective to establish the pathogenicity of Colletotrichum sp to Coffea arabica. Isolations of Colletotrichum sp. were done from material collected in the field in four States: Minas Gerais, São Paulo, Espírito Santo and Paraná. It was obtained 180 monoconidial cultures and maintained in silica-gel, for preservation. Among the 180 inoculated cultures, in branches with 3 rosettes of green fruits, there was difference in the incidence in the wounded and not wounded branches 10 days after the inoculation. The maximum incidence was of 69.5%, however check treatment atomized with sterile water, also manifested symptoms and abundant sporulation of C. gloeosporioides, the same results was observed again when the experiment was repeated. This generated doubts on the efficiency of the sanitization of the branches and green fruits collected in the field. The tissue samples went by a superficial sterilized process, used in the stages to isolate endofitic organisms. It was observed that even superficially sterile, the tissues presented abundant sporulation of C. gloeosporioides after six days of incubation in aseptic conditions. Pathogenicity tests were then accomplished, with and without wound, using three isolates in three months old coffee seedlings of tissue culture. No symptoms observed for more than 20 days after the inoculation. However, the fungus was recovered of the inoculated plants, even after they had been washed in running water, alcohol 70% and active chlorine 2%. However, C. gloeosporioides was isolate from the inoculated tissue, although no symptoms were observed. From these results it was concluded that pathogenicity tests C. gloeosporioides, should be performed in plants from tissue culture, because, even after superficial sterilization of the tissue, the organism can be in the stage latent. O experimento foi realizado em condições de casa de vegetação, utilizando-se o processo hidropônico de cultivo em areia com o método circulante de fornecimento de solução nutritiva. O delineamento experimental foi em blocos casualizados. Comparações entre médias foram realizadas pelo teste “ t ” de Student, a 5%. Foram adaptados modelos estatísticos de análise dialélica para avaliar a capacidade geral de combinação do enxerto e do porta -enxerto. Como enxerto foram utilizados os seguintes genótipos de C. arabica: as variedades Catuaí Vermelho IAC 15 e Oeiras MG 6851 e as linhagens H 419-10-3-1-5, H 514-5-5-3. Como porta- enxerto foram empregados três genótipos de Coffea canephora Pierre et Froenher: Apoatã LC 2258, Conillon Muriaé-1, Robustão Capixaba (Emcapa 8141) e um genótipo de Coffea arabica L.: Mundo Novo IAC LCMP 376-4-32. As sementes do material genético foram germinadas em caixas, contendo areia lavada e ao atingir o estádio “palito de fósforo”, efetuaram -se as enxertias, as quais foram do tipo hipocotiledonar. Após pegamento e aclimatação, as plântulas foram levadas para casa de vegetação, plantadas em vasos e irrigadas diariamente com solução nutritiva de Clark, modificada quanto às concentrações de macronutrientes. A força da solução foi aumentada para 1,5x, 2,5x e 3x da força inicial, durante condução do experimento. O pH das soluções foi mantido a 5,5 + 0,5, mediante ajustes diários com NaOH. As trocas das soluções foram efetuadas quando a condutividade elétrica atingiu 60% + 10% da solução inicial. Após 170 dias do transplantio em vaso, avaliaram-se altura das plantas, número de nós, área foliar, volume de raiz, superfície de raiz, produção de matéria seca e eficiências nutricionais. Pela enxertia em cultivo hidropônico pode perceber que nem todo porta-enxerto de Coffea canephora pode ser utilizado com vantagens na enxertia. Com relação ao volume de raiz, o porta-enxerto Mundo Novo foi o que teve melhor desempenho, proporcionando um efeito significativo quando combinado com H 514-5-5-3. Para as variáveis peso da matéria seca da raiz, peso da matéria seca da parte aérea e peso da matéria seca total, apenas as combinações H 514-5-5-3/Apoatã e H 514-5-5-3/M.Novo tiveram efeitos positivos quando comparadas com os pés -francos. Quanto à eficiência nutricional, os porta- enxertos Apoatã e Mundo Novo contribuíram para a maior eficiência de uso de nitrogênio dos genótipos de Catuaí 15 e H 514-5-5-3. O porta -enxerto Apoatã proporcionou aumento no uso de fósforo nas combinações com Catuaí Vermelho IAC 15 e H 514-5-5-3. A combinação de enxertia H 514-5-5- 3/M.Novo teve aumento no uso de potássio quando comparado com o pé- franco. Os genótipos de Coffea arabica Oeiras e H 419-10-3-1-5 mostraram ser altamente eficientes como pés -francos, não sendo beneficiados por nenhuma enxertia. A análise dialélica indicou que os melhores efeitos da capacidade geral de combinação foram para os porta-enxertos Apoatã e Mundo Novo. / The objective of this research was to evaluate in hidroponic conditions the effect of the rootstock on the growth and nutritional efficiency of young plants of Coffea arabica. The experiment was conducted at greenhouse conditions, using the sand cultive hidroponic process with the nutritive solution. It was used the random blocks experimental design. Comparisons among means was made by the Student "t" test, at 5%. It were adapted diallel analysis statistic models to evaluate the rootstocks and grafts general combination capacity. As grafts were utilized the following genotypes of C. arabica: the varieties Catuaí Vermelho IAC 15 and Oeiras MG 6851 and the lines H 419-10- 3-1-5, H 514-5-5-3. As rootstocks were utilized three genotypes of Coffea canephora Pierre et Froenher: Apoatã LC 2258, Conillon Muriaé-1 and Robustão Capixaba (Emcapa 8141) and a genotype of coffea arabica L.: Mundo Novo IAC LCMP 376-4-32. The seeds of the genetic material were germinated in boxes containing washed sand and as soon as they reached the "match" stadium it was effectuated the grafting, that were of the type hipocotyledoning. After the surviving and acclimatizing, the young plants were taken to the greenhouse, planted in a 3 L recipient and daily irrigated with Clark's nutritive solution, modified as the macronutrients concentration. The solution strength was enhanced to 1,5x; 2,5x and 3x the strength of the initial concentration, as the experiment was conducted. The solutions pH was kept at 5,5 + 0,5, by means of daily adjustment with NaOH. The exchanges of the solutions were effectuated when the electrical conductivity reached 60% + 10% of the inicial conductivity. After 170 days from the transplant into recipients, it were evaluated the plants height, number of knots, leaf area, root volume, root surface, dry matter production and nutritional efficiency. Throughout the grafting in hidroponic cultive, it was possible to perceive that neither all the rootstocks of the Coffea canephora can be utilized with advantages. Regarding the root volume, the rootstock Mundo Novo was the one that achieved the best performance, providing a significant effect when combined with H 514-5-5-3. To the variables root, aboveground part and total dry matter weights, just the combinations H 514-5-5-3/Apoatã and H 514-5-5-3/M. Novo, reached positive effects when compared with no grafted plants. Regarding the nutritional efficiency, the rootstocks Apoatã and M. Novo contributed to the greatest efficiency of the genotypes of Catuaí 15 and H 514-5-5-3 nitrogen use. The rootstock Apoatã provided phosphorus use enhancement at the combinations with Catuaí Vermelho IAC 15 and H 514-5-5-3. The grafting combination H 514- 5-5-3/ M. Novo reached pota ssium use enhancement when compared with no grafted plants. The genotypes of Coffea arabica Oeiras and H 419-10-3-15, showed to be highly efficient as no grafted plants, do not being benefited by anyone gratfting. The diallel analysis indicated that the best effects of the general combination capacity were reached with the rootstocks Apoatã and M. Novo. / Dissertação importada do Alexandria

Com o objetivo de identificar plantas e matrizes superiores para utilização per si, e para serem utilizadas na formação de uma população-base destinada a programa de melhoramento intrapopulacional, realizou-se o estudo da heterose manifestada para produção na geração F 1 e da capacidade de combinação geral e específica de seus progenitores. Foi estudado um total de 71 híbridos F 1 (total de 561 plantas). Foi realizada uma análise descritiva a partir dos dados das quatro primeiras colheitas. Alguns híbridos foram avaliados também nos seis, oito e até dez anos de colheita, conforme a disponibilidade de dados. Foi realizada também uma avaliação da reação dos híbridos F 1 e RC 1 (Catuaí como progenitor recorrente) à ferrugem do cafeeiro e um estudo, através da técnica do uso de marcadores moleculares, da natureza híbrida dos genótipos da geração F 1 mais produtivos e da distância genética existente entre vários genótipos avaliados. Na avaliação da resistência genética das plantas à ferrugem foi realizado o teste em discos de folhas, inoculados com a Raça II de H. vastatrix. 11 progenitores (Híbrido de Timor: UFV 376-2; UFV 427-15; UFV 439-2; UFV 440-22; UFV 445-46; UFV 529, e Catuaí: UFV 2143-193; UFV 2143-236; UFV 2144-32; UFV 2144-35; UFV 2144-36) serviram de testemunhas para os 18 híbridos F 1 e 107 híbridos RC 1 . A avaliação foi realizada aos 49 dias após inoculação. No estudo da diversidade genética, foi analisado um total de 51 genótipos (10 progenitores Catuaí, 12 progenitores Híbrido de Timor e 29 híbridos F 1 ). Para os melhores genótipos F 1 classificados quanto à produção, foi realizado também um estudo para a certificação da natureza híbrida dos mesmos. Em ambos os estudos, a técnica do uso de marcadores moleculares RAPD foi utilizada. Através da seleção baseada nas produções dos híbridos em geração F 1 , observou-se a superioridade de produção dos híbridos H 341-11, H 429-1, H 427-2, H 418-6, H 506-3, H 505-9, H 513-5, H 419-8, H 419-10, H 511-1, H 342-8 e H 430-1. Os progenitores Catuaí Vermelho, UFV 2144-35 e UFV 2145-113, e Híbrido de Timor, UFV 378-33 e UFV 445-46, foram os melhores quanto à capacidade geral de combinação em relação à produção. Para a capacidade específica de combinação, o cruzamento UFV 2144-36 x UFV 439-2 foi o melhor nos primeiros quatro e seis anos de produção. Na produção acumulada dos oito anos, o cruzamento UFV 2148-57 x UFV 439-2 foi o melhor. O híbrido H 429 foi o que melhor expressou, em média, o seu valor heterótico. O híbrido H 287-3 apresentou heterose de 195%, calculada com base na produção acumulada dos primeiros quatro anos. Os valores de heterobeltiose confirmaram a superioridade de produção de grande parte dos híbridos obtidos nos cruzamentos realizados. Na avaliação da reação dos genótipos à ferrugem-do- cafeeiro, os progenitores Catuaí apresentaram reação de suscetibilidade, enquanto os progenitores Híbrido de Timor, com exceção do UFV 427-15, e os híbridos F 1 , com exceção do H 415-2, apresentaram-se resistentes. Dos 107 híbridos RC 1 estudados, 81 deles foram resistentes à ferrugem-do-cafeeiro. No estudo da diversidade genética, foi realizado um total de 157 reações, com 86 primers diferentes, onde 53,5% deles apresentaram polimorfismo, totalizando 108 bandas polimórficas, polimórficas/primer. Dentre com os média primers que de 2,35 apresentaram bandas maior número de bandas polimórficas estão: OPA-8, OPC-10, OPA-5 e OPA- 10. Com um limite de dissimilaridade genética de 54%, o Híbrido de Timor UFV 427-15 permaneceu no grupo das cultivares Catuaí. Onze dos doze genótipos mais produtivos na geração F 1 , tiveram sua natureza híbrida confirmada. / This study used a population of coffee breeding program of the Plant Pathology Department of the Federal University of Viçosa, descendant of crossings between 14 ancestries of 'Catuaí' with red and yellows berries and 12 selections of the Timor Hybrid. The objective was to identify coffee plants and progenitors use to per itself, and to be used in the formation of a population-base for the intrapopulational improvement program. The heterosis of the hybrids F 1 and the specific and general combining ability of their progenitors was studied. 71 hybrids F 1 were studied (total of 561 plants).
