Analyzing Cellular Properties with Dielectrophoresis

abstract: Dielectrophoresis (DEP) is a technique that influences the motion of polarizable particles in an electric field gradient. DEP can be combined with other effects that influence the motion of a particle in a microchannel, such as electrophoresis and electroosmosis. Together, these three can be used to probe properties of an analyte, including charge, conductivity, and zeta potential. DEP shows promise as a high-resolution differentiation and separation method, with the ability to distinguish between subtly-different populations. This, combined with the fast (on the order of minutes) analysis times offered by the technique, lend it many of the features necessary to be used in rapid diagnostics and point-of-care devices. Here, a mathematical model of dielectrophoretic data is presented to connect analyte properties with data features, including the intercept and slope, enabling DEP to be used in applications which require this information. The promise of DEP to distinguish between analytes with small differences is illustrated with antibiotic resistant bacteria. The DEP system is shown to differentiate between methicillin-resistant and susceptible Staphylococcus aureus. This differentiation was achieved both label free and with bacteria that had been fluorescently-labeled. Klebsiella pneumoniae carbapenemase-positive and negative Klebsiella pneumoniae were also distinguished, demonstrating the differentiation for a different mechanism of antibiotic resistance. Differences in dielectrophoretic behavior as displayed by S. aureus and K. pneumoniae were also shown by Staphylococcus epidermidis. These differences were exploited for a separation in space of gentamicin-resistant and -susceptible S. epidermidis. Besides establishing the ability of DEP to distinguish between populations with small biophysical differences, these studies illustrate the possibility for the use of DEP in applications such as rapid diagnostics. / Dissertation/Thesis / Doctoral Dissertation Chemistry 2019

Compost Bedded Pack Barns: Management Practices and Economic Implications

Compost bedded pack (CBP) barn design and pack maintenance procedures vary considerably, making advising and problem-solving challenging. One objective of this research was to characterize herd performance and management practices employed by Kentucky CBP managers (42 farms and 47 CBP facilities). Producer satisfaction, changes in historical bulk-tank somatic cell count, and improvement in herd performance parameters after transitioning to a CBP barn support reported CBP barn system benefits. Daily milk production increased from before moving into the CBP barn to the second year after (29.3 ± 0.3 vs. 30.7 ± 0.3 kg, respectively; P < 0.05) for farms using the CBP barn as the primary housing facility (n = 8). Increasing stirring frequency, stirring depth, and ambient temperatures increased pack temperature. Increased drying rate decreased CBP moisture. Increased 20.3 cm depth CBP temperature and ambient temperatures improved cow hygiene. Mastitis-causing bacteria thrive in conditions similar to optimal composting bacteria conditions, making reduction of these bacteria difficult in an active composting environment. Producers must pay attention to other management areas where preventive measures can be employed. Identification of H. Pylori in Saliva by a Nested PCR Assay Derived From a Newly Cloned DNA Probe

A novel probe was developed from genomic DNA of Helicobacter pylori ATCC type strain 43629. It hybridized with all 73 H. pylori clinical isolates tested but not with any of 183 non-H. pylori DNAs in dot blot hybridization. Typing tests revealed 41 different HaeIII-digestion patterns from 57 H. pylori strains tested. Based on the sequence of the probe, a nested PCR was developed that detected as little as 2 fg of H. pylori DNA or approximately equivalent to one cell. No PCR products were amplified from any of 21 non-H. pylori strains tested. Using this nested PCR, H. pylori DNA was detected in 33 of 45 (73%) saliva samples collected from patients with gastric H. pylori infection. These data suggest that the probe is useful for typing H. pylori and that the nested PCR is a valuable tool for detecting H. pylori DNA in saliva.
