Aplikace systému HACCP ve společnosti "MADETA a. s." / The Application of the HACCP System in the Company "MADETA a. s."

The aim of this work was to go thoroughly throw the already existing HACCP system in the company and to evaluate if the hazard is treated in a good way and to find other more effective approach to prevent the hazard. An effort was focused on the primary processing milk and on the process of the treatment of acidified milk products. While investigating current condition the author has made several experiments focusing on the settled conditions of important process in the production (for ex. pasteurization). Chemical, physical, qualitative and microbial properties were observed on a concrete diary product. The main investigation {--} the simulation of disturbance of the cooling chain - was performed where products were exposed to various temperatures for various time periods. During this test were observed: the temperature, pH and microbial changes in exposed products. Due to results form all observations that where performed in this research its possible to proclaim that the HACCP system is settled in a good way and even if above mentioned situation happen the product is safe.

The aim of this work is to assess the current state of risk analysis and of the critical points system HACCP (Hazard Analysis and Critical Control Point) in a food processing business of the PETA Bohemia limited company. On the basis of the latest findings the new optimal HACCP system, which is based on scientific data, systematically identifies specific risks and measures for their control so as to ensure food safety. A food business operator is obliged to create a critical points system by Regulation (EC) No 852/2004 of the European Parliament and of the Council of 29 April 2004 on the hygiene of foodstuffs, Article 5 ``Hazard analysis and critical control points``, paragraph 1: ``Food business operators shall put in place, implement and maintain a permanent procedure or procedures based on the HACCP principles.`` The introductory part contains a short literature retrieval on the development of management methods and on the control of food products by means of the HACCP system. These systems are generally considered as a useful tool for food business operators serving to control risks which can occur in food. They are science-based and systematic, they identify specific risks and measures for control in order to ensure food safety. They are a tool for risk assessment and setting up of control systems which do not depend on testing of the final product but focus mainly on prevention. All HACCP systems can be accomodated to changes such as progress in external adaptation of equipment, process of manufacturing or technological development. The practical part describes in detail the technological process of producing pastry products, especially short pastry. The production is applied into a small bakery of the PETA Bohemia limited company which was founded in Soběslav in 1991. On the base of interviews and provided background information it was possible to view the food business in a complex manner and to get an insight into the current , as well as the past situation of the company and its goals. The risk analysis of the technological process of short pastry production enabled to determine a critical control point, its attributes and values of critical limits, to define the system of managed state monitoring and to determine corrective measures. A manual of the HACCP elaborated in this manner fulfills all legislative requirements. By its implentation the Peta BOHEMIA company gains a risk management system of high quality leading in its final effect towards production of high quality healthy food. The system focused mainly on practical and expert knowledge of the staff connected with sensoric knowledge of final pastry products puts emphasis on encreasing their personal responsibility and understanding of the problem.

Title: Analysis and prognosis of elite male and female triathletes performance at the ITU World Triathlon Championships in 1989-2016 in Olympic triathlon Objectives: To analyse male and female performance in individual parts of the triathlon (swim, bike, run) as well as the whole race performance during the years 1989 - 2016. To determine the performance prediction of racers using the time series analysis for Olympic triathlon in ITU World Triathlon Championship in 2028. Methods: For statistical data processing we will apply the time series analysis using SPSS Statistics 22 software. We will then add the historical content and the actual conditions of the race to the final graphs. On the basis of the processed data, we will create a performance prediction for 2028 using Excel program. Results: In swimming, women are approaching men's performance and they are now on 92.2%. In the future, women will not come closer to men's times. Performance will improve slightly. In cycling, the gap between men and women is 10%. We do not expect any major change in the future. According to the trend of development, we find deterioration in both categories, especially in men. At the moment, the performance of women in running is 88.3% of men. The thesis aims to assess the feasibility of the business plan of pre-paid model of tasting of Moravian wine and judge whether it pays off to invest into this business plan. The feasibility study serves to confirm the execution of the business plan. The study describes the establishment of limited liability company and activities that are related to its establishment. It analyzes the environment through PEST analysis and Porter's five forces model. Market research is conducted via a survey, which serves as a basis for determination of an estimate of potential customers. Marketing mix allows to describe the product, pricing, distribution, promotion plan and marketing communications. The final price is determined using the calculation formula. The opening balance sheet of the company is prepared within this thesis. The amount of capital, the costs and sources of funding are determined. The financial plan includes estimated revenue, profit, return on equity and cash flow plan for the first two years of the life of the company.

Scope of work was technical-commercial analyze with the aim of finding out if the foundation of a specialized subsidiary company by maintenance department outsourcing would have a real and economical justness. The specialized subsidiary would receive right and know-how of patented modern technology the DS Jet mixers designated for crude oil tank mixing and cleaning. Outcome of the thesis is found break-even point when applies the plan in terms of organizational-economical matters and which action would be necessary taken. Theoretical part of work explains specific of enterprise in new sphere of business, new technologies and services trends in Europe. There are discussed typical Europe crude oil market issues, explained principals of market and product analyze, internal process optimization methods and outsourcing in this part. Financial and break-even point analyzes is defined. Practical and analytical part includes the DS Jet Mixers technology description, implementation of the system in the MERO ČR, a. s. and the technology SWOT analyzes inclusive of competition benchmark. With market research had been checked EU market opportunities and marked attractive markets. The way of subsidiary foundation is proposed. The Integral part of the analytical part creates intended the break-even point. In the summary is comprehensive analyzes outcomes and recommended solution in current statute. Furthermore is advised marginal number of orders since when outsource the activities with the DS Jet mixers into subsidiary company.

The dependencies of electrophoretic mobility on pH were measured for a set of 14 markers used for isoelectric focusing that were developed by the group of Šlais and that are based on substitutions on the nitrophenol core, and for a kit consisting of 5 pI markers developed by Shimura, which have an oligopeptide structure. The dissociation constants and limiting electrophoretic mobilities of these compounds were obtained from the dependencies with the use of the program AnglerFish. The isoelectric point values of the compounds were consequently calculated using the obtained data. A comparison of the obtained pI values with the values that have been declared in literature, albeit gained by different analytical methods, has been made. The practical part focuses on the business plan itself consisting of the construction of a new building and the transfer of existing locksmiths' production into their own business premises and the extension of the offered services of metalworking. In conclusion, the business plan is assessed, considering its feasibility under realistic conditions and determining the point of reversal in the implementation of the business plan.

The subject of this diploma thesis is a study of the influence of cellulose ethers on the rheological properties of lime. The theoretical part focuses on rheological parameters, techniques and test equipment used in rheology measurements. Various types of cellulose ethers and their influence on the rheological properties of fresh mortars are described. The practical part deals with the study of the influence of different types and quantities of cellulose ethers on the properties of fresh lime pastes and mortars. The results were obtained from measurements on a rotational rheometer with a special geometry for building materials. Measuring methods used were flow and oscillatory tests. Yield stress, viscosity and flow index were determined by the flow measurement. The stability and stiffness of fresh materials were determined during the oscillation measurements. The measured results are then evaluated and compared.

The aim of the diploma thesis is to characterize the adsorption properties of probiotic bacteria. Characterization was performed using of designed and optimized spectrophotometric method of determination of adhesion of probiotic bacteria to mucin as the main viscoelastic component of mucus. In order to provide a more detailed description of the adhesion interactions involved in the adhesion of probiotic bacteria to mucin, the viability of selected probiotic bacterial strains Lactobacillus rhamnosus CCM 1825, Lactobacillus plantarum CCM 7039, Lactobacillus delbrueckii subsp. bulgaricus CCM 7190, Lactobacillus acidophilus CCM 4833, Lactobacillus casei CCM 4798, Bifidobacterium breve CCM 7825, Bacillus coagulans CCM 2658 and a potential probiotic strain of Lactobacillus zeae CCM 7069. Moreover, bacterial adhesion to carbohydrates was also determined for all the bacteria tested. The last proposed and optimized technique was dynamic and electrophoretic light scattering characterizing surface properties such as -potential, average size distribution and isoelectric point. From this method, information was obtained on aggregation of bacterial cells and on electrostatic interactions. The combination of these methods was used as a complex tool to characterize adhesion of the tested bacterial cultures as a very specific, sensitive and key parameter of a successful probiotic microorganism that is influence by the multiple effects.
