Interakce myricetinu s lidskou střevní mikroflórou / Interaction of myricetin with human gut microflora

The intestinal microbiome contributes in immune system function. It contains a large number of microorganisms that interact with each other and thus affect the host. Currently, attention is being directed towards investigating the influence of the intestinal microbiome on hosts, but also on foreign substances. Foreign substances may influence its composition and subsequent microbial metabolism. Crohn's disease patients have been found to have lower bacterial representation of beneficial bacteria. Therefore it is appropriate to examine the intestinal microbiome of these patients and so understand in greater detail the influence of bacteria on the course of the disease progression and on the medication used. The RP-HPLC method were analysed the faecal samples collected (B, C, D), which were incubated at 0, 3 and 6 hours. The incubation took place the addition of myricetin in the McDougall buffer and BHI medium. The analysis was found that myricetin degradation takes place in faecal samples during incubation, regardless of the medium used. In the faecal sample B, degradation of myricetin occurs faster in the BHI medium than in the McDougall's buffer. In faecal samples C and D, degradation is similar in both media. : Choline is being used in all mammalian cells as a precursor for synthesis of a major phospholipide phosphatidylcholine and as a donor of acetyl residues. Cholinergic neurons in addition require choline to synthesize neuromediator acetylcholine. The ability of cells to create choline via de novo synthesis is limited and therefore they need to transport choline from extracellular space. Limited availability of choline in brain leads specifically to diminished function of cholinergic neurons and in general to impaired reparation of biological membranes. Dysfunctions of cholinergic signaling in brain is characteristic for Alzheimer's disease. Aim of this work was to investigate whether gene and protein expression of high- affinity cholinergic transporters is altered in 5-6 months old APPswe/PS1dE9 mouse model of Alzheimer's disease. Expression of specific high-affinity cholinergic transporter CHT1 (responsible for transport of choline to be used for acetylcholine synthesis) and putative high-afinity choline transporter CTL1 (generally present in all cells and related to high affinity choline transport for phospholipide synthesis) in cerebral cortex was measured. More than 50 % cases of dementia are nowadays caused byAlzheimer's disease (AD). AD is a progressive neurodegenerative disease and it causes gradual memory loss, disorientation and behavioral disorders which affect patient's social and occupational life. AD is characteristic by loss of neurons in some regions of brain - for example hippocampus and cortex. Ethiopathogenesis of this disease is not completely known - that is why the treatment is still just symptomatic. Formation of β-amyloid deposits in brain tissue plays an important role - it is a protein which creates extracellular plagues around neurites and causes their degeneration and death. Intracellular tangles are made up of the changed τ-protein. These tangles also cause death of the neuronal cell. The degeneration of neurons is supported by reactive oxygen radicals too. Data used in this Diploma Thesis have been taken from foreigner scientific literature. It provides the whole summary of natural compounds with β-secretase inhibitory activity. First chapter Alzheimer's disease briefly describes a definition, risk factors and symptomatology of the disease. Most of this chapter characterizes an etiopathology of the disease focused on amyloid theory. Finally there are also mentioned current approaches to therapy of Alzheimer's disease and those, which stay in clinical trials. Next part is dedicated to physiological functions and substrates of the β-secretase. The aim of this study was to determine the frequency of adult disability caused by Forestier disease among skeletal material in monastic burial grounds and to compare it with the frequency of the lay cemeteries. Five cemeteries of the High Middle Ages to the early modern period, were divided into groups according to age and sex. Also, we evaluated the radiographs, investigated the frequency of adult disability caused by Forestier disease in the population. It was found that the disease in skeletal and X-ray material occurred more frequently among men than women, and did not occure until the age of 40. Furthermore, we found that the higher occurence was among monastic funerals. And since there is an increase Forestier disease in the present population. The question is, whether the external conditions, hence increasing obesity in the population, have an impact on the occurrence of Forestier disease.

This diploma thesis, "Alzheimer's disabilities ", deals with questions around people with Alzheimer's disabilities, which is the most common type of dementia. Alzheimer's disabilities is also very widespread in the population; and it is the 4th or 5th most common cause of death. The thesis is divided into two parts, the first part is theoretical and the second is empirical. In the theoretical part the thesis deals with the general characteristics of this disorder such as: causes, prevention, clinical manifestations, consequences, diagnostics and pharmacological approaches. The main focus of the theoretical part is in the second chapter, which deals with non-pharmacological approaches to people with Alzheimer's disabilities. In the empirical part, case studies on sufferers of Alzheimer's disabilities will be included. The empirical part focuses on substantive changes which occur in the lives of people with Alzheimer's disabilities, and whether these changes can be confirmed as being consequential of the affliction.
