Benzen v benzinech z hlediska ochrany zdraví / Benzene in petroleum-derived products and health protection

Benzene is very toxic compound, which has carcinogenic and mutagenic effects. Result of these effects is significant reduction of its use and also low hygienic limits in occupational environment and environment. The aim of this diploma thesis was to map out possible exposure to benzene and checking of clasification correctness of motor gasoline and benzine (cleaners, thinners etc.). Determination of benzene was carried out by gas chromatography with flame ionisation detector (GC/FID) and high performance liquid chromatography with diod array detector (HPLC/DAD). Marginally, the diploma thesis was focused on determination of toluene (in june 2007 became effective ordinance no.284/2006 Sb., which forbid to sell products that contain more than 0,1 % of toluene to small consumers). Toluene was determined by same methods as benzene.

Comprehensive gas chromatography (GC×GC) is a modern and advanced analytical method designed to separate very complex samples, when the separation efficiency of classical gas chromatography is not sufficient. GC×GC allows complete simultaneous analysis of a sample on two different columns, interconnected by special interface called the modulator. Three types of control units were set up and a robust pulse flow modulator (PFM) was developed in this work. PFM working parameters have been always optimized to allow combinations of columns with different sizes. Optimized PFM allows application of columns with 10-60 m length and 0.15-0.25 mm internal diameters for the first dimensions and with 1-10 m length and 0.25-0.32 mm internal diameters for the second dimension. For demonstration reasons, analyses of complex real samples, such as essential oils, oil derivatives and biological samples, were performed. PFM was compared with the commercial cryofocusation modulator Zoex for the selected column set and one sample (a mixture of volatile solvents) on a gas chromatograph equipped with both modulators. The average repeatability of retention times, expressed as a relative standard deviation, was approximately 2.0 % for PFM (cryofocusation about 0.5 %) and of the peak areas was approximately 3.5 %...

(EN) In the first part of this work, analytical methods for determination of impurities of active pharmaceutical ingredients (API) in combined pharmaceutical dosage forms were developed and validated. Development of the methods covered both the optimization of sample preparation procedure and chromatographic conditions. The methods were validated according to International Conference on Harmonization guideline and both of them were confirmed to be able to analyze stability samples. Impurities in paracetamol, codeine phosphate hemihydrate and pitophenone hydrochloride in the presence of fourth API fenpiverinium bromide were separated by using ion-pair reversed phase chromatography with gradient elution. Symmetry C18, 250 x 4,6 mm, 5 µm heated to 35 řC was used as a separation column. A diode array detector was used. The detection wavelengths were set as follows: 220 nm for paracetamol impurity K, 245 nm for paracetamol and its other impurities and 285 nm for codeine, pitophenone and their impurities. Impurities in valsartan, amlodipine besylate and hydrochlorothiazide were separated by reversed phase UHPLC method with gradient elution. Bumblebees are important pollinators, commercially used in large-scale plant growing in greenhouses. Their males produce marking pheromones for mating, which attract young bumblebee queens. These pheromones are often a complicated mixture of chemicals, which is produced in the male labial gland, and the mixture itself is specific for each bumblebee species. The regulation of bumblebee sexual pheromone biosynthesis is largely unknown, and this Master's thesis is focused on the analysis of the mechanisms which lead to the regulation of the Bombus terrestris male pheromone's fat-acid and terpenes biosynthesis, specifically on stereospecific enzymatic reduction of double bond of farnesol. This thesis studies the influence of potential neurohormones on a specific enzymatic mixture, which is involved in the pheromone biosynthesis. Methods used in this project include biochemical, analytic and molecular-biology methods.

The thesis deals with measuring the content of chosen phenolic substances in some specimen of the genera Chenopodium L. and Atriplex L. Two independent analytical methods were used to determine the content of phenolic substances, namely the Micellar Electrokinetic Capillary Chromatography (MECC) method and the High - Performance Liquid Chromatography (HPLC) method. Two cultured species of the genera Spinacia and Atriplex, three freely growing specimen of the genus Chenopodium and three freely growing species of the genus Atriplex were analysed. The analysis concerned the leaves and the inflorescence of these species.The total content of quercetin and rutin was determined by the MECC method. The highest total content of quercetin was found in the leaves of the Garden Orache (4240 mg/kg of dry matter), the lowest total content of quercetin was found in the inflorescence of the Atriplex prostrata DC. (19.6 mg/kg of dry matter). Rutin was only found in four samples, the rest of the samples contained rutin in quantities below the limit of quantification. The highest content of rutin was found in the leaves of the Lamb´s Quarters (868 mg/kg of dry matter).

Synthetic polymer materials have been a part of human life since the 20th century. A variety of their use had resulted in a growing demand for their mass production, which then led to their increasing accumulation at waste dumps. After their deposition, these materials are exposed to many environmental factors causing their decomposition and subsequent release of the degradation products into the environment. This thesis is focused on the analysis of the photodegradation products from synthetic polymer materials and particularly polyurethane foams modified with the biodegradable fillers. One dimensional GC/MS and two dimensional comprehensive gas chromatography with time-of-flight mass spectrometry (GCxGC/TOF-MS ) were used for the measurements. The most of photodegradation products of polyurethane foams are injurious to health and environment.

This master thesis deals with the potential influence of the waste dump Hrádek u Pacova and the wastewater treatment plant Brno – Modřice on surroundings using the optimized non-target screening method of organic pollutants in water and sediments. The experimental part is focused on the identification of organic compounds in samples of ground water from monitoring wells, the surface water from stream and pond and in sediment samples collected from streams in the vicinity of municipal waste dump. Organic compounds were also identified in water and sediment samples from the river Svratka near the municipal waste water treatment plant Modřice. The sequential liquid – liquid extraction was used for the isolation of organic compounds in water samples. Organic compounds in sediment samples were isolated by pressurized solvent extraction (PSE) with subsequent fractionation of the extract using column chromatography. Final analysis determination was performed using comprehensive orthogonal two-dimensional gas chromatography with mass spectrometric detection (GCxGC-TOF MS).

This thesis deals with the content of adaptogens in vegetable Schizandra chinensis. The theoretical part deals with the plant Schisandra chinensis, its occurrence, characteristics, uses and cultivation. Location of plants Schisandra chinensis are classified according to Köppen climate classification and compared with the climate in the Czech Republic, due to possible prediction of growing plants in the country. Further are described adaptogens (bioactive substances) contained in this plant and to methods for their extraction and analysis. The experimental part of the thesis deals with the identification of schisandrin in the plant Schisandra chinensis grown in our conditions. The assessment was performed by ultra-performance liquid chromatography (UPLC), high-performance liquid chromatography (HPLC) and direct injection into tha mass spectrometry (MS). Schisandrin was successfully identified in all the samples.
