Análises sobre a aprendizagem de conteúdos de genética evidenciam que nem mesmo os conceitos básicos desta ciência são compreendidos pelo público estudantil. Tal condição pode ser atribuída, dentre outros fatores, à falta de contextualização social dos conteúdos, o que normalmente configura-se em um problema educativo no ensino de Ciências. O planejamento de uma prática didática que leve em consideração os saberes cotidianos para abordar noções de genética no ensino fundamental pode ser uma estratégia promissora. Nesse sentido, sugerimos a utilização da Anemia Falciforme como referência para o ensino de conceitos de genética, visto que a doença é apontada como a alteração genética mais comum no mundo. A partir dessa perspectiva, foi realizado um estudo empírico que aliou estratégias da pesquisa etnográfica para sondagem do entendimento de moradores da Ilha de Maré sobre herança genética e doenças hereditárias, em especial sobre a Anemia Falciforme, a fim de levar em consideração seus saberes cotidianos ao mediar o conhecimento científico. Tal estudo se deu na Ilha de Maré, uma comunidade quilombola que está localizada na cidade de Salvador. Durante o ano de 2013 foram entrevistados 26 moradores de 3 comunidades da Ilha. As entrevistas foram semiestruturadas e adotou-se a técnica de amostragem por acessibilidade ou conveniência. Também foram realizados registros sistemáticos das observações em um caderno de campo. Constatou-se, entre outros fatores, um alto índice de casamento consanguíneo, fato que contribui para a alta incidência da Anemia Falciforme na região, uma vez que se trata de uma doença autossômica recessiva. Também se notou um conhecimento superficial acerca de herança genética, bem como de doença hereditária, o que pôde ser evidenciado através das respostas ambíguas e dos questionamentos realizados pelos entrevistados. Os resultados serviram como subsídio para o planejamento de um curso de capacitação para professores da educação básica, que foi oferecido na modalidade semipresencial e teve 40 horas de duração. Optou-se por este formato buscando ampliar o ambiente de integração para visando dispor mais recursos analisar a participação dos professores. Dessa forma foi possível pesquisar a visão dos professores quanto à viabilidade da transposição de conceitos de genética para o ensino de noções de genética no ensino fundamental, tendo a Anemia Falciforme como referência. Assim, a atividade principal do curso foi à construção colaborativa de uma sequência didática para aprendizagem de noções de genética por estudantes do 8º ano do ensino fundamental, a partir da abordagem contextualizada da Anemia Falciforme. Para isso os professores refletiram sobre os resultados dessa pesquisa ao contextualizar o assunto para esse público estudantil. / In studies on science teaching, arguments for the approximation of scientific knowledge to the students' daily life are common, as this strategy can generate better educational outcomes. Analyzes of the learning content Genetics show that even the basics of this science are understood by the student audience. This condition can be attributed to the lack of social contextualization of the contents, which usually sets in an educational problem in science teaching. Planning a teaching practice that takes into account the daily knowledge to address Genetics notions in elementary school may be a promising strategy. In this sense we suggest using the Sickle Cell Anemia as a reference for teaching concepts of genetics, since the disease is considered the most common genetic alteration in the world. From this perspective, we performed an empirical study that combines strategies of ethnographic research seeking to probe the understanding of residents of Ilha de Maré on hereditary genetics and hereditary diseases, in particular on the Sickle Cell Anemia in order to take into account their daily knowledge to mediate scientific knowledge. This study took place Ilha de Maré, for it is a maroon community that is located in the city of Salvador. During the year 2013 were interviewed 26 residents of three communities of the island. The interviews were semistructured and adopted the sampling technique for accessibility or convenience. Were also carried out systematic records of observations in a field notebook. In this occasion it was found, among other things, a high rate of consanguineous marriage, a fact that contributes to the high incidence of sickle cell disease in the region, since it is an autosomal recessive disease. It was also noted a superficial knowledge of genetics and hereditary disease, which can be evidenced through the ambiguous answers and questions made by participants of the interviews. The results served as input for planning a training course for teachers of basic education, which was offered in blended mode and had 40 hours of duration. We chose this format for deepening the integration environment to have more resources in order to analyze the participation of teachers. Thus it was possible to search the view of teachers as to the feasibility of implementing genetic concepts for teaching genetics concepts in elementary school, and Sickle Cell Anemia as a reference. Thus, the main activity of the course was the collaborative construction of a didactic sequence for learning genetics concepts by students of the 8th grade of elementary school, from the contextual approach to sickle cell disease. For this the teachers reflected on the results of this research to contextualize it for that student audience.

Estudo da miotonia hereditária em suínos

Orientador: Alexandre Secorun Borges / Resumo: A principal causa de miotonia não distrófica hereditária ocorre devido à mutações no gene CLCN1, codificante para a proteína CLC1 que forma o canal iônico seletivo para o íon cloreto predominante no tecido muscular esquelético. Mutações no gene CLCN1 foram descritas como causadoras de miotomia hereditária em humanos e em várias espécies animais. Não existe descrição de miotonia hereditária na espécie suína. O objetivo deste estudo foi realizar a caracterização clínica e molecular de uma forma de miotonia hereditária em suínos. A hipótese desse estudo foi que animais com sinais clínicos compatíveis apresentavam a miotonia hereditária. Esses animais foram avaliados sob aspectos clínicos, eletromiográficos, histopatológicos e moleculares. Os sinais clínicos verificados foram hipertrofia e rigidez musculares, miotonia com startle response formação de dimples e fenômeno warm-up evidentes. Não foi constatada distrofia muscular ao exame histopatológico. Ao exame eletromiográfico foram demonstradas descargas miotônicas clássicas com formação de som característico diver bomb. A nível molecular foi verificada a ausência dos nucleotídeos referentes aos éxons 15 e 16 utilizando amostras de cDNA dos animais afetados. No DNA genômico foi encontrada uma grande deleção de 4165pb (g. NC_010460.4 del 6912538_6916702) na região do gene CLCN1. Análises de expressão relativa demonstraram níveis de expressão em tecido muscular de animais wild type para um transcrito associado a miotonia hereditári... (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The major cause of hereditary non-dystrophic myotonia occurs due to mutations in the CLCN1 gene, coding for the CLC1 protein that forms the ionic channel selective for the predominant chloride ion in skeletal muscle tissue. The resulting hereditary disease is called congenital myotonia in human medicine. Mutations in the CLCN1 gene have been described as causing hereditary myotomy in several animal species, but in the swine species, no mutation in this gene has been described. The objective of this study was to perform the clinical and molecular characterization of hereditary myotonia in swine. The hypothesis of this study was that animals with compatible clinical signs had hereditary myotonia. These animals were evaluated under clinical, electromyographic, histopathological and molecular aspects. The clinical signs verifed were muscular hypertrophy and stifness, myotonia with startle response and formation of dimples. The phenomenon warm-up was evident. No muscular dystrophy was observed at the histopathological examination. Electromyographic examination showed classic myotonic discharges with characteristic sound. At the molecular level, the absence of nucleotides from exons 15 and 16 was verifed using cDNA samples of afected animals. In genomic DNA a large deletion of 4165bp (g NC_010460.4 del 6912538_6916702) was found in the region of the CLCN1 gene.
