Regulace signalizace receptorového komplexu pro IL-17 / Regulation of IL-17 receptor complex signaling.

Inflammatory immune response is essential for maintaining the defense against invading pathogens, although its aberrant activation leads to impaired self tolerance and development of autoimmune pathologies. Interleukin-17A (also known as IL-17), is a major proinflammatory cytokine, which contributes to the development and maintenance of inflammation and provides protection against several bacterial and yeast infections. However, extreme activation of IL-17 signaling leads to autoimmune pathologies. Thus, a strict regulation of IL-17 signal transduction is crucial to prevent progression of autoimmunity. Non-degradative ubiquitination is one of the main mechanisms regulating IL-17 signaling. Main E3 ubiquitin ligase within this signal transduction is TRAF6, which is also participating in several signaling pathways within the immune system. Non-degradative polyubiquitin chains created within inflammatory signaling complexes recruit signaling proteins such as IKK complex and TAK1 kinase, crucial for triggering of NF-κB and MAPKs downstream pathways. However, activation of these pathways upon IL-17 is very weak in comparison with other inflammatory stimuli, indicating a presence of a strong negative feedback loop. Simulace fyzikalních procesů v experimentu Belle II a testovaní kvality rekonstrukčního softwaru / Simulation of Belle II physics events and performance tests of reconstruction software

This thesis is about the alignment of vertex semiconductor detectors of the Belle II experiment. It is divided into several sections. The first part introduces the Belle II experiment as a new generation B- factory, with its ambitious plans of data taking and physical analysis. The second part follows with a more detailed description of Belle II detector and software tools designed for their alignment and calibration. The central part of the thesis describes physical processes used for calibration of the vertex detector, connected with the search of an optimal solution for run-time monitoring and calibration of the detector. We also tested the effect of misalignment on the Belle II tracking software. The following part gives results of the study of misalignment effects on physical observables related to analysis of selected physical channels. The last part describes the development of a data quality monitoring tool for the tracking system. The tools has to provide a run-time diagnostic of misalignment and miscalibration by monitoring the precision and accuracy of reconstruction of physical observables. Powered by TCPDF (www.tcpdf.org)

The aim of this paper is to present the development of administrative structures of the Czechoslovak State Railways (CSD) from 1918 to 1938. In the introduction I will give the history of railways in the period 1918 - 1938, including nazionalization largest private railway companies and the impact of these events on the administrative structure Following is a brief description of the management of Austrian and Hungarian State Railways on Czechoslovak territory, which came out of the structure of management for future CSD. I shall then consider the origins of the administrative structure of the independence of the CSD and its development throughout the period. This work is part of the particular division of the Ministry of Railways, each Directorate of State Railways, and other organizational units of the CSD. Added to all ministers of short vignettes railway tracks and broken down between individual Directorate of State Railways. All the work I wanted to finish the event in Munich due to the aforementioned structure, the development brief administration in subsequent periods and comparing the current situation.

Ekologie rysa ostrovida (Lynx lynx) a vlka obecného (Canis lupus) v oblasti Západních Karpat a jejich význam v lesním ekosystému

The abundance and population density of the Eurasian lynx (Lynx lynx) were studied during two winters 2011/2012 and 2012/2013 in the in Beskydy and Kysuce protected landscape areas in the West Carpathians in the Czech-Slovak borderline. A combination of camera trapping, photographic capture-mark-recapture (CMR) and simultaneously conducted snow tracking revealed 11 independent lynx in both seasons in the total area of 1500 km2 and overall population density of 0.7 lynx/100 km2. High population turnover of 54.5% was documented between the two winters, indicating a strong anthropogenic pressure at the edge of the West Carpathian population. The main prey of Eurasian lynx in winter was a roe deer (81%) and red deer (16%). The occurrence of wolves (Canis lupus) was considered to be sporadic and probably limited to only a few individuals. The nearest reproductive packs were documented at a distance of 10--50 km from the Czech border in the Slovak Carpathians.

The law of railway transportation is a phenomenon affecting several other law branches in particular European Law, Commercial Law and Administrative Law. This thesis focuses on the last mentioned branch, i.e. Administrative Law. This thesis does not cover this issue comprehensively; however, it gives fundamental and comprehensive views on the issue. In addition to information on current legal situation and its background it provides information about the history of law of railway transportation and includes the topic of public transport services. In last twenty years the initiator of changes in law of railway transportation is mainly the European Union, which gradually liberalizes the railway transportation. Both national legislation and public services reflect and adapt to these changes. After summarising the history of law railway transportation this thesis gives overview on the important European legislation which determines national legislation in the Czech Republic. The main part of this thesis is about legislation issues in law of railway transportation in the Czech Republic. It focuses on the organisation of public administration according to the Railway Act and the jurisdiction of the Rail Administrative Authority including some aspects of the administrative proceedings there. This master’s thesis deals with a design and optimization of an experimental scintillation secondary electron detector for the environmental scanning electron microscope and with a description of a detector operation principle. The experiment is founded on simulations of a gas flow in detector inner sections and on simulations of secondary electron trajectories in electrostatic fields of the detector. On the basis of the simulations, new solutions of the detector designs are proposed. For these designs, same simulations as previous are performed and designs that seem to be feasible for the secondary electron detection in environmental scanning electron microscope are selected.
