Effects of morphine on intracranial self-stimulation : the involvement of associative factors and the role of ventral tegmental dopamine neurons

A series of experiments was carried out to clarify the effects of morphine (0.3 - 10 mg/kg) on intracranial self-stimulation (ICS) and to compare these with the effects of the stimulant amphetamine on this behavior. It was shown that the enhancement of ICS by morphine requires repeated drug exposure, is prevented by pre-exposure to the drug in a non-ICS context, is mimicked by administration of vehicle, and is not reliably reversed by naloxone. In contrast, facilitation of ICS by amphetamine was immediate and remained stable over repeated days of testing. It was concluded that ICS facilitation induced by morphine, but not by amphetamine, is largely the outcome of a learned association between the drug effect and the ICS procedure, and does not appear to be a direct, opiate receptor-mediated effect. Finally, 6-OHDA lesions of ventral tegmental dopamine neurons were shown to block the facilitation of ICS by morphine but not by amphetamine. These lesions were also shown to delay the development of tolerance to morphine-induced catalepsy.

This study determines the effects of variations in suspended biomass, organic carbon, and pH on the time course of 2,2$ sp prime$,4,4$ sp prime$,5,5$ sp prime$-hexachlorobiphenyl uptake by laboratory cultures of Selenastrum capricornutum. Variations in pH had no effect, but uptake was enhanced by higher levels of biomass and depressed by higher levels of organic carbon. Mechanisms of desiccation tolerance in cryptogams.

In this study adaptations of bryophytes and lichens to desiccation stress were examined. The aim was to test whether desiccation tolerance in the selected species is constitutive or if desiccation tolerance could be induced by various hardening treatments. In addition, some putative tolerance mechanisms were investigated, including the accumulation of sugars, increase in ROS scavenging systems and other mechanisms e.g. energy dissipating processes. To determine if hardening treatments prior to desiccation stress increased desiccation tolerance, mosses and lichens were partially desiccated or treated with ABA. The effect of hardening treatments on the physiology of the moss Atrichum androgynum and lichens Peltigera polydactyla, Ramalina celastri and Telochistes capensis during a desiccation-rehydration cycle was investigated. Photosynthesis, respiration and chlorophyll fluorescence measurements were used as rapid tools to determine the metabolic activities in these lichens and moss species. In A. androgynum partial desiccation following slow drying at 52% RH increased the rate of recovery of net photosynthesis. Net photosynthesis recovered almost completely following slow drying in the material that was partially dehydrated and/or treated with ABA. This suggests that partial dehydration hardens the moss, and that ABA can fully substitute for partial dehydration. In R celastri and P. polydactyla both partial dehydration and ABA treatments displayed some improvement in desiccation tolerance depending on the duration and severity of stress. The reduction in the re-saturation respiration burst in P. polydactyla, although not quite significant, strongly suggests that hardening increases mycobiont tolerance. However, it is more difficult to establish whether the hardening treatments improve photobiont performance. In the moss A. androgynum ABA treatment increased the rate of recovery of photosynthesis and PSII activity, and also doubled non-photochemical quenching (NPQ). Increased NPQ activity will reduce ROS formation, and may explain in part how ABA hardens the moss to desiccation. In ABA treated, but not untreated mosses, desiccation significantly increased the concentration of soluble sugars in A. androgynum. Sugar accumulation may promote vitrification of the cytoplasm and protect membranes during desiccation. Starch concentrations in freshly collected A. androgynum and R. celastri were only c. 40 and 80 mg g ¯¹ dry mass respectively, and slightly rose during desiccation, but were only slightly affected by ABA pretreatment. ABA did not reduce chlorophyll breakdown during desiccation. In P. polydactyla ABA pretreatment had little effect on any of these parameters. Changes in the activities of the free radical scavenging enzymes ascorbate peroxidase, catalase and superoxide dismutase were measured during wetting and drying cycles in the moss A. androgynum and in the lichens P. polydactyla, R. celastri and T capensis. These species normally grow in the understorey of the Afromontane forest, moist, xeric, and extremely xeric miicrohabitats respectively. In A. androgyum, enzyme activity was measured shortly after collection, after 3 d storage following hardening by partial dehydration and/or 1 h treatment with ABA or distilled water and during desiccation and rehydration. In A. androgynum enzyme activities of CAT and SOD in untreated material were always higher than in the hardened treatments, while both partial dehydration and ABA treatments tended to reduce both CAT and the induction of SOD activity, although these effects were not significant between the treatments. This suggests that ABA may not be involved in the induction of free radical scavenging enzymes and probably these enzymes are not important in desiccation tolerance of A. androgynum. In lichens, the enzyme activity was measured shortly after collection, after hydration for 48 hat 100% RH, after desiccation for 14 d and 28 d, and during the first 30 min of hydration with liquid water. Enzyme activities tended to rise or stay the same following rehydration in all the species tested. After desiccation for 14 d, enzyme activities decreased, and then decreased further to very low values after 4 weeks desiccation. In all species, including T capensis from an extremely xeric habitat, the activities of all enzymes remained at very low values during the 30 min following rehydration, and were therefore unavailable to remove any reactive oxygen species accumulating in lichen tissues as a result of desiccation. Results suggests that the enzymic antioxidants are more likely to be involved in removing reactive oxygen species produced during the normal metabolic processes of lichens than having a role in desiccation tolerance. The Afromontane understorey moss Atrichum androgynum displayed an oxidative burst of H₂O₂ during rehydration following desiccation. Maximum rates of H₂O₂ production occur during the first 15 min of rehydration. While the production of H₂O₂ increases with increasing desiccation times, the moss produced significant amounts of H₂O₂ during rehydration after desiccation for times that did not inhibit photosynthesis or cause K⁺ leakage. A. androgynum may produce more H₂O₂ during desiccation than rehydration, because desiccation artificially induced using polyethylene glycol strongly stimulates production. Experiments involving inhibitors and exogenously supplied reductants indicate that peroxidases are responsible for the synthesis of H₂O₂. Factors that influence the rate of H₂O₂ production during rehydration include light and the hormone ABA. Production and physiological responses of Italian ryegrass and white clover grown in mono cultures and mixed stands.

The aim of this study was to test the hypothesis that ryegrass and clover, when grown under optimal conditions in mixed stands, interact in response to available light energy. with other resources non-limiting, pasture yield is determined by the efficiency with which solar radiation is intercepted by canopies and converted into dry matter. The clover-ryegrass interaction under varying light regimes, as experienced in the canopies of these pastures, was studied by investigating the growth and production of ryegrass and clover in relation to light harvesting abilities and photosynthetic utilization of intercepted light. Pasture canopy structure and growth were studied under a four-weekly clipping treatment. The interception of photon flux density (PFD) in the pasture canopy was monitored diurnally and seasonally in mixed and mono cultures. Light use efficiency (C02 fixed/ unit absorbed PFD) as well as photochemical efficiency of photosystem II (Fv/Fm) were studied by monitoring CO2 assimilation rates and chlorophyll fluorescence respectively. The results obtained from this study indicated that interaction did occur between ryegrass and clover, cultivated in mixed pastures. The mixture was capable of more efficient light interception than the mono cultures, which resulted in higher productivity. Light interception abilities, as manifested in canopy architecture, and not physiological utilization of light energy, were found to govern the interaction between the two pasture components. / Thesis (M.Sc.)-University of Natal, Durban, 1993.

An investigation into the ultraviolet radiation exposure of children and adolescents in Durban.

Stratospheric ozone plays an important role in absorbing ultraviolet radiation. The well-known depletion of the ozone layer has raised several concerns in terms of an expected increase in surface ultraviolet radiation. South Africa, situated in the southern mid-latitude regions, has experienced a general downward trend in total column ozone since 1979, As a result of this negative trend in total column ozone, the ultraviolet flux at the earth's surface is expected to increase. Excessive exposure to ultraviolet radiation is known to have acute and chronic effects on human health, including erythema and skin cancer. Numerous studies have acknowledged a relationship between childhood ultraviolet radiation exposure and the risk of contracting skin cancer, namely malignant melanoma, during adulthood. The aim of this study is to investigate the ultraviolet radiation dose and exposure of children and adolescents in Durban, South Africa. Polysulphone film badges were used to quantify the daily erythemal ultraviolet radiation dose of 30 individuals of varying ages and skin types, engaged in different activities, over a one-week period during summer. The results highlight the diversity of childrens' and adolescents' behavioural patterns, with behaviour being found to play an important role in determining an individual's ultraviolet radiation dose. The mean daily erythemal ultraviolet radiation dose of the children and adolescents was 1.03 MED units with a median of 0.57 MED units and a 95% range of 0.22 - 7.22 MED units. The most striking finding was that the median value was below the critical value of 1 MED unit. An explanation for the unexpectedly low erythemal ultraviolet radiation doses recorded in this study was sought in the prevailing climatic conditions. Ambient erythemal ultraviolet radiation levels recorded during the study period were high, ranging between 20.57 - 30.60 MED units. However, high temperatures (>27°C), coupled with high humidity values, may have encouraged the children and adolescents to avoid direct sunlight and find shade while outdoors. The daily erythemal ultraviolet radiation doses of the children and adolescents were also compared to the ambient erythemal ultraviolet radiation levels received on a horizontal surface by a YES UVB-l pyranometer located at the University of Natal (Durban). Children and adolescents in Durban received approximately 4.58% of the total daily ambient erythemal ultraviolet radiation incident upon a horizontal surface. This was found to be similar to a study (5 - 6%) conducted by Diffey et al. (1996) in England, as well as a study (4 - 8%) by Gies et al. (1998) in Brisbane, Australia. The personal ultraviolet radiation exposure journals of the children and adolescents were used to determine the timing of exposures, duration of exposures and nature of outdoor activities and these were then related to their daily ultraviolet radiation doses. Of all the factors considered, the nature of an individual's activity was found to have the strongest influence in determining their ultraviolet radiation dose. An activity model was derived in order to investigate the effect of activity on ultraviolet radiation dose, where three activity factors, namely swimming, walking and tennis, were calculated for a South African context and compared with those from previous international studies. It was found that the activity factors derived in this study were similar to Holman et al. (1983) and Herlihy et al. (1994) and may be used in an activity model to estimate individual erythemal ultraviolet radiation dose for a particular activity. The value of this innovative activity model lies in its ability to predict individual ultraviolet radiation dose and this may help to emphasise the importance of responsible outdoor behaviour. A mannequin was used to quantify the anatomical distribution of erythemal ultraviolet radiation under clear sky and overcast conditions. It was found that the vertex of the head and shoulders received the highest erythemal ultraviolet radiation doses under both conditions. This was then related to the erythemal ultraviolet radiation doses of the children and adolescents as recorded by the polysulphone film badges in order to identify anatomic sites susceptible to high erythemal ultraviolet radiation doses. An empirical study of the international Fisher effect.

The international Fisher effect is identified as part of the four-way equivalence model. This model outlines a relationship between exchange rates, interest rates and inflation rates. The international Fisher effect, specifically, states that the difference in interest rates between two countries is an indicator of the expected change in exchange rates of their currencies. The aim of this paper is to test the validity of the international Fisher effect between South Africa and the UK. The understanding of the exchange rate movements is vital for management decisions, investment activity and policy making for central banks and government. Data has been collected for a sampling period beginning in July 1995 and ending in April 2001. Interest rates in the UK and South Africa are recorded for this period. A record of exchange rate movements for the same period has also been compiled. Using this data, a simulation of an uncovered interest arbitrage was carried out. This was done by taking £100 from the UK, converting it to Rands and investing those Rands in a South African bank. At the same time, £100 was also invested in a UK bank. As interest accrued over the test period, interest rates in both countries changed, exchange rates fluctuated and the balance in the South African account was compared to the balance in the UK account. According to the model, the real balances in both the accounts should remain equivalent over the sampling period. It was found that interest rates in SA were higher, more volatile and less cyclic than those in the UK. As predicted by the model, the exchange rate (in R/£) constantly increased over the sampling period. Reasons for the higher interest rates in SA include a low national savings rate, high inflation, the South African economies vulnerability to events in the international market and the reserve bank's monetary policies. The simulated arbitrage was found to be profitless and the balances of the two simulated investment accounts were found to be statistically similar. There were, however, some short term deviations from the theory. The value of the SA account was lowest during times of high interest rates in SA, when there was volatility in the forex market and when the exchange rate was at peaks in the cycle. Nevertheless, the exchange rate - interest rate relationship always returned to equilibrium. The risk and unpredictability associated with the international market is high while only small chances exist to achieve economic gain from borrowing from low interest rate environments (or investing in countries where the interest rates are high). It was concluded that the international Fisher effect, between the UK and South Africa, for the period studied, had significant short term deviations but is valid over the medium term. The implication for business practice is that stakeholders should be conservative when faced with risk associated with foreign exchange exposure unless, as is the case with speculators, it is their core competence to predict macroeconomic trends and profit from beating the market. / Thesis (MBA)-University of Natal, Durban, 2001.
