Analýza toxicity průsakových vod ze skládky odpadů KuchyňkyStaňková, Jiřina January 2016 (has links)
The thesis is focused on analysis of the toxicity of the water seepage from landfill Kuchyňka. It is composed of theoretical part, which is engaged in waste management and its disposal. Then it is engaged in the process of landfill directed at water monitoring. The next part introduce into basic methods of ecotoxicology. The practical part of thesis is engaged in analysis of the water seepage and rainwater from landfill Kuchyňka. Those were tested and on semichronacal toxicity with seeds of white mustard (Sinapis alba, L.). Thesis contains a description of this method, used materials and operating procedure. Then the results of the tests are summarized and evaluated. In results there are values of root growth inhibition and seed germination capacity in individual phases of test that are graphically presented. Thesis is supplemented with photographs of landfill and interpretation of test of toxicity.

The thesis deals with landfilling, monitoring leachate from landfill and toxicity of this water. The beginning of theoretical part is focusing on the characteristics of municipal waste and description of the landfill process. The next part is focusing on description of the discipline of ecotoxicology and its basic methods. The work contains information about landfill Štěpánovice, which is located in the region Plzeň. The practical part of the thesis focuses on the description of performed testing of semichronic toxicity of leachate water from the landfill Štěpánovice with the help of seeds of white mustard (Sinapis alba L.). In conclusion, there are mentioned results of the test, which consist of calculating the root growth inhibition for a given concentration of the test substance.

Thesis on Evaluation of the toxicity of the water seepage from landfill Kuchyňky is engaged in legislation, waste management, statistical data in the beginning. Then there is a theoretical description of the process of landfill, division of landfills and characteristics of the basic methods of ecotoxicology. The next part is devoted to informations about landfill Zdounky Kuchyňky, its location, natural conditions and monitoring data on the landfill. Then the chapter is devoted to the description of the test of growth of duckweed (Lemna minor L.) on water seepage from landfill Kuchyňky. In the last part of the thesis there is the evaluation of the results of a toxicity test of water seepage and rainwater from landfill Kuchyňky.

This diploma thesis deals with the landfill and seepage waters from municipal landfill site. The first part is focused on theoretical description of landfill - description of landfill, division of landfills, landfill legislative requirements. In more details there is described the ecotoxicology and bisorption. The second part contains information about the landfill site Kuchyňky, description of the natural conditions of the area, the methodology that has been applied when determining the toxicity of leachate from the landfill and use of oyster mushrooms to reduce the toxicity of the leachate from the landfill. In the conclusion we evaluated the influence of oyster mushrooms (Pleurotus ostreatus) on the phytotoxicity of leachate from the landfill. We compared results of the experiment before applying of oyster mushrooms and after its application on the leachate waters.

Testy toxicity s perloočkou Daphnia magnaMelezínková, Petra January 2019 (has links)
The aim of this thesis was to derermine the values of toxic effect of chemical sub-stance chemical on the tested organism Daphnia magna. For testing, these materials were selected: polyaluminiumchloride (PAX 18), its aqueous solution (PAX 19) and potassium dichromate. The thesis is divided into two parts, in the first half I devote to theory of the basic terminology of the issue and the general characterization of the tested organism. In the practical part I focus on the characteristics of the used substances, the course of the tes-ting and the results of the work together with the discussion. Experimental concentrations for PAX 18 were between 1-100 mg.l-1, for PAX 19 10-300 mg.l-1 and for potassium dichromate 40-150 mg.l-1. Testing was perfomed in 3 repeatings with ten individuals + control. 24hEC50 values were: for PAX 18 45,06 mg.l-1, for PAX 19 97,82 mg.l-1 and for potassium dichromate 1,569 mg.l-1. The values of 48hEC50 for PAX 18 byly 22,62 mg.l-1, for PAX 19 37,20 mg.l-1 and for potassium dichromate 0,864 mg.l-1. In all tests, fluctuations in pH and dissolved oxygen content, as well as the number of inhibited individuals, were observed. The results of the observations were plotted and then commented verbally.

Another part is devoted to the field of ecotoxicology and describing its basic methods. The practical part describes the selected landfill and performing the assay inhibition of white mustard (Sinapis alba L.) for leachate water from landfill. The work also includes the test phytotoxicity of soil by Phytotoxkit. It focuses on the description of this method, workflow and work aids. In conclusion, the evaluation of both tests, which involve the calculation of root growth inhibition for each concentration of the test substance. The work is complemented by photographic documentation carried out laboratory tests and landfill.

Environmental effects of selected monomers, eventually aditives of polymers, will be evaluated by ecotoxicity tests. For this purpose two alternative tests of toxicity: Thamnotoxkit F TM on testing organism Thamnocephalus platyurus and Daphnotoxkit F TM on testing organism Daphnia magna will be used. Two phytotoxicity tests will be also used: Sinapis alba root growth inhibition toxicity test and Allium cepa L root growth inhibition toxicity test. Obtained results enable calculation of value LC50, EC50 and IC50 for tested substances.

This diploma thesis deals with a use of algal test in ecotoxicology. A freshwater algae, Desmodesmus subspicatus, was used to asses the ecotoxicity of selected chemical substances. A spectrophotometry method, of VIS spectrum, was used in evaluation of the EC50. A correlation, between a number of algal cells and the wavelength absorbance at 683 nm, has been determined for this purpose. Following chemicals were tested, 2-[2-[(2,6-dichlorophenyl)amino]phenyl]acetic acid, (RS)-2-[4-(2-methylpropyl)phenyl]propanoic acid and N-(4-hydro-xyphenyl)acetamide. An EC50 value was determined for all these substances and also their ecotoxicity has been evaluated.

Polymers or their modifiers may become parts of a wide range of products. After the end of life cycle useful parts of the products are recycled but significant parts still become household waste. It´s therefore desirable not only by law on chemicals and chemical products, but also according to the requirements of European directive REACH, to assess the impact not only chemical but also the additives of all products or products themselves in terms of their impact on the environment. For these purposes it serves ecotoxicity tests, which allow prediction of the influence test compounds in the environment. In this diploma thesis were evaluated polypropylene modifiers, which are commercially produced. Use of ecotoxicity tests were evaluated maleic anhydride and itaconic anhydride effects on the environment. Alternative tests have been used for toxicity and namely Thamnotoxkit FTM the organism Thamnocephalus platyurus and Daphnotokit FTM the organism Daphnia magna. The phytotoxicity test groups were selected toxicity tests: Sinapis alba root growth inhibition toxicity test, Allium cepa L root growth inhibition toxicity test and Semi-chronic toxicity test on Lemna minor L. From marine toxicity tests was performed Acute inhibition test of brine shrimps Artemia salina.
