Obsah rutinu v odpadním biologickém materiálu z výsadeb bezu černého / Content of rutin in the biological waste material from plantations of elderberry

This thesis is focused on determining the concentration of rutin in twigs elderberry (Sambucus nigra L.). The theoretical part provides an overview of the properties, occurrence and use of elderberry in folk medicine, medicine and food. Furthermore, there are the most important substances contained in elderberry, especially flavonoids, including rutin and quercetin as well. There are briefly described their properties, biological effects and possibilities of determination. The content of the experimental part of the thesis was developed cheap extraction methods to obtain the maximum yield of dried twigs rutin elderberry. Rutin content was observed in twigs cultivated elder varieties and wild elderberry. The identification and quantification of rutin in extracts was optimized and validated method of high performance liquid chromatography (HPLC). As a comparative method to develop extraction was used a pressurized hot water extraction (PHWE). There were tested three particle sizes of shredded twigs. The particles were extracted differently hot water varying duration. Most rutin was found in samples milled at 4 mm particles. The optimum time and temperature of extraction was 7 hours at 80°C. The highest amount of rutin in bred elderberry was determined in a sample Mladší 2014, digestion using 1,49 ± 0,004 mg.100g-1 and method PHWE 5,75 ± 0,015 mg.100g-1. There was determined the most rutin in a sample Září 2014 in the wild variant of elderberry. By digestion of ground twigs was obtained 0,90 ± 0,022 mg.100g-1 of rutin, using PHWE 2,98 ± 0,022 mg.100g-1.

Elementární analýza různých odrůd černého bezu / Elemental analysis of the different varieties of elderberry

This work deals with the elemental analysis of different elderberry (Sambucus nigra L.) cultivars. In the theoretical part the botanical characteristics of elderberry, including its extension and places of occurrence, the possibilities of collecting the various parts of the elderberry and its use in food and cosmetics is described. A large part is devoted to the occurrence and characteristics of minerals in food, and to the methods that are suitable for elemental analysis and decomposition of biological material. The aim of the experimental part was to analyze different cultivars of elderberry and juice made from this cultivars, and to determine concentration of selected elements (Zn, Mn, Fe, Mg, Cu, P, Ca, Na and K) using ICP–OES. The total amount of 24 cultivars was analysed from which 6 varieties were harvested in two different seasons in 2011 and 2013. Measured concentrations of elements in different varieties of elderberry were mutually compared using statistical methods. The differences between the content of elements in fruits and juice, obtained by pressing the fruit, and the effect of seasonal conditions on the content of elements in the fruit juice from different elderberry cultivars were studied.

The hereby work describes a new product - herbal liquor containing antioxidants of elderberry and sea buckthorn. It is completely designed, manufactured and tested by a sensory analysis. I performed everything using a simple, low capital technologies, and raw materials, which could easily grow on soils in LF areas. Thus would these LF areas have an appropriate industrial applications . Since LF areas are often areas with high unemplyements I think the realization of such production could bring new job opportunities.

It plays role in many biochemical reactions. Thus, iron metabolism disorders can lead to different diseases associated with lack of iron or iron overload. One of the possible treatment modalities of the latter represents the administration of iron chelators. Elderberry, Sambucus nigra L. (Adoxaceae), has been used in traditional medicine. The fruits of elderberry are a rich source of cyanidin-based anthocyanins as the main component. There are important differences, both in chemical and physical properties between several cultivars of elderberry. Anthocyanins might interact with metals in the gastrointestinal tract by formation of chelates. However, data on metal interactions with anthocyanins are sparse. The main aim of this study was to perform the analysis of interaction of iron with elderberry fruit extracts as a rich and cheap source of anthocyanins with cyanidin as the aglycon. Vybrané alkoholové extrakty bezu černého pro využití v kosmetologii / Selected alcohol extracts of Sambucus nigra for use in cosmetology

This thesis focuses on the preparation of selected alcohol extracts of elderberry and their use in cosmetology. In the theoretical part are described the botanical properties of the elderberry (Sambucus nigra). There are introduced and characterized bioactive compounds and toxic compounds contained in all vegetative parts of the plant. Attention is mostly focused on compounds with antioxidant properties. The theoretical part also contains a description of basic analytical methods, which are used in the practical part to characterize compounds contained in elder flowers. The practical part focuses on the optimization of the preparation of alcohol extracts with the aim to prepare the extract with the greatest possible content of polyphenols with respect to economy and production costs. Factors such as selection of the appropriate solvent, solvent to plant material ratio, solvent temperature, and extraction time are examined. Based on the total polyphenol concentration measured spectrophotometrically by the Folin-Ciocalteu method, the most suitable solvent is 60% ethanol, the ratio of plant material to solvent is 1:8, the laboratory temperature of solvent and the extraction time of 24 hours.Under these conditions, an ethanol extract is prepared, analyzed and determined selected properties, such as refractive index and density. Rutin, chlorogenic, caffeic and ferulic acids are identified by HPLC with a DAD detector. The pH value of alcohol extract is 5.743 ± 0.006, the refractive index is 1.365 ± 0.02 at 22.7 ° C and the density is 0.9288 gcm-3. By liquid chromatography the concentration of chlorogenic acids is quantified as 0.104 gl-1, caffeic acid as 0.086 gl-1 and ferulic acid as 0.060 gl-1. The extract is incorporated into cosmetic products, that are used in balneology. Prepared products are toilet soap and bath salt.

An important part of the drinks are simple carbohydrates, because this thesis focuses on this issue. The theoretical part describes the characteristics , production , metabolism and the use of simple carbohydrates such as glucose, fructose and sucrose. There is also the definition of drinks and their main divisions . The literature review is based on the experimental part of analyzing selected beverages by high performance liquid chromatography with refractometric detection coupled to columns or alkyl ion-exchange phases. In conclusion, the results were compared to determine and comment . Nowadays the emphasis is on the correct functioning of the digestive tract. An important factor that supports the intestinal microflora is a prebiotic carbohydrate - tagatosa This could become tagatosa whose properties , production, metabolism in the body and any use is also included in the search . In the experimental part is listed possible optimization analysis of samples containing tagatosy on both types of columns in the HPLC system. The result is again commented in conclusion.

Analýza vybraných obsahových látek v extraktu z bezových větviček / Analysis of selected substances contained in the extract from elderberry twigs

My diploma thesis deals with the optimization and verification of conditions for separation of proteins, which are contained in the twigs of elder (Sambucus nigra L.) using the technique RP-HPLC. The measurements were made with the use of a HPLC system with a UV-VIS detector. Column Zorbax 300SB-C18 300 4,6 x 250 mm and particles the size of 5 microns were used. The theoretical part describes the attributes and usage of elder and especially its proteins. The basic characteristic of the used HPLC technique is introduced. The possibilities of how to identify proteins are described. The practical part demonstrates the individual steps of optimization of the HPLC method applied to the mixture of standard proteins. The application of this method in the real sample is also reported. The final part of the diploma thesis is focused on the comparison of analysed samples taken in different seasons.

In this thesis was determinated selected chemical and nutritional parameters in the wild elderberry and 17 cultivated varieties of elderberry. The fruits was provided by the Research and Breeding Institute of Pomology Holovousy Ltd. The theoretical part deals describes elderberry, chemical composition of elderberry, especially biological active substances and its using not only food industry. Closer attention has been focused on effect of saccharides, anthocyanins and methods of their determination, specially determination by HPLC. The experimental part describes the various methods of determination of selected chemical and nutritional parameters. Total dry matter, content of total polyphenolic compounds, monomeric anthocyanin pigment and antioxidant activity was defined in all varieties of elderberry fruits. For some varieties was defined total soluble dry matter, content of organic acids and saccharides by various methods. In the end was compared the values from different methods varieties.

Drug Sambuci nigrae flos - Sambuci nigrae flos is obtained from the wild plants. The drug is primarily used to help reduce the symptoms of colds and flu. Elder flower provides antioxidant, anti-inflammatory, anticancer and diuretic effects. The main substances are flavonoids and hydroxycinnamic acid. Elderflowers from bred cultivars are not used to obtain drug so far. The aim of this work was to determine antioxidant acitivitiy of extracts of eldeflower cultivars. Cultivars ′Albida′, ′Allesö′, ′Aurea′, ′Bohatka′, ′Dana′, ′Haschberg′, ′Heidegg 13′, ′Juicy′, ′Korsør′, ′Mammut′, ′Pregarten′, ′Riese aus Voßloch′, ′Sambo′, ′Sambu′, ′Samdal′, ′Sampo′, ′Samyl′, ′Tulbing′ and ′Weinhenstephan′ were analyzed. Antioxidant activity was measured spectrophotometrically using DPPH ((2,2-diphenyl-1-picrylhydrazyl) radical method. Results of antioxidant activity were expressed as IC50 (concentration required for 50% reduction of the DPPH radical). The measured values are in the range 0.0419 - 0.0772 mg/ml. Cultivars 'Heidegg 13', 'Albida', 'Tulbing' showed the highest antioxidant activity, whereas the lowest activity was found in 'Pregarten','Korsør' and 'Allesö' clutivars.
