Systémové zóny v kapilární elektroforéze / System zones in capillary electrophoresisRiesová, Martina January 2013 (has links)
Martina Riesová Abstract of Ph.D. Thesis Capillary electrophoresis is a method of choice in many analytical laboratories for its high separation efficiency, rapidity, low consumption of chemicals and therefore low costs. Inherent to each electrophoretic separation system are system peaks, which can significantly affect or confuse the electrophoretic results. In capillary zone electrophoresis, position of system zones can be predicted easily and reliably by means of prediction software based on a theoretical description of electromigration. However, the prediction of only position of a system zone may not be sufficient for identification of system peaks in obtained electropherograms. Therefore, an existing theoretical model was significantly extended and new version of PeakMaster software (PeakMaster 5.3) was introduced in the framework of this thesis. PeakMaster 5.3 enables to predict not only the positions of system zones, but also their shapes and polarity. Thus, PeakMaster 5.3 improves the prediction possibility of overlapping or interaction of system peaks with analyte peaks. Moreover, composition of the sample can be optimized in order to obtain convenient shapes and amplitudes of system peaks. The applicability of capillary zone electrophoresis can be extended by addition of a complexation agent into...

Enantioselektivní separační systémy v kapilární elektroforéze / Enantioselective separation systems in capillary electrophoresisSvobodová, Jana January 2012 (has links)
Chiral separation systems in capillary electrophoresis are in the scope of interest of many research groups all over the world. Therefore, the need to develop reliable theoretical models, which would help to explain phenomena connected with chiral separations or optimization of separation conditions, is obvious. In this thesis several mathematical models and approaches that can fulfill these requirements are presented. First part of the thesis deals with the determination of rate constants of interconversion of enantiomers by means of dynamic capillary electrophoresis. We focused on mixtures of chiral selectors and formulated a mathematical model, which enables to determine rate constants of interconversion in such systems. Mixtures of chiral selectors are very popular in separation practice due to their enhanced enantioselectivity. The theoretical model established in the thesis is able to explain the separation mechanisms of multi-chiral selector systems and to propose and verify possible ways of their optimization. In addition, the separation mechanism in systems with simultaneous cyclodextrin and borate complexation was revealed. Finally we present the complete mathematical model of electromigration in systems with complexation agents. The model is implemented into our simulation tool Simul 5...

Matematické modelování lineárních a nelineárních jevů v kapilární elektroforéze / Mathematical modelling of linear and nonlinear phenomena in capillary electrophoresisDvořák, Martin January 2019 (has links)
Capillary electrophoresis is one of the prominent analytical separation methods. Currently, many computer programs exist which are able to predict the result of an electrophoretic experiment. Firstly, there are programs based on numerical solving of corresponding continuity equations and equations of chemical equilibria. Secondly, there are programs based on approximative models of capillary electrophoresis. Programs belonging to the first group are applicable to a wide range of modes of capillary electrophoresis and provide a precise solution. Their disadvantage is though a considerable time demand. On the other hand, the approximative models give the results almost immediately and, in addition, provide some theoretical relationships which are useful for optimization of the separation process. This dissertation thesis is focused on improvement and extension of validity of existing approximative models of capillary electrophoresis. As a part of this thesis, a model capable of a full-blown description of capillary electrokinetic chromatography is introduced. This model is implemented into program PeakMaster 6. The attention is also paid to a nonlinear model of electromigration without diffusion. This model enables a very good description of electromigration dispersion including effects related to...

Softwarové aplikace pokročilých modelů elektromigrace / Application of advanced models of electromigration by means of computer softwareMalý, Michal January 2020 (has links)
Motion of ions under the influence of electric field has been a subject of scientific interest for many decades. Capillary electrophoresis in particular benefited greatly from this research and mathematical models of electromigration applicable to capillary electrophoresis have been developed. As the sophistication of the models grew, so did the computational demands to evaluate them. In order to fully exploit the possibilities of advanced mathematical models a computer implementation capable of solving non-trivial problems at sufficient speed is necessary. This dissertation thesis explores applications of computer implementations of mathematical models re- lated to electromigration in two different areas. The main focus of this thesis is on the topic of linear theory of electromigration. We discuss the extension of the linear theory of electromigration beyond of just acid-base equilibria and computer implementation of this extented theory which is specialized to include complex-forming equilibria in order to be able to deal with affinity capillary electrophoresis prob- lems. Some technical aspects of the computer implementation are also discussed. This is followed upon by investigating certain selected affinity capillary electrophoresis systems. The purpose of this investi- gation is to re-derive...
