EARLY LIFE EXPERIENCES INFLUENCE SEIZURE SUSCEPTIBILITY OF 14-DAY OLD RAT PUPS IN A DAM-DEPENDENT AND SEX-DEPENDENT MANNER

Epilepsy is a devastating disorder characterized by recurrent seizures. The pathophysiology of the disorder is not well understood. In this study the effects of early life, including pre- and post-natal, experiences on the seizure susceptibility of offspring was determined. Sprague-Dawley rats were air transported prior to breeding (In-House), on gestation day 9 (G9), or G16. The maternal behaviour was scored from P2-P13. On P14, seizure susceptibility of pups was assessed by randomly assigning the pups into Naïve (control), Saline, lipopolysaccharides (LPS; 200 ?g/kg), Kainic acid (KA; 1.75 mg/kg) or Febrile Convulsion (FC; LPS followed by KA) groups. No effect of prenatal transport was found on seizure susceptibility. Licking and grooming (LG) maternal behaviour was associated with higher FC seizure susceptibility of offspring. Male pups were more susceptible to FC seizure than female pups. These results emphasize the dam-dependent and sex-dependent effects of early life experiences on seizure susceptibility of offspring.

Genetická analýza vybrané dědičné choroby u psů

In my thesis I concentrated on an analysis of biological samples of 60 dogs, which manifested epilepsy. The aim was to find out the relation between genotype of the selected locus and the occurrence of epilepsy. Based on the results of the analysis, I assessed, whether there is a demonstrable relation between the occurrence of epilepsy and genotype in a particular locus. According to the results, no influence of genotype on the age of first epileptic seizure was proved.

Epilepsie: Wissen und Einstellungen von Physiotherapeuten, Ergotherapeuten und Logopäden

Physiotherapists, occupational therapists, and speech therapists play a key role in the treatment of children with epilepsy. We performed a survey of therapists’ knowledge of and attitudes towards epilepsy in two regions of Germany, the city of Leipzig and the rural district of Zwickau. Therapists of 29/68 (43%) outpatient practices and 4/9 (44%) hospitals took part. In total, 195 therapists participated: 63 (32%) physiotherapists, 74 (38%) occupational therapists, and 58 (30%) speech therapist. In 65%, epilepsy was subject of vocational training. Of all therapists, 8% claimed they had not treated epilepsy patients so far. During professional life, 43% had witnessed a seizure. Of all therapists, 44% correctly assumed a seizure could result in death. During a seizure, 42% would perform the obsolete measure of placing something solid in the patient’s mouth, and 41% would administer a prescribed rescue medication. More information on epilepsy was requested by 92%. Conclusion: Most therapists treat patients with epilepsy, and almost half have already witnessed a seizure. Often, however, epilepsy is not subject of vocational training. The risk of a fatal outcome of a seizure is underestimated, and many therapists would perform obsolete measures. Detekce vysokofrekvenční EEG aktivity u epileptických pacientů / Detection of High-Frequency EEG Activity in Epileptic Patients

Tato práce se zabývá automatickou detekcí vysokofrekvenčních oscilací jakožto moderního elektrofyziologického biomarkru epileptogenní tkáně v intrakraniálním EEG, jehož vizuální detekce je zdlouhavý proces, který je ovlivněn subjektivitou hodnotitele. Epilepsie je jedním z nejčastějších neurologických onemocnění postihující 1 % obyvatelstva. Přestože jsou přibližně dvě třetiny případů léčitelné farmakologicky, zbylá třetina pacientů je odkázána zejména na léčbu chirurgickým zákrokem, pro nějž je zapotřebí přesně lokalizovat ložisko patologické tkáně. Vysokofrekvenční oscilace jsou v posledním desetiletí studovány pro jejich potenciál lokalizace patologické tkáně. Součástí této práce je shrnutí dosavadního výzkumu vysokofrekvenčních oscilací a výčet detektorů používaných ve výzkumu. V rámci práce byly vyvinuty či vylepšeny tři detektory vysokofrekvenčních oscilací, na jejichž popis navazuje evaluace z hlediska shody s manuální detekcí, přesnosti výpočtu příznaků oscilací a schopnosti lokalizace patologické tkáně. V závěru práce jsou představeny vyvinuté metody vizualizace vysokofrekvenčních výskytu oscilací a stručně uvedeny dosažené vědecké výsledky.

Convolutional Neural Networks for Epileptic Seizure Prediction

Epilepsy is the most common neurological disorder and an accurate forecast of seizures would help to overcome the patient’s uncertainty and helplessness. In this contribution, we present and discuss a novel methodology for the classification of intracranial electroencephalography (iEEG) for seizure prediction. Contrary to previous approaches, we categorically refrain from an extraction of hand-crafted features and use a convolutional neural network (CNN) topology instead for both the determination of suitable signal characteristics and the binary classification of preictal and interictal segments. Three different models have been evaluated on public datasets with long-term recordings from four dogs and three patients. Overall, our findings demonstrate the general applicability. In this work we discuss the strengths and limitations of our methodology.
