Video rasterstereography of the spine and pelvis in eight erect positions: A reliability study

Yes / To investigate the reliability and variability of Video Rasterstereography (VR) measurements of the spine and pelvis, for eight proposed standing postures, in order to help define an optimal standing position for erect pelvis radiography. Methods: Surface topography data were collected using the formetic 4D dynamic modelling (Diers) system. 61 healthy participants were recruited; each participant performed eight different standing positions. Four positions were performed with the feet shoulder width apart and parallel, and four positions were performed with the feet shoulder width apart and internally rotated. For the upper extremity, each of the (two sets of) four positions were performed with different arm positions (arms by the sides, arms crossed over the chest, arms 30° flexed and touching the medial end of the clavicle, arms 30° flexed with the hands holding a support). Three sets of surface topography were collected in the eight positions (n = 24). The variability was assessed by calculating standard error of the measurement (SEm) and the coefficient of variation (CV). Reliability was assessed using intra-class correlation coefficients (ICC ± 95% CI). Results: No significant differences in the SEm were found between the three paired measurements for all standing positions (P > 0.05). ICC values demonstrated excellent reliability for all measurements across the eight standing positions (range 0.879–1.00 [95% CI 0.813–1.00]). Conclusion: Evaluating eight standing positions radiographically would be unethical as it would involve repeat radiation exposures. Using the formetic 4D dynamic modelling (Diers) system, provides an alternative and has shown that there was only a minimal, non-statistically significant, differences between the eight different standing positions. / KA is supported by the Hashemite University/ Jordan.

Le mégalithisme est l'expression architecturale monumentale d'un ensemble de sociétés qui ont édifiées des structures funéraires et des pierres dressées. La genèse du mégalithisme, phénomène humain de la fin de la préhistoire, puise ses origines dans de multiples conjugaisons des différents courants de la néolithisation aux traditions variées, aux influences géographiques parfois lointaines, aux époques différentes. La combinaison de nombreux éléments sociaux de ces sociétés agro-pastorales a généré des générations d’architectes bâtisseurs et contribué à mettre en œuvre une diversité architecturale La volonté de mise œuvre des maisons des morts au même niveau que les maisons des vivants semble répondre à des critères sociaux et culturels. Une perception visuelle actualisée des mégalithismes, adaptée aux récentes thématiques développées amène un fil conducteur original d’étude de plusieurs mondes qui se sont combinés. L’état des connaissances depuis le milieu du XXème siècle a permis une appréhension différente des mégalithismes : - dans les années 1980, une connaissance plus complète des masses tumulaires et leurs liens étroits avec les chambres sépulcrales (Joussaume, 1997, 1999, 2003 ; Joussaume et al. 2006). - dans les années 2000 une prise en compte de l’histoire des monuments (Joussaume et al. 2006 ; Laporte, 2010 ; Laporte et al. 2004, 2011). - de nos jours, une appréhension différente des articulations entre les pierres dressées, les tumulus et les chambres sépulcrales (Laporte, 2015 ; Laporte et al. 2011). Le processus de cette monumentalisation architecturale a souvent fait l’objet d’études distinctes, les espaces sépulcraux et les pierres dressées servant de bases à deux axes de recherches séparées. Seule l’étude des stèles en remploi avait fait l’objet d’une attention particulière (L’Helgouac’h, 1983 ; Cassen, 2009b) et d’un rapprochement des deux dispositifs. Ce n’est que récemment que la complémentarité entre les espaces sépulcraux et les pierres dressées a réellement été proposée (Laporte, 2015b). Disposant d’un important corpus actualisé des mégalithes du département du Morbihan, il a donc été proposé de développer dans cette thèse cette notion de complémentarité entre les différents dispositifs qui constituent les mégalithismes. Les hypothèses formulées sont de démontrer que les processus de monumentalisation sont issus d’un croisement architectural entre les pierres dressées, les espaces sépulcraux et les masses tumulaires. Nous tenterons également de montrer les liens étroits qui semblent se dessiner entre trois mondes très différents mais intimement liés (le monde des vivants, le monde des morts et le monde naturel). / Megalithism is the monumental architectural expression of a group of societies that have built funerary structures and erected stones. The genesis of megalithism, the human phenomenon of the end of prehistory, draws its origins from multiple conjugations of the different currents of neolithization with varied traditions, geographical influences sometimes distant, at different times. The combination of many social elements of these agro-pastoral societies has generated generations of architects and contributed to the implementation of architectural diversity The desire to put the houses of the dead to the same level as the houses of the living seems to meet social and cultural criteria. An updated visual perception of megalithisms, adapted to the recent themes developed, brings an original thread of study of several worlds that have combined. The state of knowledge since the middle of the 20th century allowed a different apprehension of megalithisms: - in the 1980s, a more complete knowledge of the tumular masses and their close links with the sepulchral chambers (Joussaume, 1997, 1999, 2003, Joussaume et al., 2006). - in the 2000s, taking into account the history of monuments (Joussaume et al., 2006, Laporte, 2010, Laporte et al., 2004, 2011). - today, a different apprehension of the articulations between the erected stones, the tumuli and the sepulchral chambers (Laporte, 2015, Laporte et al., 2011). The process of this architectural monumentalization has often been the subject of separate studies, sepulchral spaces and erected stones serving as bases for two separate lines of research. Only the study of stelae in reuse had been the object of particular attention (L'Helgouac'h, 1983, Cassen, 2009b) and a comparison of the two devices. It is only recently that the complementarity between sepulchral spaces and erected stones has actually been proposed (Laporte, 2015b). It has therefore been proposed to develop in this thesis the notion of complementarity between the different mechanisms that constitute megalithisms. The hypotheses formulated are to demonstrate that the processes of monumentalisation are the result of an architectural crossing between the erected stones, the sepulchral spaces and the tumular masses. We will also try to show the close ties that seem to be taking shape between three very different but intimately linked worlds (the world of the living, the world of the dead and the natural world).

Comparing the supine and erect pelvis radiographic examinations: an evaluation of anatomy, image quality and radiation dose

Yes / Objectives: Pelvis radiographs are usually acquired supine despite standing imaging reflecting functional anatomy. We compared supine and erect radiographic examinations for anatomical features, radiation dose and image quality. Methods: 60 patients underwent pelvis radiography in both supine and erect positions at the same examination appointment. Measures of body mass index and sagittal diameter were obtained. Images were evaluated using visual grading analysis and pelvic tilt was compared. Dose–area product values were recorded and inputted into the CalDose_X software to estimate effective dose (ED). The CalDose_X software allowed comparisons using data from the erect and supine sex-specific phantoms (MAX06 & FAX06). Results: Patient sagittal diameter was greater on standing with an average 20.6% increase at the iliac crest (median 30.0, interquartile range [26.0 to 34.0] cm), in comparison to the supine position [24.0 (22.3 to 28.0) cm; p < 0.001]. 57 (95%) patients had posterior pelvic tilt on weight-bearing. Erect image quality was significantly decreased with median image quality scores of 78% (69 to 85) compared to 87% for the supine position [81 to 91] (p < 0.001). In the erect position, the ED was 47% higher [0.17 (0.13 to 0.33) mSv vs 0.12 (0.08 to 0.18) mSv (p < 0.001)], influenced by the increased sagittal diameter. 42 (70%) patients preferred the standing examination. Conclusion: Patient diameter and pelvic tilt were altered on weightbearing. Erect images demonstrated an overall decrease in image quality with a higher radiation dose. Optimal acquisition parameters are required for erect pelvis radiography as the supine technique is not directly transferable. / College of Radiographers Industry Partnership Scheme grant.

Spatio-Temporal Control Of Drosophila Indirect Flight Muscle Development And Maintenance By The Transcription Factor Erect Wing

Muscle development involves concerted action of a repertoire of mechanisms governing myoblast proliferation, migration, fusion and differentiation. Subsequently, there are cellular events administrating proper muscle function and maintenance of muscle integrity. Chapter 1 covers what is known about muscle development, building up of mass and maintenance in vertebrates and Drosophila, highlighting the myogenic programs and factors that play a role in them. The formation of vertebrate skeletal muscles can be recapitulated in Drosophila indirect flight muscles (IFMs), making IFMs an interesting model to dissect and understand the mechanisms of muscle development and maintenance. The cellular and developmental events that occur during IFM development have been discussed in detail along with their genetic control which encompasses both cell autonomous and cell non-autonomous mechanisms. The fly resources and tools used for experimentations have been described in Chapter 2. One of the hallmark events during muscle development is myoblast fusion. Myoblasts are kept in undifferentiated state until they fuse through a balanced action of anti-differentiation and pro-differentiation factors. The swarming myoblasts are in semi-differentiated state and just prior to fusion should exit cell cycle to achieve terminal differentiation. The mechanisms of cyclin/CDK complexes and their regulation via CKI (CDK inhibitor) are known in a cell. However, tissue specific factors exerting additional control on molecules that participate in cell cycle have been proposed but have not been shown in vivo. Chapter 3 uncovers a novel role played by the transcription factor, Erect wing (Ewg) in IFM development and patterning. Despite the fact that Ewg is known to express in fusing myoblasts and nuclei of developing IFMs and has long been used as a nuclear marker for IFMs, the mechanism(s) behind Ewg‟s function has remained enigmatic. Historical perspective of Ewg has been presented in Chapter 1. One set of IFMs; dorsal longitudinal muscles (DLMs) require larval templates for their formation and the other set; dorsal ventral muscles (DVMs) form de novo. Chapter 3 shows that Ewg is required in a spatio-temporal fashion to initiate myoblast fusion process. In the absence of Ewg, the number of fusion events in a given time is reduced. In addition de novo fusion is observed in the region of DLM development just like DVM and overall IFM development is delayed resulting in an aberrant adult IFM pattern. Genetic studies undertaken reveal a requirement for Ewg in exerting a temporal control on myoblast fusion. This is achieved by down-regulating Cyclin E levels, as a result of which the myoblasts exit cell cycle at G1/S stage. Through this study the proposal for DLM development and pattern has been put forth as follows: i) appropriate progression of DLM development commences on synchronous exit of myoblasts from cell cycle. This function is facilitated by Ewg expression in fusing myoblasts assisting symmetrical DLM formation in hemithoraces. ii) DLM pattern of six muscles in each hemithorax is dependent on template survival which requires fusion of enough myoblasts and further subsequent fusion events to support the splitting of three larval templates or presumptive DLM. The muscles that develop should preserve their structural integrity for efficient functional output. Muscles perform extensive activities warranting high energy requirements. IFMs are widely utilised for thorax movements that aid flight. IFMs are exclusively oxidative in nature with upto 40% mass contributed by large mitochondria themselves. Chapter 4 describes yet another novel finding for Ewg function in IFM maintenance. Vertebrate homolog of Ewg is nuclear respiratory factor 1 (NRF1) known for its role in mitochondrial biogenesis. This prompted an investigation on the role of Ewg, if any, in mitochondrial function and IFM maintenance. In this chapter, Ewg null adult IFMs are shown to undergo degeneration. Mitochondria in these muscles show rounder and smaller phenotype. Mitochondria morphology is traced throughout Drosophila pupal DLM development and extensive fusion is observed in last one-fourth of pupal phase. In Ewg null condition transcripts of Opa1-like required for inner mitochondrial membrane fusion is found to be absent, suggesting lack of mitochondrial fusion behind the smaller mitochondrial morphology. This emerged as an intriguing problem since Ewg expression follows until sarcomerogenesis (formation of sarcomeres) initiates at mid pupal stage. Developmentally extending Ewg‟s expression beyond mid pupal stage is not observed to increase Opa1-like levels pointing an indirect regulation by Ewg. However, Opa1-like knock-down beyond mid pupal stage is not observed to result in any muscle or flight defect. It is thus proposed that Ewg expression early during muscle development helps to up-regulate Opa1-like levels needed to support mitochondrial growth and fusion. In addition, this chapter provides additional data on requirement of Opa1-like for maintenance of mitochondrial as well as muscle integrity. This is the first ever report of tissue specific temporal regulation of Opa1-like by Ewg. Chapter 3 and Chapter 4 conclude spatially segregated functional requirements of Ewg which are also mechanistically distinct. Expression in fusing myoblasts channelizes fusing myoblasts to exit cell cycle and undergo timely fusion saving the larval template, subsequent fusion assists template splitting thus forming the appropriate adult DLM pattern. On the other hand expression until mid pupal stage up-regulates Opa1-like expression, facilitating mitochondrial fusion during the late pupal stage. This as a result helps maintain structural integrity of muscles in the adult. Vertebrate skeletal muscle contains multiple muscle fibres that provide appropriate mass and size to muscles. As DLM share similarity in development to that of the vertebrate skeletal muscle, DLM organisation is studied to get insights into the mechanisms which regulate the process. Chapter 5 discusses role of nuclear number and nuclear activity in determining the DLM organisation. In order to alter nuclear number, myoblast population is reduced to amounts lesser than that of the wild type and to alter nuclear activity, two nuclear encoded genes Opa1-like and Marf , involved in inner and outer mitochondrial membrane fusion respectively have been knocked down in IFMs. First, the DLM organisation is established by comparing it to the vertebrate skeletal muscle organisation. This organisation is affected on lowering the number of myoblasts destined to fuse and form IFMs, without affecting the differentiation. On the other hand, when nuclear encoded mitochondrial fusion genes are knocked down, not only DLM organisation but their differentiation is also affected. A proposal for achieving DLM organisation has been presented which should also apply to vertebrate skeletal muscle given their developmental similarity. In conclusion, the studies decipher a novel mechanism by which a transcription factor, Ewg exerts a temporal control on myoblast fusions directly influencing progression of DLM formation, and thereby, symmetry and pattern. Moreover, Ewg is also shown here to regulate mitochondrial fusion during later pupal stages helping muscles to attain greater function and maintain structural integrity. Discovery of such regulatory mechanisms controlling mitochondrial dynamics in vertebrates can open up new avenues to understand and design new therapeutic approaches to tackle mitochondrial diseases. Additionally, myoblast fusion and hence myonuclear number and their efficient functioning are shown to be important determinants of muscle organisation. This has further implications in understanding and using stem cell science in dystrophic or atrophic or ageing related muscle loss and therapy.

Canopy Architecture and Plant Density Effect in Short-Season Chickpea (Cicer arietinum L.)

Chickpea (Cicer arietinum L.) production on the semi-arid Canadian Prairies is challenging due to a short growing season and low and variable moisture. The current recommended chickpea population density of 44 plants m-2 is based on preliminary studies and a narrow range of 20 to 50 plants m-2. The aims of this study were to i) determine optimum population density of varying chickpea canopy types, i.e., leaf type and growth habit, by investigating seed yield responses at 30 to 85 plants m-2 and ii) identify desirable parental traits for breeding programs by assessing growth and yield parameter responses to varying leaf types and growth habits at a range of population densities. Field experiments were conducted from 2002 to 2005. Canopy measurements and calculated variables included light interception, biomass, growth rate, seed yield, harvest index, ascochyta blight severity and radiation- and water use efficiencies. The plant density which produced the highest seed yield when averaged over years for each location for each treatment revealed that a plant density of at least 55 plants m-2 produced a 23% to 49% seed yield increase above that of the currently recommended plant density. This indicates that a higher seed yield average over the long term in spite of periodic low seed yield episodes will be more profitable to producers. Increasing plant density increased lowest pod height significantly in all except one location-year but did not explicitly increase ascochyta blight severity or decrease individual seed size. This suggests that increasing the recommended chickpea plant density on the Canadian Prairies will increase seed yield but would neither negatively impact individual seed size nor ascochyta blight severity, especially, when combined with good agronomic practices. Fern-leaved cultivars had significantly higher maximum intercepted light (62 to 91%), seed yield (136 to 369 g m-2), harvest index (0.33 to 0.53), yield-based water use efficiency (0.56 to 1.06 g m-2 mm-1) and lower ascochyta blight severity (3 to 27%) than the unifoliate cultivars in all location-years. The fern-leaved cultivars also tended to show significantly higher cumulative intercepted radiation (221 to 419 MJ m-2) and biomass (306 to 824 g m-2) but leaf type showed no consistent effect on radiation use efficiency. Cultivars with bushy growth habit generally performed better regarding maximum intercepted light (62 to 90%), cumulative intercepted radiation (233 to 421 MJ m-2), biomass (314 to 854 MJ m-2), seed yield (120 to 370 g m-2), harvest index (0.37 to 0.50), yield-based water use efficiency (0.56 to 1.06 g m-2 mm-1) and ascochyta blight severity (7 to 36%) than the erect cultivars. The overall performance of the spreading cultivar was generally intermediate between the bushy and erect cultivars except for ascochyta blight severity where the spreading cultivar exhibited significantly lower disease severity (3 to 36%). Radiation use efficiency was generally not influenced by growth habit. Increasing plant population density generally increased intercepted light, biomass and cumulative intercepted radiation on each sampling day after seeding resulting in a general increase in seed yield. Harvest index, however, remained constant and ascochyta blight severity was generally stable but radiation use efficiency decreased with increasing population density. Chickpea cultivars with fern leaves and bushy growth habit at higher than currently recommended population densities would best utilize the limited resources of the short-season Canadian prairie environment to maximize and stabilize seed yield.
