(English) Sex determination plays an important role in the viability of populations and species evolvability. This is one of the reasons why sex determination has become an important subject of many studies during more than the last 100 years. The thesis focuses on the evolution of sex determination systems in toxicoferan reptiles. Toxicofera is a group of squamate reptiles containing more than 6000 species. Their species richness is also reflected in the diversity of their sex determination systems. The presence of environmental sex determination (ESD) as well as genotypic sex determination (GSD) with either XX/XZ or ZZ/ZW sex chromosomes, was reported among the toxicoferan species; however, the current knowledge on sex determination in toxicoferan reptiles is not equally distributed across their lineages. The main aim of the theses is to expand our knowledge on sex chromosome evolution using cytogenetic methods in snakes, chameleons and anguimorphan lizards. The first part of the thesis deals with the sex chromosome evolution in caenophidian and henophidian snakes. It is focused mainly on the variability in the distribution of repetitive content as well as heterochromatinization of the W chromosome of caenophidian snakes. This thesis focuses on the storage location assignment problem using modern meta-heuristic techniques combined with realistic simulation. A graphical tool implemented as part of this work is capable of warehouse model creation, generation of synthetic customer orders, optimization of product allocation using state of the art techniques, extensive warehouse simulation, and a pathfinder capable of finding the shortest path for orders going through the system. The work presents the comparison between different approaches based on many parameters to reach the most efficient allocation of products to warehouse slots. The author conducted tests on an experimental warehouse featuring almost twice the throughput -- 57%. The benefit of this work is a possibility to create model of an already built warehouse and its simulation and optimization, driving impact on the throughput of the warehouse, saving the user's resources, or helping him in planning and bottle-neck identification. Furthermore, this thesis introduces a new approach to warehouse optimization and new optimization criteria.

This thesis deals with an evolutionary solving of the Rubik's cube. The worldwide known puzzle has been for several decades not only a toy for children and adults, but also almost a lifestyle for crowds of fans and definitely a big challenge in the world of computation, where scientists seek to find an effective automated solution. The potential for its solution could also be borne by evolutionary algorithms. The author of this thesis has developed an application employing, apart from genetic algorithms, also many advanced technics, such as linear genetic programming or local search. The goal of this special technics is to make the evolutionary process more effective. There have also been made tests of the crossover, the population size and the mutation probability influence. All the tests have been statistically evaluated.

Evolution of sex determining mechanisms in squamate reptiles (Reptilia: Squamata) Martina Pokorná Ph.D. thesis Abstract This Ph.D. thesis is focused on the evolution of sex determining mechanisms and genomes in squamate reptiles. It is based on three published articles and two manuscripts. The evolution of sex determining mechanisms, sex chromosomes and genomes, and their organisation, was studied on a wide phylogenetic scale of the whole group of squamate reptiles and some lineages of other Sauropsids, as well as on the small phylogenetic range as a detailed comparative study inside individual lineages of squamates. This thesis is based upon the use of classical cytogenetic methods, methods of molecular cytogenetic (especially fluorescent in situ hybridisation) and the results were analysed using phylogenetic approaches. The results and outputs of this study represent an important contribution to the general knowledge of the principals of sex determination and the evolution of these phenomena not only in squamate reptiles but also in the whole group of amniotes. Using the results obtained during the work on this thesis we can conclude that sex chromosomes evolved in particular lineages of amniotes independently. In the past decade XML became a wide-spread information exchange data model. Many XML users have many XML formats described by XML schemas. To ease the management of multiple XML schemas modeling similar reality, the conceptual model for XML was defined. With its definition came various challenges that needed to be further researched. This thesis focuses on two of those challenges. The first challenge is to manage the evolution of the multi-level conceptual model as the modeled reality, XML schemas and applications evolve in time. The second challenge is to allow the majority of users who already use XML schemas in their system without the conceptual model to use their schemas to semi-automatically create one. In addition a step towards integration of the conceptual modeling of XML and semantic web techniques was taken.

Fylogeneze a fylogeografie kaprovitých ryb rodu Pelasgus (Teleostei: Cyprinidae) / Phylogeny and phylogeography of the cyprinid fish genus Pelasgus (Teleostei: Cyprinidae)

The genus Pelasgus (Cyprinidae) is endemic to the southern part of the Balkan Peninsula and includes seven species. In this work, a multilocus approach has been applied to study phylogenetic relationships between the species and their populations and to revise their distribution areas. 180 specimens from 47 localities from 30 river drainages were analyzed, comprehensively covering the distribution range of the genus. Moreover, samples from type localities of all species were included in the analyses. Mitochondrial (cytochrome b) and nuclear markers (the first intron of ribosomal protein S7, recombination activating gene RAG1 and rhodopsin) were used. Existence of seven well supported lineages was revealed based on cytochrome b, which is the most variable marker. These lineages correspond to P. laconicus, P. marathonicus, P. minutus, P. stymphalicus, P. thesproticus, P. prespensis and Pelasgus sp. The most variable nuclear marker was first intron of S7, which provides almost the same results as cytochrome b, revealing six well supported lineages, whereas RAG1 and rhodopsin appear to be less informative, revealing only four well supported clades. These markers did not separate several species (P. marathonicus, P. stymphalicus, P. thesproticus, and Pelasgus sp.) due to low variability of the markers...

Snakes (Serpentes) are a group of squamate reptiles (Squamata) including more than 3600 species. The vast majority of snakes are from the group Caenophidia, which includes approximately 90% of all extant snake species and represents the most studied lineage. Squamate reptiles are variable in sex determination and genome organisation, however, caenophidian snakes are quite stable in these respects. The typical - and probably ancestral - snake karyotype is composed of 36 chromosomes with 16 macro- and 20 microchromosomes. In all snake species, genotypic sex determination (GSD) is expected and it was assumed until recently that all snakes possessed ZZ/ZW sex chromosomes. The main reason for this is that most of the studied snakes were from the group Caenophidia and in the rest of the snake lineages it was believed that their sex chromosomes were homologous. In fact, the sex chromosomes of non-caenophidian snakes have not yet been identified - with the single exception of Acrantophis dumerili. Nevertheless, a recent study showed that there was an independent evolution of XX/XY sex chromosomes in pythons and boas.
