Breeding of coniferous trees in the Czech Republic is undergoing an important development during last decades, especially thanks to molecular-genetic methods, which refine and simplify mapping of tree genotypes and the selection of superior genotypes. Recently, in the Czech Republic superior genotypes are selected based on forestry parameters (tree height, trunk diameter, and timber quality) what does not always correlate with the ability of a tree to resist abiotic and biotic stresses. Recently, there is an effort to include in the breeding also physiological parameters and select superior genotypes using nonspecific stress indicators, which are able to correspond better to tree fitness than the forestry growth parameters. The present thesis deals with genotypes of Scots pine (Pinus sylvestris L.) growing in seed orchards Doubrava and Silov in the Pilsen region in the Czech Republic. Seed orchards are tree plantations, which serve as a reserve of the genetically valuable reproduction material, they are parts of breeding programs. Pine needles were collected in July 2015 and analyzed for non-specific stress indicators - photosynthetic pigments, phenolics, lignin, cellulose and proline contents and leaf reflectance and fast chlorophyll fluorescence measurements. The thesis deals with the content of selected phenolic compounds in some species of the genus Amaranthus, in black elderberry (Sambucus nigra L.) and buckwheat (Fagopyrum esculentum M.). Phenolic compounds are a group of natural compounds exclusively vegetable character. Flavonoids represent only one group of phenolic compounds. Flavonoids show many positive biological effects, in particular act as antioxidants. Natural flavonoids may cause to prevent from coronary- heard diseases and other diseases associated with older age. In recent years the increased attention is paid to flavonoid investigation due to its biological effects. For the determination of phenolic substances there were used two independent analytical methods. There are the high-performance liquid chromatography (HPLC) and micellar electrokinetic capillary chromatography (MECC). The MECC method was used for determination rutin and free quercetin. The highest content of rutin was found in leaves of buckwheat (76,400 mg/kg of dry weight) and the lowest content of rutin was determined in buckwheat hulls. The highest content of rutin was observed in teas from buckwheat leaves and inflorescence. This amount of rutin corresponds with rutin content in more than two pills of Ascorutin (the most favourite flavonoid medicament in the Czech Republic) The HPLC method was used for quantitative determination of phenolic acids. The content of free quercetin was monitored in all samples. No free quercetin was found both in plant material and in samples of teas. The ethanolic extract from the elderberry inflorescence didn´t contain any free quercetin. Free quercetin wasn?t found in any further samples of teas, which were prepared by described methods.

The aim of the study was to determine the influence of phenolics content and inoculation with soil extract on microbial respiration, on the phenoloxidase (PhOx), peroxidase (PerOx) and newly Mn-peroxidase (MnP) activity in two dominating litter samples (Calamagrostis villosa and Picea abies) differing in phenolics content from Plešné and Čertovo lake watersheds. At PhOx and PerOx activity, the dependence on incubation temperature with L-DOPA was estimated. PhOx and MnP activities significantly increased with higher content of hardly decomposable phenolics, but decreased with water extractable phenolics content. Inoculation with soil extract had no influence on microbial respiration, enzyme activity, nor on decomposition of phenolics. Microbial respiration was significantly higher at 10°C, but average enzyme activity was comparable at 0 and 10°C. PhOx activities had temperature optimum higher than 22°C, while PerOx activities had temperature optimum at 0 - 15°C.

Extrakce anthokyanových barviv z bezinkových výlisků / Extraction of anthocyanin pigments from elderberries pomaceLošková, Tereza January 2018 (has links)
This Diploma thesis deals with the production of the concentrate from the Elderberry marc extract (Sambucus nigra L.). The basic chemical characteristics in this concentrate were determined. The theoretical part of the thesis deal with the botanic characteristic of the Elderberry and its berries. The characteristic of the phenolic compounds, mainly anhocyanins was summarized. The use of the anthocyanin pigments in the food industry, the possibilities of their extraction and the concentrate production was discussed. Thereafter the methods of the anthocyanin content and the phenolic content determination were described. In the experimental part of study, the production of the anthocyanin rich extract from Elderberry marc was optimized. The extraction of the marc by the 1:1 ethanol/water (V/V) solvent with the ratio marc/solvent 1:2 (w/V) showed the most convenient. The production of the concentrate followed. During the concentration on the rotary evaporator the pigment degradation was investigated. The degradation decreased with the decrease of the temperature. The final concentrate was prepared from the extract on the rotary evaporator by 45 °C for 15 minutes. This product had the 7times lower volume compared to the initial extract. In the final concentrate come of the chemical characteristic were determined. By liquid chromatography (HPLC) the major pigments cyanidin-3-sambubiosid and cyanidin-3-glucosid were determined. Their total content was determined to 34,9 gl-1. The total anthocyanin content was determined by pH differential method to 7,1 gl-1 (expressed as cyanidin-3-glucoside equivalent). The total phenolic content was determined to 27,1 gl-1 (expressed as gallic acid equivalent) by Folin-Ciocalteu method. By electron paramagnetic resonance the increase in antioxidant activity with the increase in concentration was found. Up to 42 aromatic compounds was determined in Elderberry marc, its extract and the final concentrate.

This diploma thesis deals with the stability and basic properties of combined beverages and fruit concentrates. The theoretical part describes the production and use of fruit concentrates. Furthermore, the work deals with anthocyanic pigments and phenolic substances. The principle and instrumentation of liquid chromatography and electron paramagnetic resonance were described. In the next subchapter, the methods of determination of total anthocyanins and phenolic substances were described. The experimental part of the thesis described the determination of soluble solids, viscosity and antioxidant activity. The total content of phenolic compounds were determined by the Folin-Ciocalteua method and the total anthocyanins were determined by the pH-differential method. In the samples were determined cyanidine-3-glucoside and cyanidin-3-galactoside by liquid chromatography.

Základní chemické vlastnosti plodů vybraných odrůd třešní / Basic chemical properties of fruits of selected cherry varietiesChmil, Vojtěch January 2019 (has links)
Diploma thesis is focused on chemical characteristics of nine selected sweet cherry varieties in nine parameters. Total and soluble solids were analyzed with these results 13.07–16.58 % for total and 14.67–19.50 °Brix for soluble solids, ash content 0,31–0,39 %, titratable acidity 5.44–9.91 g of malic acid per 1 kg of fresh cherries and formol number 14.45–23.73 ml of 0.1 M NaOH per 100 g of cherries. Then there were analyzed crude proteins content calculated from total nitrogen content determined by Kjeldahl method with results 0.41–0.68 % of crude proteins. Molecular absorption UV/VIS spectrophotometry was used for determination of total phenolic substances 0.30–0.89 g of gallic acid per 1 kg of cherries and total content of anthocyanins 128–848 mg of cyanidine-3-glucoside in 1 kg of cherries. Saccharides were determined by high performance liquid chromatography (HPLC) with ELSD detector. Content of glucose was 45.45–59.49 g/kg and fructose 46.88–60.01 g/kg. All results are compiled using tables and graphs and discussed. Experimental part of the thesis also describes principles and procedures of every analysis, so it can be reproduced. In theoretical part there is described botanical characteristic of cherry tree (Prunus avium L.), active substances contained in its fruits and their use in food industry. At the end of the theoretical part there is described instrumentation of high-performance liquid chromatography.

This Diploma thesis talks about the production of concentrates and lyophilisate from the extract of elderberry moldlings (elderberry, Sambuvus nigra L.) and by determination by chosen chemical characteristics of these products. The theoretical part includes the botanical characteristics of the plant species Sambucus nigra L. Next is the part that is dedicated to biologicaly active substances found in elderberry and it‘s possible usage in the food industry. Next is the part where the extraction, purification and indentification of anthocyanic dyes are described. In the next part is the analyrical method HPLC described, including the description of instrumentation. The last part talks about the description of the used methods: methods for the concentration of the extract, UV-VIS spectrophotometry, determination of phenolic compounds, determination of dry matter, determination of active acidity and the overall titrating acidity. The experimental part contained the optimalization of the process of producing the extract from elderberry moldings. The extract obtained was then concentrated for preservation and characterization purposes based on selected chemical properties. The total content of anthocyanin dyes was chosen as a quality parameter. The most suitable process for the preparation of the extract was chosen to be the ratio of moldings and solvent 1:2, 4 (m/V). The most suitable solvent was chosen to be a mixture of water and ethanol in a ratio of 1:1 (V/V). According to the needs of the submitter, an extract with the use of water as the extraction agent is also provided. The prepared extract was then concentrated on a vacuum evaporator or lyophilized for storage purposes. The dye degradation while using the evaporator was the lowest in a 40 ° C water bath. The final products were characterized by the determination of soluble solids, pH, titration acidity, total phenolic compounds and total anthocyanin dyes. The anthocyanin pigments contained identified by liquid chromatography are: cyanidine-3-sambubioside, cyanidine-3-glucoside, cyanidine-3-sambubioside-5-glucoside, and cyanidine-3,5-diglucoside The total content of anthocyanins in the concentrate was determined to be equivalent to cyanidine-3-glucoside at 2.8 gl-1 and in the lyophilisate 2.5 gl-1.

Diploma thesis is focused on the chemical analysis of the juices of black, red and white currant and jostaberry (Ribes a Ribes x culverwellii). The theoretical part is focused to the definition of fruit, fruit importance in human nutrition, description of Ribes and Ribes x culverwellii genuses, active substances in the currants and the jostaberry, description of selected chemical parameters and the determination of some of them. The procedures of determining for the following chemical parameters are described in the experimental section: determination of reducing sugars, determination of total and soluble solids, formol number, pH, titratable acidity, determination of total phenols and anthocyanins and determination of vitamin C in two samples jostaberry, six varieties of black currants, one variety of white currant and two varieties of red currants. Based on the evaluation of the obtained results, the comparison was made with different varieties and the thesis that "overall comparison of the different varieties in the content of phenolic compounds, vitamin C and anthocyanins appears to be the most promising variety of blackcurrant demon. It should however be noted that the highest content of phenolics and anthocyanins were determined in jostaberry„ has been suggested.

In this work, the application of boron-doped diamond electrodes with various surface pre- treatment were tested on selected oxidizable and reducible model compounds, specifically para and ortho cresols and 2'-deoxycytidine and 7-deazaadenosine labeled by 4-nitrophenyl triazole. The aim of the study was the investigation of electrochemical and adsorptive processes with respect to the development of electroanalytical methods of detection of the selected compounds, or utilization of boron-doped diamond electrodes as detectors in liquid-flow systems. Cresols were used as the model oxidizable compound. On O-terminated and polished boron- doped diamond electrodes, cresols provide oxidation signal owing to their oxidation to methylphenoxy radical at comparable potentials as on other carbon-based electrodes used as a reference in this work, i.e., glassy carbon electrode, pyrolytic graphite electrode, and carbon paste electrode. These carbon electrode materials show relatively high propensity to adsorption of reaction products, while such adsorption is minimal on boron-doped diamond. In situ anodic activation allows for a rapid regeneration of boron-doped diamond surface prior to each scan. Auto-fluorescence emission of plant tissues can be a powerful reporter on plant biochemistry and physiology since it originates in substances inherent to primary or secondary metabolism. Plant bodies contain a plethora of intrinsic fluorescent compounds emitting practically all wavelengths of visible light. Moreover, the spectrum of fluorescent reporter signals was recently extended by a variety of fluorescent proteins that provide a new tool to mark whole cells or sub-cellular structures, study protein localization and monitor gene expression and molecule interactions. The imaging of such fluorescence signals reveals a possibility to acquire the information from as many as millions of points simultaneously, in vivo and in a non-invasive way thereby preserving integrity of cells and whole organisms. Imaging is particularly suited to visualize heterogeneity such as a localized immune response to invading pathogens. It can be applied both at macro- as well as micro-scales in two and three dimensions. The recent advancement in microscopy, the multi-photon microscopy, has made possible to monitor fluorescence signals, such as NAD(P)H fluorescence from intact leaf interior, that have been hidden to single-photon techniques.
