Adaptive population shifts in response to climate change Ing. Michaela Horníková, Doctoral thesis Abstract Understanding of species' reactions to past climate and environmental changes is a hot topic in many fields of biology as it is relevant also for addressing species' future under the contemporary climate change. Using an emerging model species, the bank vole, I combine genomic phylogeographic data with information on known intraspecific functional variability and environmental niche modelling and aim to elucidate the particular role of intraspecific variation and ultimately selection in shaping the species' response to the climatic and environmental changes after the end of the last glaciation. Based on the mtDNA markers, bank voles exhibit a complex phylogeographic pattern suggesting population replacement events during the postglacial recolonization of Europe and thus possible involvement of selection in the process. An extensive dataset of more than 6000 SNPs was used to search for signs of population replacement in the bank vole genomic DNA and to investigate the species' postglacial recolonization history throughout its European distribution range. Sheep breeding is nowadays a developing branch of agriculture again. There is a lot of different breed and they react distinctly on the same conditions of the enviroment. Sheep of breeds charollais, suffolk, šumavská ovce and valaška bred in similar conditions were observed in spring and autumn of years 2007 and 2008. In these seasons were taking blood samples (from {$\pm$}7{--}24) ewes and lambs and were analyzed in hematology laboratory. There were determined haemoglobin level, haematocrit indicator, erytrocytes and leucocytes levels, glucose, cholesterol, triglycerides levels, urea and plasmatic proteins, activity of ALP and GMT enzymes, phosphor, calcium, magnesium, zinc and copper levels. The main objective of this project was determine seasonal changes in observed parametres. Next objectives were determine different changes in blood parametres in different breeds and different aimes of breeds. There were recognized that all the observed breeds don`t react the same way on similar conditions. There were not provably determined seasonal changes in observed parametres, but average Hb level was higher in autumn than in spring. Urea level was conversely higher in spring season than in autumn. The demostrable fact is, that the similar conditions induce different answers not only in different breeds but also in different aimes of breeds.

Aims: 1. Determination of AGEs (Advanced Glycation End products) in patients with various types of nephropathy. 2. Association AGEs with nutritional parameters and anemia. 3. Influence of renal parameters on sRAGE (soluble form of Receptor for Advanced Glycation End products) levels. 4. Technics and proceeding methods of the podocytes cultivation. 5. Determination of urine podocytes. Methods: We determined fluorescent AGEs by spectrofluorometry, sRAGE by Enzyme-Linked ImmunoSorbent Assay (ELISA). Podocytes were passaged and identified immunocytochemically. Podocytes in urine were specified by flow cytometry method. Results: 1. We did not find significant differences in AGEs serum levels among various types of nephropathy, even though the pathogenesis differs. 2. The albumin and prealbumin levels positively and haemoglobin levels negatively correlate with AGEs in patients with CKD grade 1-5, without necessity of dialysis. 3. Serum sRAGE levels are increased in patients with decreased renal function independently on the course of renal disease. 4. We implemented the methods and technics of podocyte cultivation. 5.
