Yes / Context: Urinary biomarkers are promising as simple alternatives to cystoscopy for the diagnosis of de novo and recurrent bladder cancer. Objective: To identify a highly sensitive and specific biomarker candidate set with potential clinical utility in bladder cancer. Materials and methods: Urinary biomarkers concentrations were determined by ELISA. The performance of individual markers and marker combinations was assessed using ROC analysis. Results: A 5-biomarker panel (IL8, MMP9, VEGFA, PTGS2 and EN2) was defined from the candidate set. Discussion and conclusion: This panel showed a better overall performance than the best individual marker. TRAITS UNDERLYING INVASIVENESS: A COMPARISON OF WIDESPREAD AND ENDEMIC SPECIES IN THE GENUS GAMBUSIA (POECILIIDAE)

Due to the irreversible nature of biological invasions, prediction has been a key area of emphasis in invasion biology. Specifically, the degree to which species-specific traits may help us predict invasion success is a core issue in the field. My research examined a series of traits and asked whether they were good predictors of invasion success, particularly establishment success. I compared traits among four species of the poeciliid fish Gambusia, two of them highly invasive (G. affinis and G. holbrooki) and two of them non-invasive (G. hispaniolae and G. geiseri).I examined abiotic tolerances, feeding behavior, behavioral responses to novel predation and competition, life histories, and dispersal tendencies. I found the invasive Gambusia species to be more tolerant of low temperatures and to exhibit higher feeding rates and dispersal tendencies than non-invasives. Invasive species were more likely to respond appropriately to novel predation by reducing foraging and activity level and by increasing refuge use, and less likely to show lower foraging success when faced with competitors. Invasives exhibited higher fecundity and juvenile growth rates, and consequently reached maturity sooner than non-invasives. I found no differences in the species' diet breadth or aggressiveness.I then simulated the invasions of simplified pond communities and measured establishment success (with and without novel competitors) and community impact by tracking population trajectories over several months. As predicted from the trait comparisons, I found that in both simulations invasive Gambusia outperformed non-invasives by achieving andvmaintaining larger populations. In the first experiment, only invasive Gambusia were able to successfully establish (non-invasive populations had zero survival). In the second experiment, invasive Gambusia populations were better able to cope with competition and had greater community impact on lower trophic levels than the non-invasives.Overall, species traits were good predictors of establishment success. A species' ability to cope with the abiotic conditions of the invaded community seemed particularly important to whether or not establishment occurred in the study communities. Life history traits and the species' ability to cope with biotic interactions were important to determining the level of establishment species achieved if invaders survived the novel abiotic element.

The Thin Green Line: A Framework for Evaluating the Invasive Potential of Bioenergy Crops

The 2007 Energy Independence and Security Act mandates the production of 135 billion liters of transportation fuel achieved through the use of alternative energy sources. The most economically and sustainably competitive bioenergy crops will need to be high yielding, perennial species which require minimal inputs coupled with the ability to grow on marginal land. Grower adoption will be imperative to the success of this industry. Our results indicate that after three years of growth, several species including Arundo donax and Miscanthus × giganteus are able to produce up to 48.2 and 61.4 Mg ha⁻¹ dry weight of feedstock respectively. We also found that weed management may not be necessary under ideal growing conditions, but a group of herbicides appear promising for preemergence and postemergence application timings, tested under greenhouse conditions. The herbicides pyroxasulfone, sethoxydim and nicosulfuron may prove to be useful tools for the control of bioenergy crops if they escape cultivation. The potential for many of these exotic and highly competitive species to become invasive has hindered advancement of this industry. We develop a beginning framework to evaluate the potential for seeded Miscanthus × giganteus to become invasive, as no proven formula currently exists. We developed a novel methodology to assess the invasive potential of bioenergy crops across a continuum of invasiveness, which proved to be critical to the interpretation of our results. This methodology allows us to make relative comparisons of risk, evaluating our otherwise isolated results in broader context. We chose to evaluate M. × giganteus in a direct comparison with known exotic invasives, as well as with species that are known not to be invasive. We use a tiered approach, combining qualitative risk assessment models with quantitative field trials. Results from two weed risk assessment models indicate that many bioenergy feedstocks are of high risk, but agronomic crops such as Oryza sativa were also found to be high risk. Subsequent field trials indicate that seedling establishment for M. × giganteus in perennial systems may be low as only 0.1% of emerged seedlings survived after six months, while up to 10% of the invasive Microstegium vimineum survived. Spread of fertile M. × giganteus seedlings into annual systems with low competition had greater success; we observed 316 culms m⁻², at a distance of 1.5 m from the planted plot, after three years of growth. The observed spread of fertile M. × giganteus, in the low competition environment, did not differ from that of our positive control species; however, spread of fertile M. × giganteus seedlings into areas of high competition was significantly lower than our positive controls, up to 3 m from the cultivated plot. Importantly, A. donax and sterile M. × giganteus never spread from the cultivated plot. Results from our study and application of this novel methodology will help to prepare for the expansion of this burgeoning industry while providing useful information for the implementation of best management practices and improved stewardship. / Ph. An in vitro study of human melanoma tumor cell metastasis: Cytological and molecular events during extravasation.

In order to study the process by which human melanoma cells achieve invasion of basement membranes, a modification of the Membrane Invasion Culture System was developed to allow the in vitro collection of invasive tumor cells from heterogeneous tumor cell populations. A significant increase in the number of double minute chromosomes was observed in metaphase nuclei of the low metastatic A375P human melanoma cells which had invaded 2 consecutive amniotic membranes over that of cells in the control groups. After 25 days in culture, the incidence of double minutes had dropped below the control range. These data indicate that an unstable gene amplification event may be part of the process by which melanoma cells execute invasion through basement membranes. A375P cells which had invaded 1, 3 and 5 consecutive basement membrane-coated filters were established and compared with the parental cell line and a highly metastatic subclonal line for the following characteristics: (a) in vitro invasive potential, (b) mRNA expression of several oncogenes, (c) expression of laminin receptor, at the (cell surface) protein and mRNA levels, and, (d) secretion of endogenous laminin. There was a progressive increase in invasive potential and expression of endogenous laminin and laminin receptor which correlated with the number of membrane-coated filters through which the A375P cells had been selected. There were significant increases in the steady-state mRNA expression of c-myc and c-fos, a decrease in c-jun, and no change in Ha-ras, that correlated with increases in the invasive and metastatic potential of the cells. A novel in vitro adhesion assay was developed to study the interaction of tumor cells with lymphatic endothelium, the first step of extravasation from the lymphatic vessel. Human tumor cells from: one primary Ewing sarcoma, two melanoma, two colon and two breast carcinomas were assayed for their ability to attach to monolayers of lymphatic endothelium. There was a clear positive correlation between the metastatic potential and attachment potential of the melanoma cell lines. Overall, these data suggested that highly fibroblastic established tumor cell lines were more adaptive in rapid adhesion than primary tumor cell cultures with a more rounded morphology.
