This thesis examines from theatrical point of view the Janáčkian dramaturgy in German speaking countries within the context of local reception of Czech and Slovak opera and with special consideration of the Bavarian State Opera in Munich. The main referential base of research is German daily critique through which the Author presents an overview of more important productions of Leoš Janáček's operas. The Author then discusses the historical context of these productions, the stage history, German Janáčkian programme and roots of modern-day social-critical expressive staging of Janáček in German speaking area as compared to the more lyrical realistic Czech approach (to simplify). The discussed period is concluded with the year 1998.

The diploma thesis "Cross-Curricular topics in the teaching of mathematics at primary schools" describes possible ways of involving personality and social education, education of a democratic citizen, education towards thinking in European and global contexts, multicultural education, environmental education and media education in primary school mathematics. The thesis is divided into a theoretical, practical and an evaluative section. The theoretical part of this thesis analyses domestic and foreign findings on cross-curricular topics, outlines the historical development of the curriculum in relation to this issue and characterizes the educational area of "Mathematics and its application RVP ZV". It further focuses on interconnecting particular cross-curricular subjects with mathematics and examines their integration in current mathematics schoolbooks. The practical part contains proposals of activities (methodical activity sheets) connecting the area of "Mathematics and its application" with cross-curricular topics. It further presents organizational teaching forms and work methods used in activities including their descriptions. The last part evaluates the application of the activities in practice in terms of the development of pre-assigned mathematical skills and knowledge and the involvement of thematic units of cross-curricular topics.

My thesis ,,Possibilities of solving of unemployment in the Czech Republic and in France" has the objective of insight to the issue of solving of unemployment from the perspective of the state and its employment policy, especially to instruments of active employment policy in our republic and in France. My thesis is divided into nine chapters. The research part of the thesis deals with theoretical knowledge from areas as labour market, employment, unemployment and employment policies on the level of European Union and on the level of given surveyed states. At its end I made a short preview to the issue of macroeconomical indicators and national economy that I later used in the practical part for brief characteristics of economical situation of the states and their chosen parts. As I have mentioned above, the practical part of my thesis deals with economical characteristics of both surveyed states and analysis of unemployment in them. Its main part is formed by my focusing on the comparison of possibilities of solving of unemployment in selected higher territorial self-governing units of the Czech Republic and France. Here I perform analysis of the labour market according to different criteria in the Czech region Highlands and in the French region Limousin. Then I define and create a comparison of the particular ways of solving of unemployment in these areas. In my thesis I further express my own assessment of effectiveness of the Active employment policy operating both in the Czech Republic specifically in the Highland region and in France in mentioned Limousin region. The very end contains mainly summary of knowledge gained at the practical part and statement how the set target was achieved.

Vliv malých DNA virů na regulaci tvorby interferónu / Effect of small DNA viruses on regulation of interferon production

Plasmacytoid dendritic cells (pDC) represent innate immune cells capable to detect viruses in their endosomal environment via Toll-like receptors (TLRs). Viral nuclear acid recognition leads to the massive production of type I interferon (IFN I) and induction of the antiviral state in uninfected cells. Crosslinking of the surface regulatory receptors, such as BDCA-2, with monoclonal antibodies or with some viruses leads to the activation of MEK1/2- ERK signaling pathway and inhibition of IFN I production in pDC. In this study, the role of MEK1/2 kinase has been highlighted. Its inhibition reversed the inhibitory effect of BDCA-2 crosslinking and its direct activation with PMA led to the inhibition of IFN-α production. Yet an unclear role of pDC in sensing of BK polyomavirus virus (BKV) responsible for kidney transplant rejection was investigated as a major topic of this thesis. Experiments with the pDC cell line Gen2.2 and HRPTEC primary cell line showed that pDCs were not able to detect BKV particles, however, exposure of activated Gen2.2 cells to BKV inoculum dramatically upregulated production of IFN-α. Most importantly, coculture of Gen2.2 cells with BKV- infected HRPTEC cells resulted in IFN-α and TNF-α production, which was prevented by Bafilomycin. These results suggest that BKV-infected...

Gama záření geologických objektů a jejich lokalizace radiometrickým měřením / Gamma radiation of geological objects and their localization by radiometric measurement

This thesis deals with gamma-ray field of size-limited uranium objects and with possibilities of their localization by a field radiometric measurement. The gamma-ray field attenuates progressively with distance from the ground source of radiation. The applied method of radiometric survey plays an important role in localization of a radioactive object. The gamma- ray field of uranium anomalous objects was studied for variable signatures of ground anomalous objects and different methods of gamma-ray survey by using mathematical modelling. The method of modelling was derived from fundamental theoretical laws describing the gamma-ray field of a point source. The verification of applied mathematical modelling was carried out by the comparison of calculated model of the gamma-ray field at three real uranium anomalous objects with experimental data of ground and mini-airborne measurement. Mini-airborne measurement was realized by the prototype of a gamma spectrometer Georadis D230A. This instrument is characterized by relatively high sensitivity compared to previously used similar devices. Airborne platform was a hexacopter Kingfischer. Vliv malých DNA virů na funkci plasmacytoidních dendritických buněk / Effect of small DNA viruses on function of plasmacytoid dendritic cells

Plasmacytoid dendritic cells (pDC) are a highly specialized subset of immune cells that sense viral nucleic acids by endosomal toll-like receptors 7 and 9 (TLR7/9). Activation of TLR7/9 leads to the production of type I interferons (IFN-I). Moreover, pDC contribute to the antiviral response by presenting viral antigens to T lymphocytes and link innate and adaptive immunity. pDC need to be properly regulated in order to limit excessive production of IFN-I that is associated with autoimmune diseases. Therefore, pDC possess a battery of regulatory receptors (RR) that limit TLR7/9-mediated cytokine production. This thesis focuses on the mechanism of RR-mediated inhibition of IFN-I production in pDC and explores interactions between pDC and two enveloped viruses, that possess the ability to hijack RR in pDC: hepatitis B virus (HBV) and human immunodeficiency virus (HIV). We showed, that MEK-ERK signaling pathway plays an active role in RR-mediated inhibition of IFN-I in pDC. Our results indicate that in line with other studies of our group, pharmacological targeting of MEK1/2-ERK signaling could be a strategy to re-establish immunogenic activity of pDC. 1 Abstract The Podgorica Assembly 1918, its causes and consequences This dissertation deals with the causes and consequences of the Podgorica Assembly (Serbian: Подгоричка скупштина / Podgorička skupština) held in late 1918. As this work is a continuation of my doctoral thesis entitled The Origins of Constitutionalism and Parliamentarianism in Montenegro in the 1918, let me briefly outline the content of my doctoral thesis as it already analyzed some of the primary causes of the Podgorica Assembly (1918). The doctoral thesis deals with the Constitution of the Principality of Montenegro in 1905, its model rules, the process of drafting and adoption of the Constitution as well as with the analysis of the particular provisions of the Constitution and its comparison with the relevant provisions of other selected European Constitutions of that time.
