In the experimental part of the work will be carried out techno-economic analysis of the suitability of each technology for curing parts in a conventional and catalytic infrared oven.

Katalytické a adsorpční vlastnosti papainu a jeho derivátů / Catalytic and adsorption properties of papain and its derivatives

The aminoacid sequence of papain (EC 3.4.22.2) consists of 212 aminoacids. It has only one free sulfhydryl group, which is located in the active site of the protein. Some organometallic complexes could be bonded only to this free -SH group due to their structure. The artificial metalloproteins synthesised by this way may have different electrochemical properties. In this work, we have studied the electrochemical properties of papain and its derivatives. We compared the ability of papain and its three artificial derivatives to catalyse the hydrogen evolution by the chronopotenciometry. The work was completed by the study of the electrochemical properties of the organometallic complexes of ruthenium, which were used for the artificial metalloprotein preparation. The electrochemical properties of the compounds were never studied before. The process of the hydrogen evolution catalysed by the proteins is held in the adsorbed state of the catalyst. Due to this fact we have also studied the adsorption properties of papain on the substrates with different level of hydrofobicity. (In Czech)

Příprava nízkodimenzionálních III-V polovodičů / Preparation of low-dimensional III-V semiconductors

Tato diplomová práce se zabývá přípravou nanostruktur z indium arsenidu (InAs) pomocí metody molekulární svazkové epitaxe (MBE). Důraz je kladen na výrobu struktur ve formě nanodrátů na křemíkovém substrátu. V úvodní části práce je popsána motivace pro studium III-V polovodičů a konkrétně InAs. Následující kapitoly vysvětlují dva základní princpy tvorby nanodrátů. Experimentální část práce diskutuje možnost přípravy indiového katalyzátoru pro samokatalyzovaný růst InAs nanodrátů v konkrétní aparatuře MBE. Následuje prezentace výsledků růstu InAs nanodrátů mechanismem selektivní epitaxe (SAE). Nanodráty byly vyrobeny na substrátu s termálně dekomponovaným oxidem a rovněž na substrátech s litograficky připravenou oxidovou maskou.

Predikce koroze trubek pece s využitím provozních dat / Prediction of furnace tubes corrosion using operating data

The thesis deals with the modeling and prediction of corrosion of radiation tube snake in the heating furnace. Specifically it is focused on vertical cylindrical furnace which is included in the catalytic hydrocracking unit and serves for heating aggressive circulation gas which is the cause of high temperature corrosion. An important basis for the creation of computational models are available records about the operation of the furnace and about the corrosion and degradation mechanisms during the lifetime of the tube system in furnace. Such information enables the creation of a computational model which is based on the prediction of high-temperature corrosive damage of radiation tube snake. The computational model involving all relevant factors may serve as the basis for a predictive life management system of radiation snakes in the heating furnace.

Studium ligandů fosfatas z rodiny haloacidních dehalogenas / Study of Ligands for Phosphatases from the Haloacid Dehalogenase Superfamily

Phosphatases of the haloacid dehalogenase superfamily are one of the cell's tools for dephosphorylation of many diverse endogenous and exogenous compounds. This work is aimed at enzymes Tt82 and cytosolic purine 5'-nucleotidase II (cN-II), two members of this large enzyme superfamily. The Tt82 originates in the hyperthermophilic archaeon Thermococcus thioreducens. Up to date, there is only a small amount of knowledge about properties and biological function of this enzyme. Based on its sequence and structure, it was predicted that the Tt82 should possess a phosphatase catalytic activity. Consequently, potential substrates of the Tt82 were proposed by the molecular docking. In this work, the phosphatase activity of the Tt82 was confirmed together with several of its substrates: AMP, D-glucose 1-phosphate, D-glucose 6-phosphate and p-nitrophenyl phosphate (pNPP). Activity towards AMP and pNPP was then characterized by steady-state kinetics at 37 řC and 60 řC. In consistence with its thermophilic origin, the Tt82 showed markedly higher activity towards both substrates at 60 řC. Nonetheless, the effectivity of the Tt82 catalytic activity towards these substrates was actually very low.
