The present work maps a specific set of the traditional subsistence techniques by way of example of the abandoned village named Nová Ves u Klikova (1792-1953). In the 18th century occure changes in methods of forest exploitation in Central Europe in general. The increased demand for fuelwood and building wood gave rise to the "rationalization" of the forest management. Another source of energy for the developing industry was peat at that time. Nová Ves u Klikova was an example of the so called peat colonization. Small farmsteads were founded on the infertile peaty soil that was not able to ensure sufficient means of subsistence. As a consequence it was developed an important centre of production of straw baskets and chip baskets in the second half of the 19th century. The existing ethnological literature pursued the local production of baskets, whereas the topic of the cutting of peat and its further treating in relation to folk culture is still untreated. In view of that fact this text lays stress on the "cultural history of peat".

1 ABSTRACT The work deals with the infiltration process and the emphyteutic law evidence in the Čáslav province in the Middle ages. This area can be delimitated with the range of the Iron Mountains from the north, and then with the river Sázava from the south. Several significant territorial suzerains (authorities) acted here such as Vilémov monastery, Sedlec monastery and Želivka monastery, especially significant was the colonization activity of the Lichtenberk family which was developing in the area from the half of the 13th century. The Lichtenberks came to the area from north-western Bohemia, from the contact area of the Czech state, where their relatively early-ripening emphyteutic contracts are documented, and infiltration of early forms of the German law is presumed in general. This medieval law, sometimes designated as "stable and firm" in written sources, was brought from the area of present western Europe (also form the Netherlands) by new settlers, colonists who were bringing with them excluding new knowledge of landscape cultivation (drainage, forest transformation into fields) also juridical habits, which were used to adjust legislative relations while establishing villages. Anotation The Bohemian kingdom lost its ruler and shortly after that the throne of St. Adalbert lost its bishop; Tobias of Benešov (bishop 1278-1296, known as Tobias of Bechyně) succeeded Jan III. in hardly enviable conditions under the regency of Ota's V. of Brandenburg. Despite that (or maybe thanks to that) he succeeded in continuing the reform of the Diocese of Prague not only in the ecclesiastical management, but also in settling the individual bishopric's districts' issues of property and economic. A set of more than 200 formularies, also known as The Formulary of Tobias of Bechyně, as well as further first-hand or indirect sources, allow us to recognize those issues. This section comprises of an economic history in a narrower sense and also the history of settlement, archaeology and other related branches. As the name suggests, I intend to view Tobias' personality mainly from three essential points of view - firstly as an administrator of the diocese who is responsible for the condition of his subordinate and entrusted clergy, secondly as a responsible administrator of a large property domain and finally as a politically active person. Abandoned industrial-waste deposits (incl. fly ash or tailings ponds) represent very specific and mostly extreme habitat conditions. They offer unique opportunity to study primary succession in cultural landscape. Substrate of the material deposited here shows extreme properties, such as low pH, overheating of the open surface, salinization etc. The aim of this work is to map the colonization of different successional stages of vegetation by ants after years of abandonment of ore-waste deposits in Chvaletice (Eastern Bohemia, CR) and to compare the present state with analogous study made in 2001. Particular aims of both studies (Jarešová 2001, and present study, 2011-12) is to test the influence of ants on vegetation succession. During the years 2011-2012 several prospections were made to record species diversity of ants. On the surface plateau of the sedimentation basin two types of habitats were chosen - the areas with present ant nests and the other areas without them. Each of both types of habitats exhibits different plant species richness: separately the species-area curves were constructed and explained. In my dissertation, I inquire into the phenomenon of monastic architecture and its plastic decoration in 16th century New Spain. The aim of this work is to present Mexican monasteries and their artistic significance. In my analysis, I build on the interpretation of monastic architecture from the perspective of early colonial society, I also reflect on the individual level, assessing its impact on the conquest and colonization of America. The methodology of this research project combines analysis of historical (e.g. study of historical sources) and art historical processes (iconological analysis of E. Panofsky) with analysis of anthropological concepts (e.g. rituals of transition of A. van Gennep, thick description of Cl. Geertz, concepts of liminality, structure and communitas of V. Turner). During my PhD studies, I conducted a field trip in Central Mexico (2013), supported by a grant from the Government of Mexico. I visited and documented one hundred and sixteen monasteries and the photographic documentation is the main empirical source of my analysis which further draws from the study of historical sources and academic literature. Adaptive population shifts in response to climate change Ing. Michaela Horníková, Doctoral thesis Abstract Understanding of species' reactions to past climate and environmental changes is a hot topic in many fields of biology as it is relevant also for addressing species' future under the contemporary climate change. Using an emerging model species, the bank vole, I combine genomic phylogeographic data with information on known intraspecific functional variability and environmental niche modelling and aim to elucidate the particular role of intraspecific variation and ultimately selection in shaping the species' response to the climatic and environmental changes after the end of the last glaciation. Based on the mtDNA markers, bank voles exhibit a complex phylogeographic pattern suggesting population replacement events during the postglacial recolonization of Europe and thus possible involvement of selection in the process. An extensive dataset of more than 6000 SNPs was used to search for signs of population replacement in the bank vole genomic DNA and to investigate the species' postglacial recolonization history throughout its European distribution range. Vliv mikrobiomu na karcinogenezi / Microbiota as a modulator of carcinogenesis

Many studies show the ability of gut microbes to modulate the anti-tumour immune response by direct triggering the immune cells or by bacterial metabolites. Interestingly bacteria may even migrate to the tumour tissue and orchestrate the immune response on site. These anti-tumour effects can be improved by the administration of immune checkpoint inhibitors (ICI). Notably, some microbial effects occur only in the presence of ICI. On the contrary, microbiota may also promote tumour growth and negatively impact the effects of ICI therapy. We have disrupted the gut microbiota homeostasis by antibiotics (ATB) to study the effects of gut microbiota on the ICI. This disturbance led surprisingly to reduced tumour growth and enhanced pro-inflammatory immune response not only in the gut but also within the tumour tissue, where especially IFN-γ orchestrated the anti-tumour immune response. Importantly the anti-tumour immune response could be transferred through colonisation of germ-free mice by ATB-changed gut microbiota if concomitantly anti- programmed cell death protein 1 (αPD-1) monoclonal antibody was administrated. The use of probiotics has emerged in the last decades as a promising strategy when it comes to the treatment of inflammatory diseases. Through modulation of composition of the intestinal microbiota and the signalling it provides, probiotics can favourably tune the immune system. Beneficial effects of probiotic treatment have been documented in multiple animal inflammatory disease models. The effect of probiotic treatment on uveitis-a sight- threatening disease-has however not yet been described. In our study, we have tested two commercially available probiotics-Escherichia coli Nissle 1917 (EcN) and Escherichia coli O83:K24:H31 (EcO)-in the treatment of experimental autoimmune uveitis (EAU). The disease severity was assessed by ophthalmoscopy and histology, proportions of leukocyte populations and intracellular expression of cytokines were evaluated by flow cytometry and the gut immune environment was analysed by tissue culture and ELISA. We found that prophylactic and early oral treatment with EcN reduces the severity of EAU. However, EcO treatment does not. Kořeny moravské urbanistické struktury / Roots of Moravian Urban Structure

The thesis has been written on the basis of main architectural concepts and their application in real life within a historical context investigation. Main architectural concepts are represented in a harmony of architectural composition that deals with relations among form, structure and space in the unique art work. Genius loci play very important role in the architectural creation - it represents a special and extra-ordinary urban locality and its architectural value in the historical, geographical and cultural context. The origin of Ostravice village within the historical frame concept of Moravia domain is the topic of this thesis. Other historical documents gave information about Bruneswerde as the Brno Castle. That means Brno Castle had to be founded not in Brno (as it has been consid-ered for many years) but it was located in Beskydy at Ostravice. The sacred city – Civitas Dei had been located in Bruneswerde region in early ages as the centre of European culture, education and spiritual life. Civitas Dei – divine Jerusalem in the transcription of St. Augustine's book De civitate Dei – is not only glorification of God and religious fantasy. It is also one of significant clues for recognition of historical architecture. The depiction of unknown settlement from the book of unde-fined origin is a superb testimony about extinct architectural works. They are legible from cadastral maps. Brno Castle - residence of nobility and power served shelter to St. Vojtěch, St. Prokop, St. Václav and St. Ludmila as it is obvious for the mentioned picture. Three major temples, three com-position axes symbolized by three towers on coins are in analogical relation to Brno triangle of four saints - the Saint family of Brno temples. Powerful ambitions of Brno City principals and clergy are inscribed into the urban structure in the way of composition relations which are legible to them who devoted themselves to the mystery of harmony. The absence of historical continuity affects personal attitudes and also identity of the whole community. The architecture truly reflects the past state of polis and it is eloquent even after its death. The architectural composition relations influence the natural environment for long time, longer than the architectural work existence. The geometric order of Renaissance Brno existence has not been in attention of architects so far. The features were discovered in characteristic paintings by Albrecht Durer. They are evidently secret works of the genius. A meaningful collaborator and follower in the extensive project was also his friend Jan Čert from Brno and lately from Vienna. His noble genealogy played a significant role in history of Silesia and Moravia for centuries. It is tendency to consider him as Austrian or even German architect. It is because of the fact that the genealogy tree of his noble family had roots in Moravia. It is supposed that Durer with Jan Čert´s support created the extraordinarily monumental architectural and urban works in Brno. A remarkable consensus in the urban composition of two squares and transition of traditional urban structure of Ostravice Civitas Dei into the modern Brno is also confirmed due to the identification of noble creators and owners, who were at the foundation, transformation and extinction of elements of the Moravian urban structure.
