Mechanické vlastnosti materiálů připravovaných pomocí procesu SLM / Mechanical properties of materials prepared by SLM process

The diploma thesis deals with the selection of process parameters used for manufacturing of high-strenth materials using SLM technology. The feedstock material was powder with a chemical composition according to standard DIN X3NiCoMoTi 18-9-5. Influence of change in process parameters on mechanical properties was examined by hardness tests and tensile tests. Metallographic and fractographic analysis were conducted with an aim to understand mechanisms of failure present in this type of material. Selection of optimal process parameters was based on the analysis of mechanical properties of manufactured samples. Possible future steps related to the improvement of the process were proposed. Results of this experiment were compared with literature regarding parts produced by SLM technology and conventional methods.

This diploma thesis deals with properties of multi-materials interface composed of pure iron and Cu7Ni2Si1Cr alloy produced by SLM process. The theoretical part of thesis is focused on selective laser melting technology, and on description of defects connected with the production of SLM parts. Furthermore, one section deals with the production of multi-materials prepared by the SLM process. The experimental part of this thesis deals with selections of the SLM process parameters appropriate for bulk samples preparation. Mechanical properties were determined by the tensile tests at room temperature. Metallographic and fractographic analyses were performed for evaluation of the microstructure and description of the fracture mechanisms.

Vliv hybridizačních směsí na intenzitu fluorescence při in situ hybridizaci / Influence of a composition of hybridization mixtures on fluorescence intensity during the in-situ hybridization

Fluorescenční in situ hybridizace (FISH) je široce používaná metoda pro detekci určité sekvence DNA na chromozomech. Cílem práce je porovnání tří různých chemických sloučenin (formamidu, ethylenkarbonátu a sodných kationtů) používaných ve směsích pro in situ hybridizaci. Složení těchto směsí ovlivňuje renaturaci DNA a je důležité porovnat jejich fyzikální vlastnosti. Práce je rozdělena do dvou hlavních částí. První část se zabývá otázkou termodynamických parametrů používaných pro experimenty FISH, jako je teplota tání, entalpie přechodu DNA ze dvoušroubovice na vlákno nám dává přehled o energii potřebné k tomto přechodu a interakcích mezi bázemi a každou složkou směsi. Kromě toho hodnoty entropie určují poř uvnitř směsi - systém DNA. Druhá část porovnává intenzitu fluorescenčního signálu při optimalizovaných teplotách tání sondy použité pro in situ hybridizaci. Jako sonda byla použita sub-telomerní repetice X43.1, která je umístěna na Y chromozomu rostlinného modelového organismu Silene latifolia. Směs obsahující formamid má nejlepší výkon při delším postupu hybridizace, zatímco ethylenkarbonát poskytuje vyšší intenzitu signálu, a proto je vhodnější pro rychlé FISH protokoly.

Vývoj skenovací strategie laseru pro výrobu mikro-prutové struktury technologií SLM / The developement of SLM laser strategy for lattice structure fabrication

Additive technology (AT) is increasingly used to design unique parts, mainly due to the ability to produce complex structures such as lattice structures. However, this also includes the need to modify the process parameters or the production strategy of the AT, which is usually set to produce volume geometry. Using samples corresponding to the geometries of the lattice structures, the exact input values were measured, which were used to design the SLM (Selective laser melting) production process using a contour strategy. Thanks to this, vertical and angled (35.26°) struts with low porosity (up to 0.2 %), low surface roughness and high dimensional accuracy were produced. Porosity was measured on µCT, surface roughness and dimensional accuracy were measured on STL data. The results show that if the parameters of the SLM process are set correctly, it is possible to produce struts with low porosity and surface roughness using different combinations of laser power and scanning speed. The above findings were used in the creation of script that allow the selection of suitable process parameters to produce lattice structures.

This diploma thesis deals with development and design modifications of tire mould segment which will be batch produced by additive technology Selective Laser Melting. Material for its production is maraging steel 1.2709. Lattice structure was used inside the segment construction. The geometry of the lattice cell was chose based on two main factors – eliminating production costs and providing sufficient stiffness. Strength of the segment was calculated by FEM. The functional sample was made and its distortion was analyzed by optical digitalization.

The diploma thesis deals with the study of the influence of process parameters of AlSi7Mg0.6 aluminum alloy processing using the additive technology Selective Laser Melting. The main objective is to clarify the influence of the individual process parameters on the resulting porosity of the material and its mechanical properties. The thesis deals with the current state of aluminum alloy processing in this way. The actual material research of the work is carried out in successive experiments from the welding test to the volume test with subsequent verification of the mechanical properties of the material. Material evaluation in the whole work is material porosity, stability of individual welds, hardness of the material and its mechanical properties. The results are compared with the literature.

Immunomagnetic separation is based on binding of antibody with antigen, where antibody is bound to magnetic particle. In this thesis there were used particles of magnetic pearl cellulose with antiLactobacillus and antiBifidobacterium antibodies. Immunomagnetic separation method was optimalized and verified for its efficiency and specifity with bacterial and yeast cells. This cells were identified by polymerase chain reaction. Efficiency of immunomagnetic separation was verified on probiotic meat product, where Lactobacillus cells were isolated. With DNA from isolated Lactobacillus cells the high resolution melting was performed. The results show presence of several bacterial strains of Lactobacillus species.

The paper deals with the creation of interface based on multiple input signals, i.e. multimodal interface. For this purpose analyzes the benefits of the approach to communicate with the device that way. The work also includes an overview of the level at which you can perform data fusion, and different approaches to the layout of the system architecture for multimodal data processing. The important part is the actual design of the system, where for the interface was chosen distributed architecture using software agents for processing inputs. As a method for data integration was picked hybrid fusion based on dialog driven and unification strategy. The result should be an interface for media center control and interaction with other devices around the user.

The aim of this diploma thesis was to introduce real-time PCR with high-resolution melting analysis for Bifidobacterium species. Currently a small number of publication, dealing with identification of Bifidobacterium species using high-resolution melting analysis, is available. According to publications dealing with identification of lactic acid bacteria were selected primers P1V1 and P2V1, LAC1 and LAC2, LsppUPF and LsppUPR, V3F and V3R, V6F and V6R. Using this primers bacterial DNA was amplified by real-time PCR with high-resolution melting analysis. After evaluation of the measured results efficiency of selected primers was verified on DNA izolated from complex sample of probiotic product. After further optimisation real-time PCR with high-resolution melting analysis could be suitable using selected primers for Bifidobacterium species.
