Le mucus recouvre et protège les épithéliums des tractus respiratoire, gastro-intestinal et reproducteur. Les mucines sécrétées sont des molécules mosaïques de très grande taille moléculaire, fortement O-glycosylées et responsables des propriétés rhéologiques du gel de mucus. MUC5B est l’une des cinq mucines gélifiantes et est principalement sécrétée dans le tractus respiratoire. Chez l’Homme, MUC5B est impliquée dans le cancer mammaire et la mucoviscidose. MUC5B est exprimée dans plus de 80% des cancers mammaires alors que MUC5B n’est pas exprimée dans le tissu mammaire sain. Par ailleurs, MUC5B pourrait jouer un rôle majeur dans le poumon de patients atteints de mucoviscidose. Pour étudier la fonction de MUC5B, nous avons utilisé des protéines recombinantes composées de différents domaines de MUC5B et des modèles animaux. Nous avons montré que MUC5B favorise la prolifération cellulaire et l’invasion in vitro et favorise la croissance tumorale et la dissémination métastatique chez des souris immunodéprimées. MUC5B apparait donc comme une cible thérapeutique intéressante pour ralentir la prolifération cellulaire et la dissémination métastatique dans le cancer du sein. Nous avons également montré que l’expression de Muc5b et le nombre de cellules Muc5b positives sont plus élevés dans un modèle murin de mucoviscidose (Cftr–/–) que chez les souris frères-sœurs contrôles, ce qui suggère que les souris Cftr–/– sont prédisposées à former des bouchons bronchiques. Les souris Cftr–/– infectées expérimentalement par Pseudomonas aeruginosa développent des bouchons bronchiques AB-PAS positifs principalement composés de la mucine Muc5b et d’ADN. MUC5B semble être une cible thérapeutique intéressante pour diminuer la viscosité du mucus dans la mucoviscidose. Enfin, nous avons montré que la protéine Muc5b est exprimée très précocement (dès E8.5) au cours du développement embryonnaire murin. A E11.5, Muc5b est exprimée dans le bourgeon pulmonaire, dans le bourgeon formant la trachée et l’œsophage et dans l’estomac. De manière inattendue, Muc5b est exprimée au niveau baso-latéral des cellules épithéliales suggérant un rôle clé de MUC5B dans la morphogenèse. / Respiratory, gastrointestinal and reproductive tracts are protected by a mucus layer. Secreted mucins are large, high molecular weight and heavily O-glycosylated mosaic proteins that are responsible for the rheological properties of mucus gel. MUC5B is one of the five secreted gel-forming mucins and is mainly secreted in the respiratory tract. In human pathology, MUC5B has been implicated in breast cancer and cystic fibrosis (CF). This ‘non mammary’ mucin is expressed in more than 80% of breast cancer and it has been suggested that MUC5B may represent the main mucin to be implicated in the lung disease of CF.To investigate the function of MUC5B, we used recombinant proteins of several MUC5B domains and animal models. We show that MUC5B promotes cell proliferation and invasion in vitro and tumor growth and metastasis using SCID mice suggesting that MUC5B may represent a therapeutic target to slow down the tumor growth and dissemination in breast cancer. We show also that Cftr–/– mice harbor more Muc5b positive Clara cells than do control littermates suggesting that CF mice are predisposed to develop mucus plugs. Experimental infection with Pseudomonas aeruginosa increased the number of Muc5b-positive cells and the formation of mucus plugs in bronchi or bronchioles of Cftr–/– mice. These plugs are mainly composed of Muc5b and DNA suggesting that MUC5B may be a valuable target for decreasing mucus viscosity in CF. Finally, Muc5b protein was detected very early in mouse embryogenesis (at day 8.5). At E11.5, Muc5b is expressed in pulmonary bud, in tracheo-esophagal groove and in stomach. Unexpectedly, Muc5b is expressed at the basal and lateral membrane of epithelial cells of the buds suggesting a key role in epithelial morphogenesis.

1. Abstract (EN) This thesis is collection of work focused mainly on the understanding of mast cell activation and its regulation by Orm1-like (ORMDL) proteins. ORMDL family is a group of endoplasmic reticulum (ER) membrane resident proteins that are highly conserved amongst mammalian species. ORMDL proteins can be found in diverse range of organisms from plants through fungi to animals. ORMDL proteins were first discovered in yeasts and the interest in these proteins skyrocketed after the discovery that ORMDL3 is associated with childhood onset asthma in genome wide association studies. Following research connected ORMDL3 also with allergic inflammation and inflammatory bowel disease. Since mast cells are mainly known for their role in allergy and allergen induced inflammation, we decided to investigate the role of ORMDL proteins in regulation of mast cell activation and signaling. In our first study we focused on the role of ORMDL3 in mast cell activation via the high affinity IgE receptor 1 (FcεRI). We prepared bone marrow-derived mast cells with decreased (ORMDL3-KD) or increased (ORMDL3-OE) ORMDL3 expression. The role of mucin in establishment of Escherichia coli in porcine small intestineAimutis, William R. January 1985 (has links)
Mucin was isolated for incorporation in bacteriological media by reduction and proteolysis of mucous gel from porcine small intestine. Mucin prepared in this study contained (by weight) 37.2% protein, 58.7% carbohydrate (4.6% fucose, 9.4% mannose, 10.7% sialic acid, 13.5% galactose, and 20.5% hexosamine), and 3.7% ester sulfate. Fractionation of mucin on Sepharose CL-4B yielded one peak which eluted at the void volume. However, sodium dodecyl sulfate gel electrophoretic patterns contained 9 polypeptide bands of which 6 stained with periodic acid—Schiff reagent. Proline, serine, and threonine residues accounted for 26% (by weight) of the total protein in the preparation. Half-cysteine residues made up another 1%. Intrinsic viscosity of mucin prepared by reduction and proteolysis was 135 ml/g.
Mucin was incorporated into a minimal bacteriological medium as the sole-source of carbon and nitrogen. Enterotoxigenic and non-enterotoxigenic Escherichia coli grew equally well in mucin medium at levels comparable to growth in 3 mM glucose medium. Growth did not appear to be limited by availability of metabolizable substrates. Spent mucin medium supported growth in comparable numbers. E. coli P-155 produced heat-stable and heat-labile enterotoxins during growth in both mucin medium and fresh mucosal scrapings medium. E. coli utilized total hexose and protein in mucin medium at comparable levels (6 to 10%). Increases in reducing end groups (0.28 μ moles/ml) and free amino sugar end groups (0.04 μmoles/ml) during growth were detected. E. coli used approximately 15% of the total carbohydrate in mucin medium including 36% of the galactose, 15% of the fucose, and 27% of the mannose. Utilization of mucin by QL ggli produced minor changes in gel filtration patterns on Sepharose CL-4B.
Twelve strains of E. coli were examined for glycosidase activity during growth on mucin. All twelve produced a cell-bound and an extracellular α-fucosidase although the majority of activity was cell-bound. Although α-fucosidase was a constitutive enzyme of E. coli P-155, maximum activity was observed during exponential growth in mucin medium. Eleven strains produced cell-bound α-galactosidase. No extracellular activity of this enzyme was detected. Maximum levels of induced α-galactosidase activity were obtained in late exponential to early stationary growth of E. coli. E. coli ATCC 23723, a mutant of E. coli K12 lacking the galactoside permease gene, did not produce α-galactosidase activity during growth on mucin. No α-mannosidase activity was detected using nitrophenylmannoside as substrate.
Porcine small intestinal mucin was a positive chemoattractant for E. coli in capillary assays. Optimal chemotactic response by E. coli P-155 in capillary experiments was obtained at a mucin concentration of 1 mg dry wt/ml at a pH of 7.0. Spent mucin was still a positive chemoattractant for E. coli P-155 and 123 despite losing 15% of the total mucin carbohydrate. / Ph. D.

MUC5B, une des 5 mucines gélifiantes chez les mammifères, constitue la matrice de gels de mucus et est responsable des propriétés visco-élastiques de ces gels. Cette macromolécule est sécrétée par des cellules caliciformes dont la densité reflète l'homéostasie des épithéliums sécrétoires.L'objectif principal de ce travail de thèse était la création et la caractérisation d'une souris transgénique afin de suivre facilement la production in vivo de Muc5b. Le second objectif était de montrer, chez des souris mucoviscidosiques CftrΔF508, qu'une nutrition enrichie en acides gras polyinsaturés à longues chaînes (n-3) module la production de la mucine Muc5b, confirmant des résultats précédemment publiés sur la régulation transcriptionnelle du gène Muc5b chez des souris de type sauvage par ce régime alimentaire.Une souris transgénique Muc5b-GFP a été créée par insertion d'une séquence codant la GFP (pour « Green Fluorescent Protein ») à la place du codon Stop de Muc5b. Le profil d'expression de Muc5b a été déterminé ex vivo sur tissus et chez la souris anesthésiée. Muc5b est produit dans l'oreille moyenne, l’œil, la cavité nasale, la trachée, la vésicule biliaire, le cervix, ainsi que dans le poumon embryonnaire à E12,5. Nous apportons la preuve de concept que Muc5b-GFP est un biomarqueur de la densité des cellules à mucus dans un modèle de syndrome de l’œil sec où nous mesurons, chez la souris anesthésiée, une diminution de la densité en cellules caliciformes qui est restaurée par application topique d'interleukine 13. La souris Muc5b-GFP est donc un modèle pré-clinique pour suivre la production de la mucine et la densité de cellules caliciformes. Dans le modèle murin CftrΔF508, nous montrons qu'une supplémentation à long terme en acides gras polyinsaturés à longues chaînes (n-3) n'influence pas le taux d'expression de Muc5b suite à une inflammation pulmonaire aiguë, mais améliore l'histologie pulmonaire et module la réponse inflammatoire. / MUC5B, one of the 5 gel-forming mucins in mammals, is part of the mucus gels and responsible of the visco-elastic properties of these gels. The density of goblet cells, which secrete this macromolecule, reflects the homeostasis of secretory epitheliums.The main goal of this thesis was the creation and characterization of a transgenic mouse to easily follow in vivo the production of Muc5b. The second goal was to show that, as previously reported by the laboratory in wild-type mice, a (n-3) long chain polyunsaturated fatty acid enriched diet modulates the production of Muc5b in the cystic fibrosis mouse model CftrΔF508.A Muc5b-GFP transgenic mouse was created by replacing the Stop codon of Muc5b with a GFP (Green Fluorescent Protein) coding sequence. Muc5b expression pattern was studied ex vivo in tissues and in anaesthetized mouse. Muc5b is produced in middle ear, eye, nasal cavity, trachea, gallbladder, cervix, and in embryonic lung at E12.5. As a proof-of-concept, we show that Muc5b-GFP is a biomarker of goblet cell density in a dry eye model. In anaesthetized mice, we measured a decreased goblet cell density after dry eye induction, which was restored by interleukin 13 topical application. Muc5b-GFP mouse is a preclinical model to follow the mucin production and goblet cell density. In the cystic fibrosis mouse model CftrΔF508, we show that a long term (n-3) long chain polyunsaturated fatty acid supplementation has no effect after an acute pulmonary inflammation on the Muc5b expression level, but induces an improvement of lung histology and a modulation of the inflammatory response.

Com objetivo de analisar criticamente os tipos histológicos propostos na classificação da Sociedade Brasileira de Patologia para o adenocarcinoma gástrico, estudamos os tipos histológicos e o perfil imuno-histoquímico de mucinas em 272 adenocarcinomas gástricos operados no Hospital das Clínicas da Faculdade de Medicina da Universidade de São Paulo. A casuística pode ser dividida em 237 tumores únicos, 4 tumores múltiplos e 31 tumores com mais de um tipo histológico. Foram avaliados idade, gênero, localização, aspecto, estadiamento tumoral, padrões de expressão de mucinas intestinais pelos marcadores MUC2 e CD10 e gástricas por MUC5AC e MUC6. A coexistência de focos de padrão pouco diferenciado e de padrão gástrico em adenocarcinoma gástrico com mais de um aspecto e em casos com mais de um tumor e a ausência de tal concomitância com o padrão intestinal favorece sua compreensão como padrões independentes. As variáveis histológicas \"favoráveis\" predominaram nos adenocarcinomas gástricos de padrão intestinal, seguido dos de padrão tubular gástrico. Estes representam o cerne para estudos futuros por compreender o tipo túbulo-papilífero cujos aspectos clínicos se assemelham aos intestinais, o tipo tubular com expressão de mucina incaracterística e o tipo microtubular pela primeira vez individualizado, com aspectos morfológicos menos favoráveis e expressão imuno-histoquímica própria da mucosa gástrica. Os adenocarcinomas mucinosos mostraram marcante dicotomia quer nos aspectos clínicomorfológicos, quer no padrão de expressão de mucinas. Mucinous adenocarcinomas have shown a stricking dichotomy in both clinical and morphological aspects, as in the expression pattern of mucins. Data indicate the need to understand them as disparate entities. The mucocelular type (signet ring cells) was detected in cases found inearly tumor stages, but includes the highest fraction of advanced tumors, Borrmann IV with predominant expression of gastric mucins. Muconodular type was only detected in advanced stages and co-expressed intestinal and gastric mucins. The poorly differentiated cases were frankly distinct from the other types, by both clinical and morphological data, and by the scarcity of mucin expression.

Impact des facteurs micro-environnementaux de l'hôte sur la colonisation instestinale des Escherichia Coli adhérents et invasifs. / Impact of microenvironmental host factors on the gut colonization of adherent-invasive Escherichia coliGibold-Lyonne, Lucie 28 September 2016 (has links)
La maladie de Crohn (MC) est une affection inflammatoire chronique du tube digestif dont l’étiologie est multifactorielle. Les lésions intestinales des patients atteints de MC sont anormalement colonisées par des souches pathogènes d’Escherichia coli appartenant au pathovar AIEC pour «Adherent-Invasive Escherichia coli ». Ces souches sont capables d’adhérer et d’envahir les cellules épithéliales intestinales, et ont la capacité de survivre et de se multiplier en macrophages en induisant une synthèse intense de cytokines pro-inflammatoires. Les AIEC pourraient ainsi être impliquées dans l’induction et/ou l’entretien de l’état inflammatoire de la muqueuse intestinale.L’objectif de ce travail est d’identifier les déterminants bactériens des AIEC qui vont intervenir dans les étapes précoces de l’implantation des AIEC au niveau intestinal et de définir quel est le rôle des facteurs micro-environnementaux de l’hôte dans cette implantation.Nous montrons que l’AIEC LF82 possède une activité mucinolytique qui est portée par le gène vat-AIEC et qui favorise la traversée du mucus et la colonisation intestinale. Nous avons retrouvé ce gène chez 42% des souches AIEC isolées de patients atteints de MC, et chez 97% des souches AIEC appartenant au phylogroupe B2. Par ailleurs, nous avons montré que les sels biliaires augmentent l’expression de cette mucinase.Nous avons ensuite étudié l’influence des sels biliaires sur l’expression globale des gènes de la souche LF82. Les sels biliaires modifient profondément le métabolisme de la souche, induisant une diminution globale des voies de biosynthèse (protéines, lipides) et une augmentation des voies de dégradation (alcools, acides carboxyliques, polyamines, …). L’étude du catabolisme de l’éthanolamine et du propanediol indique que les AIEC pourraient se servir de ces substrats pour s’implanter au sein de la flore iléale. De plus, les analyses transcriptomiques révèlent que les sels biliaires augmentent l’expression de gènes codant des facteurs de virulence comme l’invasine IbeA, les systèmes de sécrétion de type VI et la yersiniabactine. Nous montrons également qu’ils favorisent la formation de biofilm chez les souches AIEC.Ces données indiquent que les sels biliaires constituent un signal permettant à la souche AIEC LF82 de mettre en place différentes voies métaboliques et déterminants bactériens nécessaires à son implantation au niveau intestinal.Mots-clé : Escherichia coli, maladie de Crohn, mucines, serine protéase, mucinase, AIEC, / The etiology of Crohn's disease (CD) involves disorders in host genetic factors and intestinal microbiota. Ileal mucosa of CD patients is often abnormally colonized by adherent-invasive Escherichia coli (AIEC). These strains isolated from the intestinal mucosa of CD patients are able to adhere to intestinal epithelial cells (IECs). This adhesion to IECs promotes the invasion of cells by the bacteria. Furthermore, the invasive ability of AIEC strains allows bacteria to translocate across the human intestinal epithelium, move into the deep tissues and activate immune cells continuously upon arrival. Thus AIEC could be involved in the inflammatory state of the intestinal mucous membrane. The aim of this study was to identify components of AIEC virulence, which might favor their implantation in the gut of CD patients and to define the role of several chemical factors from the ileal environment. Here, we reported a protease called Vat-AIEC from AIEC which favors the penetration of AIEC through the mucus layer and enhances gut colonization. The screening of E. coli strains isolated from CD patients revealed a preferential vat-AIEC association with AIEC strains belonging to the B2 phylogroup. Besides, Vat-AIEC transcription was increased with bile salts from the ileum environment. Then a global RNA sequencing (RNA-seq) of E. coli LF82 has been used to observe the impact of bile salts on the expression of bacterial genes. The results demonstrate the explosive effect of bile salts with a dysregulation of about 40% of the genome, with a global upregulation of genes involved in degradation and downregulation of those implicated in several biosynthesis. Our results show that LF82 use ethanolamine as a nitrogen source and propane diol as a carbon source, which can favor their colonization in the gut compared to the other bacteria. We also studied virulence genes expression in the presence of bile salts. They increase the expression of several virulence factors like the IbeA invasion, the type 6 secretion systems and the yersiniabactin. Furthermore, we noticed an increased expression of genes implicated in biofilm formation. These results improve the understanding of the global regulatory network in the presence of bile salts and thus of AIEC implantation in the human gut of CD patients.

Mucinous adenocarcinomas have shown a stricking dichotomy in both clinical and morphological aspects, as in the expression pattern of mucins. Data indicate the need to understand them as disparate entities. The mucocelular type (signet ring cells) was detected in cases found inearly tumor stages, but includes the highest fraction of advanced tumors, Borrmann IV with predominant expression of gastric mucins. Muconodular type was only detected in advanced stages and co-expressed intestinal and gastric mucins. The poorly differentiated cases were frankly distinct from the other types, by both clinical and morphological data, and by the scarcity of mucin expression.

<p>Akrilamid je toksična hemijska supstanca koja ima vrlo široku primenu u hemijskoj industriji, a 2002. godine otkriveno je njegovo prisustvo u namirnicima bogatim skrobom koje se pripremaju na visokim temperaturama. U poslednjh desetak godina primećen je veliki porast gastrointestinalnih tegoba u ljudskoj populaciji. Cilj istraživanja bio je ispitati patohistološke promene u tkivu želuca pacova soja Wistar izazvanih peroralnim aplikovanjem akrilamida i na taj način povući paralelu sa mogućim gastrointestinalnim tegobama nastalim kao posledica konzumiranja hrane bogate akrilamidom. U istraživanju je ispitivano 6 grupa od po 5 eksperimentalnih životinja (pacovi soja Wistar). Dve kontrolne grupe kojima je peroralno aplikovana destilovana voda i koje su žrtvovane posle 24h i 72h; dve eksperimentalne kojima je peroralno aplikovan akrilamid u dnevnoj dozi od 25 mg/kg i koje su žrtvovane posle 24h i 72h; dve eksperimentalne grupe kojima je peroralno aplikovan akrilamid u dnevnoj dozi od 50 mg/kg i koje su žrtvovane posle 24h i 72h. Na histološkom materijalu tkiva želuca primenjena je kvalitativna histološka analiza pod svetlosnim mikroskopom, semikvantitativna procena tipa mucina u epitelnim ćelijama sluznice želuca, prisustvo limfocita i granulocita u sluznici želuca, stereološka merenja pojedinih kompartmana zida želuca, linearna merenja broja i veličine ganglijskih ćelija u Maissner-ovom i Auerbach-ovom nervnom pleksusu, kao i broj mastocita u lamini propriji sluznice i podsluznici želuca. Dobijene vrednosti merenih parametara su potom statistički obrađene. Nastale promene na tkivu želuca pacova soja Wistar se ogledaju u vidu blagog direktnog oštećenja površnog epitela sa propratnom blagom inflamatornom reakcijom i blagom degranulacijom mastocita. U Maissner-ovom i Auerbach-ovom nervnom pleksusu su smanjene volumenske gustine nervnih vlakana i ganglijskih ćelija, kao i broj i veličina ganglijskih ćelija. Direktno toksično delovanje na epitel dovodi do posledične obnove epitela, te je potvrđeno prisustvo nezrelijih oblika mukoproduktivnih ćelija koje sadrže kisele, AB pozitivne mucine. Ispitani inflamatorni i degenerativni parametri pokazuju pozitivnu korelaciju u odnosu na dozu i/ili dužinu ekspozicije akrilamidu. Primena akrilamida peroralno pokazala je da postoje patohistološke promene na tkivu želuca u vidu direktnog toksičnog oštećenja epitela, inflamatorne reakcije i oštećenja nervnih pleksusa. Poznavanjem mehanizma delovanja ove toksične materije moguće je primeniti adekvatnu prevenciju u ishrani i izvršiti odgovarajući izbor terapijskih metoda.</p> / <p>Acrylamide is a toxic chemical substance with wide implementation in chemical industry. In 2002 it was discovered the presence of acrylamide in foods rich in starch which are prepared at high temperatures. In the last ten years there is a large increase in gastrointestinal illnesses in human population. The aim of this study was to investigate the histopathological changes in the gastric tissue in Wistar rats induced with injection of oral acrylamide and thus draw a parallel with possible gastrointestinal problems arising as a result of the consumption of foods rich in acrylamide. The research was carried out 6 groups of 5 experimental animals (Wistar rats). Two control groups that are orally concomitant application of distilled water and which were sacrificed after 24h and 72h; two experimental groups which are orally administrated acrylamide in a daily dose of 25 mg / kg and that were sacrificed after 24h and 72h; two experimental groups which were orally administrated acrylamide in a daily dose of 50 mg / kg and that were sacrificed after 24h and 72h. On histological gastric tissue material is applied qualitative histological analysis by light microscopy, semi-quantitative assessment of the type of mucin in epithelial cells of the stomach lining, the presence of lymphocytes and granulocytes in gastric mucosa, stereological measurements of individual compartments of the stomach wall, linear measuring the number and size of ganglion cells in the Maissner and Auerbach's nerve plexus, and the number of mast cells in the lamina propria of the mucosa and in the submucosis of the stomach. Obtained values of measured parameters were statistically processed. Histological changes in the stomach tissue of Wistar rats are seen as a direct slight damage of the surface epithelium, with accompanynig mild inflammatory reaction and the degranulation of mast cells. The Meissner's and Auerbach's nerve plexus decreased volume density of nerve fibers and ganglion cells, as well as the number and size of the ganglion cells. Directly toxic effect on epithelium leads to the result of the reconstruction of the epithelium, which is confirmed by the presence of immature form of mucoproductive cells which contain acid, AB positive mucins. Examined inflammatory and degenerative parameters show a positive correlation with respect to dose and / or a time of exposition to acrylamide. Acrylamide oral application revealed that there are histologic changes in the stomach tissue in the form of a direct toxic damage to the epithelium, inflammatory reaction and damage to the nerve plexus. Knowing the mechanism of action of these toxic substances allows to apply adequate prevention in nutrition and make an appropriate choice of therapeutic methods.</p>
