Molekuly "DASH systému" v lokálních a systémových patogenetických procesech revmatoidní artritidy / "DASH molecues" in local and systemic pathogenetic processes of rehumatoid arthritis

The biological half-life of several pro-inflammatory mediators involved in the pathogenesis of rheumatoid arthritis (RA) is controlled by molecules exhibiting dipeptidyl peptidase-IV (DPP-IV)-like enzymatic activity (Dipeptidyl peptidase-IV activity and/or structure homologues- DASH). The aim of this thesis was to identify the molecular source of the DPP-IV-like enzymatic activity in the peripheral blood and synovial fluid in patients with rheumatoid arthritis as compared to control patients with osteoarthritis (OA), and to evaluate the association of DPP-IV with the disease activity. We found that the main source of the DPP-IV-like enzyme activity in the plasma and in the synovial fluid in patients with RA is the canonical DPP-IV. DPP-IV-like enzymatic activity and canonical DPP-IV were also detected on the cell surface of blood and synovial fluid mononuclear cells. Significantly lower DPP-IV-like enzymatic activity and DPP-IV expression in the synovial fluid mononuclear cells was found in RA as opposed to OA patients. In the synovial fluid of RA patients there was also a negative correlation between the concentration of the pro-inflammatory DPP-IV substrate SDF (stromal cell-derived factor-1 and the proportion of the DPP-IV+ T cells. The topic of this thesis is the situation of families with children having severe visual or multiple disabilities, examined from the father's standpoint. The aim of this thesis is to map out (explore) the lives of fathers whose children have severe visual impairments or multiple disabilities from their subjective point of view. The focus will be on the process of accepting disabled children, the fathers' experiences, their behavior and expectations in relation to the children, their spouse / partner and other family members. In addition, the focus will be on their view of the relationship between the larger society and individuals with disabilities, including organizations dealing with this issue. For the purposes of qualitative research "What it is like to be a father whose child has a severe visual impairment," a method of semi-structured interviews for data collection will be used. The next part addresses the legislation of this system in detail, also sums up the state of its implementation and the plan of implementation. Our conclusion shows the problems connected with the system , offers their solution and views for the future.

Presented master thesis deals with description of the gradual development and positions of the church schools in CR during the last 20 years. These schools are then complemented by the schools of theology at CR universities. This thesis is in a certain way a continuation of a bachelor thesis, successfully defended in 2007 at the School of Education of Charles University. The thesis describes in brief the history of education and pedagogy in general, church schools including. Then there are specified all the pertinent valid legal regulations for operation of church schools based on experience of the author from a managerial position of such a school for many years. The role of the general management and management of human resources on one side and the theological reflection of the church schools on the other side is discussed. In detail, the equipment of the facilities and their economy is analyzed as well as the goals and calling of these schools. In a wide spectrum of opportunities open to church schools in 21st century their potential can be used through new and fresh methods for evangelizing, mission and pastoral care.
