Soybean downy mildew, caused by the oomycota Peronospora manshurica, is one of the diseases of major distribution in all regions of the world where soybean is cultivated. Soybean downy mildew can damage leaves and in some cases pods. This research aimed to evaluated the incidence and severity of downy mildew at field, and its impact on soybean yield. Two experiments were carried out in Dourados-MS, one in Maracaju-MS and another in Ponta Porã-MS, from October, 2006, to March, 2007, where BRS 133, BRS 240 and BRS 245 RR cultivars were cultivated, respectively. Treatments were established as randomized block experimental design, with five replications and four treatments, as following: fenamidona (500 g i.a. L-1), iprovalicarbe (55 g i.a. kg-1) + propinebe (613 g i.a. kg-1), cloridrato de propamocarbe (722 g i.a. L-1), which were applied every two weeks, and a control, without applying of fungicide for downy mildew control. For all treatments, soybean rust (Phakopsora pachyrhizi) was controlled by tebuconazole (150g i.a.ha-1). In the conditions that essays were carried out, it was not detected the effect of the disease on soybean yield. are necessary for elucidating the effect of the disease on soybean yield / As doenças foliares que incidem na cultura da soja constituem-se um dos principais fatores que limitam a obtenção de maiores rendimentos na cultura. O míldio da soja, causado pelo estraminópila Peronospora manshurica, é uma das doenças de maior distribuição em todas as regiões onde se cultiva soja no mundo, podendo causar lesões nas folhas e vagens, em alguns casos. Este trabalho teve como objetivo avaliar a incidência e a severidade do míldio no campo, assim como a produtividade de grãos da soja. Dois ensaios foram conduzidos em Dourados-MS, um em Maracaju-MS e outro em Ponta Porã-MS, no período de outubro de 2006 a março de 2007, utilizando as cultivares BRS 133, BRS 182, BRS 240 e BRS 245 RR. Os tratamentos foram dispostos segundo o delineamento experimental de blocos ao acaso, com cinco repetições e quatro tratamentos, conforme segue: fenamidona (500 g i.a. L-1), iprovalicarbe (55 g i.a. kg-1) + propinebe (613 g i.a. kg-1), cloridrato de propamocarbe (722 g i.a. L-1) aplicados quinzenalmente, e testemunha; sem aplicação de fungicida para controle do míldio. Em todos os tratamentos, a ferrugem asiática da soja foi controlada pela aplicação de tebuconazole (150g i.a. ha-1). Nas condições em que foram conduzidos os ensaios, não foi detectado o efeito da doença sobre a produtividade da soja, porém ensaios adicionais acrescentando novos fungicidas e conduzidos por várias safras são necessários para elucidar o efeito da doença sobre a produtividade de soja

PHYLLOPLANINS: NOVEL ANTIFUNGAL PROTEINS ON PLANT LEAF SURFACES

Secreted surface proteins are an innate immune defense component employed by animals to inhibit invading microbes. Surface proteins have not been documented in plants, even though the aerial leaf surface, or phylloplane, is a major site of pathogen ingress. We have discovered novel proteins, termed phylloplanins, which accumulate on leaf surfaces of Nicotiana tabacum, and we have isolated the gene Phylloplanin that is unique in gene databases. Natural and E. coli-expressed phylloplanins inhibit spore germination and limit leaf infection by the oomycete pathogen Peronospora tabacina. We investigated the site of phylloplanin biosynthesis using biochemical techniques. These techniques included radiolabeling of detached trichome glands, radiolabeling of epidermal peels, analysis of leaf water washes of various Nicotiana plants, and examination of guttation fluid, leaf vein contents, and extracellular fluid. From these experiments, we tentatively conclude that phylloplanins are produced by hydathodes, or an unknown surface secreting system, but not by glandular secreting trichomes. Future experiments with the phylloplanin promoter, whose elucidation is described herein, and its fusion to a reporter gene (GUS or GFP), will undoubtedly provide further insight into the location of phylloplanin biosynthesis and deposition. We suggest that the hydrophobic nature of phylloplanins aids in their dispersal over the leaf surface. Phylloplanins constitute a first-point-of-contact, rapid response, innate immune deterrent to pathogen establishment on N. tabacum leaf surfaces, and are the first studied representatives of a novel protein class in the plant kingdom. Further study of leaf surface proteins is justified to understand further their roles in plant defense, and to investigate their potential in agricultural biotechnology. Additionally, we describe miscellaneous observations we have made during the course of this research. Low molecular mass proteins (as yet uncharacterized) are washed from leaf surfaces of sunflower, soybean, and other plants. Pathogenesis-related (PR-)-5a, a known antifungal protein, was found to be present on the leaf surfaces of healthy plants, although its function there remains unknown. A phylloplanin homologue from Arabidopsis appears to be antibacterial. Further study of this protein is warranted. We note that proteins can also be recovered from N. tabacum root surfaces, or the rhizoplane, but we have not further characterized these proteins. In summary, novel surface-accumulated proteins, termed phylloplanins, and the gene encoding these have been discovered in N. tabacum. An antifungal function for phylloplanins is reported, and evidence was found for a unique mechanism of surface deposition.

Baigiamajame darbe 2012–2013 m. tirta netikrosios miltligės (Peronospora viciae (Berk) Caspary) įtaka skirtingos geografinės kilmės miškinio pelėžirnio (Lathyrus sylvestris L.) cenopopuliacijų morfologiniams ir sėklų produktyvumo rodikliams. Darbo objektas – iš skirtingų Lietuvos rajonų surinktos dvylika miškinio pelėžirnio cenopopuliacijų, augančių vienodos agrotechnikos sąlygomis. Darbo tikslas – įvertinti netikrosios miltligės įtaką skirtingų cenopopuliacijų miškinio pelėžirnio morfologinias ir sėklų produktyvumo rodikliams. Darbo metodai – įvertinti skirtingų miškiniopelėžirnio cenopopuliacijų stiebo aukštis, žiedynų, ankščių, sėklapradžių ankštyje skaičius, ankšties produktyvumo, vieno stiebo sėklų produktyvumas. Atsparumas netikrajai miltligei įvertintas 5 balų sistema, atsparumo įtaka ankščiau išvardintiems rodikliams įvertinta naudojantis programa STATISTIKA 8. Rezultatai. Tyrimo metų duomenimis, miškinio pelėžirnio stiebo aukščiui, žiedynų skaičiui ir ankščių produktyvumui lemiamą įtaką turėjo cenopopuliacijos genotipas ir tyrimo metų meteorologinės sąlygos. Didesnis miškinio pelėžirnio stiebo aukštis teigiamai įtakojo ankščių (r=0,7539) ir sėklų skaičių (r = 0,6223). Stabiliausias iš tirtų rodiklių tyrimo metais buvo sėklapradžių (vid. 12,4 vnt.) skaičius ankštyje (V = 4,7), labiliausi – ankščių (vid. 21,6 vnt.; V = 36,5) ir subrandintų sėklų skaičiaus (vid. 151,1 vnt., V = 39,8) rodikliai. Miškinio pelėžirnio atsparumas netikrajai miltligei įtakojo... Subject of the research – 12 flat pea (Lathyrus sylvestris L.) cenopopulations from different geographical origins of Lithuania, grown on equal agrotechnic conditions. Aim of the research – estimate an influence of downy mildew on flat pea morfological and seed produktivity parameters. Methodology – to evaluate stem height, number of inflorescence, pods, ovules per pod and pod productivity, one stalk of seed productivity of different flat pea coenopopulations. Resistance to downy mildew is rated by 5-point scale, impact of resistance on above listed parameters estimated using the statistics program STATISTICA 8. Results. The research data shows, that genotype of coenopopulation and meteorological conditions had a decisive influence on flat pea stem height, number of inflorescences and pod productivity. Height of stem influenced positively pod (r = 0,7539 ) and seed number (r = 0,6223 ). The most stabile from the investigated parameters were number of ovules (mean 12,4 pcs.) per pod (V = 4,7 ), most labile – number of pods (mean 21,6 pcs., V = 36,5 ) and the number of mature seeds (mean 151,1 pcs., V = 39.8).
