Využití metody phage display při zkoumání povrchových antigenů Leishmania mexicana / The use of phage display to investigate Leishmania mexicana surface antigens

Leishmania is a protozoan parasite of vertebrates transmitted by the bite of infected phlebotomine sandflies. In humans, it causes a disease called leishmaniasis, which ranks as one of the most serious neglected tropical diseases. In the vectorial part of the life cycle, the crucial moment is when the flagellate forms (promastigotes) attach to the midgut epithelium of the sandfly. For most leishmania species, little is known about which types of phlebotomine receptors and leishmania surface antigens participate in the binding. Phage display was used to screen for Leishmania mexicana peptide ligands which may play a role in such binding. By affinity selection of phages incubated with promastigote cells, 16 unique peptides were identified. Fluorescent labelling of peptide-bearing phages indicated their putative binding sites on the leishmania surface. Based on the hypothesis that the identified peptides may be a part of receptors found in the phlebotomine midgut, experiments were performed where the sandflies were infected with promastigotes whose binding sites were blocked by two different peptide-bearing phages. This thesis deals with the processing and comparing information on the thickness of A0 horizon (humus mulch) forming anthropogenic soils in forestry reclamation Sokolov brown coal basin. Comparison of data found in Bažantnice Bohemie reclaimed forestry in 1923 with newer areas of forestry reclamation dumps Velký Riesl, Dvory, Gustav and data from previous studies on the Velká podkrušnohorská výsypka. This thesis deals with finding major differences, especially power, humus horizons A0, depending on the time of the establishment of forest reclamation and the reclamation age of trees. To unify and minimize variables are all shallow pits removed under alder sticky. Based on field research was collected from each of five shallow soil probes on each of the dumps. Probes were taken evenly over the surface of the dump and always under identical reclamation trees. Were identified basic characteristics of humus horizon A0 and the individual probes was a proper photographs. Based on removing shallow soil pits were identified basic characteristics of humus horizon A0. The test was conducted in the laboratory granularity to better implement the results of the work in the coming years. Identified the basic characteristics of humus layers A0 are mutually compared and evaluated basic statistical methods. From the data obtained were created layers in ArcGIS, which were interpreted in this thesis and stored on a data carrier, mainly because of their possible use for further research needs in the coming years.

Adsorpční procesy Cr(VI) v syntetické směsi půdních minerálů / Cr(VI) adsorption to synthetic mixture of soil minerals

Due to its toxicity, the Cr(VI) is known as a substance with negative effects on the environment. Understanding of Cr mobility, speciation and its toxicity helps to prevent negative impacts of this contaminant in soils, natural waters and atmosphere. This diploma thesis deals with a mechanism of Cr(VI) adsorption on to mixture of selected soil minerals as quartz, kaolinite and ferrihydrite based on mineralogical and chemical composition of the real contaminated soil. Main goal of diploma thesis is to test the selected surface complexation model, the DLM (diffuse layer model), which could be able to capture the process of Cr(VI) adsorption in natural heterogenic systems like soils. Component additivity approach (CA) was tested. The adsorption experiments were performed with three different concentrations of Cr(VI) (10-4, 10-5, 10-6) as a function of pH at different ionic strengths. Obtained data were used for modelling of adsorption edges and for DLM testing. Results confirmed, that Cr(VI) adsorption in synthetic mixture of enlarged soil minerals, is controlled mainly by presence of Fe-(oxy)hydroxides. The mechanistic surface complexation models used for modelling of Cr(VI) adsorption onto synthetic mineral mixtures, based on real mixed ratios of representative mineral phases in soils, can be useful for prediction of processes responsible for transport and retention of the Cr(VI) in soils.

The thesis aims for the surface treatment of materials using electron beam. It includes a literary research describing physical principle and properties of the electron beam. This research also presents possible influencing of the beam and describes the construction of the device that generates the beam. Various applications of the electron beam for the material treatment are presented in the second part of the paper. Within this thesis an experiment with the heat treatment of the steel surface by the electron beam has been realized. The experiment was aimed at creating a layer as deep as possible without melting the surface. The results and their evaluation are presented in the experimental part of the thesis. The attention is also paid to selected process parameters.
