Effect of washing on the quality, desirability and stability of drum-dried machine separated rockfish flesh

The effect of washing machine separated rockfish flesh on composition, quality and desirability was investigated. Washing machine separated rockfish flesh resulted in a 36.88% loss of solids. The greatest reductions were found in the ash (80.32%) and lipid (65.13%) levels in the washed flesh. Most of the sarcoplasmic proteins were lost during washing and only 77.29% of the protein (N X 6.25) was recovered. Washing the machine separated fish flesh did not appreciably alter the amino acid composition. The content of essential amino acids of the drum-dried treatments was similar to casein and this was reflected by the high protein efficiency ratios compared to the casein reference. The mineral composition of the unwashed machine separated flesh was similar to that of the fillet. Washing the machine separated flesh greatly altered the mineral composition in the flesh. The levels of calcium, magnesium, barium, strontium, boron, and manganese were not appreciably altered by washing. Phosphorus, potassium, and sodium levels were reduced while the iron, copper, zinc and chromium levels were increased in the washed flesh. The rapid formation of trimethylamine (TMA) in the drumdried rockfish fillet and unwashed machine separated flesh treatments may be related to the development of off-odors and off-flavors formed during storage at 30°C. The over-all desirability of bland wafers consisting of 10% drum-dried washed flesh increased slightly during storage but was not appreciably higher than the wafers containing the drum-dried fillet and unwashed fish flesh. Over-all desirability may not be directly linked to the formation of trimethylamine (TMA) since washing drastically reduced the level of trimethylamine oxide (TMAO) in the washed flesh and the subsequent formation of trimethylamine (TMA) during storage at 30°C. The much higher levels of malonaldehyde found in the drumdried washed machine separated rockfish flesh may implicate the oxidation of the lipids as being responsible for the lower over-all desirability of the washed flesh. The rapid formation of malonaldehyde may also indicate that naturally occurring antioxidants are leached from the flesh during the washing process. / Graduation date: 1976

Role of eIntermediaries in the United States Paper Supply Chain

In an attempt to streamline the paper supply chain, paper industry vertical business-to-business eMarketplaces were established to help industry players to decrease inefficiencies in their supply chains, to minimize negative effects of economic cyclicality, and to achieve better visibility. eMarketplaces rode the hype of revolutionizing the way in which industries conduct business, citing the cost savings achieved by expanded market reach, operational efficiencies, aggregated purchasing, and finding the least expensive suppliers. Adoption of eIntermediaries has been lower than expected in the paper industry and many of the paper vertical start-ups failed when the economy softened and the dot.com bubble burst. This thesis examines the expectations, experience and role of eIntermediaries in the paper supply chain. eIntermediaries failed to fulfill paper suppliers expectations regarding key promises, such as improving cash flow, reducing cycle time, and reducing errors. None of the paper supplier respondents achieved the expected benefits from eIntermediary implementation. Overall paper suppliers lack commitment and trust for eIntermediaries. Paper buyer and paper supplier attitudes and expectations on eIntermediaries do not differ significantly. Both suppliers and buyers are concerned that using eIntermediaries would lead to loss of contact with exchange partners, but paper suppliers have a greater level of concern. Suppliers are most concerned with profitability, security of sensitive information, technical resources, costs, and the need to restructure established business processes in the context of using eIntermediaries. B2B exchanges seem to have underestimated the complexity of the paper industry and overestimated companies ability to adopt eCommerce. Organizational changes, changes in business processes, development of industry standards, and improvements in integration technology systems are all needed to capture benefits from B2B exchanges.

We product test the structured financial products offered in Mangold Fondkommission AB’s issue nr. 7, according to guidelines set up by the European Securities and Market Authority. The constructed model is a real world economic scenario generator (ESG) that forecasts future performance of the assets that underlies each product. In the model, we assume that stock paths follow a geometric Brownian motion where volatility is time dependent, heteroscedastic and auto correlated. Given a forecast, we test a product’s performance to receive understanding of the risks that a holder might face. The results have led to the conclusion that the potential return corresponds to the level of risk taken by the investor. Thus, product testing would serve as good practice for all distributors in order to find the most suitable structured product for a given customer due to their risk- and reward profile.

Separation and structural characterization of alpha-lactalbumin and beta-lactoglobulin from whey products

In most food applications, whey proteins are used, rather than the individual proteins and this accounts for the high functional variability among commercially available whey protein products, and limits their applications. The overall objective of this study was to investigate the structural and thermal properties of individual alpha-lactalbumin (alpha-Lac) and beta-lactoglobulin (beta-Lg) fractions isolated from different whey protein sources. / A common non-chromatographic process that isolate alpha-Lac and beta-Lg, with relatively high purity and yield from liquid whey (LW), whey protein concentrate (WPC) and whey protein isolate (WPI) using different chelating agents, was developed. The use of sodium citrate (NaC) and sodium hexametaphosphate (SHMP) were more effective than other chelating agents. Yield results indicated that 47 to 69% of beta-Lg originally present in the whey preparations was recovered, with purities ranging from 84 to 95%, and protein contents ranging from 40 to 99%, while the yields of alpha-Lac were 23 to 89%, with purities ranging from 83 to 90%, and protein contents ranging from 65 to 96% depending on the source of whey protein preparations and type of chelating agents. / Structural and thermal properties of beta-Lg and alpha-Lac isolated fractions were studied using polyacrylamide electrophoresis (native and SDS), RP-HPLC, differential scanning calorimetry (DSC), Fourier transform infrared spectroscopy (FTIR) and electrospray ionization mass spectrometry (ESI-MS). Results showed that all beta-Lg and alpha-Lac isolated fractions exhibit increased thermal stability and reversibility over standard proteins and difference in thermal properties were dependent on protein source. The relative intensity of the 1692 cm-1 band in the beta-Lg isolated fractions was dependent on the nature of the chelating agent, and disappearance of this band occurred at temperature higher than that of beta-Lg standard, indicating increased thermal stability of beta-Lg isolated fractions. Denaturation of apo-alpha-Lac was related to the gradual decrease in the alpha-helix band and accompanied by the gain in intensity of 1653 and 1641 cm-1 bands, while denaturation of holo-alpha-Lac was associated by breakdown of beta-sheet structure and increase in turns and unordered structures. / Changes in charge state distribution (CSD), as measured by ESI-MS of beta-Lg and alpha-Lac in response to pH and storage time, were only qualitative and were of relatively low resolution at basic pH. The hydrogen/deuterium (H/D) exchange results demonstrated that the conformation of holo-alpha-Lac was more stable than that of apo-alpha-Lac and conformation of beta-Lg variant B was more stable than beta-Lg variant A. Kinetics of H/D exchange indicated that alpha-Lac and beta-Lg fractions isolated from different whey protein sources have the same or improved conformational stabilities compared to that of alpha-Lac and beta-Lg standard. The covalent binding of 3 or more hexose residues to alpha-Lac enhanced its conformational stability, but covalent binding of two hexose residues to beta-Lg resulted in less stable conformation.
