Expressão e purificação de uma proteína multiepítopo recombinante desenhada a partir de proteínas do vírus da hepatite C

Considering, principally, the increase of preoccupation with the detection of precocious hepatitis C and that the diagnostic methods of this infection are of the highest clinical importance, the presence study proposes the expression and the purification of one codified protein through a synthetic gene (MEHCV Multiepitope HCV). To achieve this objective, the new recombinant protein multiepitope was designed on the basis of epitopes linear, immunodominant and phylogenetically conserved from the virus of hepatitis C (HCV), including the immune dominating sequences of the genotypes that are more prevalent in Brazil (1a e 3a) and a His-tag in C-terminal to facilitate the purification of the recombinant protein express in bacteria. These epitopes (core, NS3, NS4A, NS4B e NS5) are considered among the most important for the diagnosis of the illness utilized for many HCV detection tests. The MEHCV gene (~720pb) possesses restrictive of sites (NdeI e XhoI) in yours extremities that permits the clonage in phase in the expression of vector bacterial pET-21a. To the synthesize of this gene was made the optimization to the codon usage of E. coli. The protein of interest (~29kDa) was detected by SDS-PAGE and Western blot. The purification of the protein express was realized by centrifugal in resin Ni-NTA by affinitive chromatography. Inasmuch as the purification was not total, a new purification was made of this protein by means of chromatography of affinity in column with resin Ni-NTA. However, did not having this purification owe a presence of cellular proteins. In such case, in the trial of obtain a total purification of this protein multiepitope, would be interesting the utilization of the one new protocol, with the extraction of the inclusion body, wherein all the dissolvable proteins cellular would be remove of the solution. / A hepatite C é considerada um dos maiores problemas de saúde pública no mundo inteiro, inclusive no Brasil, pois uma vacina preventiva não está disponível, o número de infectados com a forma assintomática é elevado e para o controle da doença, há somente kits importados. Considerando, principalmente, o aumento da preocupação com a detecção precoce da hepatite C e que os métodos de diagnóstico desta infecção são de grande relevância clínica, o presente trabalho propõe a expressão e purificação de uma proteína codificada por um gene sintético (MEHCV Multiepítopo HCV). Para atingir este objetivo, uma proteína multiepítopo recombinante foi desenhada a partir de epítopos lineares, imunodominantes e filogeneticamente conservados do vírus da hepatite C (HCV), com a inclusão de seqüências imunodominantes dos genótipos mais prevalentes no Brasil (1a e 3a) e uma His-tag no C-terminal para facilitar a purificação da proteína recombinante expressa em bactéria. Estes epítopos (core, NS3, NS4A, NS4B e NS5) são considerados entre os mais importantes para o diagnóstico da doença e utilizados em muitos testes para detecção do HCV. O gene MEHCV (~720pb) possui sítios de restrição (NdeI e XhoI) nas suas extremidades que permitem a clonagem em fase no vetor de expressão bacteriano pET-21a. No entanto, não houve esta purificação devido à presença de proteínas celulares. Sendo assim, na tentativa de alcançar a purificação total desta proteína multiepítopo, seria interessante a utilização de um novo protocolo, com a extração dos corpos de inclusão, onde todas as proteínas celulares solúveis seriam retiradas da solução.

PADRONIZAÇÃO DE UM ENSAIO IMUNOENZIMÁTICO (ELISA) UTILIZANDO UMA PROTEÍNA MULTIEPÍTOPO RECOMBINANTE PARA PRODUÇÃO DE KIT PARA DIAGNÓSTICO DA HEPATITE C

Due to its high genetic variability was classified in genotypes (1-6) and subtypes (a, b, c etc.). The genotypes 1a, 1b, 2a, 2b and 3a, have cosmopolitan distribution; whereas in Brazil, genotypes 1 and 3 are the most prevalent. The incubation period for acute hepatitis C varies around 6 to 10 weeks and approximately 80% of the acute infections are asymptomatic. Between 10 and 20 years, about 20% of the chronic infections evolve for cirrhosis and these can evolve for death due to complications. In accordance with the World Organization of Health, 3% of the world-wide population (about 180 million individuals) is infected for the HCV. Hepatitis C can be diagnosed by means of serological tests and analyses-based techniques of Molecular Biology. The assay more commonly used for detention of antibodies anti-HCV is ELISA. This test consists of antigens fixed in the wells of a microtiter plate. Specific antibodies against these antigens will adhere to the plate and will be detected by antibodies anti-human immunoglobulin colorimetric markers. This study aims the confection of ELISA using a multiepitope protein (MEHCV). The samples used in the assays are from blood banks. Tests with nickel resin-purified MEHCV and column-purified MEHCV (desalting and non-desalting) had been carried through. The tests aimed standardize the concentration of the protein for well and the dilutions of the serum; to evaluate unspecific reactions through tests with positive serum for other infections (positive anti- HBV and HAV) and with negative serum; and to compare ELISA in house with commercial ELISA. In accordance with the standardization results tests, how much bigger the protein pureness degree biggest was the protein concentration necessary in the microplates sensitization to evaluate possible crossed-reactions. The tests to evaluate unspecific reactions using the resinpurified MEHCV had presented higher sensitivity (92.86%) and specificity (82.89%) than the column-purified MEHCV (Sensitivity = 80%; Specificity = 47.5%). In contrast of ELISA in house, commercial ELISA only presented a result false-positive for sample 5 (anti-HBV positive). To evaluate the interference of other proteins, it was carried through, an assay using microplates sensitized with unspecific proteins. These results suggest that MEHCV protein don t possess yet the ideal pureness degree to be used as antigen and that possibly, the false-positive reactions occurred had been caused by unspecific proteins present with the recombinant protein MEHCV. / O vírus da hepatite C (HCV), descrito em 1989, pertence ao gênero Hepacivirus e à família Flaviviridae e possui um RNA genômico contendo aproximadamente 9.600 nucleotídeos. Devido à sua alta variabilidade genética foi classificado em genótipos (1-6) e subtipos (a, b, c etc.). Os genótipos 1a, 1b, 2a, 2b e 3a, têm distribuição cosmopolita; enquanto que no Brasil, os genótipos 1 e 3 são os mais prevalentes. O período de incubação para hepatite C aguda varia em torno de 6 a 10 semanas e aproximadamente 80% das infecções agudas são assintomáticas. Entre 10 e 20 anos, cerca de 20% das infecções crônicas evoluem para cirrose e estas podem evoluir para óbito devido a complicações. De acordo com a Organização Mundial de Saúde, 3% da população mundial, cerca de 180 milhões de indivíduos, está infectada pelo HCV. A hepatite C pode ser diagnosticada laboratorialmente por meio de testes sorológicos e por meio de análises baseadas em técnicas de biologia molecular. O ensaio mais comumente utilizado para detecção de anticorpos anti-HCV é o ELISA. Esse teste consiste de antígenos virais que são fixados nas cavidades de uma placa de microtitulação. Anticorpos específicos contra esses antígenos irão se aderir à placa e serão detectados por anticorpos anti-imunoglobulina humana contendo marcadores colorimétricos. Esse estudo tem como objetivo a confecção de um ELISA utilizando uma proteína multiepítopo (MEHCV). As amostras utilizadas nos ensaios vieram de bancos de sangue. Foram realizados testes com MEHCV purificada em resina e em coluna de níquel (dessalinizada e não-dessalinizada). Os testes consistiam em padronizar a concentração da proteína por cavidade e as diluições dos soros; avaliar reações inespecíficas através de testes com soros positivos para outras infecções (anti-HBV e HAV positivos) e com soros negativos; e comparar o ELISA in house com um ELISA comercial. De acordo com os resultados dos testes de padronização, quanto maior o grau de pureza da proteína, maior foi a concentração de proteína necessária na sensibilização das microplacas para avaliar possíveis reações cruzadas. Os testes para avaliar reações inespecíficas utilizando a proteína purificada em coluna apresentaram sensibilidade (92,86%) e especificidade (82,89%) superior aos realizados com a proteína purificada em coluna (Sensibilidade = 80%; Especificidade = 47,5%). Ao contrário do ELISA in house, o ELISA comercial apresentou somente um resultado falso-positivo para a amostra 5 (anti-HBV positiva). Para avaliar a interferência de outras proteínas, foi realizado ainda, um ensaio utilizando microplacas sensibilizadas com proteínas inespecíficas. Esses resultados sugerem que a proteína MEHCV ainda não possui o grau de pureza ideal para ser utilizada como antígeno e que possivelmente, as reações falso-positivas ocorridas foram causadas por proteínas inespecíficas presente junto à proteína recombinante MEHCV.
