Partindo da hipótese de que a linguagem cinematográfica não replica a linguagem literária, mesmo porque são artes diferentes, invalidando, assim, qualquer comparação entre as duas sob a perspectiva da hierarquização, discutiremos os códigos fílmicos que transformaram o conteúdo verbal do romance gótico num filme de terror. Baseadas em teóricos dos Estudos da Tradução como Roman Jakobson (1959) que adotou o termo tradução intersemiótica ou transmutação para a interpretação dos signos verbais por meio de sistemas de signos não-verbais, Gideon Toury (1995) que concentra sua pesquisa na obra traduzida para observar o processo que determina sua concretização e Rosemary Arrojo (2003) que desconstrói a idéia de estabilidade e autenticidade na tradução, examinamos a impossibilidade da fidelidade no deslocamento do livro para tela, à medida que identificamos a mescla de narrativas anteriores diluída em intertextos que se entrelaçam com a criatividade dos artistas envolvidos na adaptação. Ao apontar as narrativas que ecoam, influenciam e alimentam a obra de Jordan, atestamos que tais histórias partem de fontes múltiplas, heterogêneas, sem uma origem única, fato que comprova que até mesmo a obra de partida consiste numa colcha de retalhos. Teóricos como Gerárd Genette (2006) que trata da intertextualidade como a presença de um texto em outro, Tiphaine Samoyault (2008), que trata dos diálogos e mosaicos de citações que constroem textos nos rastros de outros textos e Robert Stam (1979) que estende a abordagem de Genette para o universo fílmico ao considerar a adaptação uma ramificação intertextual que não alude apenas ao romance no qual foi inspirado, mas possivelmente a outros que vieram antes dele, nos proporciona ferramentas valiosas para a efetuação da nossa análise. A ação de interpretar ou reescrever a história gótica nos leva a aludir o ato do tradutor/cineasta com o do vampiro,já que o tradutor “sangra” sobre sua obra ao mesmo tempo em que “bebe” de diferentes veias a fim de dar vida ao seu produto audiovisual e o vampiro além de drenar sua vítima, também a alimenta com seu sangue sobrenatural, tornando-a imortal. Para desenvolvermos a metáfora tradução/vampirização, o filósofo Jacques Derrida (2006) e a autora Cristina Rodrigues (2000) nos oferecem reflexões que elucidam a idéia da alternância de empréstimos ou sucessivas suplementações realizadas no universo vampiresco, contribuições que prolongam a eternidade das criaturas da noite a cada geração. Ao tempo em que descrevemos a trajetória da entidade vampiresca desde suas raízes folclóricas à sua ascensão a ícone pop, ressaltamos as revisitações ou atualizações que o bebedor de sangue sofreu, através dos meios de comunicação. Apoiadas em autores como J. Gordon Melton (2011) e Rosemary Guiley(2005), verificamos que a ficção vampiresca seja ela fílmica ou literária, está longe de obter seu descanso definitivo, ou ser indiferente a presença dos vestígios de sangue derramado por predecessores que marcaram o palimpsesto vampírico ao longo dos séculos. / Universidade Federal da Bahia. Ectopic expression of an Arabidopsis glutaredoxin increases thermotolerance in maize during reproductive developmental stages

Abiotic stress response pathways are complex and consist of osmotic adjustors, macromolecule stabilizers, and antioxidants to counteract the damaging nature of abiotic stress induced reactive oxygen species (ROS) accumulation. In this work, we studied the effect of overexpression of an Arabidopsis glutaredoxin, AtGRXS17, on heat tolerance in maize (Zea mays L.) and drought tolerance in rice (Orzya sativa L.). Glutaredoxins (GRXs) are proteins cable of reducing disulfide bonds, therefore regulating the cellular redox status, and require glutathione for regeneration. Ectopic expression of AtGRXS17 in maize resulted in increased heat stress tolerance during flowering. AtGRXS17 enhanced heat tolerance by increasing kernel set and total grain yield during heat treatments, compared to wild type controls. Our results indicated that AtGRXS17-expressing maize plants produce heat tolerant pollen with higher germination rates than wild type when challenged during heat treatments. Furthermore, AtGRXS17-expressing plants were less susceptible to post pollination heat induced kernel abortion. Rice plants expressing AtGRXS17 were also tolerant to abiotic stress. AtGRXS17-expressing rice was more tolerant to drought stress challenges and consistently survived drought treatments. A nontargeted metabolomics study revealed distinct changes in profiles of key metabolite groups in response to drought stress. Soluble sugars and amino acids accumulate as osmotic adjustors while antioxidants, such as glutathione, accumulate to mediate ROS accumulation and regulate redox activity. All genotypes accumulated amino acids, soluble sugars, and raffinose family oligosaccharides in response to drought stress. Our results indicated AtGRXS17-expression affected several pathways known to increase drought tolerance. Altered sugar metabolites suggested a redox modulation of sucrose synthase activity and significant increases in the secondary sulfur assimilation pathway metabolites suggested altered sulfur metabolism. This research provides new insights into ability of GRXs to improve heat tolerance and crop yield in maize and functions of GRXs in affecting metabolite profiles contributing to increased drought tolerance in rice.

Genetic mapping of rooting in rice : exploiting a high throughput phenotyping in plants

Meeting future demands of food security will require enhanced rice production that is more environmentally sustainable. To achieve this it is important to know the genetic and molecular mechanism controlling the root traits. High throughput phenotyping which can keep pace with genotyping is needed, but for many researchers this needs to be cheap as well as meaningful. Here a very simple, low cost and reliable method of assessing root depth of seedling using a layer of diuron-soaked filter paper buried 25 cm deep in a soil-filled box has been developed which is suitable for screening of hundreds of accessions. The assumption is that deep-rooting plants die quicker. This method was then used to screen five established rice panels. Deep rooted cultivars were screened from a panel of an aus panel from IRRI and a panel of Brazilian and Japanese cultivars by using this method. Root QTLs were detected by using bi-parental mapping population and GWA study was performed in two panels, the rice diversity panel (RDP-1) and Bengal Assam Association Population. Assessing 139 RILs from Bala x Azucena bi-parental population revealed heritability of 55% for herbicide symptoms where eleven QTLs were detected, many of which were co-localised with previously reported root QTLs in this population. A GWA study was performed using RDP1) of 356 accessions with 44k SNP markers. Analysis revealed 17% of phenotypic variation of herbicide score was attributable to rice sub-population where the aus showed the deepest rooting systems. A number of QTLs have been identified and a number of positional candidate gene lists were produced. A further 298 cultivars from Bengal and Assam were screened and GWA was performed using 2 M SNP database available from sequencing. ANOVA revealed 37% variation for herbicide score explained by genotype. Soil-filled rhizotron were used to assess 12 of these cultivars, revealing strong xx correlations between deep root traits and herbicide score, confirming the reliability of this method. GWA revealed a number of significant SNPs associated with the traits in this population. Finally a set of mutant gene (LOC_Os09g31478, LOC_Os05g40330, LOC_Os11g34140) which are functional candidate gene for root growth QTLs were studied. Here hydroponic phenotypic screening approach were used to identify the T-DNA mutant lines. However, no convincing mutants were revealed. The herbicide screening method has been shown to be a quick and robust system for the assessment of deep rooting rice plants in soil. This method can now be used for screening large number of cultivars and the identification of QTLs and candidate genes.
